Isolation, Purification and Characterization of Chitosanase from Bacillus subtilis CH1

  • Bacillus subtilis strain with highly active chitosanase was isolated from the intestine of Sebastiscus marmoratus (scorpion fish). It was named as Bacillus subtilis CH1 by morphological, biochemical and 165 rRNA gene analysis. The optimal conditions for chitosanase production were investigated. The optimum carbon and nitrogen sources for Bacillus stibtilis CH1 were 2% starch and 1% yeast extract respectively. Unlike other chitosanases, the expression of this chitosanase was not induced or slightly induced with chitosan. The chitosanase secreted into the medium were concentrated with ammonium sulfate precipitation and purified by gel permeation chromatography. The molecular weight of purified chitosanase was 30 kDa. The optimum pH and temperature of purified chitosanase were 5.5 and $60^{\circ}C$ respectively. The purified chitosanase was continuously thermostable at $40^{\circ}C$ and showed stable activity between pH 6.0 and 8.0. Chitosanase activity of Bacillus subtilis CH1 under optimum condition was 4.1 units/ml.

Relationship of Habitual Alcohol Consumption to the Nutritional Status in Middle Aged Men (음주습관이 중상류층 중년남성의 영양상태에 미치는 영향)

  • The effect of habitual alcohol consumption on the nutrient intake. body weight and serum values were studied in 194 upper middle class men aged 35-55yr. living in Seoul. Each subject was interviewed to get information of alcohol and nutrients intake. Blood samples were taken. Mean values for each variable were compared by fertile of alcohol intake. The average alcohol consumption of the subject was 32.2g/day. Most of times foods were taken along with drinks. The subjects preferred food items such as meat. fish, nuts. dry fishes when they drink. High alcohol consumers had a significantly higher intake of calorie. protein, fat. Fe, and riboflavin compared to nondrinkers and moderate drinkers. Body weight was significantly higher in high alcohol drinkers followed by moderate and nondrinker groups. They also showed the higher serum values of triglyceride and r-GTP. High alcohol consumption resulted in an increased intakes of most nutrients especially calorie and fat which leads to higher body weight and higher serum concentrations of triglyceride and r-GTP. Therefore it seems that heavy alcohol drinkers are more prone to obesity and related chronic diseases.

Improvement of Broiler Meat Quality (브로일러 육질의 향상)

  • The consumer demands for quality of meats has become diverse in recent years. The present paper describes mainly the technology to improve the broiler meat quality with special reference to reducing fat contents in edible meats which is the heart of the quality constraints. Abdominal fat deposition in broilers was reduced by feeding of medium-chain triglycerides(MCT), suggesting MCT feeding is useful to produce broiler meat with low fat content. A phase feeding system to aim at improving meat quality that is mainly comprised with partial replacement of dietary protein into phase during 4∼6 weeks increased edible meat yields and reduced abdominal fat deposition and fat contents in breast and thigh meats. Whiteness of fat tissue was intensified by feeding beef tallow or lard in place of yellow grease. Feeding Phaffia yeast containing astaxanthin increased redness of breast and thigh meats and improved visual appearance of meats which may be preferential for consumers. Feeding fish oil reduced abdominal fat deposition and increased EPA and DHA contents of fat tissues. These procedures could be used for manipulation of meat quality to meet consumer demands.

Antimicrobial, Antioxidant and Hemolytic Activity of Water-soluble Extract of Mottled Anemone Urticina crassicornis

  • We evaluated the biological activities of five water extracts of tissue of the mottled anemone Urticina crassicornis. Most extracts exhibited broad-spectrum antimicrobial activity as determined by ultrasensitive radial diffusion assay (URDA) against gram-positive and -negative bacteria, including a fish pathogen, Aeromonas hydrophila, but no activity against fungi. The activity of the extracts was abolished by tryptic digestion, indicating that protein compounds were responsible for the antimicrobial activity. Furthermore, in a 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical-scavenging activity assay, only the visceral tissue extract showed activity. However, no extract had hemolytic activity against human red blood cells. Consequently, this study suggests the water-soluble extract of mottled anemone to be a promising source of proteinaceous antimicrobial compounds that can be utilized for development of novel antibiotics.

  • The giant croaker, Nibea japonica is a large, estuarine sciaenid distributed around southem Korea and Japan. In Repubic of Korea, giant croaker is a highly desired commercial and recreational species and there is interest in develoment of techniques for production of fish for enhancement of wildstocks and aquaculture since the giant croaker possesses some desirable characteristics for aquaculture including tolerance to widely varying salinities and receptivity to induced spawning in captivity. Ingestion of Polystyrene Microplastics Acutely Induces Oxidative Stress in the Marine Medaka Oryzias javanicus

  • Larvae from the marine medaka fish Oryzias javanicus were exposed with polystyrene microplastics (MPs) for 24 h. Exposure to waterborne fluorescent MPs showed clear ingestion and egestion in feces. Under constant MPs, the concentration of dissolved oxygen significantly decreased in 24 h compared to the control. Significant intracellular reactive oxygen species and malondialdehyde contents were detected in larvae, indicating oxidative stress and lipid peroxidation. Significant elevations in mRNA expressions of heat shock protein 70 and antioxidant defense system genes (glutathione reductase, glutathione peroxidase, catalase, and superoxide dismutase) were measured with increases in enzymatic activity of oxidative stress-related proteins. Taken together, the alterations to the molecular and biochemical components suggested that waterborne MPs had an oxidative stress effect on marine medaka larvae.

