Analysis of body size selectivity of by-catch using the cover net method for a shrimp beam trawl

  • Selectivity and by-catch are the main features used to define fish catch for fisheries management or to determine fishing gear efficiency. A fish girth distribution analysis was carried out to determine the retention rate of juveniles discarded and to establish common selectivity for a multispecies catch. Total body length, body weight, and girth of juvenile fish retained in the cod-end and cover net separately were collected using an 18-mm mesh cover net during 12 fishing trials with a 41-mm cod-end beam trawl. The by-catch weight ratio in the cod-end was twice that of the by-catch ratio in number. The 50% selectivity of body length and 50% girth of redfin velvet fish were not significantly different between the cod-end and cover net, whereas those of other fish species were significantly different between the cod-end and cover net. The difference in 50% selectivity girth of other fish between the cod-end and cover net was similar between cod-end mesh size and cover net mesh size. Furthermore, the difference in 50% body length selectivity of other fish between the cod-end and cover net was almost double the difference in girth. Girth selectivity in a multispecies catch using towed fishing gear was effectively used to determine fish and net mesh size.

Effect of Korean Mistletoe (Viscum album Coloratum) on the Non-Specific Immune Responses in Japanese Eel (Anguilla japonica)

  • In the present paper, the immunostimulatory effects of Korean mistletoe (Viscum album Coloratum) on the non-specific immune responses of Japanese eel (Anguilla japonica) were examined. Eel were innoculated with mistletoe, Freund's complete adjuvant (FCA), or phosphate-buffered saline (PBS) as a control into their peritoneal cavities. The number of nitrobule tetrazolium (NBT)-positive cells in the head kidney of fish was significantly increased by the second day post-injection of mistletoe. ROI products were more enhanced in mistletoe-injected fish kidney leucocytes than in FCA-injected ones. The level of lysozyme activity detected in the serum of fish 2 days after injection with mistletoe was also significantly higher than that found in the serum of the control fish. The appropriate concentration of mistletoe to induce the highest level of serum lysozyme activity was revealed to 1000${\mu}g$/200 g of fish. In phagocytic activity assay, mistletoe-sensitized eel kidney phagocytes captured more zymosan than did the control fish. Korean mistletoe appeared to be a good activator of the non-specific immune responses of Japanese eel.

Studies on the Tripartiella sp from three species of freshwater freshwater fish, Cyprinus carpio, Crassius auratus and Parasilurus asotus in Korea (한국산 담수어에 기생하는 섬모충 Tripartiella sp에 관한 연구 -잉어, 금붕어 및 메기를 중심으로-)

  • To investigate the species and infection rate of Tripartiella sp in three species of freshwater fish, carp(Cyprinus carpio), goldfish(Crassius auratus) and Korean catfish(Parasilurus asotus), fish cultured in the fish farms next to Yosu and in the western area of Chunbuk province in Korea was examined from April to June, 1996. The infection rate was evaluated after observing the gill and mucous from 130 fish under the microscope. The species of Tripartiella was identified on the basis of the shape and number of denticle according to methods of Bychowsky(1985) after staining the parasites with 2% methyene blue solution. All the detected-parasite was classified as Tripartiella differed from Trichodina reported previously in our country. T carassii, T californica, T rhombi from Goldfish, T rhombi from common carp, T californica from Israeli carp and T bychowsky from Korean catfish were observed. In the fish from the fish farms in the western area of Chunbuk province, the infection rate of Tripartiella sp was ranged from 6.7% to 100%, but in neither carp nor goldfish from the farms next to Yosu the parasite detected.

