Growth, Feed Utilization and Blood Metabolic Responses to Different Amylose-amylopectin Ratio Fed Diets in Tilapia (Oreochromis niloticus)

  • A feeding trial was conducted in tilapia to determine the growth performance, nutrient digestibility, digestive enzymes, and postprandial blood metabolites in response to different dietary amylose-amylopectin ratios. Five isonitrogenous and isolipidic diets containing an equal starch level with different amylose-amylopectin ratios of 0.11 (diet 1), 0.24 (diet 2), 0.47 (diet 3), 0.76 (diet 4) and 0.98 (diet 5) were formulated using high-amylose corn starch (as the amylose source) and waxy rice (as the amylopectin source). Each diet was hand-fed to six tanks of 15 fish each, three times a day over a 6-wk period. After the growth trial, a postprandial blood metabolic test was carried out. Fish fed diet 2 exhibited the highest percent weight gain and feed efficiency and protein efficiency ratio, whereas fish fed with diet 5 showed the lowest growth and feed utilization among treatments. The digestibility for starch in fish fed diet 1 and 2 was higher than those in fish fed with other diets (p<0.05). The highest activities for protease, lipase and amylase were found in fish fed the diet 2, diet 1, and diet 1 respectively among dietary treatments, while the lowest values for these indexes were observed in fish fed the diet 3, diet 5 and diet 4, respectively. The liver glycogen concentrations in fish fed diets 4 and 5 were found higher than in fish fed other diets (p<0.05). The feeding rate, hepatosomatic index, condition factor, and plasma parmeters (glucose, triglyceride, total cholesterol, high-density lipoprotein cholesterol, and low-density lipoprotein cholesterol) did not differ across treatments. In terms of postprandial blood responses, peak blood glucose and triglycerides were lower after 3 or 6 h in the fish fed with diets 3-5 than in the fish fed diet 1, but delayed peak blood total amino acid time was observed in fish fed with the diets 1 or 2. The lowest peak values for each of the three blood metabolites were observed in fish fed diet 5. The results indicate that high-dietary amylose-amylopectin ratio could compromise growth, but help in reducing the blood glucose stress on fish caused by postprandial starch load.

  • This study compared the density of fish determined using three different echogram methods: the frequency-difference, time variable, and threshold modification methods. An acoustic survey was conducted off the coast of Jeju Island after sunset. Data at 38 and 120 kHz frequencies were collected using a commercial fishing vessel. As a reference point, the value of ${\Delta}MVBS_{120-38kHz}$ that distinguished fish from zooplankton using the 38 and 120 kHz frequencies was set at < 2 dB. The estimated density of fish along the survey line was 0.1-30.4, 0.1-64.3, and $0.1-51.7m^2/nmi^2$ using the frequency difference, time variable threshold, and threshold modification methods, respectively. The results of this study constitute basic research for estimating fish densities.

고성군 용산리의 두 저수 수계에서 어류의 다양성과 밀도 효과 (Density Effect and Diversity of Fish in Water System at Both Reservoirs in the Youngsan-ri, Goseong-gun)

  • 경상남도 고성군 거류면 용산리 신은저수지 수계 4곳과 정촌 저수지 1곳 수계에서 어류 다양성 분석을 실시하였다. 인위적 교란과 환경변화에 따른 집단조절을 유도하는 밀도 의존성을 평가하였다. SMATR freeware를 사용하여 어류 밀도에 영향을 미치는 환경 요인 분석을 실시하였다. 2012년에 4과 8속 9종 158개체가 동정되었다. 비교적 풍부한 어종은 참붕어(Pseudorasbora parva)로 빈도는 33.1%였다. 그 다음은 송사리(Oryzias latipes)로 28.8%였다. 2012년에는 4곳에서 밀도 효과가 감소하였다. 신은저수지의 Shannon-Weaver의 다양도 지수(H’)는 정촌 저수지의 다양도와 유사하였다. 종 다양도는 0.645에서 2.105로 다양하게 나타났다. 상류지역의 다양도(H’)는 중류와 하류지역보다 높았다. 풍부도 지수 역시 상류가 하류보다 높았다. 강의 하류 두 지점을 제외한 최대 가능성 분석을 이용하면 한 지점에서 다른 지점으로 이동한 5종의 가능성은 평균 0.623이였다. 특히 신은저수지와 정촌저수지 간 이동 가능성은 높았다(평균 0.681). 이는 두 저수지간 지리적으로 짧은 탓에 기인한다(50 m). 나머지 4종에 대한 최대 이동가능성은 유의성이 없어 지점간 유사함을 시사한다.

