신안산 손바닥선인장의 첨가비율 및 부위별에 따른 약주의 품질특성 (Quality Characteristics of Yakju (a Traditional Korean Beverage) after Addition of Different Tissues of Opuntia ficus indica from Shinan, Korea)

  • 손바닥선인장 약주의 pH 변화는 발효가 진행됨에 따라 감소하는 경향을 나타내었으나, 총산 함량은 증가 하는 경향을 보였다. 환원당 함량은 발효 2일에는 모든 시료구가 증가하였으나 발효 4일 경과후 감소하는 경향을 나타냈으며, ethanol은 발효 2일째부터 함량이 급속히 증가하여 발효 6일 부터는 완만한 증가를 보였고 발효 종료 시점인 14일에는 16.23~17.10%의 최대치를 나타냈다. 약주에서 확인된 유리당은 sucrose, maltose, fructose 및 glucose였고 주요 유리당은 glucose로 나타났으며 시료부위별로는 열매보다 줄기를 첨가해서 담금 한 약주에서 함량이 높았다. 유기산은 총 5종의 유기산이 확인되었고 주요 유기산은 lactic acid였으며 줄기보다 열매를 첨가해서 담금 한 약주에서 유기산 함량이 높았다. 손바닥선인장 약주의 향기성분은 총 16종의 물질이 동정되었고, 주요 향기성분으로는 iso-amyl alcohol, butyl alcohol 및 methyl ester로 나타났다. DPPH free radical scavenging activity는 열매보다 줄기를 첨가하여 담금 한 약주에서 항산화활성이 높게 나타났으며, 열매와 줄기의 첨가량이 증가할수록 항산화활성도 증가하는 경향을 나타냈다. 관능평가 결과는 열매를 첨가하여 담금 한 약주의 경우 20% 첨가구가 우수하였고, 줄기를 첨가하여 담금 한 약주의 경우는 10% 첨가구에서 높게 나타났다. 시료 부위별 기호도는 줄기보다 열매로 담금 한 약주가 우수하였다.

  • 신안산 손바닥선인장의 수분함량은 열매 71.18%, 줄기 74.85%로 나타났고, 조단백질 함량은 줄기가 3.48%로 높았으며, 조섬유 또한 줄기가 2.58%로 열매 1.54%보다 높은 함량을 보였고, 조회분, 조지방은 열매와 줄기 모두 비슷한 함량을 보였다. 신안산 손바닥선인장의 유리당은 fructose, glucose, sucrose으로 구성되어 있었으며 maltose는 소량 검출되었고, 유기산은 tartaric acid, malic acid, citric acid가 검출되었고 citric acid의 함량이 가장 높게 나타났다. 부위별 함량은 유리당, 유리산 모두 열매가 줄기보다 높게 나타났다. 신안산 손바닥선인장의 무기성분은 K의 함량이 178.61~254.71 mg%로 가장 높았고 열매가 줄기보다 높게 나타났다. 아미노산 함량은 구성아미노산 및 유리아미노산 모두 Glutamic acid의 함량이 357.10~654.82 mg%와 218.25-263.32 mg%로 가장 높았으며 열매보다 줄기에서 높은 함량을 나타내었다.

백년초 분말 첨가가 소시지의 품질 특성에 미치는 영향 (Effect of Opuntia ficus-indica var. saboten Powder Addition on Quality Characteristics of Sausage)

