유가식 배양에서 재조합 대장균으로부터 Interferon ${\alpha}$-1 생산에 산소 공급이 미치는 영향 (Effect of Oxygen Supply on the Production of Interferon ${\alpha}$-1 by Recombinant Escherichia coli in Fed-batch Fermentation)

  • 산소를 많이 소비하는 발효공정일수록 배양액중의 용존산소의 농도가 목적생산물의 생산성에 많은 영향을 주는 경우가 많다. 때문에 고농도 발효에 앞서, 발효조의 sparging hole로부터 임펠러 높이에 따른 산소전달 능력을 알아본 결과 공기공급이 1 vvm, 교반속도가 600 rpm에서 산소전달계수($K_La$)는 2.67($min^{-1}$)으로 가장 높았다. 배양 시 용존산소 농도를 20% 이상 유지시켰을 때 온도에 따른 k6ub/IFN-${\alpha}1$ 생성은 $30^{\circ}C$에서 세포증식을 하고 $25^{\circ}C$에서 IPTG로 Induction 하였을 때 발현율이 6.43mg/ml로 total protein의 37%로 가장 많은 양이 발현되는 것을 알 수 있었다. 용존산소 농도에 따른 k6ub/IFN-${\alpha}1$의 발현양은 용존산소 농도가 35%일 때 가장 높은 수율을 나타냈다. 용존산소량은 산소소비 속도를 측정함으로써 정확한 임계점을 찾을 수 있었는데 용존산소량이 35% 유지될 때 산소 전달 속도와 비교하여 가장 적당한 산소공급량임을 확인할 수 있었다.

동결농축 Muscat Bailey A 포도 과즙으로 제조한 무가당 적포도주의 품질 특성 (Properties of Red Wine Fermented Using Freeze-Concentrated Muscat Bailey A Grape Juice)

  • 국산 포도주의 품질을 향상시키고 개선하기 위한 발효방법의 일환으로 국내산 Muscat Baily A 포도 과즙을 약 $24^{\circ}Brix$가 되게 동결농축한 후 포도주 발효를 수행하였다. 효모로는 산업용 포도주 효모인 S. cerevisiae $OC_2$, S. cerevisiae Fermivin, S. cerevisiae W-3를 이용하였으며 이들에 의한 농축 과즙의 알코올 발효를 행하면서 발효 특성과 발효 후 여과하여 제성한 포도주의 품질 특성을 조사하였다. 발효가 끝날 시점의 알코올 함량은 12.6-13.0%로 비슷하였으며 환원당의 함량은 거의 흔적 정도로 검출되었다. pH는 약 3.4-3.7, 총산 함량은 0.62-0.71%로 모든 시험구에서 정상적으로 발효가 일어났음을 알 수 있었다. 발효 종료 후 여과한 포도주의 아세트알데히드, 메탄올, fusel oil의 함량은 식품공전에 명시된 과실주의 기준치보다 매우 낮게 나왔으며 총 폴리페놀 함량은 1.42-1.56 mg/mL 정도를 나타내었다. 각 시험구의 포도주는 모두 농축으로 인하여 진한 색을 띄었고 특히 적색을 나타내는 520 nm에서의 흡광도가 높았으며 이는 색도인 L, a, b 값을 통해서도 확인하였다. 관능검사를 실시한 결과는 색, 향, 전반적인 기호도에서 S. cerevisiae OC2$OC_2$ 포도주가 가장 우수한 것으로 나타났으며 향과 맛은S. cerevisiae Fermivin 포도주가 가장 우수한 것으로 나타났다.

양고추냉이와 겨자 분말을 첨가한 고추장의 발효특성 (Fermentation Characteristics of Kochujang Containing Horseradish or Mustard)

