Fish Myogenic Regulatory Protein LUC7L: Characterization and Expression Analysis in Korean Rose Bitterling (Rhodeus uyekii)

  • Serine-arginine-rich nuclear protein LUC7L plays an important role in the regulation of myogenesis in mice. In the present study, we isolated and characterized the Korean rose bitterling Rhodeus uyekii Luc7l cDNA, designated RuLuc7l. The RuLuc7l cDNA is 1,688 bp long and encodes a 364-amino-acid polypeptide containing serine/arginine-rich region at the C-terminus. The deduced RuLuc7l protein has high amino acid identity (71-97%) with those of other species including human. Phylogenetic analysis revealed that RuLUC7L clustered with fish LUC7L proteins. The expression of RuLuc7l mRNA was high in the brain, kidney, and stomach of Korean rose bitterling. Expression of the RuLuc7l mRNA was detected from 1 day post-fertilization (dpf) and moderately increased until 21 dpf during the early development. Further investigations are required to elucidate the functional role of RuLUC7L in myogenesis in R. uyekii.

  • Economic development involves increase in life expectancy as well as human health care. Consequently, demand for fish meal and fish oil is rapidly growing. In particular, Krill (Eupausia superba) oil product is in high demand due to its rich unsaturated-fatty acid, and thus stable supplies are necessary in the krill oil market. It is required for captured krills to be immediately frozen and stored during ship transport, since proteins of the krill are quickly denatured in natural temperature condition. However, the transportation cost has been sharply increased, which encourages researchers to involve in studies for development of efficient oil extraction process. In this study, a solid/liquid separation technique on boat for the krill oil was developed through triple separation tests using only a separator or using either brush or crusher prior to the separator. The separation tests revealed that the efficiency were 46.2, 60.2 and 60.4 % by the separator, combination with brush, and combination with crusher, respectively. In addition, it was found that byproduct, extracted cake, derived from the separation process could be used as a feed stuff. These results suggest that smashing using the brush or crusher prior to the separator is more efficient than using only the separator.

Isolation and purification of chicken egg yolk immunoglobulin against Edwardsiella tarda (Edwardsiella tarda에 대한 계란난황항체의 분리와 정제)

  • The present study compared purification methods of hen egg yolk immunoglobulin (IgY) from the hen immunized with Edwardsiella tarda. The purification of anti-E. tarda IgY was performed by four different methods, polyethylene glycol (PEG), chloroform polyethylene glycol (Chloroform-PEG), ammonium sulfate and purification kit. Purified IgY had heavy chain of 64 kDa and light chain of 27 kDa size. IgY purified from the hen immunized with E. tarda showed higher ELISA values and agglutination titers than those with IgY purified from the non-immunized hen as a negative control. In addition, purified IgY recognized similar E. tarda proteins to those with anti-E. tarda rabbit serum by western blotting. Purified IgY had an agglutination titer of 1:512 by PEG method and ammonium sulfate method, and 1:128 by chloroform-PEG method and purification kit. Moreover, PEG method was the most rapid method among the four different IgY purification methods. These results indicate that PEG method is effective purification method maintaining biological activity of the IgY.

  • The Interferon stimulated gene 15 (ISG15) is strongly induced in many cell types by IFNs, viral infections, and double-stranded RNA (poly I:C). The ISG15 homologue cDNA was isolated from the rock bream LPS stimulated leukocyte cDNA library. The rock bream ISG15 homologue was found to consist of 833 bp encoding 157 amino acid residues. Compared with other known ISG15 peptide sequences, the most conserved regions of the rock bream ISG15 peptide were found to be the tandem ubiquitin-like domains and a C-terminal LRLRGG conjugating motif, characteristic of mammalian and non-mammalian ISG15 proteins. Phylogenetic analysis based on the deduced amino acid sequence revealed a homologous relationship between the ISG15 sequence of rock bream and that of Atlantic salmon, Atlantic cod, northern snake head, black rockfish and olive flounder. The expression of the rock bream ISG15 molecule was induced in the peripheral blood leukocytes (PBLs) from 1 to 24 h following poly I:C stimulation, with a peak at 3 h post-stimulation. The rock bream ISG15 gene was predominantly expressed in the PBLs, spleen and gill.

