This study was conducted to evaluate the effects of dietary arachidonic acid (AA, 20:4n-6) levels on growth performance and body composition in juvenile eel, Anguilla japonica. Six semi-purified experimental diets were formulated to be isonitrogenous and iso-caloric containing 55.0% crude protein and 15% crude lipid (18.3 kJ of available energy $g^{-1}$). Six different levels of AA were added to the basal diet, with 0, 0.2, 0.4, 0.6, 0.8 or 1.2% on a dry matter (DM) basis, respectively ($AA_{0.07},\;AA_{0.22},\;AA_{0.43},\;AA_{0.57},\;AA_{0.78}\;or\;AA_{1.23}$). After a conditioning period, fish initially averaging 27${\pm}$0.5 g (mean${\pm}$SD) were randomly distributed into each aquarium as triplicate groups of 20 fish each. One of six experimental diets was fed on a DM basis to fish in three randomly selected aquaria at a rate of 2-3% of total body weight twice a day. At the end of the 12-week feeding trial, weight gain (WG) and feed efficiency (FE) of fish fed $AA_{0.78}$ and $AA_{1.23}$ diets were significantly higher than of fish fed $AA_{0.07},\;AA_{0.22},\;AA_{0.43}$ diets (p<0.05). Specific growth rate (SGR) of fish fed the $AA_{0.78}$ diet was significantly higher than of fish fed $AA_{0.07},\;AA_{0.22},\;AA_{0.43}$ diets (p<0.05). However, there were no significant differences in WG, SGR and FE among fish fed $AA_{0.57},\;AA_{0.78}\;or\;AA_{1.23}$ diets (p>0.05). Whole body AA deposition of fish fed the $AA_{1.23}$ diet was significantly higher than for the other diets (p<0.05). Broken-line model analysis on the basis of WG and SGR indicated that the dietary AA requirement could be greater than 0.69% but less than 0.71% of the diet in juvenile eel. The growth-promoting activity of AA observed in the present study provides strong support for the contention that dietary AA is essential for juvenile eel.

This study was designed to re-evaluate the dietary methionine requirement by means of the plasma methionine and ammonia concentrations in surgically modified rainbow trout, Oncorhynchus mykiss. A total of 35 rainbow trout averaging $505{\pm}6.5$ g (initial body weight, mean${\pm}$SD) were randomly distributed into seven groups with five fish in each group. After 48 h of feed deprivation, each group of fish was fed one of seven L-amino acid based diets containing 0.5% cystine and graded levels of methionine (0.25, 0.40, 0.50, 0.60, 0.70, 0.80 or 0.95% of diet, dry matter bases) by intubation at 1% body weight on dry matter basis. Blood samples were taken at 0, 5 and 24 h after intubation. Post-prandial plasma free methionine concentrations (PPmet, 5 h after intubation) and post-absorptive plasma free methionine concentrations (PAmet, 24 h after intubation) of fish fed diets containing 0.60% or higher methionine were significantly (p<0.05) higher than those of fish fed diets containing 0.50% or lower methionine. PPmet and PAmet in fish fed diets containing 0.60% or higher methionine were not significantly different except PPmet of fish fed diet containing 0.95% methionine. Post-prandial plasma ammonia concentrations (PPA, 5 h after intubation) of fish fed diets containing 0.70% or higher methionine were significantly higher than those of fish fed diets containing 0.60% or lower methionine, and PPA of fish fed diets containing 0.25 and up to 0.60% methionine were not significantly different from each other. Broken-line model analyses on PPmet, PAmet, and PPA indicated that the dietary methionine requirement of rainbow trout was between 0.59 (1.69) and 0.67 (1.91) % of diets (% dietary protein bases) when the diets contained 0.5% cystine.
This study was conducted to investigate the effect of dietary oxidized oil and ${\alpha}$-tocopherol level on growth and body composition of juvenile flounder. To prepare oxidized diets, squid liver oil was oxidized by aeration at $25^{\circ}C$ for 30 days. The six diets were prepared to contain 6% fresh or oxidized squid liver oil as the lipid sources in combination with three levels of ${\alpha}$-tocopheryl acetate at 0, 80 and 800 mg/kg diet. Triplicate groups of fish ($3.9{\pm}0.1$) were fed to apparent satiation twice a day for 8 weeks. Survival was not significantly different among treatments. Weight gain, feed efficiency, daily feed intake, protein efficiency ratio and condition factor of fish fed the fresh oil diets were significantly higher than those of fish fed the oxidized oil diets (P<0.05). The increase of the vitamin E level in diets did not result in any significant improvement on growth performance of fish fed both oil diets. The vitamin E content of the liver and dorsal muscle increased with increasing dietary vitamin E level at both oil diet groups. A decreasing trend in vitamin E content of the tissues was observed in fish fed the oxidized oil diets at the same dietary vitamin E level. Significantly higher moisture content and lower crude lipid content were observed in the whole body of fish fed the oxidized oil diets than fish fed the fresh oil diets (P<0.05). Dietary lipid source affected the fatty acid content of the whole body; higher contents of saturated and monoenoic fatty acids, and lower n-3 HUFA contents such as 20:5n-3 and 22:6n-3 were observed in fish fed the oxidized oil diets than those of fish fed fresh oil diets. The results of this study suggest that the dietary oxidized oil may impair the growth performance, and an increase in ${\alpha}$-tocopheryl acetate supplementation have no beneficial effect on growth and feed efficiency of juvenile flounder.
