Hc nuclear polyhedrosis virus DNA 제한효소절편의 molecular cloning 과 외래 유전자 발현 (Molecular cloning and foreign gene expression of restriction endonuclease fragments of the Hc nuclear polyhedrosis virus DNA)

  • HcNPV DNA genome 을 제한효소 EcoRI 으로 절단하여 그들의 일부 절편을 pUC8 vector 에 cloning 한 후 E. coli JM 83 세포에 형질 전환시켰다. 이 결과 24 개의 EcoRI 절편중 12 개의 절편이 cloning 되었다. 이들 제조합체중 4 개는 eNP-O, eNP-Q, eNP-R, eNP-S 라 명명하였다. 또한 이들 제조합체의 외래 유전자 발현을 SDS-PAGE 에 의해 단백질 패턴을 분석하였다. 그 결과 제조합체 eNP-O, eNP-Q, eNP-R 에서는 E. coli JM 83 숙주세포의 단백질 밴드와 비교하여 다른 분자량을 갖는 밴드가 나타났다.

Genotype distribution of infectious haematopoietic necrosis virus (IHNV) in Korea

  • Infectious haematopoietic necrosis virus (IHNV) is an important fish pathogen that infects both wild and cultured salmonids. Since the first isolation of IHNV from rainbow trout and masu salmon in 1991, a series of IHN disease outbreak has been reported in Korea. In 2011, we isolated two IHNV isolates from rainbow trout cultured in Korea. The full open-reading frame (ORF) encoding the glycoprotein (G) of them were sequenced and the amino acid sequences were phylogenetically analyzed. Phylogenetic analysis of the G revealed that both IHNV isolates were grouped into an Asian genogroup containing Korean IHNV isolates and Japanese IHNV isolates. However, based on their sequence variation, they were divided into different subgroup. While one isolate was similar to other Korean isolates, the other isolate showed a high level of similarity with Japanese isolates, suggesting the possibility of influx of new IHNV strain into Korea.

양식 넙치에서 Parvicapsula anisocaudata의 감염 사례에 대한 보고 (A case report of farmed olive flounder Paralichthys olivaceus infected with Myxosporean Parvicapsula anisocaudata)

  • Parvicapsula anisocaudata, a myxosporean parasite, is presumably one of causative agents of emaciation in olive flounder Paralichthys olivaceus in Korea. In this study, we report a case of unusual abdominal distension due to exceptionally enlarged liver in farmed olive flounder. For the identification of the causative agent, bacteria and nucleic acids of virus that are possibly present were attempted to isolate from internal organs of five fish sampled from a fish farm in Jeju. Although a few bacterial colonies were isolated from some samples, there was no evidence that fish were primarily affected by virus and/or bacteria. From histopathological analysis, myxosporean were found in almost all internal organs, particularly in the stomach. The causative agent was identified as P. anisocaudata by sequencing a part of small subunit rRNA. This study contains a very unusual case of olive flounder heavily and systemically infected with P. anisocaudata, showing excessively enlarged liver with a small amount of ascitic fluid.

코로나 방전 플라즈마 처리수에 의한 어류 병원체 소독 효과 (Disinfection effect of corona discharged plasma water on fish pathogens)

  • Fish culture is constantly threatened by various infectious diseases which are largely transmitted by water. Plasma technology is being used to sterilize polluted water in many industries. In this study, two bacterial pathogens Aeromonas salmonicida and Streptococcus iniae, and a virus (viral hemorrhagic septicemia virus, VHSV) were subjected to plasma water that was produced by a corona discharge system. Growth of A. salmonicida was greatly inhibited from 105.61 CFU/ml in positive control to 103.51 CFU/ml in treated group by only 60 sec contact with plasma water. Similarly, S. iniae was inhibited from 105.85 CFU/ml to 103.40 CFU/ml. VHSV titer also decreased from 104.1 TCID50/ml to 101.45 TCID50/ml by the same treatment. Activation of water by the plasma was confirmed by the existence of ozone in the plasma water. These results suggest that plasma water could efficiently disinfect fish pathogens, possibly by the action of reactive oxygen species contained in the plasma water.

2005년부터 2007년 사이 양식 넙치, Paralichthys olivaceus 질병에 대한 통계 자료 (Statistical data on infectious diseases of cultured olive flounder Paralichthys olivaceus from 2005 to 2007)

  • 2005년부터 2007년 사이 매분기별로 포항, 울산, 기장, 거제, 완도 지역에서 채집한 양식넙치를 대상으로 기생충, 세균 및 바이러스에 대한 병원체 모니터링 결과를 분석하였다. 총 2,238마리 중 1,528마리(68.3%)에서 병원체가 검출되었다. 연도별 검출률을 비교한 결과, 2005년도에는 60.6%, 2006년도에는 66.7%, 2007년도에는 72.3%의 병원체 검출률을 나타내어 해마다 증가하는 것으로 나타났다. 조사시기별 총 병원체 검출률은 2월에 63.5%, 5월에 67.3%, 8월에 75.1%, 11월에 64.2%로 나타났다. 기생충, 세균 및 바이러스의 검출률은 각각 36.7%, 32.8%, 31.4%로 나타났다. 병원체의 단독감염률 및 혼합감염률은 각각 33.6%와 34.6%로 나타났다. 질병별 분포 조사에서 가장 높은 검출률을 나타낸 병원체는 Trichodina sp. (28.2%), viral nervous necrosis virus (24.3%), Vibrio spp. (11.6%), viral hemorrhagic septicaemia virus (10.5%) 순으로 나타났다.

