Prevalence of Enterobius vermicularis Infection among Preschool Children in Kindergartens of Taipei City, Taiwan in 2008

  • The prevalence of Enterobius vermicularis infection among preschool children was reported to be low based on a 5-year screening program in Taipei City, Taiwan. The Taipei City government intended to terminate the E. vermicularis screening program among preschool children. Thus, we were entrusted with confirming whether pinworm infections among preschool children in Taipei City had truly declined. From each of 12 administrative districts 2-3 kindergartens were randomly selected for investigation. In total, 4,349 children were examined, of which 2,537 were boys and 1,812 were girls. The cellophane tape adhered to a glass slide was used, and all examinations were done by certified medical technologists. Results indicated that the overall prevalence rate of pinworm infections was 0.62% (27/4,349). Although the infection rate was higher among boys (0.67%, 17/2,537) than in girls (0.55%, 10/1,812), no significant difference was found ($X^2$=0.399, P=0.62). According to the administrative district, the infection rate ranged from no positive cases of E. vermicularis infection in the Xinyi, Zhongzhen, and Wanhua Districts (0%; 0/299, 0/165, and 0/358, respectively), to 0.26% (1/131) in Songshan District, with the highest rate of 1.88% (7/373) in Wenshan District. Because the overall infection rate (0.62%, 27/4,349) in the present study was unchanged compared to that (0.40%, 197/49,541) previously reported in 2005, we propose that regular pinworm screening and treatment programs should be continued in some parts of Taipei City.

Mini-Array of Multiple Tumor-associated Antigens (TAAs) in the Immunodiagnosis of Esophageal Cancer

  • Sera of cancer patients may contain antibodies that react with a unique group of autologous cellular antigens called tumor-associated antigens (TAAs). The present study aimed to determine whether a mini-array of multiple TAAs would enhance antibody detection and be a useful approach in esophageal cancer detection and diagnosis. Our mini-array of multiple TAAs consisted of eleven antigens, p53, pl6, Impl, CyclinB1, C-myc, RalA, p62, Survivin, Koc, CyclinD1 and CyclinE full-length recombinant proteins. Enzyme-linked immunosorbent assays (ELISA) were used to detect autoantibodies against eleven selected TAAs in 174 sera from patients with esophageal cancer, as well as 242 sera from normal individuals. In addition, positive results of ELISA were confirmed by Western blotting. In a parallel screening trial, with the successive addition of antigen to a final total of eleven TAAs, there was a stepwise increase in positive antibody reactions. The eleven TAAs were the best parallel combination, and the sensitivity and specificity in diagnosing esophageal cancer was 75.3% and 81.0%, respectively. The positive and negative predictive values were 74.0% and 82.0%, respectively, indicating that the parallel assay of eleven TAAs raised the diagnostic precision significantly. In addition, the levels of antibodies to seven antigens, comprising p53, Impl, C-myc, RalA, p62, Survivin, and CyclinD1, were significantly different in various stages of esophageal cancer, which showed that autoantibodies may be involved in the pathogenesis and progression of esophageal cancer. All in all, this study further supports our previous hypothesis that a combination of antibodies might acquire higher sensitivity for the diagnosis of certain types of cancer. A customized mini-array of multiple carefully-selected TAAs is able to enhance autoantibody detection in the immunodiagnosis of esophageal cancer and autoantibodies to TAAs might be reference indicators of clinical stage.

Cutting Efficiency Using Phragmites australis Culms According to Content and Timing of Indole-acetic Acid Treatment (옥신 처리 농도 및 시기에 따른 갈대 지상경 삽목 효율)

  • In this study, we tried to overcome a limit in cutting timing of reed culm by applying a plant hormone, indole-acetic acid (IAA) as a growth regulator with various contents ($10^{-3}$ M, $10^{-6}$ M, $10^{-9}$ M, $10^{-12}$ M). 19 shoots emerged from 240 segments of hardened reed culm from montane fen and eight out of the 19 shoots emerged by $10^{-6}$ M IAA treatment as the most in $5^{th}$ Sep. 2012. 50 shoots emerged from 60 segments of non-hardened reed culm from a population in Seoul National University transplanted from Mt. Odae by $10^{-6}$ M IAA treatment despite the cutting was performed about two weeks later ($19^{th}$ Sep.). Via third cutting experiment performed about 40 days later ($29^{th}$ Oct.), only two shoots out of 60 segments were observed by the same experimental condition except atmospheric temperature. It seemed likely that it was too low temperature in third experiment ($10^{\circ}C$) than the former experiments (about $20^{\circ}C$) to form adventitious buds from culm segment. We recommend to utilize the thick reed culm in culm cutting as possible because the thicker culm segment we used, the thicker emerged shoot we could observe (i.e., diameters of emerged shoots were about 20% of the planted segment's diameters).

