Identification of Vibrio species isolated from cultured olive flounder (Paralichthys olivaceus) in Jeju Island, South Korea

  • Olive flounder (Paralichthys olivaceus) is the major species developed for aquaculture in South Korea. Over the long history of olive flounder aquaculture, complex and diverse diseases have been a major problem, negatively impacting industrial production. Vibriosis is a prolific disease which continuously damages olive flounder aquaculture. A bacterial disease survey was performed from January to June 2017 on 20 olive flounder farms on Jeju Island. A total of 1710 fish were sampled, and bacteria from the external and internal organs of 560 fish were collected. Bacterial strains were identified using 16 s rRNA sequencing. Twenty-seven species and 184 strains of Vibrio were isolated during this survey, and phylogenetic analysis was performed. Bacterial isolates were investigated for the distribution of pathogenic and non-pathogenic species, as well as bacterial presence in tested organs was characterized. V. gigantis and V. scophthalmi were the dominant non-pathogenic and pathogenic strains isolated during this survey, respectively. This study provides data on specific Vibrio spp. isolated from cultured olive flounder in an effort to provide direction for future research and inform aquaculture management practices.

Correlation of virus replication and spleen index in rock bream iridovirus infected rock bream Oplegnathus fasciatus

  • Rock bream iridovirus (RBIV) is a member of the Megalocytivirus genus that causes severe mortality to rock bream (Oplegnathus fasciatus) with characteristic clinical signs of spleen enlargement. In this study, we assessed spleen size and RBIV copy number patterns in RBIV-infected rock bream to determine lethal and safe levels of virus copy number/spleen index that may define disease progress. We found that rock bream infected with RBIV ($1.1{\times}10^7virus\;copy\;number/100{\mu}l$) and held at 29, 26, 23 or $20^{\circ}C$ exhibited significantly higher levels of spleen size compared to $17^{\circ}C$. In dead condition (100% mortality at $20{\sim}29^{\circ}C$), the spleen index ($spleen\;weight/fish\;weight{\times}100$) and virus copy number were 3.00~5.38 and $10^6{\sim}10^8/{\mu}l$, respectively. Conversely, in survived condition (0% mortality at $17^{\circ}C$), spleen index and virus copy number was as low as not-infected control ($0.34{\sim}1.22/10^0{\sim}10^1/{\mu}l$, respectively). These findings suggest that spleen index can be an indicator of disease severity of RBIV disease.

2010년 방류용 수산종묘에 대한 병원체 검출 (Detection of fish pathogens in cultured juveniles for stock enhancement in 2010)

  • 배양장에서 생산된 양식생물은 자원조성에 필요한 종묘를 제공한다는 측면에서 매우 중요한 역할을 담당하고 있으며, 방류종묘의 질, 특히 개체의 건강도 는 방류이후 서식처에서 생존하고 자원조성 효과를 결정짓는데 매우 중요한 요소가 된다. 2010년도 방류 품종(해면품종 33종과 내수면품종 12품종)을 대상으로 수산동물전염병의 감염 여부를 검사하였다. 검사 품종 중에서 해면품종으로는 전복이 20.0%로 가장 많았으며, 그 다음 해삼(15.6%), 넙치(8.4%), 조피볼락(6.7%), 감성돔(6.3%), 꽃게(6.1%) 순으로 나타났다. 내수면품종 중에서는 붕어가 19.4%로 가장 많았으며, 그 다음으로 뱀장어(11.8%), 동자개(10.9%), 쏘가리(10.8%), 다슬기(8.4%), 메기(7.7%)의 순으로 나타났다. 총 45개 품종을 대상으로 1,120회의 검사가 의뢰되었으며, 검사 항목별로 2,105건의 검사를 실시한 결과 30건에서 koi herpesvirus (KHV), red sea bream iridovirus (RSIV), white spot syndrome virus (WSSV) 또는 viral haemorrhagic septicemia virus (VHSV)와 같은 병원체가 검출되어 불합격 처리되었다.

