Antioxidant activities of solvent extracts from different Glehnia Radix parts and their inhibitory effect against nitric oxide production in Raw 264.7 cell (해방풍 부위별 용매추출물의 항산화 활성 및 nitric oxide 생성 억제)

  • In this study, we compared the antioxidant and immuno-modulatory effects of water (HR, HL, HS), 70% ethanol (ER, EL, ES), and 70% methanol (MR, ML, MS) extracts of different parts (root, leaf, and seed) from Glehnia Radix. Yields were 17.40-30.17% for water extract, 11.47-28.67% for 70% ethanol extract, and 10.73-30.57% for 70% methanol extract, respectively. The total polyphenol and flavonoid contents of EL were 10.79 g/100 g and 2.01 g/100 g, respectively. The DPPH and ABTS radical scavenging activities of EL at $1,000{\mu}g/mL$ were 84.70% and 57.64%, respectively. The superoxide radical scavenging and ferric-reducing antioxidant power of EL at $1,000{\mu}g/mL$ were 84.05% and $975.28{\mu}M$, respectively. Moreover, 70% ethanol and 70% methanol extracts of root from Glehnia Radix significantly inhibited production of NO in LPS-stimulated macrophage RAW 264.7 cells without cytotoxicity. These results suggest that 70% ethanol and 70% methanol extracts of Glehnia Radix leaf may be a useful functional food material in the food industry.

The Skin Protecting Effects of Ethanolic Extracts of Eggplant Peels (가지 외피 에탄올 추출물의 피부보호효과)

  • We investigated the in vitro antioxidant and antimelanogenesis effects of ethanol extracts from eggplant (Solanum melongena L.) peels. The total phenolics and chlorogenic acid in ethanol extracts were 2,465 mg/100 g and 2.08 mg/100 g, respectively. ABTS radical scavenging activity, ferric reducing/antioxidant power assay, and malondialdehyde (MDA) inhibitory effect of the extracts increased in a dose-dependent manner. In addition, the extracts generally showed strong UV absorption in the range of UV-B (290-320 nm). The $IC_{50}$ of mushroom tyrosinase inhibitory activity of ethanol extracts from eggplant peels was 870 ${\mu}g/mL$. Importantly, the melanin syntheses of B16/F10 melanoma cells were decreased by extracts in a concentration-dependent manner. Overall, these results suggest that eggplant peels can be potentially applied as a anti-melanogenic agent as well as an antioxidant resource.

Quality Characteristics and Antioxidant Activities of Cookies Added with Pleurotus eryngii Powder (새송이버섯 분말을 첨가한 쿠키의 품질 및 항산화활성)

  • This study examined the quality characteristics and antioxidant activities of cookies added Pleurotus eryngii powder. There were no significant differences in bulk density or water content between the doughs. Spread factors and leavening rates of cookies decreased with increasing Pleurotus eryngii powder content. However, the loss rates of cookies showed no significant differences. L values decreased with increasing Pleurotus eryngii powder content while a values were gradually increased. b values showed no significant differences among the samples. Hardness, total phenol compound content, ferric reducing antioxidant power (FRAP) and DPPH radical scavenging activity of cookies were significantly increased with increasing Pleurotus eryngii powder content. In a descriptive test, the color, flavor, palatability, hardness and graininess were all increased with increasing Pleurotus eryngii powder content. In a preference test, color and flavor were the highest in the 10% group without significant differences. Texture was preferred in the control-20% groups without significant differences. However, taste and overall acceptability were the highest in the 10% group.

  • The purpose of this study was to examine the physicochemical and antioxidant properties of cookies with mealworm (Tenebrio molitor) and black soybean powder. The leavening rates, and loss ratios were significantly increased with adding up the amount of mealworm powder (p < 0.05). The total polyphenol, DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging ability, FRAP activity (ferric reducing antioxidant power) and ABTS radical scavenging ability (2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid) were all significantly increased as mealworm powder contents increased. In addition, cookies containing both mealworm and black soybean powder were higher in total polyphenol, DPPH and ABTS radical scavenging abilities than cookies containing only mealworm powder (p < 0.05).

