To investigate the effects of the nucleotide sequences in Shine-Dalgarno (SD) and the spacer region (SD-ATG) on bovine growth hormone (bGH) gene expression, the expression vectors under the control of the T7 promoter (pT7-7 vector) were constructed using bGH derivatives (bGH1 & bGH14) which have different 5'-coding regions and were induced in E. coli BL21 (DE3). Oligonucleotides containing random SD sequences and a spacer region were chemically synthesized and the distance between the SD region and the initiation codon were fixed to nine bases in length. The oligonucleotides were annealed and fused to the bGH1 and bGH14 cDNA, respectively. When the bGH gene was induced with IPTG in E. coli BL21(DE3), some clones containing only bGH14 cDNA produced considerable levels of bGH in the range of $6.9\%\;to\;8.5\%$ of total cell proteins by SDS-PAGE and Western blot. Otherwise, the bGH was not detected in any clones with bGH1 cDNA. Accordingly, the nucleotide sequences of SD and the spacer region affect on bGH expression indicates that the sequences sufficiently destabilize the mRNA secondary structure of the bGH14 gene. When the free energy was calculated from the transcription initiation site to the +51 nucleotide of bGH cDNA using a program of nucleic acid folding and hybridization prediction, the constructs with values below -26.3 kcal/mole (toward minus direction) were not expressed. The constructs with the original sequence of bGH cDNA also did not show any expression, regardless of the free energy values. Thus, the disruption of the mRNA secondary structure may be a major factor regulating bGH expression in the translation initiation process. Accordingly, the first stem-loop among two secondary structures present in the 5'-end region of the bGH gene should be disrupted for the effective expression of bGH.
This paper is contented with packet transmission scheduling problem for repeating all-to-all broadcasts in WDM optical passive-star networks in which there are N nodes and k wavelengths. It is assumed that each node has one tunable transmitter and one fixed-tuned receiver, and each transmitter can tune to k different wavelengths. The tuning delay represents the time taken for a transmitter to tune from one wavelength to another and represented as ${\delta}$(>0) in units of packet durations. We define all-to-all broadcast as the one where every node transmits packets to all the other nodes except itself. So, there are in total N(N-1) packets to be transmitted for an all-to-all broadcast. The optimal transmission scheduling is to schedule In such a way that all packets can be transmitted within the minimum time. In this paper, we propose the condition for optimal transmission schedules and present an optimal transmission scheduling algorithm for arbitrary values of N, k, and ${\delta}$ The cycle length of the optimal schedules is $max{[\frac{N}{k}](M-1)$, $k{\delta}+N-1$}.

This study investigated the performances of precipitable water vapor (PWV) retrieval from the sets of ground global positioning system (GPS) signals, each of which had different length of observing-session duration, for the purpose of obtaining as short session duration as possible that is required at the least for appropriate retrieval of the PWV for meteorological usage. The shorter duration is highly desirable to make the most use of the GPS instrument on board the mobile observation vehicle making measurements place by place. First, using Bernese 5.0 software the PWV retrieval was conducted with the data sets of GPS signals archived continuously in 30 seconds interval during 2-month period of January and February, 2012 at Bukgangneung site. Each of the PWVs produced independently using different session durations was compared to that of radio-sonde launched at the same GPS location, a Bukgangneung site. Second, the same procedure was done using the data sets obtained from the mobile observation vehicle that was operating at Boseong area in Jeonnam province during Changma observation campaign in 2013, and the results were compared to that at Bukgangneung site. The results showed that as the observing-session duration increased the retrieval errors decreased with the dramatic change happening between 3 and 4 hours of the duration. On average, the root mean square error (RMSE) of the retrieved PWV was around 1 mm for the durations of greater than 4 hours. The results at both the Bukgangneung (fixed site) and Boseong (mobile vehicle) seemed to be fairly comparable with each other. From this study it is believed that at least 4 hours of observing-session duration is needed for the retrieval of PWV from the ground GPS for meteorological usage using Bernese 5.0 software.