Effect of Seeding Rate on Forage Quality Components and Productivity of Alfalfa in Alpine Area of Korea

  • The optimal determination of seeding rate is critical to minimizing uncertainties about the large variations observed in forage quality and productivity when alfalfa is cultivated under different geographical areas and growing conditions. The objective of this investigation was to provide information about the proper seeding rate according to harvest timing for alfalfa cultivation in the Northern regions of Korea. Alfalfa was sown in September 2018 at a seeding rate of 20, 30 or 40 kg/ha and harvested four times in 2019: May 3, July 2, September 11, and October 13. Regardless of seeding rate, alfalfa plant height was longest at the third harvest (113 cm) and the shortest in the last annual harvest (43.8 cm). However, seeding rate had no effect on alfalfa plant height at any harvest. Forage relative feed value was increased in the first cutting but decreased in the third cuttings as seeding rate increased. However, seeding rate had slight effect on alfalfa forage quality components at the second and fourth cuttings. Total annual DM and crude protein production (in 4 harvests) was greater at higher seeding rates. Plots seeded at a rate of 40 kg/ha produced on average 1,257 and 2,620 kg/ha more forage (DM basis) than those seeded at a rate of 30 or 20 kg/ha, respectively. Forage DM production at the first, second, third, and fourth harvests accounted for 36.1, 24.0, 27.1, and 12.8 % of total annual DM production, respectively. Overall, small differences were seen when alfalfa seeding rate was different but maximum forage DM production (in four harvests) was detected when seeding rate was 40 kg/ha. These data could be useful to the alfalfa growers by allowing them to make more accurate trade-offs between seed price and the expected magnitude of forage yield gains in order to select the best seeding rate.

Clinical Effect of Traditional Chinese Medicine Shenhuang Granule in Critically Ill Patients with COVID-19: A Single-Centered, Retrospective, Observational Study

  • The coronavirus disease 2019 (COVID-19) pandemic has become a public health emergency of global concern. In China, traditional Chinese medicine has been widely administered to COVID-19 patients without sufficient evidence. To evaluate the efficacy of Shenhuang Granule (SHG) for treating critically ill patients with COVID-19, we included in this study 118 patients who were admitted to the ICU of Tongji Hospital between January 28, 2020 and March 28, 2020. Among these patients, 33 (27.9%) received standard care plus SHG (treatment group) and 85 (72.1%) received standard care alone (control group). Enrolled patients had a median (IQR) age of 68 (57-75) years, and most (79 [67.1%]) were men. At end point of this study, 83 (70.3%) had died in ICU, 29 (24.5%) had been discharged from ICU, and 6 patients (5.2%) were still in ICU. Compared with control group, mortality was significantly lower in treatment group (45.4% vs. 80%, p < .001). Patients in treatment group were less likely to develop acute respiratory distress syndrome (ARDS) (12 [36.3%] vs. 54 [63.5%], p = 0.012) and cardiac injury (5 [15.1%] vs. 32 [37.6%], p = 0.026), and less likely to receive mechanical ventilation (22 [66.7%] vs. 72 [84.7%], p = 0.028) than those in control group. The median time from ICU admission to discharge was shorter in treatment group (32 [20-73] days vs. 76 [63-79] days, p = 0.0074). These findings suggest that SHG treatment as a complementary therapy might be effective for critically ill adults with COVID-19 and warrant further clinical trials.

Association analysis of polymorphisms of G protein-coupled receptor 54 gene exons with reproductive traits in Jiaxing Black sows

  • Objective: The aim of this study was to detect single nucleotide polymorphisms (SNP) of G protein-coupled receptor 54 (GPR54) gene and explore association of this candidate gene with reproductive traits in Jiaxing Black sows. Methods: Six pairs of primers of the gene were designed to amplify all exons thus sequences of which were detected by means of direct sequencing and then SNP loci were scanned. The effects of SNPs on total number of piglets born (TNB), number of piglets born alive (NBA), number of still born piglets (NSB), and litter weight at birth (LWB) of Jiaxing Black sows were analyzed. Results: Three SNP loci, including T3739C, C3878T and T6789C, were identified via comparison of sequencing and two genotypes (AB, BB) at each SNP site were observed. T3739C resulted in the change of amino acid ($Leu{\rightarrow}Pro$) in corresponding protein, and C3878T resulted in synonymous mutation ($Ile{\rightarrow}Ile$). Statistical results demonstrated that allele B was the preponderant allele at the three SNP loci and Genotype BB was the preponderant genotype. Meanwhile, Chi-Square test of these three SNPs indicated that all mutation sites fitted in Hardy-Weinberg equilibrium (p>0.05). For GPR54-T3739C locus, Jiaxing Black sows with genotype BB had 1.23 TNB and 1.28 NBA (p<0.01) that were more than those with genotype AB, respectively. Jiaxing Black sows that had the first two parities with genotype BB had additional 2.23 TNB, 2.27 NBA (p<0.01), and 1.94 LWB (p<0.05) compared to those with genotype AB, respectively. However, for other two loci, no significant difference was found between TNB, NBA, NSB, and LWB, and different genotypes of Jiaxing Black sows. Conclusion: In conclusion, the polymorphisms of GPR54-T3739C locus were significantly associated to TNB, NBA, and LWB and could be used as a potential genetic marker to improve reproductive function of Jiaxing black sows.