  • Fish mortality is the most important success factor in aquaculture management. To order fingerlings considering the effect of mortality is a important problem in aquaculture farm. This study is aimed to decision the number and size of fry in aquaculture farm. This study build the mathematical model that finds the value of decision variable to minimize total cost that sums up the fingerling purchasing cost, aquaculture farm operating cost and feeding cost under mortality constraint. The proposed mathematical model involve biological and economical variables: (1) number of fingerlings (2) fish growth rate (3) mortality (4) price of a fry (5) feeding cost, and (6) possible order period. Numerical simulation model presented here in. The objective of numerical simulation is to provide for decision makers to analyse and comprehend the proposed model. When extensive biological and cost data become available, the proposed model can be widely applied to yield more accurate results.

  • Fish mortality is the most important success factor in aquaculture management. To analyze the effect of mortality considering biological and economic condition is a important problem in land-based aquaculture. This study is aimed to analyze the effect of mortality for duration of cultivation in land-based aquaculture. This study builds the mathematical model that finds the value of decision variable to minimize cost that sums up the water pool usage cost, sorting cost, fingerling cost and feeding cost under critical standing corp constraint. The proposed mathematical model involves many aspects, both biological and economical: (1) number of fingerlings (2) timing and number of batch splitting event, based on (3) fish growth rate, (4) mortality, and (5) several farming expense. Numerical simulation model presented here in. The objective of numerical simulation is to provide for decision makers to analyse and comprehend the proposed model.   • This study describes the optimum size of a reef set in Korean coastal waters. Data were analyzed in relation to the weight and variety of fish species caught with a three-layer gill net. The results did not indicate a significant difference in the mean catch and mean number of fish species among the three different reef sets, 800, 1,600 and 2400 $m^3$.

  • Twelve black seabreams (Acanthopagrus schlegeli) were tagged with acoustic transmitters and their movement and behavior patterns were tracked using acoustic telemetry from 12 December 2006 to 15 August 2007. Seven of the 12 fish stayed within 500 m of the release point for over a month. In the spring tide, the number of detected signals and swimming depth of tagged fish changed at 12-hour intervals with the ebb tide. The number of detected signals of tagged fish decreased dramatically with the passage of time and had decreased by over 80% in May 2007, compared with January 2007.

  • Raw-fish food contains a lot of moisture and is a high-protein food. It is a first-stage processed food taking a lot of manual work. Therefore, it is classified as a PHF food, very liable to cause a bacterial food-poisoning. But its manufacturers are usually small-sized and a systematic sanitation management is difficult to expect. But the manufacturer participating in this study produces chilled fresh raw-fish food. Fish are sliced into two fillets, which are packaged under vacuum, kept and distributed in refrigerators, and sold within a day. It is a newly-developed kind of raw-fish food, and a more improved kind of raw-fish food making possible a systematic sanitation management. The HACCP (Hazard Analysis and Critical Control Point) is a systematic and continuous process-control method which is very efficient for controling food sanitation and reducing the expenses. A new HACCP model has been developed to be applied to a large-sized chilled fresh raw-fish food manufacturer. To ascertain its efficiency, the baterial examination was done to its workplace and products. The significance test was done on its data by "SPSS 12.0 for Window" and "Mann-Whitney U Test". The numbers of bacteria on its final products were significantly different in flatfish and porgy. The number of bacteria tended to decrease in each time-differential sampling (P<.00l). The final food products showed no food-poisoning bacteria in all the time-differential tests and in all the samplings, which proves that the CCP of the HACCP system is under control. After the SSOP program was applied, no pathogenic bacteria were found in the work-place, and the kinds and numbers of bacteria decreased. The numbers of general bacteria and colon bacilli also showed a significant difference from those before the SSOP program in the filleting board (P<.05), in the skinning board (P<.0l), in the neck-removing knife (P<.05), and in the filleting knife (P<.01). The working equipments, periodically disinfected, also showed a significant difference in sanitary conditions (in the dehydrator, P<.05). The number of bacteria found on the food-touching surface was within the standard (below 500/l00 cm$^2$) After the SSOP program was applied, the general bacteria and colon bacilli were not found. The quality of water used in the food processing was also within the standard. The numbers of bacteria falling from the air in the work-place were negligible in all the samplings (<30CFU/l000ι). The staphylococci and fungi were not found.