  • 본 연구는 백년초 분말 첨가가 아질산염의 대체를 통한 소시지의 품질 특성에 미치는 영향을 조사하기 위하여 C(아질산염 및 백년초 분말 무 첨가구), T1(0.014% 아질산염 첨가구), T2(0.014% 아질산염 + 0.5% 백년초 분말 첨가구), T3(0.014% 아질산염 + 1.0% 백년초 분말 첨가구), T4(0.5% 백년초 분말 첨가구) 및 T5(1.0% 백년초 분말 첨가구)로 처리하며 소시지를 제조하였으며 $4^{\circ}C$에서 15일간의 저장을 통하여 실험을 실시하였다. 명도를 나타내는 CIE $L^*$값은 대조구가 다른 모든 처리구들에 비해 높게 나타났으며, 적색도를 나타내는 CIE $a^*$값은 T5 처리구가 T1과 T3 처리구의 중간 값인 10.84-11.30 범위를 나타내었다(p<0.05). 황색도를 나타내는 CIE $b^*$값과 보수력 및 조직감을 나타내는 응집성, 탄력성 그리고 씹힘성은 T5 처리구에서 다른 처리구들에 비해 유의적으로 높게 조사되었다(p<0.05). 이에 반하여, 수분함량, pH 및 백색도를 나타내는 W값은 T1 처리구에서 가장 높게 나타났다. 관능적 특성에서는 T1과 T5 처리구 간 유의적인 차이가 나타나지 않았다(p>0.05). 따라서 본 실험 결과에 의하면 0.014% 아질산염의 소시지내 첨가는 1.0%의 백년초 분말로 대체시 조직감에 부정적인 영향을 주지 않으면서도 적색도를 높일 수 있는 소시지 제조가 가능할 것으로 판단되며 이를 실용화하기 위해서는 제품의 유통기한 설정에 관한 추가 연구가 필요할 것으로 사료된다.

  • 항생제 및 항균 물질 등의 오, 남용으로 인하여 항생제 내성균들이 등장, 사회적으로 큰 문제가 되고 있으며, 최근에는 식물이나 probiotics 등과 같이 항생제를 대체할 수 있는 물질에 대한 연구가 활발히 진행되고 있다. 이에 본 연구에서는 국내에서 오래 전부터 민간 요법으로 사용되어 오던 백련초 (Opuntia ficus-indica var. saboten)를 이용하여 in vitro와 in vivo에서 Salmonella enteyica serova. Enteritidis (S. enteritidis), S. enterica serova. Typhimurium (S. Typhimurium) DT104 및 Escherichia coli O157:H7에 대한 항균능을 평가하였다 백련초의 수용액이 포함된 Moulter Hinton agar에 Salmonella spp. 및 S. coli O157:H7를 접종한 결과 이들 균에 대한 억제능(inhibitory activity)을 확인할 수 있었다. 백련초 제제를 경구투여한 군에서 분변으로 배출되는 S.typhimurium DT104의 수가 급격히 감소하였고, 낀 후에는 더 이상 분변으로 균이 배출되지 않았으며, intestinal IgA가 유의적으로 증가함이 확인되었다. 본 연구를 통하여 백련초가 in vitro에서 S. enteritis, S. Typhimurium DT104와 E. coli O157:H7대한 항균력을 나타낼 뿐만 아니라 마우스의 gastrointestinal tract에서 역시 Salmonella에 대한 항균력을 보임을 확인할 수 있었다. 본 연구를 통해 백련초 및 그 성분을 이용한 병원성 세균의 제거 및 면역 증진 효과 등과 같은 연구의 필요성이 확인되었고, 그 응용 가능성도 매우 높을 것으로 사료된다.

백년초열매 추출물의 염색성 (The Dyeability of Extract from Opuntia Ficusindica var. Saboten fruits)

  • Opuntia Ficus has been an epiphyte plant since it was introduced in Cheju island as an ornamental plant two hundred years ago. And now it is grown as many as it is designated to local souvenir No.35. The fruit of this plant contains red-purple sap, which is due to be used in coloring clothes. This study aims at confirmation of use possibility as eco-friendly dyeing stuffs on extract from opuntia ficus fruits and contributing to the practical use of natural dyeing. Thus this study was investigated the dyeability of extract from opuntia ficus fruits on the silk fabric. Antioxidants were used ascorbic acid, L-cysteine, prophyl gallete. After dyeing, K/S and dye fastness were measured. The results were as follows; Maximum absorption band of extract from opuntia ficus fruits was 534.5nm. The appropriate dyeing condition was bath ratio 50:1, dyeing temperature $30^{\circ}C$, dyeing concentration 50%, dyeing time 50min, pH 3,concentration of all antioxidants 3%. Addition of antioxidants brought increase of K/S value. And K/S value and dye fastness had the greatest value when ascorbic acid was added