The purpose of this thesis is that the students understand the sentence stated math problems closely related to the real life and adapted the right solving strategies try to find the solution to a problem. The following research problem were proposed. 1. How repeated thinking lessons develop the understanding of problems and influence the usage of correct problem solving strategies and extensions of problem solving. 2. There are how much differences of achievement for each type of sentence stated problems by using comparative analysis of upper class, intermediate class, and lower class for each level between the experimental and comparative classes. In order to conduct this research the classes were divided into three different level - upper class, intermediate class and lower class. Each level include an experimental class and a comparative class. The two classes (experimental class and comparative class) of the same level were tested on the basis of class division record with the experimental class repeated learning papers for two weeks were used to guide the fixed thinking algorism for each sentence stated math problems. Eight common problems were chosen from a variety of textbooks : number calculation problems, velocity-distance-time problems, the density of a mixture, benefit problems, distribution problems, problems about working, ratio problems, the length of a figure problems. After conducting this research experiment The differences in achievement level between the experimental class and comparative class, were compared and analyzed through achievement tests made from the achievement test papers with seven problems, which were worth seventy points (total score). The conclusions of this thesis are as follows: Firstly, leaning activities through the usage of repeated learning papers for each level class produce an even development of achievement level especially in the case of the upper class learners, they have particular differences (between experimental class and comparative class) compared to the intermediate level and lower classes. Secondly, according to the analysis about achievement development each problems, learners easily accept the strategies of solution through the formula setting up to the problem of velocity -distance-time, and to the density of the mixture they adapted the picture drawing strategies interestingly, However each situation requires a variety of appropriate solution strategies. Teachers will have to employ other interesting solution strategies which relate to real life.
The present study was performed to study the genetic relationship of productive life with production and type traits of Korean Holsteins at first three lactations. The data for the analysis from 56,054, 28,997, and 11,816 animals of first, second and third parity cows which were born from 2006 to 2011 were collected by Dairy Cattle Improvement Center, National Agricultural Co-operative Federation. Milk, protein and fat yields adjusted for 305 days and average somatic cell score considered as production traits and analyzed type traits were stature, strength, body depth, dairy form, rump angle, rump width, rear leg side view, foot angle, front attachment placement, rear attachment height, rear attachment width, udder cleft, udder depth, front teat placement and front teat length. A multi trait genetic analysis was performed using Wombat program with restricted maximum likelihood animal model composed of fixed effect of birth year, farm and the random effect of animal and random residual effect according to the traits. Heritability estimates of productive life were between 0.06 and 0.13. Genetic and phenotypic correlations between production and productive life traits ranged from 0.35 to 0.04 for milk, 0.16 to 0.05 for protein and 0.18 to 0.02 f 15-0034 (2nd) 150520 or fat. Somatic cells score showed a negative genetic and phenotypic correlation with productive life and also udder type traits, indicating that the selection for higher udder traits will likely to improve resistance to mastitis and persistence in the herd. Among all dairy form type traits, udder characters such as udder cleft showed a significant relationship with productive life. However, a specific change of heritabilities or correlations were not observed with the change of parity. Moreover, further studies are needed to further confirm the significance of the above traits and the effect of parity on above relationships in order to minimize both voluntary and involuntary culling rates while improving herd health and maintaining high yielding dairy cows.

In this study, the applicability and external/internal stability of a MSEW abutment with a slab were investigated. Structural analysis of slab bridges between 10 ~ 20.0 m and thicknesses of 0.7 ~ 0.9 m was carried out to calculate the reaction forces due to dead and live loads acting on the bridge supports. The slab bridge with a length of 20.0 m satisfied the allowable contact pressure of 200 kPa for the true MSEW abutment. Because the external stability of the true MSEW abutment was dominated by the geometry of the MSE wall, the change in the factor of safety due to the load of the super-structure is small. Because the stiffness of the foundations is fixed and the load of the super-structure is increased, the factor of safety of the bearing capacity was reduced. As the load of the super-structure was increased, the horizontal earth pressure of the true MSEW abutment increased greatly. As a result, the pullout and fracture of the uppermost reinforcement, which are the factors of safety, did not meet the design criteria. Therefore, it is necessary to increase the pullout resistance and the long-term allowable tensile force of the reinforcement placed on the top of the reinforced soils to ensure efficient design and performance of a true MSEW abutment.
Large scale secular registry or surveillance systems have been accumulating vast data that allow mathematical modeling of cancer incidence and mortality rates. Most contemporary models in this regard use time series and APC (age-period-cohort) methods and focus primarily on predicting or analyzing cancer epidemiology with little attention being paid to implications for designing cancer registry, surveillance or evaluation initiatives. This research models age-specific cancer incidence rates using logistic growth equations and explores their performance under different scenarios of data completeness in the hope of deriving clues for reshaping relevant data collection. The study used China Cancer Registry Report 2012 as the data source. It employed 3-parameter logistic growth equations and modeled the age-specific incidence rates of all and the top 10 cancers presented in the registry report. The study performed 3 types of modeling, namely full age-span by fitting, multiple 5-year-segment fitting and single-segment fitting. Measurement of model performance adopted adjusted goodness of fit that combines sum of squred residuals and relative errors. Both model simulation and performance evalation utilized self-developed algorithms programed using C# languade and MS Visual Studio 2008. For models built upon full age-span data, predicted age-specific cancer incidence rates fitted very well with observed values for most (except cervical and breast) cancers with estimated goodness of fit (Rs) being over 0.96. When a given cancer is concerned, the R valuae of the logistic growth model derived using observed data from urban residents was greater than or at least equal to that of the same model built on data from rural people. For models based on multiple-5-year-segment data, the Rs remained fairly high (over 0.89) until 3-fourths of the data segments were excluded. For models using a fixed length single-segment of observed data, the older the age covered by the corresponding data segment, the higher the resulting Rs. Logistic growth models describe age-specific incidence rates perfectly for most cancers and may be used to inform data collection for purposes of monitoring and analyzing cancer epidemic. Helped by appropriate logistic growth equations, the work vomume of contemporary data collection, e.g., cancer registry and surveilance systems, may be reduced substantially.