  • There occurred a large quantity of death of the cultured carp (Cyprinus carpio) in net pens in Lake Taechong and Lake Chungju(Chungcheongbuk-do) from July through Sep., 1987. The carp were two-year-old and 500g to 2,500g in body weight. As a result of biopsy of dying carp, 1 through 10 egg-shaped giant cysts with 12 through 60mm in diameter were observed in their intestine per one carp. The author suggested that the carp were died from blockade of intestinal tract and following enteritis by the giant cysts. The giant cysts were filled with numerous mature Thelohanellus genus spores confirmed to Thelohanellus kitauei after following examination. In this case the author could find out this disease was spread by transverse infection following infestation in net pens of fingerling fish farms.

Effects of methylmercury on the infants

  • In the natural course of events, most human exposure to MeHg is through fish/shellfish consumption. The methylmercury exposure levels depends on the amount and species of fish/shellfish consumed daily. The developing brain in the late gestation period is known to be most vulnerable. Further, more methylmercury accumulates in the fetuses than in mothers. Therefore, efforts must be made to protect the fetuses from the risk of methylmercury, especially in populations which consume a lot of fish/shellfish.

남해지역 자연산 해산어의 Trichodina 감염 현황 (Survey of Trichodina infection in wild populations of marine fish caught from Namhae region, southen coast of Korea)

  • The ectoparasite ciliate Trichodina has been recorded from above 100 species of freshwater and marine fishes. In April 2009, we investigated the trichodia infection in 13 species of marine fishes captured by emplacement net and gill net in order to understand trichodina infection status among the natural fish populations along the Namhae-Gun coast area, the southen coast of Korea. Trichodina infection was observed in 10 out of 10 sea basses Lateolabrax japonicus, 2 out of 5 finespotted flounder Pleuronichthys cornutus and 1 out of a stone flounder Kareius bicoloratus. The sea bass infected with the parasite showed hypertrophy and exfoliation in gill epithelium. In addition, hyperplasia and lamellar fusion of gill filament were noticed among naturally affected sea bass.

한국의 양식넙치에서 분리한 Tenacibaculum maritimum의 특성과 자생식물 추출물에 대한 감수성 (Biological characterization of Tenacibaculum maritimum isolated from cultured olive flounder in Korea and sensitivity against native plant extracts)

  • Tenacibaculum maritimum (formerly Flexibacter maritimus)은 tenacibaculosis라 불리며, 해산어에서 궤양 및 괴사성 증상을 나타내는 질병원인체 이다. Tenacibaculosis는 다양한 양식 어종에서 있어서 경제적 측면에서도 중요한 질병 중 하나이며, 피부, 입, 지느러미, 꼬리 등과 같은 어체 표면에 심각한 궤양과 괴사로 인한 손상을 유발시 킨다. 본 연구에서는 tenacibaculosis의 증상을 나타내는 넙치 병어로부터 A-7 균주를 분리하였고, 형태학적 특성, 생화학적 성상 및 유전자 분석 결과를 바탕으로 T. maritimum으로 동정하였으며, 침지법을 통하여 넙치에 인위적으로 감염되는 것을 확인하였다. Tenacibaculosis를 제어하기 위해 약욕에 의한 항생제 투여가 많이 이루어지며, 그 중 oxytetracycline이 주로 사용되고있다. 그러나, 대부분의 양식어민과 소비자들은항생제 잔류와 환경오염 등의 문제를 우려하고있으며, 비싼 항생제의 사용으로 인하여 양식어의 생산 비용도 높아지고 있다. 이러한 문제점들을 극복하기 위하여 본 연구에서는 천연 자생식물 추출물을 항생제 대체제로 사용하기 위한 연구를 수행하였다. 총 35 종류의 천연식물의 항균 활성을 검색하였고, 많은 천연식물 추출물이 T. maritimum에 대하여 높은 항균 효과를 나타낸다는 것을 밝혔다.