하천에 설치된 횡구조물의 영향 및 섬진강 수계의 어류 풍부도 예측 (Impact of the Crossed-Structures Installed in Streams and Prediction of Fish Abundance in the Seomjin River System, Korea)

  • 섬진강 수계 210개 지방하천을 대상으로 하천연장과 보 밀도 대비 어류 출현종 현황을 분석하였다. 하천연장에 따른 어류 출현종수는 비선형지수함수 (Nonlinear exponential relationship) 관계를 보였다. 모델에 따른 계수값 (b)은 0.03이며, 결정계수(R2)은 약 59.0%인 것으로 나타났다. 모델식에 따른 예측기대종수는 실제 출현종수와 차이를 보였으며, 대상하천 가운데 약 110개 하천 (약 52.4%)에서 실제 출현종수는 예측기대종수보다 낮게 나타났다. 한편 섬진강 수계 하천에서 나타난 평균 보 밀도는 약 2.7개/km였으며, 이는 타 수계 하천과 비교시 월등히 높은 수치였다. 보 밀도 지표 (Index of Weir's Density, IWD)에 따른 어류출현종수간의 2-DKS 분석 결과 어류 다양성에 영향을 주는 보 밀도 역치값 (Threshold value)은 약 2.5개/km인 것으로 나타났다 (Dmax=0.048, p<0.05). 실제 보 밀도 지표가 역치값 이상인 경우 어류 출현종수 대비 기대 종수비율은 70% 미만으로 낮아졌다. 따라서, 향후 수생태계 연속성 확보를 위한 보 밀도는 역치값 이하로 관리하는 것이 필요하다.

배합사료의 크기, 공급횟수 그리고 사육밀도가 조피볼락(Sebastes schlegelii) 치어의 성장에 미치는 영향 (Effect of Food Particle Size, Stocking Density and Feeding Frequency on the Growth Performance of Juvenile Korean Rockfish Sebastes schlegelii)

  • We performed four sets of feeding trials to establish optimum feed size (Exp-I), stocking density (Exp-II) and feeding frequency (Exp-III and IV) for Korean rockfish. In Exp-I, three different particle sizes of a commercial diet (small, 2.0-2.1 mm; medium, 2.4-3.2 mm; and large, 4.0-5.3 mm) were fed to four replicate groups of fish ($22.8{\pm}0.1g$), each of which was fed to apparent satiation for six weeks. In Exp-II, fish ($44.3{\pm}0.4g$) were reared at four stocking densities (1.5, 3.0, 4.5 and 6.0 $kg/m^3$) and fed a commercial diet to apparent satiation for four weeks. In Exp-III, fish ($14.8{\pm}0.1g$) were fed a commercial diet once, twice, thrice, $4{\times}$ or $5{\times}$ a day at a feeding ratio of 3.25% of body mass. Another group of fish was fed the same commercial diet $5{\times}$ a day to apparent satiation; this treatment was designated FS (five times satiation/day). In Exp-IV, fish ($31.3{\pm}0.1g$) were fed a commercial diet once, twice, thrice or $4{\times}$ a day to apparent satiation. Another group of fish was fed to apparent satiation once every 48 hours. In Exp-I, fish fed the large particle (4.0-5.3 mm) diet had a significantly higher feed conversion ratio and lower protein efficiency ratio than fish fed the small particle diet. In Exp-II, groups of fish reared at densities of 4.5 and 6.0 $kg/m^3$ had significantly higher feed intake and growth performance than fish reared at 1.5 and 3.0 $kg/m^3$. In Exp-III, fish fed to apparent satiation had significantly higher growth performances than fish fed once or $4{\times}$ a day. A significantly higher feed conversion ratio and a lower protein efficiency ratio were obtained in the FS group. In Exp-IV, growth performance and feed utilization efficiency were not significantly affected by experimental variation in feeding frequency. Fish fed to apparent satiation once every 48 hours had better feed utilization than those in other treatments and growth performances of those were comparable. Therefore, the optimum feed particle size, stocking density and feeding frequency for Korean rockfish under conditions we used were 2.0-3.2 mm, 4.5-6.0 $kg/m^3$, and once a day or once every 48 hour, respectively.