Antioxidant and growth inhibitory activities of Mesembryanthemum crystallinum L. in HCT116 human colon cancer cells (아이스플랜트의 항산화 및 HCT116 인체 유래 대장암세포 성장억제 활성)

  • Purpose: This study examined the antioxidant and cancer cell growth inhibitory activities of an ethanol extract and different solvent fractions of Mesembryanthemum crystallinum L. (ice plant). Methods: The ice plant was freeze-dried, extracted with 99.9% ethanol, and then fractionated with hexane, ethyl acetate, butanol, and water. The total polyphenol content (TPC), total carotenoid content (TCC), 2,2-diphenyl-1-picrylhydrazyl radical-scavenging activity (RSA), and ferric reducing antioxidant power (FRAP) were measured. Assays using 2',7'-dichlorofluorescin-diacetate and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide were performed to measure the intracellular reactive oxygen species (ROS) and cell growth, respectively. Annexin V/propidium iodide staining and cell cycle analysis were performed for the detection of apoptosis and cell cycle arrest. Results: TPC, TCC, RSA, and FRAP of the ethanol extract (EE) were 3.7 mg gallic acid equivalent/g, $13.2{\mu}g/g$, 21.0% (at a concentration of 5 mg/mL), and 21.0% (at a concentration of 5 mg/mL), respectively. Among the different solvent fractions, the butanol fraction (BF) showed the highest TPC (5.4 mg gallic acid equivalent/g), TCC ($86.6{\mu}g/g$), RSA (34.9% at 5 mg/mL), and FRAP (80.8% at 5 mg/mL). Treatment of HCT116 human colon cancer cells with EE and BF at concentrations of 250 and $500{\mu}g/mL$ reduced the levels of intracellular ROS. Concomitantly, EE and BF resulted in the dose-dependent inhibition of cell growth (at the concentrations of 125, 250, and $500{\mu}g/mL$ for 24 ~ 48 h) and the induction of apoptosis (at the concentrations of 250 and $500{\mu}g/mL$ for 48 h) in HCT116 cells. An increased G2/M cell population was also found in the BF-treated cells. Conclusion: These results suggest that ice plant possesses antioxidant and growth inhibitory activities in colon cancer cells.

Biological Activities of Various Solvent Extracts of Seomaeyakssuk (Artemisia argyi H.) (섬애약쑥 용매별 추출물의 생리활성)

  • Artemisia argyi H. has been used for centuries as a traditional medicine and food supplement in Asian countries. The objective of this study was to investigate the physiological activities of Artemisia argyi H. extracts prepared by butanol, chloroform, ethyl acetate, ethyl ether, hexane, and methanol extraction. We evaluated total phenol and flavonoid content, antioxidant activity, nitric oxide (NO) and reactive oxygen species (ROS) release, and osteoclastogenesis inhibition. The total phenolic and flavonoid contents were highest in the methanol extract (49.46 mg GAE/g and 24.32 mg QE/g, respectively). The methanol extracts also had the highest antioxidant activity (DPPH and ABTS radical scavenging ability and ferric reducing antioxidant power), while the hexane extract had the lowest. The release of NO and ROS was dose-dependently decreased by pre-treatment with all solvent extracts. At the same concentrations, the ethyl acetate and butanol extracts showed higher inhibition of NO and ROS production when compared with the other extracts. The butanol extract, at a concentration of $20{\mu}g/ml$, inhibited about 89% of the activity of the osteoclast marker, tartrate resistant acid phosphatase (TRAP). These results suggest that butanol extracts of Artemisia argyi H. may be effective natural medications for the prevention and treatment of osteoporosis.

Analysis of Antioxidant Activity and Serotonin Derivatives in Safflower (Carthamus tinctorius L.) Germplasm Collected from Five Countries (국외 수집 홍화 유전자원의 항산화 활성 및 세로토닌 유도체 함량 분석)

  • In order to select potential plant resources as functional materials and natural antioxidants, we evaluated antioxidant activity and serotonin derivatives of safflower germplasm collected from five countries. N-(p-Coumaroyl) serotonin (CS) and N-feruloylserotonin (FS) were analyzed by using Ultra Performance Liquid Chromatography (UPLC). Total polyphenol content (TPC) was determined by Folin-Ciocalteu method and antioxidant activities were estimated by 2,2-diphenyl-1-picryl-hydrazil (DPPH), 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) diammonium salt (ABTS), Ferric reducing antioxidant power (FRAP) and Reducing power (RP) assays. The TPC showed a range of 28.25 to $90.53{\mu}g$ gallic acid equivalent (GAE)/mg dried extract (DE). DPPH, ABTS, FRAP and RP assay were in the range of 18.76 to 93.98, 48.91 to 163.73, 3.80 to 132.29 and 26.32 to $80.08{\mu}g$ ascorbic acid equivalent (ASCE)/mg DE, respectively. Among the five countries, safflower seed collected from Iran had the highest levels of serotonin derivatives and antioxidant activities than other countries (p<0.05). CS showed high correlation with TPC, ABTS and DPPH (r=0.673,0.727,0.820), and FS showed high correlation with DPPH (r=0.740). Accessions IT321214 and IT321215 could be useful for development of new functional materials and could be used as a source of valuable natural antioxidant materials.