  • 우수균주를 접종한 고추장에 항균성 물질을 함유하고 있는 양고추냉이와 겨자 분말을 첨가하여 $25^{\circ}C$에서 발효시키면서 미생물과 성분의 변화를 비교하였다. 향신료 첨가로 인해 고추장 효모의 증식이 억제됨으로서 유통중 문제가 되는 가스가 발생되지 않았고 세균수에는 큰 차이가 없었다. 아미노산성 질소는 발효가 진행되면서 계속하여 증가하였으며 발효 120일째 P-2 코지와 겨자 분말을 함께 첨가한 실험구의 함량이 유의적으로 높았다. 효소 역가의 경우 ${\alpha}-amylase$활성은 매우 낮았으나 ${\beta}-amylase$는 향신료 첨가구의 활성이 높았고, protease(산성, 중성)는 발효가 진행되면서 계속 증가하였으나 실험구간의 뚜렷한 차이는 보이지 않았다. 고추장의 색과 향기는 실험구간의 유의적 차이를 보이지 않았고 맛과 전체적 기호도의 경우 양고추냉이 첨가구와 P-2 코지만을 첨가한 실험구가 우수하였다. 양고추냉이 및 겨자를 첨가하고 선발된 균주를 이용하여 발효시킨 고추장은 저장 중 가스발생이 완전히 억제되었고 품질 또한 우수하였다.

Influence of Mentha×piperita L. (Peppermint) Supplementation on Nutrient Digestibility and Energy Metabolism in Lactating Dairy Cows

  • The characteristic smell of cow milk was suppressed when herbs were consumed by lactating dairy cows. But it is unclear whether or not peppermint ingestion affects the nutritional and milk production parameters in lactating dairy cows. The objective of this study was to examine the effect of peppermint feeding to lactating dairy cows on nutrient digestibility, energy metabolism, ruminal fermentation and milk production. Eight Holstein cows were given a diet supplemented with or without 5% of dried peppermint per diet on a dry matter basis. The digestion of nutrients from cows fed the diet with peppermint was significantly lower than that of the control group. Energy loss as methane and methane released from cows receiving the peppermint treatment was significantly lower than that in the control cows. Peppermint feeding to cows resulted in the promotion of thermogenesis. However, ruminal fermentation and milk production were not affected by peppermint feeding. In conclusion, peppermint ingestion by lactating dairy cows reduces the nutrient digestibility and methanogenesis, and changes energy metabolism.

Effects of Feeding System on Rumen Fermentation Parameters and Nutrient Digestibility in Holstein Steers

  • In order to compare effects of feeding systems on rumen fermentation characteristics and nutrient digestion, steers were fed either total mixed ration (TMR) or separate concentrate-roughage ration (CR). Total tract digestibility of nutrients was higher in steers receiving TMR. Especially, DM, ADF and NDF in TMR were digested to a greater extent than those in CR. Rumen pH was not influenced by the feeding systems. Holstein steers on TMR had higher ruminal $NH_3$-N than those on CR. Feeding system did not alter VFA production but TMR feeding resulted in lower A/P ratio. TMR feeding tended to increase the number of bacteria and protozoa in the rumen fluid. Also steers fed TMR generally had higher fiber degrading enzyme activities, which might be the result of increased number of cellulolytic microbes in the rumen of animals on TMR. Our results indicate that TMR may provide more favorable condition for nutrient digestion both in the rumen and in the total tract of steers.

Effects of Daily and Interval Feeding of Sapindus rarak Saponins on Protozoa, Rumen Fermentation Parameters and Digestibility in Sheep

  • Several researchers have demonstrated that the rumen microbial community rapidly adapts to saponins and proposed interval feeding to prevent this rapid adaptation. An in vivo experiment was carried out to examine the effect of daily versus application every third day (interval feeding) of Sapindus rarak saponins (SE) on rumen fermentation end products, protozoal counts and nutrient digestibility. Thirty sheep were allocated into 5 groups. Sheep were fed daily or every third day with two levels of SE (0.48 and 0.72 g/kg body mass). One group received no saponin and served as control. All sheep received the same diet, a mixture of elephant grass and wheat pollard (65:35 w/w). Independent of the feeding regime and the level of inclusion, the addition of SE decreased protozoal counts and rumen ammonia concentrations (p<0.01). Microbial N supply and N retention were not affected by the high feeding regime. Daily feeding negatively influenced rumen xylanase and cellulase activity, but only when the high level of saponins was fed. However, these negative effects on rumen cell wall degradation were not reflected in decreasing total tract digestibility of the organic matter or the plant cell walls. Our results show that rumen microorganisms do not rapidly adapt to S. rarak saponins.