Comparative Analysis of the Complete Genomes of Three Ficus L. (Moraceae) Species and Its Implication

  • The genus Ficus L., containing approximately 850 species, is by far the largest genus in the Moraceae. They are mainly distributed worldwide, mainly in tropical countries. In South Korea, there are three native Ficus (including F. erecta Thunb, F. sarmentosa var. nipponica (Franch. & Sav.) Corner, and F. thunbergii Maxim.). Among them, F. erecta is effectively natural resources for the improvement of senile cognitive impairment. However, the chloroplast (cp) genome sequences and information of F. erecta have not been addressed. Therefore, in this study, we provide the complete cp genome of F. erecta and its allied species using next-generation sequencing technology. The chloroplast of Ficus species has typical structure which includes large and small single copy regions and a pair of inverted repeats (IRs). The sizes of cp genomes range from 160,276 bp to 160,603 bp. To determine the phylogenetic positions of these species, we conducted a maximum likelihood analysis using common protein-coding genes in chloroplast sequences. Also, we describe a newly developed single nucleotide polymorphism (SNP) markers using multiplex PCR to identify F. erecta based on amplification-refractory mutation system (ARMS) technique. We analyzed matK, atpB of the chloroplast genes and ITS from F. erecta and three related taxa, F. carica, F. sarmentosa var. nipponica and F. thunbergii. It provides useful information for molecular identification between F. erecta and related Korean native species.

제주도 손바닥선인장 추출물의 표면소독제로써의 항균효과 (Effect of Antimicrobiotic of Opuntia ficus-indica on Surface Disinfection)

  • Purpose: To verify the anti-microbiotic activity and to develop eco-friendly surface disinfectants using the Opuntia ficus-indica extract. Methods: The stem and fruits of the Opuntia cultivated in Jeju Island were extracted with hot water, 70% EtOH and 100% MeOH, respectively. To examine the antimicrobial activity of the extracts, gram positive bacteria (Bacillus subtilis, Staphylococcus aureus), gram negative bacteria (Escherichia coli, Salmonella typhimurium), yeast (Candida albicans), mold (Aspergillus flavus) and antibiotic resistant bacteria (E. coli, S. aureus) were measured to form a clear zone. Results: The antibacterial activity of the fruit extract was higher than that of the stem extract, and the antibacterial activity of the 100% MeOH (FM) extract and the 70% EtOH (FE) extract of the palm cactus fruit was suitable for the surface disinfectant overall, and clear-zone formed even at relatively low concentrations (250 mg/mL). The FM and FE have antibacterial activity against various bacteria, therefore they can be used as disinfectants. Conclusion: The antimicrobial activity of Opuntia ficus-indica extract from Jeju Island by stem and fruit was determined at relatively low concentrations (250 mg/mL). The Opuntia ficus-indica extract will be useful for surface disinfectants dissolving the patients' bath, oral gargling, and powdered powder in water or ethanol in appropriate concentrations.

제주 손바닥 선인장의 초음파 추출을 통한 면역활성 증진 (Enhancement of Immune Activities of Opuntia ficus-indica L. Miller by Ultrasonification Extraction Process)

  • Immune activities of Opuntia ficus indica L. Miller were investigated through aqueous extracts associated with ultrasonification process at $60^{\circ}C$, and showed the highest promotion of human B and T cell growth, up to 52% and 41%, respectively, compared to the control. The secretion of TNF-${\alpha}$ and IL-60 was also enhanced by the addition $(0.5mg/m{\ell})$ of the extracts, up to 48%. NK cell activation was significantly improved up to 1.3 times higher than the case of adding other extracts. It was also found that extracts from O. ficus indica could yield higher nitric oxide production from macrophage than Lipopolysaccaharides (LPS). It can be concluded that, in general, the extracts treated with ultrasonification has higher Immune acitivities than others, possibly by higher yielding immune-modulatory than conventional extraction process. The optimum condition for the extraction of O. ficus-indicais water extraction associated with ultrasonification.