Two consecutive feeding trials investigated the effects of feeding rate and pellet expansion by water-soaking on the growth performance, blood components and histology of olive flounder Paralichthys olivaceus. The first two experiments were carried out to determine the effects of pellet expansion and feeding rate. In the first experiment, growth performance, feed utilization and survival of fish were not significantly affected by pellet expansion for six weeks. There were no significant differences in hematocrit, hemoglobin, aspartate aminotransferase, alanine aminotransferase, glucose and total protein of fish fed the expanded pellet. However, whole-body lipid content of fish in the non-expanded group was significantly higher than that in the expanded group. Histological analysis of the anterior intestine revealed that fish in the expanded group had shorter and smaller mucous folds. These results indicate that pellet expansion had no beneficial effect in terms of growth performance, feed utilization and fish health. In the second experiment, weight gain and feed efficiency were significantly increased as feeding rate increased from 0 to 2.5% body weight per day (BW/d), but there were no significant differences in weight gain or feeding efficiency in fish fed the expanded pellet at ratios of 2.5% BW/d and satiation for three weeks. Plasma aspartate aminotransferase activity of fish fed the expanded pellet at a ratio of 2.5% was significantly lower than that of starved fish. Histological analysis of the anterior intestine revealed that fish in the 0% group had shorter mucous folds. Broken-line regression analysis suggested that the optimum juvenile olive flounder feeding rate was 3.5% BW/d during the low temperature season ($16-17^{\circ}C$).
Magnesium hydrogen phosphate (MHP, $MgHPO_4$) recovered from swine manure was prepared as an alternative phosphorus (P) source. Conventional P additives, monocalcium phosphate (MCP), dicalcium phosphate (DCP), and tricalcium phosphate (TCP) were compared with the MHP in terms of growth and P availability by juvenile carp Cyprinus carpio. A basal diet as a negative control was prepared using practical feed ingredients without P supplementation to which four supplemental P sources were added at the level of 2%. Five groups of 450 fish having mean body weight of 6.5 g following 24 h fasting after 2 weeks of adaptation period were randomly distributed into each of 15 tanks (30 fish/tank). Fish were hand-fed to apparent satiety twice a day for 9 weeks. Fish fed the MHP had weight gain (WG), feed conversion ratio (FCR), protein efficiency ratio (PER), and specific growth rate (SGR) comparable to those fed the MCP. Those values of both the MHP and MCP groups were significantly different (p < 0.05) from the other groups. Fish groups fed control and the TCP showed the lowest WG, PER, and SGR and the highest FCR among treatments. No fish were died among treatments during the experimental period. Fish fed control and the TCP showed hematocrit and hemoglobin significantly lower (p < 0.05) than fish fed the MHP. The lowest inorganic P (Pi) in plasma was found in the control group. Even though Pi was not significantly different (p > 0.05) from other phosphate groups, fish fed the MCP and MHP retained higher P in whole body than the other groups. P availability was determined to be 93.2, 62.4, 6.1, and 98.0% for MCP, DCP, TCP, and MHP, respectively. The present results suggested that the MHP recovered from wastewater stream could be used as an alternative P source in carp diet.