Comparison of Digestive Organ and Body Composition among the Cultured, Wild and Released Fish, 1-Year Black Sea Bream (Acanthopagrus schlegeli) (양식, 방류 및 자연산 감성돔 1년어의 소화기관 및 체성분 비교)

  • In order to evaluate adaptation to the environment in released fish, digestive organ and body composition were compared among 1-year old cultured (CUL), released (REL) and wild (WIL) Black Sea Bream, Acanthopagrus schlegeli. Hepatosomatic index (HSI) of the REL was significantly lower than those of the CUL and WIL fish, but intestine weight index (IWI) of REL was significantly higher than those of the CUL and WIL fish. There were no significant differences in stomach weight index (SWI) and condition facto. (CF) among those different fish groups. The number of pyloric caeca of the REL and WIL fish were 4, but the $30{\%}$ of CUL fish was 3 counts. The moisture content of whole body in the WIL fish was significantly higher than the CUL and REL fish, but significant lowest in the crude lipid of whole body. The content of moisture and crude protein of dorsal muscle were no significant differences among the CUL, REL and WIL fish, but crude lipid content of WIL fish was significantly lower than the CUL and REL fish. Moisture content of CUL fish in the liver was significantly lower than the REL and WIL fish, and crude lipid was significant lowest in the WIL fish. Amino acid content of dorsal muscle in the WIL fish was highest in the total amino acid, EAA and E/A ratio, and CUL fish was highest EAA and E/A ratio in liver. EPA content in dorsal muscle of WIL fish was lower than CUL and REL fish, and the REL fish was highest in DHA content of the fatty acid. EPA content in liver of WIL fish was lower than CUL and REL fish, and DHA content in REL and WIL fish was highest and lowest. Crude protein content of CUL and WIL fish in the scale was significantly higher than REL fish, but there were no significant differences in contents of crude lipid and ash. Ca and P of scale were formed most of mineral and content of P in CUL fish was significantly higher than those of REL and WIL fish. There were no significant differences in Mg, K, Na and S of scale, but CUL fish was significantly lower than REL fish in C1. The results suggest that difference of digestive organs and body composition concluded it from differences of inhabitation environment and feed formulations.

Efficacy of Primed In Situ Labelling in Determination of HER-2 Gene Amplification and CEN-17 Status in Breast Cancer Tissue

  • Considerable attention has been given to the accuracy of HER-2 testing and the correlation between the results of different testing methods. This interest reflects the growing importance of HER-2 status in the management of patients with breast cancer. In this study the detection of HER-2 gene and centromere 17 status was evaluated using dual-colour primed in situ labelling (PRINS) in comparison with fluorescence in situ hybridization (FISH). These two methods were evaluated on a series of 27 formalin fixed paraffin embedded breast carcinoma tumours, previously tested for protein overexpression by HercepTest (grouped into Hercep 1+/0, 2+ and 3+). HER-2 gene amplification (ratio${\geq}2.2$) by PRINS was found in 3:3, 6:21 and 0:3 in IHC 3+, 2+ and 1+/0 cases, respectively. Comparing FISH and IHC (immunohistochemistry), showed the same results as for PRINS and IHC. Chromosome 17 aneusomy was found in 10 of 21 IHC 2+ cases (47.6%), of which 1 (10%) showed hypodisomy (chromosome 17 copy number per cell${\leq}1.75$), 7 (70%) showed low polysomy (chromosome 17 copy number per cell=2.26 - 3.75) and 2 (20%) showed high polysomy (chromosome 17 copy number per cell ${\geq}3.76$). The overall concordance of detection of HER-2 gene amplification by FISH and PRINS was 100% (27:27). Furthermore, both the level of HER-2 amplification and copy number of CEN17 analysis results correlated well between the two methods. In conclusion, PRINS is a reliable, reproducible technique and in our opinion can be used as an additional test to determine HER-2 status in breast tumours.