이미지 조사를 통한 자생수종 활용에 관한 연구 -상층목을 중심으로- (A Study on the Utilization of Interior Lanscape Plant through the Investigation of Image -Focused on the Tall Trees-)

  • This study was conducted to investigte the visual preference and response between introduced and native trees for utilization of the south native trees in interior space. 1. The intensity of illumination was below 500lux at 24 buildings of interior spaces surveyed. Generally the temperature was 24~$26^{\circ}C$, and humidity was 60~70%. 2. Total number of introduced tree species were 9 familiar, 16 genus, 18 species, especially palm genus were the largest tree among them at these surveyed interior spaces, but native trees were only 2 species. The introduced tree of 2 to 3m in height was 46 percent, and the case of 1.2 to 2m was 42 percent. 3. The mean of visual preference was the highest at Rhapis excelsa and howeia belmoreana, and followed with Neolitsea sericea, Daphniphllum macropodum, Listsea japonicum. In the mean of visual preference for selected tree groups, native tree was not different as compared with introduced tree. 4. Visual image factor of the native and introduced trees at interior spaces was classified by individual factor and emotional factor. These 2 factors were shown 66.5% total variance. The native tree was strongly recognized on individual factor, as , and introduced tree was also strongly recognized on emotional factor. 5. In the analysis of visual preference and image, a difference between the selected groups of the introduced and native trees were not shown significant, and the introduced trees were similar to the native trees on the visual image. This result was as follow There was an analogy between Cinnamomum camphora, Ligustrum japonicum and Ficus retusa, Ficus benjamina. There is an analogy between Castanopsis cuspidata var. sieboldii, Cinnamomum japonicum and Ficus retusa, Ficus benjamina. There was an analogy between Elaeocarpus sylvestris var. ellipticus, Fatsia japonica and Rhapis excelsa, Howeia belmoreana. There was an analogy between Neolitsea sericea, Daphniphyllum macropo여, Listsea japonica and Rhapis excelsa, Howeia belmoreana. There was an analogy between Elaeocarpus sylvestris var. ellipticus, Fatsia japonica and Dracaena fragranse, Ficus elastica, Monstera deliciosa.

Induction of apoptosis by methanol extracts of Ficus carica L. in FaDu human hypopharynx squamous carcinoma cells

  • Ficus carica L. (fig) is one of the first cultivated crops and is as old as humans. This plant has been extensively used as a traditional medicine for treating diseases, such as cough, indigestion, nutritional anemia, and tuberculosis. However, the physiological activity of fig leaves on oral cancer is as yet unknown. In this study, we investigated the anticancer effect of methanol extracts of Ficus carica (MeFC) and the mechanism of cell death in human FaDu hypopharyngeal squamous carcinoma cells. MeFC decreased the viability of oral cancer (FaDu) cells but did not affect the viability of normal (L929) cells, as determined by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide assay and Live and Dead assay. In addition, MeFC induced apoptosis through the proteolytic cleavage of procaspase-3, -9, poly (ADP-ribose) polymerase (PARP), downregulation of Bcl-2, and upregulation of Bax, as determined by 4′,6-diamidino-2-phenylindole dihydrochloride staining and western blot analysis. Moreover, a concentration of MeFC without cytotoxicity (0.25 mg/mL) significantly suppressed colony formation, a hallmark of cancer development, and completely inhibited the colony formation at 1 mg/mL. Collectively, these results suggest that MeFC exhibits a potent anticancer effect by suppressing the growth of oral cancer cells and colony formation via caspase- and mitochondrial-dependent apoptotic pathways in FaDu human hypopharyngeal squamous carcinoma cells. Therefore, the methanol extract of Ficus carcica leaves provide a natural chemotherapeutic drug for human oral cancer.