Effects of Temperature and Stocking Density on the Ammonia Excretion Rate of Red Seabream, Pagrus major

  • An experiment was conducted to investigate the effects of temperature and stocking density on daily patterns and rates of total ammonia nitrogen (TAN) excretion in juvenile red seabream Pagrus major (mean body weight: 29.0 g) under fasting and feeding conditions. Fish were acclimated over 7 days under four different temperatures (10, 15, 20, and $25^{\circ}C$) and at two different densities (5.5 and $11.0\;kg\;m^{-3}$). Each treatment had three replicates and a total of 216 fish were used. After 72 hours starvation, endogenous TAN excretion was measured for each temperature and density. To investigate exogenous TAN excretion, fish were handfed a commercial diet containing 51.6% crude protein twice a day for 7 days, at 08:00 and 16:00. Water was sampled from both inlets and outlets of chambers every 2 hours over a 24 hour period. Both endogenous and exogenous TAN excretion increased with increases in temperature and density (P<0.05). Mean daily endogenous TAN excretion rates at 10, 15, 20, and $25^{\circ}C$ were 88.8, 101.1, 125.0, and $143.3\;mg\;TAN\;kg^{-1}\;d^{-1}$ at low density, and 105.2, 119.2, 141.5, and $168.8\;mg\;TAN\;kg^{-1}\;d^{-1}$ at high density, respectively. Mean daily exogenous TAN excretion rates at 10, 15, 20, and $25^{\circ}C$ were 343.5, 403.7, 535.7, and $601.7\;mg\;TAN\;kg^{-1}\;d^{-1}$ at low density, and 391.9, 479.7, 611.9, and $683.4\;mg\;TAN\;kg^{-1}\;d^{-1}$ at high density, respectively. The exogenous TAN excretion rate peaked 10~12 hours after the first feeding under all temperatures and densities. The TAN loss for ingested nitrogen increased with increases in temperature and density (P<0.05), ranging from 27.9 to 50.1% at low density and 31.7 to 56.9% at high density. This study provides empirical data for estimating ammonia excretion and managing the culture of red seabream under the given temperatures and densities.

Toxicogenomic analysis of Effects of Bisphenol A on Japanese Medaka fish using high density-functional cDNA microarray

  • With the introduction of DNA microarrays, a high throughput analysis of gene expression is now possible as a replacement to the traditional time-consuming Southern-blot analysis. This cDNA microarray should be ahighly favored technology in the area of molecular toxicology or analysis of environmental stresses.In this study, therefore, we developed a novel cDNA microarray for analyzing stress-specific responses in japanese Medaka fish. In the design and fabrication of this stress specific functional cDNA microarray, 123 different genes in Medaka fish were selected from eighteen different stress responsive groups and spotted on a 25${\times}$75 mm glass surface. After exposure of the fish to bisphenol A which is the one of the well-known endocrine disrupting chemicals (EDCs), over 1 or 10 days, the responses of the DNA chip were found to show distinct expression patterns according to the mode of toxic actions from environmental toxicants. As a results, they showed specific gene expression pattern to bisphenol A, additionally, the chemical spesific biomarkers could be suggested based on the chip analysis data. Therefore, this chip can be used to monitor stress responses of unknown and/or known toxic chemicals using Medaka fish and may be used for the further development of biomarkers by utilizing the gene expression patterns for known contaminants.