Effects of dietary fish oil and trans fat on rat aorta histopathology and cardiovascular risk markers

  • Fish oil and shortening have been suggested to have opposite effects on cardiovascular disease (CVD). This study investigated the effect of shortening and fish oil on CVD risk factors and aorta histopathology, and the association between risk factors and aorta histopathology. Male Wister rats (n=30) were fed an AIN-93G diet containing 20% fat in the form of fish oil, shortening, or soybean oil for 4 weeks. Total cholesterol (TC), triacylglyceride (TG), and C-reactive protein levels were significantly (P<0.001) lower in the fish oil than in soybean oil and shortening groups. HDL-cholesterol concentrations were significantly different (P<0.001) between groups. In addition, LDL-cholesterol levels were significantly (P<0.001) lower in the fish oil and shortening groups than in the soybean oil group. Insulin and glucose concentrations did not differ among groups. Effect of dietary fat on tissue fatty acid composition significantly differed in abdominal fat and brain compared with RBC, heart, kidney and liver. The aortic wall was significantly (P=0.02) thinner in the fish oil group than in the soybean oil and shortening groups. The aortic wall thickness was positively correlated with TG and TC, but negatively with EPA + DHA levels of all tissues. These results suggested that fish oil had protective effects on aorta histopathology by hypolipidemic action in this rat model.

양식 어류에 질병을 유발하는 연쇄구균증의 특성 및 진단 방법 (Characteristics and Diagnostic Methods of Streptococcosis Causing Disease in Aquaculture)

  • 본 논문은 양식 어류에게 많은 경제적 피해를 입히고 있는 다양한 어류질병세균 중에서 연쇄구균증에 대한 일반적인 특성과 종류, 진단 방법에 대하여 기존에 연구 보고된 논문을 기반으로 알아보고자 한다. 대표적인 어류연쇄구균증의 원인체로는 Lactococcus garvieae, Streptococcus iniae, S. parauberis가 있다. 연쇄구균증에 감염 시 나타나는 증상으로는 체색변화, 안구이상, 아가미 퇴색, 출혈, 복부팽만, 신장과 비장의 종대 등의 보이다 폐사가 일어난다. 또한 수온이 상승하는 6월부터 10월까지 주로 일어나며 집단적으로 발병하여 폐사가 일어난다. 현재 연쇄구균증을 진단하기 위한 기술로는 16S rRNA, 16S-23S rRNA intergenic spacer region (ISR), Random Amplified polymorphic DNA (RAPD), Ribotyion (RT), Loop-mediated isothermal amplification (LAMP) 등이 있다. 이중에서 현장에서 적용 가능성이 높은 LAMP법이 각광을 받고 있지만 결과 확인 등의 문제로 인하여 현재는 어려움을 겪고 있는 실정이다. 이에 현장에서 진단부터 결과 확인까지 손쉽게 사용할 수 있도록 문제점을 보완하면 연쇄구균증에 대한 경제적 손실을 최소화 할 것으로 사료된다.

해조류(톳, 감태) 혼합물의 사료 내 첨가가 넙치의 선천성 면역과 질병저항성에 미치는 영향 (Effects of Dietary Supplementation of Alga Mixtures (Hizikia fusiformis and Ecklonia cava) on Innate Immunity and Disease Resistance Against Edwardsiella tarda in Olive Flounder (Paralichthys olivaceus))

  • We report non-specific immune responses and its disease resistance against Edwardsiella tarda by alga mixture (HE; Hizikia:Ecklonia) in olive flounder for the first time. Five isonitrogenous (44% crude protein) and isocaloric (17.1 MJ $kg^{-1}$) diets were formulated to have 0%, 2%, 4%, 6% and 8% of the alga mixture. One of five experimental diets was fed triplicate groups of fish (30 fish/group) to apparent satiation in a flow through system. After a two week feeding, blood was sampled at 3, 6, 12, 24 h after the last feeding for a kinetic measurement of nitroblue tetrazolium (NBT) activity and healthy fish with similar sizes in each tank were selected and injected with 1 mL of E. tarda suspension ($1.0\times10^7$ CFU/mL) to evaluate the disease resistance of the fish. Dietary supplementation of alga mixtures resulted in significantly higher non-specific immune responses compared with the fish fed the control diet. The cumulative mortality was significantly lower in the fish groups fed alga mixture containing diets than control group in the challenge test with E. tarda. Therefore, the results in this study indicate that dietary supplementation of Hizikia and Ecklonia mixtures enhance the non-specific immune responses and a disease resistance of olive flounder.