Preoperative Long Course Chemoirradiation in a Developing Country for Rectal Carcinoma: Kuala Lumpur Hospital Experience

  • Background: The use of preoperative chemoirradiation is the commonest treatment strategy employed in Malaysia for locally advanced rectal cancer. We need to determine the local control and survival rates for comparison with established rates in the literature. Materials and Methods: This retrospective study analyzed all newly diagnosed patients with rectal adenocarcinoma who underwent long course preoperative radiotherapy (RT) at the Department of Radiotherapy and Oncology, Kuala Lumpur Hospital (HKL) between $1^{st}$ January 2004 and $31^{st}$ December 2010. The aim of the study was to determine the radiological response post radiotherapy, pathological response including circumferential resection margin (CRM) status, 3 years local control, 3 years overall survival (OS) and 3 years disease free survival (DFS). Statistical analysis was performed using the SPSS software. Kaplan-Meier and log rank analysis were used to determine survival outcomes. Results: A total of 507 patients with rectal cancer underwent RT at HKL. Sixty seven who underwent long course preoperative RT were eligible for this study. The median age at diagnosis was 60 years old with a range of 26-78 years. The median tumour location was 6 cm from the anal verge. Most patients had suspicion of mesorectum involvement (95.5%) while 28.4% of patients had enlarged pelvic nodes on staging CT scan. All patients underwent preoperative chemo-irradiation except for five who had preoperative RT alone. Only 38 patients underwent definitive surgery (56.7%). Five patients were deemed to be inoperable radiologically and 3 patients were found to have unresectable disease intraoperatively. The remaining 21 patients defaulted surgery (31.3%). The median time from completion of RT to surgery was 8 weeks (range 5.6 to 29.4 weeks). Fifteen patients (39.5%) had surgery more than 8 weeks after completion of RT. Complete pathological response was noted in 4 patients (10.5%). The pathological CRM positive rate after RT was 18.4%. With a median follow-up of 38.8 months, the 3 year local control rate was 67%. The 3 years rate for CRM positive (<2 mm), CRM clear (>2 mm) and pCR groups were 0%, 88.1% and 100% respectively (p-value of 0.007). The 3 year OS and DFS were 57.3% and 44.8% respectively. Conclusions: In conclusion, the approach of long course preoperative chemoirradiation for rectal cancer needs to be re-examined in our local setting. The high rate of local recurrence is worrying and is mainly due to patient defaulting post-preoperative chemoirradiation or delayed definitive surgery.

Anticancer Activity of Acanthopanax trifoliatus (L) Merr Extracts is Associated with Inhibition of NF-κB Activity and Decreased Erk1/2 and Akt Phosphorylation

  • Acanthopanax trifoliatus (L) Merr (AT) is commonly used as an herbal medicine and edible plant in some areas of China and other Asian countries. AT is thought to have anticancer effects, but potential mechanisms remain unknown. To assess the anticancer properties of AT, we exposed prostate cancer cells to AT extracts and assessed cell proliferation and signaling pathways. An ethanol extract of AT was suspended in water followed by sequential extraction with petroleum ether, ethyl acetate and n-butanol. PC-3 cells were treated with different concentrations of each extract and cell viability was determined by the MTT and trypan blue exclusion assays. The ethyl acetate extract of the ethanol extract had a stronger inhibitory effect on growth and a stronger stimulatory effect on apoptosis than any of the other extracts. Mechanistic studies demonstrated that the ethyl acetate extract suppressed the transcriptional activity of NF-${\kappa}B$, increased the level of caspase-3, and decreased the levels of phospho-Erk1/2 and phospho-Akt. This is the first report on the anticancer activity of AT in cultured human prostate cancer cells. The results suggest that AT can provide a plant-based medicine for the treatment or prevention of prostate cancer.