Evaluation of feed types based on growth performance, survival, hematology, and resistance in celebes rainbow (Marosantherina ladigesi)

  • Celebes rainbow (Marosantherina ladigesi) is one of Indonesia's exported ornamental fish commodities, but the exploitation of this fish only relies on wild catches. The rise of unlimited fishing, especially those using poison, has changed the aquatic environment, threatening sustainability and causing fish extinction. This study aimed to evaluate the effectiveness of several types of feed in improving the absolute growth rate (AGR), specific growth rate (SGR), survival rate (SR), feed conversion ratio (FCR), feed efficiency (FE), hematology, and immune response of Celebes rainbow. The fish used in this study were male ornamental Celebes rainbow (M. ladigesi) weighing 1.32 ± 0.21 g/ind, reared in 54 L-aquariums at a stocking density of 30 individuals/aquarium for six weeks. The fish were fed according to the test diet consisting of live Tubifex sp worms, dry Tubifex sp worms, Spirulina platensis, and crumble pellets. The parameters observed were AGR, SGR, SR, FCR, FE, hematology, intestinal histology, liver histology, and a challenge test with the pathogenic bacteria Aeromonas hydrophila. The results showed that fish-fed live Tubifex sp worms had better AGR, SGR, SR, FCR, FE, hematology, and disease resistance compared to all other treatments. These results indicate that live Tubifex sp worms are the best feed for rearing Celebes rainbow.

댐예정지를 중심으로 한 웅천천 수계의 어류군집에 관하여 (The Fish community of the Ungchon Stream Around the New Dam Intended Area)

  • 1994년부터 1996년도까지 계류성 독립하천인 웅천천 수계의 보령댐 수몰예정지역을 중심으로 하여 6개지점에서 어류상, 주목할만한 어종 및 어류군집에 대하여 조사하였다. 본 조사지역에서 94년에서 96년 사이에 채집을 통하여 서식이 확인된 종은 11과 25속으로 총 29종이었으며 이중 9종은 한국특산어종이었다. 이 중 멸종위기종인 감돌고기와 퉁사리, 특산어종인 꺽지는 본 지역에서 확인되지 않아 절멸된 것으로 생각된다. 우세어종은 갈겨니, 피라미, 긴몰개가 우세한 그룹의 어종으로 나타났다. 지점별 평균풍부도는 0.68개체/$m^2$, 평균체적밀도는 1.45개체/㎥로 나타났으며, 종의 다양도는 상류와 하류보다는 중류가 가장 높게 나타났다. 군집간 유사도는 1군집과 4군집, 5군집과 6군집이 유사하였으나 유의한 차이는 나타내지 않아 전체가 유사한 군집으로 나타났다.

하절기 사육수온 및 밀도가 터봇 Scophthalmus maximus 미성어의 성장과 생존에 미치는 영향 (Effect of Water Temperature and Culture Density on Growth and Survival of Juvenile Turbot Scophthalmus maximus during Summer Season)

  • 유럽과 중국에서 양식 대상종으로 각광을 받고 있는 터봇 Scophthalmus maximus의 하계 국내 사육 가능성을 평가하고자 사육수온과 밀도에 대한 성장 특성을 비교하였다. 수온별 실험 결과 성장은 $20^{\circ}C$에서 가장 빨랐으며, 생존율은 $20^{\circ}C$$23^{\circ}C$ 모두 98.4%, 비만도는 $3.96{\sim}4.14$, 사료계수는 $0.84{\sim}0.89$로 차이가 없어 $23^{\circ}C$에서도 사육이 양호하였다. 수온 $26^{\circ}C$에서는 20일까지 일간성장률이 양호하였으며 50일 이후 폐사개체가 증가하여 60일에는 60.9%를 나타내었다. $29^{\circ}C$에서는 실험시작 7일 후에 전개체가 폐사되었다. 밀도별 사육실험 결과 생존율은 200마리/$m^2$ 실험구가 99.4%, 160 마리/ 실험구가 100%, 120 마리/실험구가 97.9%, 80 마리/$m^2$ 실험구가 98.4%로 각 실험구 모두 양호한 생존율을 보였다. 사육실험 결과 200 마리/$m^2$ 실험구는 다른 저밀도 실험 실험구와 비만도, 일간성장률, 사료계수의 차이가 없었고, 최종사육밀도는 $10\;kg/m^2$로 넙치보다 고밀도로 사육이 가능함이 확인되었다. 이상의 결과로부터 하절기에 $25^{\circ}C$ 이상의 수온이 거의 나타나지 않는 울진 이북 동해안 지역은 터봇을 사육하는데 적합한 수온 조건을 지니고 있으며, 수온 $26^{\circ}C$가 20일 이내인 지역이라면 터봇 종묘를 양성하는데 지장이 없을 것으로 사료되었다.
