  • Two experiments were conducted to investigate the dietary effects of conventional or lutein fortified chlorella on lutein absorptions, the tissue distributions and the changes in lutein content of eggs in laying hens. In Exp 1, a total of one hundred and fifty, 70 wk-old Hy-Line brown layers were divided into three groups with five replicates and fed with each experiment diet (control diet, diet with 1% conventional chlorella or lutein fortified chlorella) for 2 wk, respectively. The egg production in groups fed diets containing both chlorella powders were higher than that of the control group (p<0.01). With chlorella supplementations, the yolk color significantly increased, although there were no significant differences in the eggshell qualities. The lutein contents of serum, liver and growing oocytes were greatly increased by feeding conventional or lutein fortified chlorella (p<0.01). In Exp. 2, a total of ninety 60 wk-old Hy-Line brown layers were assigned into three groups with three replicates per group (10 birds per replicate). The birds were fed with one of three experimental diets (0, 0.1 or 0.2% lutein fortified chlorella) for 2 wk, respectively. The egg production was not affected by dietary treatments. The egg weight in the group fed with diet containing 0.2% of lutein fortified chlorella was higher than that of the control (p<0.05). As the dietary chlorella levels increased, the daily egg mass linearly increased, although not significantly. The yolk colors in groups fed diets containing lutein fortified chlorella were dramatically increased as compared to the control (p<0.001). The lutein in chicken eggs significantly increased when fed with 0.2% of lutein fortified chlorella (p<0.01). These results suggested that the dietary lutein derived from chlorella was readily absorbed into the serum and absorbed by the liver with growing oocyte for commercial laying hens. Particularly, the lutein fortified chlorella was a valuable natural source for the production of lutein enriched chicken eggs.

  • This study was conducted in order to investigate the growth and morphological development of Cypselurus hiraii larvae and to obtain basic data for taxonomic research. Specimens for this study were collected using a scoop net (aperture Ø60 cm) at the Geomun Island in Yeosu-si, Jollanam-do in July and August of 2013, and the mean water temperature was $20.6{\pm}1.84^{\circ}C$ and the mean salinity was $32.1{\pm}0.46$‰ during the collection. In post-larvae of total length 9.58~9.69 mm (average $9.63{\pm}0.04mm$, n=5), the mouth was opened, and the maxilla was shorter than the mandible. The length of the dorsal fin did not exceed that of the ventral fin, and the dorsal fin was on the same line as that of the ventral fin. In post-larvae of total length 18.0~18.5 mm (average $18.2{\pm}0.22mm$, n=5), with the growth accelerated, the pectoral fin was developed broadly and their caudal fin was differentiated into the heterocercal tail. These characteristics suggest the development of swimming ability and resultant active feeding activities. In post-larvae of total length 22.4~23.0 mm (average $22.7{\pm}0.21mm$, n=5), striped melanophores deposited in the pectoral fin, and small spot-like melanophores expanded in the processes formed on the mandible. The dorsal and anal fins were on the same line, and the anal fin was situated behind the dorsal fin.

  • This study aimed to investigate the effects of reducing feed intake on performance and meat quality in old laying hens. A total of 200 Hy-Line Brown laying hens (100 weeks old) were randomly allotted to five dietary treatments: control (100% daily feed intake), 90%, 60%, 50%, and 20% daily feed intake. Each treatment was replicated four times with 10 birds per replication and two birds per cage. Ten-bird units were arranged according to a randomized block design. The feeding trial lasted for 4 weeks under a 16L:8D lighting regimen. The results indicated that the daily feed intake correlated with hen-day egg production and feed conversion ratios (P<0.05). The carcass yields and partial ratios were also correlated with daily feed intake (P<0.05). The levels of leukocytes (without basophils) were higher in the 50% and 20% daily feed intake groups than in the other groups. The concentrations of dry matter, crude ash, crude fat, and crude protein, water holding capacity, cooking loss, and fatty acids in the breast meat did not decrease as the daily feed intake decreased. In conclusion, reducing daily feed intake decreased laying performance and carcass yield but had no effect on breast meat quality.

  • This study was conducted to evaluate dietary effects of Marigold extract on laying performance, egg quality, oxidative stability of egg yolk and lutein transfer into chicken eggs. A total of one-hundred eighty nine 55-wk-old Hy-Line Brown layers were divided into seven groups and fed control diet or each experimental diet containing 0.1, 0.3, 0.5, 1.0, 1.5 or 2.0% Marigold extract. Egg production, egg weight and daily egg mass were not affected by dietary treatments. The yolk colors in groups fed diets containing Marigold extract were significantly higher than that of control. The Haugh unit were tended to be improved by feeding of diets containing Marigold extract although there were no significant difference in egg shell strength and thickness. The MDA (malondialdehyde) contents in groups fed diets containing Marigold extract above 0.5% were significantly reduced than that of control. After 14d of storage, the Haugh unit values in groups feed diets containing 0.3 and 1.0% Marigold extract were significantly higher than that of control (p<0.05). The concentration of lutein in egg yolk increased by feeding of Marigold extract. When 2% Marigold extract was supplemented to the diet, lutein content of egg was increased as much as 1.71 mg/60 g. These results indicated that the use of Marigold extract in layer diets was effective in egg quality and for the production of lutein fortified eggs.

  • CpBV (Cotesia plutellae bracovirus) is a polydnavirus and encodes a cystatin (CpBV-CST1) gene. Its overexpression suppresses insect immunity and alters insect developmental processes. This study aimed to construct a genetically modified (GM) tobacco to further explore the physiological function of the viral cystatin and to apply to control insect pests. To this end, the transgenic tobacco lines were screened in expression of the target gene and assessed in insecticidal activity. A recombinant vector (pBI121-CST) was prepared and used to transform a bacterium, Agrobacterium tumefasciens. The transformed bacteria were used to generate transgenic tobacco lines, which were induced to grow callus and resulted in about 92% of shoot regeneration. The regenerated plants were screened by PCR analysis to confirm the insertion of the target gene in the plant genome. In addition, the expression of the target gene was assessed in the regenerated plants by quantitative real-time PCR (qRT-PCR). The qRT-PCR analysis showed that the transgenic line plant expressed the target gene about 17 times more than the control tobacco, indicating a stable insertion and expression of the target gene in the transgenic tobacco line. The insecticidal activity was then analyzed using the screened transgenic tobacco lines against the teneral 1st instar larvae of the oriental tobacco budworm, Helicoverpa assulta. Though there was a variation in the insecticidal efficacy among transgenic lines, T9 and T12 lines exhibited more than 95% mortality at 7 days after feeding treatment. These results suggest that CpBV-CST1 is a useful genetic resource to be used to generate GM crop against insect pests.