Development of High Throughput Screening Techniques Using Food-borne Library against Anti-asthma Agents (식품소재 라이브러리를 이용한 천식 완화용 물질의 초고속스크리닝 기법 개발)

  • Oxidant stress is a well-known pivotal parameter for the degenerative immune diseases including asthma, atopic dermatitis, and rhinitis. In order to screen for anti-asthma agents effectively, we first established the infrastructure of high throughput screening(HTS) for anti-oxidant agents from agricultural products and/or oriental medicine library extracted with water, methanol, dimethyl sulfoxide, ethyl acetate and juice, Using the screening system, we found that Chaenomelis langenariae, Rhus javanica L., Camellia sinensis, Helianthus annuus and Angelica utilis Makino had strong anti-oxidant activity. Moreover, Helianthus annuus, Rehmannia glutinosa Libo and Angelica utilis Makino have protection activities by treatment of an oxidant hydrogen peroxide. Together, these results suggest that screened agents could be potential agents against asthma, although the in vivo studies should be clearly tested.

Anti-Proliferative Activities of Vasicinone on Lung Carcinoma Cells Mediated via Activation of Both Mitochondria-Dependent and Independent Pathways

  • Vasicinone, a quinazoline alkaloid from Adhatoda vasica Nees. is well known for its bronchodilator activity. However its anti-proliferative activities is yet to be elucidated. Here-in we investigated the anti-proliferative effect of vasicinone and its underlying mechanism against A549 lung carcinoma cells. The A549 cells upon treatment with various doses of vasicinone (10, 30, 50, $70{\mu}M$) for 72 h showed significant decrease in cell viability. Vasicinone treatment also showed DNA fragmentation, LDH leakage, and disruption of mitochondrial potential, and lower wound healing ability in A549 cells. The Annexin V/PI staining showed disrupted plasma membrane integrity and permeability of PI in treated cells. Moreover vasicinone treatment also lead to down regulation of Bcl-2, Fas death receptor and up regulation of PARP, BAD and cytochrome c, suggesting the anti-proliferative nature of vasicinone which mediated apoptosis through both Fas death receptors as well as Bcl-2 regulated signaling. Furthermore, our preliminary studies with vasicinone treatment also showed to lower the ROS levels in A549 cells and have potential free radical scavenging (DPPH, Hydroxyl) activity and ferric reducing power in cell free systems. Thus combining all, vasicinone may be used to develop a new therapeutic agent against oxidative stress induced lung cancer.

Antioxidant activity of Green Tea Fermented with Monascus pilosus

  • Green tea leaves were fermented for 15 and 30 days with Monascus pilosus which is known to produce functional statins (TMs), and the content of various biochemical constituents such as total polyphenol (TP), total flavonoid (TF), theaflavin, and thearubigin were analyzed and compared with that of non-fermented green tea (GT) and Pu-erh Chinese post-fermented tea (PU). In addition to the electron donating ability (EDA), ferric iron reducing power (FIRP), xanthine oxidase (XO) inhibitory activity, superoxide dismutase (SOD)-like activity, iron chelating activity (ICA) and hydrogen peroxide contents were also measured and compared with that of GT and PU. Content of TP and TF in the water and ethanol extracts in TMs were lower than those in GT and PU. Theaflavin and thearubigin contents of water and ethanol extracts in TMs were higher than those of GT. And, these components were increased depending on the period of fermentation. While, EDA and FIRP of TMs were lower than those of GT, XO inhibitory activity of TMs was higher than non-fermented tea. While, ICA of TMs was slightly higher than GT and PU, the content of hydrogen peroxide in TMs was markedly lower than GT. This results suggested that the green tea fermented by M. pilosus was valuable for oxidative stress-induced diseases by decreasing hydrogen peroxide, and forming theaflavins and thearubigins with functionality of genus Monascus.