도다리를 이용한 뼈째썰기회는 봄철 대표적인 생선회로 비교적 양식이 어려운 어종이다. 따라서 대부분 자연산 도다리를 이용하여 뼈째썰기회를 만들어야 하지만 최근 자연산 도다리의 수급과 비용의 문제로 인해 유사어종인 양식산 소형넙치 및 중국산 양식 돌가자미를 도다리로 둔갑시켜 판매 되는 실정이다. 양식을 통하여 계절에 관계없이 구입이 가능한 소형넙치와 중국에서 수입되는 양식산 돌가자미는 도다리에 비해 가격이 저렴하고, 회로 만들면 동일한 가자미과에 속해있어 구별이 어려운 어종이다. 따라서 봄철 소비가 많은 도다리 뼈째썰기회를 저렴한 돌가자미 및 소형넙치로 둔갑시켜 파는 경우가 많으며, 도다리회 전문점의 수조에서 돌가자미 및 소형넙치가 도다리 대신 판매되기 위해 볼 수 있는 경우가 많다. 따라서 이러한 도다리 유사어종에 대한 판별법이 중요하기 때문에 본 연구에서는, 도다리 및 도다리 유사어종의 근육 중 SDS 전기영동과 지방함량을 통해 어종을 판별하였다. 즉, 자연산 도다리와 양식산 소형넙치, 중국산 양식 돌가자미 근육의 식품학적 성분을 분석, 비교를 통해 도다리의 크기에 따른 성분 함량의 차이를 알아보고, 시판 뼈째썰기회를 구입하여 SDS 전기영동 및 지방함량을 이용하여 분석, 비교하였다. 본 연구를 바탕으로 얻어진 결과는 외식산업에 자연산 도다리 뼈째썰기회에 대한 기초지식을 제공 할 것으로 사료되어지지만 분자량과 성분적인 측면에서만 연구된 사항에 대해 좀 더 명확한 결과를 위하여 차후 필수 연구인 정밀분석적인 측면에서 연구가 필요할 것으로 판단되어지며 검증을 통한 확실한 방법 확립 등이 추가적으로 필요할 것으로 판단된다.
This experiment was conducted to determine RDP values of locally available feedstuffs that are commonly used in ruminant rations in Bangladesh. Four cattle were fistulated in the rumen for the in situ nylon bag studies. Seventeen different feedstuff sample (9 roughages and 8 concentrates) were evaluated in $4{\times}14cm$ nylon bags and incubated in the rumen for different periods of time (2, 6, 12, 24, 48 and 72 h). The variation in crude protein (CP) contents reflected on the average CP disappearance value throughout the rumen incubation. Soluble fraction (a), insoluble but degradable fraction (b) along with the rate of degradation also varied widely among the various feedstuffs. Under 2% of rumen outflow rate, the percentages of the calculated protein degradabilities of roughages were rice straw, 16.7; maize grass, 70.6; oat grass, 70.8; dhal grass, 71.1; sunhemp, 78.4; napier grass, 62.4; matikalai grass, 72.1; khesarikalai grass, 76.9 and daincha browse, 78.4, respectively. The results in the protein degradabilities (%) in 8% ruminal outflow rate of concentrates were wheat bran, 61.6; rice polish (red), 61.3; rice polish (auto), 30.9; mustard oil cake, 71.8; sesame oil cake, 74.2; coconut oil cake, 57.9; soybean meal, 49.2 and fish meal, 37.9, respectively.
Immunomagnetic separation of virus coupled with .reverse transcription-polymerase chain reaction (IMS-PCR) was performed with infectious hematopoietic necrosis virus (IHNV). A DNA fragment of expected size was synthesized in the RT-PCR with total RNA extracted from IHNV inoculated CHSE-214. In a SDS-PAGE analysis, a protein band of over 70kDa was detected from non-infected cells and cells inoculated with IHNV and infectious pancreatic necrosis virus (IPNV). This protein was detected in the Western blot analysis probably because of non-specific reaction to monoclonal antibody against IHNV nucleocapsid protein. In the immunomagnetic separation, magnetic beads coated with monoclonal antibody against the IHNV nucleocapsid protein was incubated with supernatant from IHNV inoculated CHSE-214 cells. During this process, the non-specifically reacting protein could be removed by washing the magnetic bead with PBS in the presence of an external magnetic field, and viral proteins were detected from the remaining, cleaned magnetic beads. It was necessary to extract viral RNA from the captured virus particles before RT-PCR, and no DNA product was detected when the captured virus was only heated 5 min at $95^{\circ}C$. A PCR-product of expected size was synthesized from IMS-PCR with magnetic beads double coated either by goat anti-mouse IgG antibody -monoclonal antibody or streptavidin - biotin conjugated monoclonal antibody.

But the nutritional quality of milk lipids cannot be inferior to those of margarine, fish oil, and vegetable oil. Milk has been produced commercially since 1936 in Korea. The most popular milk product is a drink milk now in Korea. But according to the change of dietary pattern the consumption of yoghurt and cheese has been enormously increased during the last 20 years. As the soy sauce, tofu, and soy been paste have been the fundamental seasoning and source of protein in Korea, milk and cheese have consisted the essential flavor of western cuisine. But the basic idea for the usage of protein, peptides, and amino acids are the same. We found that milk and milk products can be nicely added in many Korean dishes such as Juk, Mandoo, Jeon, and Bindeatuk for the diversity of traditional flavor.