Protein Consumption Market Trends in Korea: Focusing on Meat, Fishery, and Plant-based Protein Foods (국내 단백질 소비시장 동향: 축산물, 수산물, 식물성 단백질 식품을 중심으로)

  • This study aims to investigate protein consumption market trends in Korea. Protein consumption was divided according to the protein source into meat, fishery, and plant-based protein. To accomplish the goal of this study, food purchase data from 525 households panels collected by the Rural Development Administration over the last 10 years were used. The results of the study showed an increase or decrease in protein consumption by protein type over the last 10 years, and a reason to explain this change has been suggested. Specifically, this study found a dramatic increase in the consumption of several proteins, including beef sirloin, beef tenderloin, seasoned beef & steak, pork belly, pork shoulder, pork neck, seasoned pork, pork cutlet, sweet and sour pork, canned ham, chicken drumstick, chicken breast, dak gangjeong, Chinese fried chili chicken, salmon, eel, abalone, squid, octopus, webfoot octopus, octopus minor, canned whelk, tofu, cold bean soup,and plant-based milk. Some items showed no increase in consumption (such as beef jerky, pork rib, sausage, bacon, whole raw chicken, cutlass fish, oyster, fish cake, crab stick, surimi sausage,and canned fishery), whereas a few items showed decreased consumption (e.g., mackerel, pollack, cod,and canned tuna)

  • Insulin-like growth factor-I (IGF-I) is a mitogenic peptide with a molecular mass of 7 kDa. It is produced mainly in the liver and has important functions in the regulation of development and somatic growth. Moreover, Serum IGF-I concentration is regulated by the quantity and the nutritional quality of dietary protein. To determine the IGF-I level in Korean rockfish, Sabastes schlegeli, were fed four experiment diets that contained different protein quantities, namely $30\%,\;40\%,\;50\%\;and\;60\%$ for 70 days. Weight gain of the fish increased depending dietary protein quantity, Also, IGF-I concentrations increased according to dietary protein quantity, Feeding experiments were conducted to examine the effects of dietary protein sources on the serum IGF-I level in Korean rockfish, Fish meal (CO), soybean meal (SM), corn-gluten meal (CGM), meat meal (MM) and feather meal (FM) were used as variable protein sources of the formulated diet. IGF-I concentrations of the CO and MM groups ($277.7\pm23.2,\;291.5\pm41.2\;ng/mL$) were higher than those of the CGM and FM groups ($208.9\pm21.3,\;217.2\pm38.2\;ng/mL$). And IGFBP-3 levels by western blot analysis increased in good protein diets such as in the CO and MM groups. In conclusion, IGF-I may be a sensitive indicator the protein metabolism in fish as well as mammalian.

  • Telomeres consist of TTAGGG tandem repeated DNA sequences with specific proteins and locate at chromosome ends. Telomeres are essential for chromosome stability and are related with cell senescence, apoptosis and cancer. Telomerase is a ribonucleoprotein which has a template for the synthesis of telomeric DNA. This study was carried out to analyze the amount of telomeric DNA and telomerase activity in cattle cells. Analysis of the quantity of telomere in lymphocytes was done at different ages, sex and among Korean cattle and Holstein breeds. The telomerase activity was also analyzed in liver, brain, heart, kidney, and testis tissues of fetal calf and of 18 month old cattle. The amount of telomeres in lymphocytes and other tissue cells was analyzed by Quantitative-Fluorescence in situ Hybridization (Q-FISH) technique using a telomeric DNA probe. Telomerase activity was analyzed by Telomeric Repeat Amplification Protocol assay (TRAP). The amount of telomeric DNA on the lymphocytes during the whole life span was decreased along with age. Quantity of telomeres in Korean cattle was significantly higher than that in Holstein breed. The amount of telomeric DNA in males was significantly higher than that in females. Telomerase activity was up-regulated in most bovine tissues during fetal stage, but was down-regulated in most tissues at mature 18 month age except the testis cells. This study indicates that the amount of telomeres and telomerase activity of cells can be used as an age marker or/and a physiological marker of cattle.