Effect of Different Levels of Rumensin in Diet on Rumen Fermentation, Nutrient Digestibility and Methane Production in Cattle

  • Twelve rumen fistulated cross-bred calves were divided into three groups and fed wheat straw and concentrate mixture according to their maintenance requirement. Animals in group II and III were fed 50 and 100mg rumensin per day, in addition to basal feed. Supplementation of rumensin in the diet decreased the dry matter intake significantly (p<0.05) along with a significant decrease in the straw intake. Digestibility coefficients of all the nutrients were not affected significantly except that of CF digestibility which was lower (p<0.05) in groups II and III as compared to group I. Among N-parameters in the rumen fluid, mean $NH_3-N$ was significantly lower in groups II and III (19.13 and 18.63 mg N/100 ml respectively) than in group I (22.68); total-N and TCA-ppt-N did not differ among the three groups. Total VFA concentration did also not differ among the three groups, however, propionate increased from 24.33 molar % to 32.73 while acetate and butyrate decreased respectively from 65.85 to 58.81% and 9.79 to 8.46%. Total VFA, bacteria and protozoa production rates were not affected significantly due to rumensin in diet. Methane production per kg DDM as well as % of methane in total gas were reduced at both the levels of rumensin on different concentrate ratios with wheat straw as roughage. Similar trend was also observed with rice straw and concentrate mixture as substrate with rumensin addition.

Optimization and Evaluation of Organic Acid Recovery from Kraft Black Liquor Using Liquid-Liquid Extraction

  • Liquid-liquid extraction (LLE) can be used for the recovery of acetic acid from black liquor prior to bioethanol fermentation. Recovery of value-added chemicals such as acetic-, formic- and lactic acid using LLE from Kraft black liquor was studied. Acetic acid and formic acid have been reported to be strong inhibitors in fermentation. The study elucidates the effect of three reaction parameters: pH (0.5~3.5), temperature ($25{\sim}65^{\circ}C$), and reaction time (24~48 min). Extraction performance using tri-n-octylphosphine oxide as the extractant was evaluated. The maximum acetic acid concentration achieved from hydrolyzates was 69.87% at $25^{\circ}C$, pH= 0.5, and 36 min. Factorial design was used to study the effects of pH, temperature, and reaction time on the maximum inhibitor extraction yield after LLE. The maximum potential extraction yield of acetic acid was 70.4% at $25.8^{\circ}C$, pH=0.6 and 37.2 min residence time.

Zymomonas mobilis에 의한 알코올 발효 I. 발효 환경이 생육에 미치는 영향 (Alcohol Fermentation by Zymomonas mobilis Part. 1. Effects of environmental conditions on the growth kinetics of Zymomonas mobilis)

  • The effect of various environmental conditions on the growth kinetics of Zymomonas mobilis were studied and the kinetic parameters were evaluated. The value of ${\mu}m$ was $0.45hr^{-1}$ and Ks was 0.23 g/L. Inhibition of growth at high glucose concentration was found to follow the threshold substrate inhibition. Threshold substrate concentration was 102 g/L and substrate inhibition constant was 196 g/L. The effects of yeast extract concentrations were found to follow the Monod equation. ${\mu}m$ value was $0.45hr^{-1}$ and Ks was 0.3 g/L at 20 g/L of glucose and $0.24hr^{-1}$ and 0.24 g/L respectively at 200 g/L of glucose. The optimum temperature was found to be $35^{\circ}C$ and the activation energy of growth was 7.7 Kcal/mole below $35^{\circ}C$ and -29 Kcal/mole above $35^{\circ}C$.

녹색 홀그레인 보리, 밀, 찹쌀, 멥쌀 분말을 첨가한 발효유의 특성 (Characterization of fermented milk added with green whole grains of barley, wheat, glutinous rice and common rice powders)

  • Yogurt was prepared from skim milk added with 2, 4 or 6% of green whole grain of barley, wheat, glutinous rice and common rice, respectively. Changes in pH, titratable acidity, viable cell counts and viscosity during fermentation were monitored and its sensory evaluation was also performed. The optimum level of additives such as green whole grain of barley, wheat, glutinous rice and common rice for yogurt manufacture was selected to 2%. The samples added with green whole grain of barley and wheat powders reached pH 4.5 in 12 hours. After 4 hours of fermentation, pH, the viable cell counts and viscosity in samples added with 2% green whole grain of barley, wheat, glutinous rice and common rice powders were the highest. Especially, in overall sensory evaluation using 5 parameters, the sensory scores of glutinous rice and common rice 2% yogurts were significantly high. From this experiment, the additives optimum level of glutinous rice and common rice powders was selected to be 2%.