State invariant is a property that holds in every reachable state. It can be used not only in understanding and analyzing complex software systems, but it can also be used for system verifications such as checking safety, liveness, and consistency. For these reasons, there are many vital researches for deriving state invariant from finite state machine models. In previous works every reachable state is to be considered to generate state invariant. Thus it is likely to be too complex for the user to understand. This paper seeks to answer the question `how to simplify state invariant\ulcorner`. Since the complexity of state invariant is strongly dependent upon the size of states to be considered, so the smaller the set of states to be considered is, the shorter the length of state invariant is. For doing so, we let the user focus on some interested scopes rather than a whole state space in a model. Computation Tree Logic(CTL) is used to specify scopes in which he/she is interested. Given a scope in CTL, mixed reachability analysis is used to find out a set of states inside it. Obviously, a set of states calculated in this way is a subset of every reachable state. Therefore, we give a weaker, but comprehensible, state invariant.
In recent years, the number of traffic accidents caused by car accidents has been decreasing steadily due to traffic accident prevention activities in Korea. However, the number of accidents in Seoul is higher than that of other regions. Various studies have been conducted to prevent traffic accidents, which are human disasters. In particular, previous studies have performed the spatial analysis of traffic accidents by counting the number of traffic accidents by administrative districts or by estimating the density through kernel density method in order to identify the traffic accident cluster areas. However, since traffic accidents take place along the road, it would be more meaningful to investigate them concentrated on the road network. In this study, traffic accidents were assigned to the nearest road network in two ways and analyzed by hotspot analysis using Getis-Ord Gi* statistics. One of them was investigated with a fixed road link of 10m unit, and the other by computing the average traffic accidents per unit length per road section. As a result by the first method, it was possible to identify the specific road sections where traffic accidents are concentrated. On the other hand, the results by the second method showed that the traffic accident concentrated areas are extensible depending on the characteristic of the road links. The methods proposed here provide different approaches for visualizing the traffic accidents and thus, make it possible to identify those sections clearly that need improvement as for the traffic environment.
The effect of circumvaginal muscle(CVM) exercises to improve sexual function in married women has not been investigated by currently acceptable research methods, nor have appropriate instruments and techniques to carry out such investigation been available. The purpose of this research was to study the effect of CVM exercise on sexual function, and of measuring CVM function after CVM exercises. The research tools used were a modified Derogates Sexual Function Inventory questionnaire and a pressure sensitive intravaginal balloon device. This research was conducted in Kwangju-city and Chonnam province, Korea from July, 1994 to July, 1995. The research used a non-equivalent control pre-post test quasi-experimental design. Forty-five healthy married female volunteers, aged 30-58, and were randomly assigned by age using the matching fixed-length blocks to two groups. The experimental group consisted of 21 women who were assigned a 25-minute per day CVM home exercise program for six weeks. The control group of 24 women did not do the CVM home exercises. The CVM home exercise was developed by Dougherty(1989a) and adopted to Korea by Lee (1993). Data were analyzed by $x^{2-}$test, Paired t-test, Spearman product-moment correlation using SAS /P $C^+. The results are summarized as follows : 1. There were no significant differences in the characteristics of the subjects between the experimental and control groups before the CVM home exercises. 2. Hypothesis 1 that married women who participated in CVM home exercises would have higher mean scores on the sexual function(SF) than in those who did not participate in home exercise was supported. 3. Hypothesis 2 that married women who participated in CVM home exercises would have higher vaginal pressure on SF than in those who did not participate in home exercises was supported (mean maximum pressure, t=-7.338, P<.0001, peak maximum pressure, t=-11.164, P<.0001). 4. Hypothesis 3 that the more often(number of days) and the more frequent (numbers of time per day) that married women do CVM home exercise, the higher their mean scores on SF and vaginal pressures was supported(r=0.233, P<.01 ; r=0.352, P<.05). A six week CVM home exercise program using a tape recording showed that SF can be improved. Results of this study showed that married women who exercise on a regular basis for six weeks improve their sexual function and increase the mean vaginal pressure and peak maximum pressure (tested by electronic monitor). In conclusion, CVM exercise is effective in increasing SF.SF.