Cutlassfish allergy in children: Usefulness of serum cod specific IgE and cross-reaction with cod (소아 갈치알레르기: 대구 특이 IgE 검사 활용성 및 대구와의 교차반응)

  • Purpose: Fish allergy is the ninth common food allergy, and cutlassfish is one of the common allergenic fishes in Korean children. However, there is no commercial diagnostic tool for testing cutlassfish allergy in the world. We evaluated the usefulness of serum cod specific IgE (cod-sIgE) to diagnose cutlassfish allergy and cross-reaction between cutlassfish and cod. Methods: Nineteen children who experienced immediate type reactions after consumption of cutlassfish were enrolled. Cod-sIgE was measured by ImmunoCAP, and serum samples were obtained from 11 allergic patients and 11 controls. Using our own homemade crude extracts, sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), anti-parvalbumin (PV) immunoglobulin G immunoblot, enzyme-linked immunosorbent assay (ELISA), and ELISA inhibition were performed. Results: Thirteen patients were clinically allergic to both cutlassfish and cod, and 6 were allergic to cutlassfish alone. The median age and cod-sIgE concentrations were not significantly different between the 2 groups. The clear fish protein bands and PVs were identified on SDS-PAGE and immunoblotting. Serum cod-sIgE was positive in 4 out of 6 cutlassfish mono-allergic patients, however, there was no significant correlation between cod-sIgE by ImmunoCAP and cutlassfish-specific IgE by ELISA. The cutlassfish IgE ELISA was profoundly inhibited by cutlassfish, while the cod IgE ELISA was profoundly inhibited by cod but partially inhibited by cutlassfish. Conclusion: We found a potential diagnostic value of cod-sIgE to diagnose cutlassfish allergy and the asymmetric cross-reaction between cutlassfish and cod. These results could help diagnose and provide a dietary guidance in cutlassfish allergic children.

Yeast Single-Cell Protein Production Using Potato Processing Waste Water

  • Four species of yeast, Saccharomyces cerevisiae, Candida utilis, Saccharomycopsis flbuligera, and Schwanniomyces castellii were evaluated for their ability to bioconvert potato processing waste water into microbial protein and the resulting single-cell proteins were evaluated as protein sources for rainbow trout, using in vitro analyses. The studies indicated that Schwanniomyces castellii, which utilizes starch dircetly and converts it into cell mass efficiently, was suitable for the bioconversion. In the single-stage continuous bioconversion, the yield S. castellii cell mass, which contained approximately 37% protein, was 77%, at dilution rate 0.25 $h^{-1}$. Reduction of total carbohydrate was 81%. During batch fermentations, cell mass yield was about 72% and total carbohydrate reduction was 81%. Among the yeasts tested, S. castellii possessed the most fragile cell wall and had a favorable amino acid profile for salmonid fish; protein score of 86% (Met). In an in vitro pepsin digestibility test 80% digestibility (23~38% above control) was observed when cells were pre-heated in a steam bath for 30 min. Results presented should be regarded as being preliminary in nature because they were derived from single experiments.

Effect of Dietary Proteins without Vitamin Premix Supplementation on the Growth and Body Composition of Juvenile Black Rockfish Sebastes schlegeli (비타민프리믹스 무첨가시 사료 중 단백질원이 조피볼락(Sebastes schlegeli) 치어의 성장과 체성분에 미치는 영향)

  • A feeding experiment was conducted to determine the nutritional value of various dietary protein sources without a vitamin premix supplementation in juvenile black rockfish, Sebastes schlegeli. Diets containing similar quantities of lipid and carbohydrate with 50% crude protein from fish meal (FM)/soybean meal (SBM)/corn gluten meal (CGM), FM only, casein/gelatin, high FM/low casein and low FM/high casein were fed to rockfish along with a commercial rockfish diet. The diet containing only 65.6% FM and the commercial rockfish diet produced significantly greater weight gain (WG) and feed efficiency (FE) than all other semipurified diets (SPDs); the economical diet with 52.8% FM, 10% SBM and 5% CGM produced lower WG than the diet containing FM and commercial rockfish diet, but FE and survival were similar both diets. Intermediate responses were observed for rockfish fed the SPD containing 32.8% FM and 27.4% casein, while SPDs containing 10% FM and 41.8% casein and 38.5% casein and 10% gelatin yielded the lowest WG and FE. Some differences in tissue indices and body composition of rockfish were induced by the diets containing various protein sources and levels without the vitamin premix supplementation.