  • Organisms constituting a large proportion of marine ecosystems, ranging from bacteria to fish, exhibit fluorescence and bioluminescence. A variety of marine organisms utilize these biochemically generated light sources for feeding, reproduction, communication, and defense. Since the discovery of green fluorescent protein and the luciferin-luciferase system more than a century ago, numerous studies have been conducted to characterize their function and regulatory mechanism. The unique properties of fluorescent and bioluminescent proteins offer great potential for their use in a broad range of applications. This short review briefly describes the functions and characteristics of fluorescent and bioluminescent proteins, in addition to summarizing the recent status of their applications.

마우스 수정란의 발생단계별 telomeric DNA의 분포양상과 telomerase activity 분석

  • Telomere란 진핵세포에 존재하는 DNA-protein 복합체로서 염색체의 말단부에 tandem repeated DNA 서열 (TTAGGG)과 특정 단백질로 구성되어 있으며 세포 분열이 진행함에 따라 이의 길이가 짧아지게 되고 일정 길이 이하가 되면 세포의 사망이 유발된다. 반면 telomerase는 ribonucleoprotein으로서 telomeric DNA의 합성에 관여하는 것으로 염색체의 말단에 telomeric DNA의 소실을 보충하는 역할로 알려져 있다. 최근 암, 노화 등과 관련하여 telomere 및 telomerase의 연구들이 활발히 진행되고 있으며, 다양한 세포들에 있어 이들의 존재와 역할에 대해서도 많은 연구들이 수행되고 있다. 포유동물의 초기 배자에 있어 telomere의 분포 양상과 telomerase의 activity의 분석은 배 발생의 기작과 배자의 세포적 특성을 구명하는데 매우 중요한 과제라 사료된다. 따라서 본 연구에서는 마우스의 초기 배 발생 단계별 수정란의 telomeric DNA의 분포 양상과 각 단계별 배자들의 telomerase activity를 제시하고자 하였다. 시험에 공시된 마우스는 4-6주령된 ICR계통으로 이들을 과배란 처리 후 자연 교배시켜 얻은 2-, 4-, 8-세포기배, 상실배 및 배반포배를 대상으로 하였다. Telomeric DNA의 양적 분석은 각 발생단계별 수정란의 표본을 제작하고 human telomere repeat probe를 이용하여 FISH (fluorescence in situ hybridization)를 시행하였으며, 분리된 할구들을 형광현미경으로 관찰 후 상을 포착하고 image analyser program (MataMorph, UIC, USA)을 이용하여 한 개의 세포내 telomere의 상대적 함량을 분석하였다. 발생 단계별 배자의 telomerase activity의 분석은 TRAP (telomeric repeat amplofication protocol) assay로 분석한 바 각 발생 단계별 30개의 수정란으로부터 핵 단백질을 추출하여 telomerase를 신장시키고 PCR을 시행한 후 15% PAGE gel loading하여 이의 activity를 확인하였다. 분석 결과, telomeric DNA의 함유율은 발생단계별 다소의 차이를 나타내었으며 telomerase activity는 모든 발생단계의 수정란에서 확인할 수 있었고, 특히 상실배부터 높게 나타남을 확인하였다.

Optimum Feeding Rates in Juvenile Olive Flounder Paralichthys olivaceus Fed Practical Expanded Pellet at Low and High Water Temperatures (저수온기 및 고수온기에 있어서 배합사료를 공급한 넙치(Paralichthys olivaceus) 치어의 적정 공급율)

  • Two feeding trials were conducted to investigate the optimum feeding rates in juvenile olive founder Paralichthys olivaceus fed practical expanded pellet(EP) containing 58.1% crude protein, 10.7% crude lipid, and 10.8% ash at low and high water temperatures. In the first experiment, triplicate groups of 15 fish with average weight of 7.7 g were fed at one of seven feeding rates(0, 1, 1.5, 2, 2.25, or 2.5% of body weight/day or to satiation) for 4 weeks at low water temperature. In the second experiment, quadruplicate groups of 20 fish with average weight of 5.5 g were fed at one of eight feeding rates(0, 3, 3.5, 4, 4.25, 4.5, or 4.75% of body weight/day or to satiation) for 4 weeks at high water temperature. Based on growth performance, we estimated that the optimum feeding rates for juvenile olive flounder were 1.97-2.51% and 4.82-6.36% of body weight/day at low and high water temperatures, respectively.