Anti-proliferative Effects of Atractylis lancea (Thunb.) DC. via Down-regulation of the c-myc/hTERT/Telomerase Pathway in Hep-G2 Cells

  • Atractylis lancea (Thunb.) DC. (AL), an important medicinal herb in Asia, has been shown to have anti-tumor effects on cancer cells, but the involved mechanisms are poorly understood. This study focused on potential effects and molecular mechanisms of AL on the proliferation of the Hep-G2 liver cancer cell line in vitro. Cell viability was assessed by MTT test in Hep-G2 cells incubated with an ethanol extract of AL. Then, the effects of AL on apoptosis and cell cycle progression were determined by flow cytometry. Telomeric repeat amplification protocol (TRAP) assays was performed to investigate telomerase activity. The mRNA and protein expression of human telomerase reverse transcriptase (hTERT) and c-myc were determined by real-time RT-PCR and Western blotting. Our results show that AL effectively inhibits proliferation in Hep-G2 cells in a concentrationand time-dependent manner. When Hep-G2 cells were treated with AL after 48h,the $IC_{50}$ was about 72.1 ${\mu}g/mL$. Apoptosis was induced by AL via arresting the cells in the G1 phase. Furthermore, AL effectively reduced telomerase activity through inhibition of mRNA and protein expression of hTERT and c-myc. Hence, these data demonstrate that AL exerts anti-proliferative effects in Hep-G2 cells via down-regulation of the c-myc/hTERT/telomerase pathway.

Satzmodelle im Koreanischen und Deutschen im Hinblick auf ihre didaktische Verwertbarkeit ($\cdot$독 문형의 비료설정 시론)

  • In der vorliegenden Arbeit werden die deutschen Satzmodelle $f\"{u}r$ Zwecke des DaF-Unterrichts in Korea untersucht. Dabei gehen wir davon aus, dass man die deutschen Satzmodelle vergleichend mit den koreanischen neu aufbauen sollte, um sie im Deutschunterricht fur Koraner recht benutzen zu konnen. Dafur haben wir hier zunachst untersucht, wie man die koreanischen Satzmodelle im Hinblick auf den didaktischen Zweck modifizieren muss. Unter $Ber\"{u}cksichtigung$ des dabei erzielten Ergebnisses wurden $anschlie{\ss}end$end die deutschen Satzmodelle rekonstruiert. Im Koreanischen wurden die Satzmodelle ab Ende der 1960er Jahre verschiedenartig untersucht. Zwar gibt es bisher noch keine Arbeit, die Ergebnisse dieser Forschungen zusammenfasst, dennoch versuchen wir hier aus einigen bisherigen Untersuchungen einen begrenzten Rahmen von koreanischen Satzmodellen herauszufinden. Als Resultat haben wir funf grundlegende Satzmolle hergestellt: (1) Subjekt + $Pr\"{a}dikat$, (2) Subjekt + $Pr\"{a}dikativ$ + $Pr\"{a}dikat$, (3) Subjekt + Objektiv + $Pr\"{a}dikat$, (4) Subjekt + Adverbiale + $Pr\"{a}dikat$, (5) Subjekt + Obj. + Obj./$Pr\"{a}dikativ$/Advb. + $Pr\"{a}dikat$. Das Modell (1) $umfa{\ss}t$ drei $Pr\"{a}dikate$, die sich nach den Wortarten unterscheiden: (intransitives) Verb, Adjektiv und Substantiv. Auch beim Fall (5) $k\"{o}nnen$ drei verschiedene Untermodelle nach der $Kombinationsm\"{o}glichkeit$ auftreten: Sub + Obj. + $Pr\"{a}d$., Sub. + Obj. + $Pr\"{a}dikativ$ + $Pr\"{a}d$., Sub. + Obj. + Advb. + $Pr\"{a}d$. Auch im Deutschen kann man ahnlich wie im Koreanischen didaktisch geeignete Satzmodelle herausarbeiten. Auch dabei wurden bisherige Untersuchungen zu den deutschen Satzmodellen herangewgen. Zu der $d\"{a}fur$ $ausgew\"{a}hlten$ Terminologie $geh\"{o}ren$ Subjekt(S), $Pr\"{a}dikat(P)$, $Pr\"{a}dkativerganzung(PE)$, $Objekterg\"{a}nzung(OE)$, $Adverbialerg\"{a}nzung(AE)$. In der deutschen Sprache werden nach den $Valenzm\"{o}glich­keiten$ des $Pr\"{a}dikativteils$ folgende Modelle unterschieden: S + P, S + P + PE, S + P + OE, S + P + AE, S + P + OE + OE/PE/AE. Unter die OE werden okk. OE, dat. OE, $pr\"{a}d$. OE, refl. OE usw. subsumiert. Um die Stichhaltigkeit der in dieser Schrift neu festgestellten Satzmodelle zu $\"{u}berpr\"{u}fen$, sollten sie anhand eines Textkorpus heutiger Texte in weiteren Untersuchungen validiert werden. Ferner ist es zu empfehlen, die Anwendbarkeit der hier vorgestellten Satzrnodelle im Deutschunterricht $f\"{u}r$ Koreaner $gr\"{u}ndlich$ zu testen