  • We identified a microsatellite marker, Poli121TUF, which appears to be significantly linked (P<0.001) with a lymphocystis disease virus (LCDV)-resistance gene in the olive flounder, Paralichthys olivaceus. The olive flounder is an economically important food fish, that is widely cultured in Korea, Japan, and China. Lymphocystis disease has spread in these countries and has seriously reduced the economic value of the fish. LCDV causes lymphocystis cells (LC) to form on the body surface, fins, gills, mouth, and intestine. Fish with LC lose commercial value due to their deformed appearance. The identified micro satellite marker can be used as a candidate locus for marker-assisted selection (MAS) in order to enhance the efficiency of selection for LCDV resistance in the olive flounder.

  • Lymphocystis is a viral disease of fish primarily in marine and brackishwaters. Here we report the cloning, expression, and the serological applications of the lymphocystis disease virus (LCDV) major capsid protein (MCP). The MCP gene was amplified by PCR from the genomic DNA of LCDV isolated from Schlegel's black rockfish, Sebastes schlegeli, and expressed in E. coli. Mouse antisera raised against the purified recombinant MCP (rMCP) reacted with the viral MCP in an immunofluorescence assay, indicating that this rMCP would be useful for serological studies of field samples.

  • 한국에서 분리된 전염성 조혈괴저바이러스(infectious hematopoietic necrosis virus, IHNV)인 IHNV-PRT의 nucleocapsid(N)를 암호화하고 있는 cDNA를 클로닝하여 분석하였다. N 유전자는 391개의 아미노산으로 구성된 42.3 kDa의 분자량을 가진 단백질을 암호화하는 1,176 bp 크기의 open reading frame을 가지고 있었다. N 단백질의 아미노산 서열을 다른 외국에서 분리한 IHNV들의 N과 비교한 결과 75-90% 정도의 유사성을 보였으나 다른 종의 fish rhabdovirus인 hirame rhabdovirus(HRV) 및 viral hemorrhagic septicemia virus(VHSV)와는 각각 43%와 38%의 유사성을 보였다. 그러나 아미노산 서열 214-265의 52개의 아미노산들은 모든 종의 fish rhabdovirus간에 매우 높은 유사성을 보여 주었다.

Identification of antigenic proteins of lymphocystis disease virus (LCDV) by MALDI-TOF mass spectrometry

  • The antigenic proteins of Lymphocystis disease virus (LCDV) from tumors of olive flounder, Paralichthys olivaceus, are described following characterization by mass spectrometry. In SDS-PAGE, predominant protein bands were observed at 114, 88, 70, 54, 52, 47, 42 and 24 kDa. Western blot analysis showed that antisera reacted strongly at molecular weights of 114, 67 and 54 kDa, and reacted weakly at molecular weights of 74, 70, 36, 24 and 22 kDa. In the identification of LCDV antigenic proteins by matrix-assisted laser desorption ionization (MALDI) TOF mass spectrometry, 10 of 14 excised bands consisted mostly of proteins with amino acid sequences that matched LCDV-C (lymphocystis disease virus isolate China) ORFs. Strong antigens with molecular weights of 114, 67 and 54 kDa were identified as LDVICp236 (chromosome segregation ATPase), LDVICp033 (membrane bound metallopeptidase) and LDVICp157 (hypothetical protein), respectively. Minor antigens with molecular weights of 70, 36, 24 and 22 kDa proteins were identified as LDVICp160 (acetyl-coA hydrolase), LDVICp213 (hypothetical protein), LDVICp039 (hypothetical protein) and LDVICp213 (hypothetical protein). However, the major capsid protein (LDVICp043) did not react with the polyclonal antibody.

국내 양식 흰다리새우에서 검출된 노랑머리바이러스(YHV-8)의 유전학적 특성 (Detection and genetic characteristic of Yellow-head virus genotype 8 (YHV-8) Cultured Litopanaeus vanamei, in Korea)

  • This study was performed for screening yellow head virus(YHV) complex in 252 including 235 white leg shrimps (Litopenaeus vannamei) and 17 oriental shrimp (Fenneropenaeus chinensis) collected from 18 farms located in southwestern province of Korea. The virus complex was detected by nested reverse-transcriptase polymerase chain reaction (RT-PCR) assay. In the assay, amplicons were resulted in RNAs exracted from 38 shrimps (21 white leg shrimps and 17 oriental shrimps) obtained from 7 farms. In phylogenetic analysis using sequences of ORF1b gene, all 38 sequences obtained in this study formed an independent lineage with YHV-8 genotype firstly isolated in China, belonged to an YHV-8 clade.
