Fruits and vegetables reportedly have a protective effect against hyperlipidemia and oxidative disease. Accordingly, this study aimed to investigate the lipid-lowering effect and antioxidative capacity of persimmon leaf extract (PLE) in rats fed a high-cholesterol diet. Male rats were fed a high-cholesterol (1% wt/wt) or high-cholesterol diet supplemented with Lovastatin (0.02% wt/wt) or PLE (0.2% wt/wt) for 5 weeks. The concentration of plasma total cholesterol was significantly lower in the PLE group than in the lovastatin group. However, the concentration of plasma HDL-cholesterol and the ratio of HDL-cholesterol/total-cholesterol (%) were significantly higher in the PLE group than in the control group. The PLE supplement also significantly lowered the contents of hepatic cholesterol and triglyceride. In comparing fecal sterol contents, the PLE group saw a significant increase of both neutral and acidic sterol compared to the other groups. The PLE supplement significantly lowered plasma GOT and GPT activity, which ave indices of hepatic toxicity. Plasma TBARS concentration was significantly lower in the PLE group than in the control group, while hepatic TBARS level was not significantly different between the groups. In a comparison of hepatic antioxidant parameters, SOD, catalase and GSH-Px activity were significantly higher in the PLE group than in the control group. However, the PLE supplement significantly towered antioxidant enzyme activity in the erythrocyte. Furthermore, these results suggest that supplementation of PLE promoted the excretion of fecal sterols, thereby leading to decreased absorption of dietary cholesterol. In addition, PLE may play an important role in regulating antioxidative capacities by altering SOD and ChT activity.
Surface sediment samples from 31 stations in Jinhae Bay were analyzed to evaluate the pollution by sewage from the spatial distribution of the fecal sterol, coprostanol. The sums of eight sterols (coprostanol, epicholestanol, epicoprostanol, cholesterol, cholestanol, brassicasterol, stigmasterol and $\beta$-sitosterol) were in the range of $2,703-27,154{\cal}ng/{\cal}g$ dry weight. The concentrations of the coprostanol in the sediments, which is a good tracer of sewage-derived organic contamination, were in the range of $76-3,964{\cal}ng/{\cal}g$ dry weight. The levels of coprostanol were much higher (almost one order of magnitude) at stations close to the big cities such as Masan and Jinhae. However, those levels were comparable to or lower than those previously reported in other foreign locations. Some ratios of coprostanol to other sterols and multivariate analysis could provide us with the information on the origin of the sterols being from sewage, plankton, and/or benthos. Those showed sewage dominance at the stations near the cities Masan and Jinhae, plankton dominance at the center of Jinhae Bay and mainly marine fauna origin at the mouth of Jinhae Bay. These results suggest that the contents of sterols and some ratios of these components are quite powerful tracer for the detection of the origin of the organic contaminants in the coastal environments.
Coprostanol is a metabolic product of cholesterol, formed by microbial action in the mammalian gut. This chemical compound is the major sterol in human and has been routinely studied as a biomarker of sewage pollution in marine and lacustrinesediments. This has led to the search for coprostanol as a biomarker in archaeologicalsoils, in order to detect the presence of fecal material. In this study, five samples of archaeological soils excavated at Wanggung ri, Iksancity, were used to assess the possibility of using coprostanol as indicators of ancient human activity in archaeological areas. The sampled soils were analyzed MXRD,EDXRF for their physical and chemical properties. And coprostanol was analysed byGC/MSD, using SIM method to detect and quantify specific compound. The results showed the soils were composed of quartz and feldspars, inorganicelement such as $SiO_2$, $Al_2O_3$, $Fe_2O_3$ etc. Moreover, the result from the analysis wasindicated that the specific compound is coprostanol. The coprostanol was determined at $0.16~1.01\mug$/g in the range of concentrations. This finding indicate that clear promise exists for the exploitation of coprostanol as biomarker of ancient human activity inarchaeological survey. Therefore such studies can serve to increase the confidence we place on biomarker-based methodologies for assessing fecal pollution. The application of this methodology has proved a simple and effective way of searching for that pattern in successively more aged deposits either known or suspected to contain fecal material. And the more scientific analysis of the soils will be showed to utility of the area ancient dietary life style, ancient environment.
고콜레스테롤 식이 흰쥐에서 xylooligo당이 간조직중의 UDP-glucronyl transferase 활성과 분변을 통한 지질, 담즙산 및 중성 sterol류의 배설 및 장기능개선 효과를 관찰하였다. 실험동물은 100$\pm$10g의 Sprague-Dawley 횐쥐를 이용하여 정상 식이군과 1% 고콜레스테롤 실험군으로 나누고 다시 실험군은 1%의 고콜레스테롤 식이에 xylooligo당을 0%, 5%, 10% 및 15% 농도 첨가군(C, C5XO, C10XO 및 C15XO군)로 나누어 각군마다 10마리씩 4주간 자유섭식 시켰다. 분변중의 총지질 및 TG함량은 C군에 비해 xylooligo당 공급군에서 현저하게 증가되었다. 간 microsome의 UDP-glucuronyl transferase의 활성은 정상군에 비해 C군에서 35% 증가되었고 또 C5XO, C10XO 및 X15XO군은 C군에 비해 15%, 41% 및 21%씩 각각 증가되었다. 분변중의 bile acid 함량은 C군에 비해 C5XO, X10XO 및 C15XO군에서 3.1배, 3.6배, 2.8배씩 각각 증가되었다. 분변중 중성 스테롤류인 cholesterol, coprostanol, coprostanone 함량은 C군에 비해 xylooligo당 공급군에서 현저하게 증가되었다. 이상의 결과에서 xylooligo당은 고콜레스테롤 식이 횐쥐에서 간조직의 UDP-GTase활성을 증가시키고 분변으로 총지질 및 중성지방 그리고 스테롤류의 배설을 증가시키는 효과가 있었고 또 분변량도 증가시키는 장기능 개선 효과도 뚜렷하였다.

Polycyclic aromatic hydrocarbons (PAHs), butyltins (BTs), nonylphenol (NP), and fecal sterols concentrations in sediments were investigated from Gunsan coast of Korea to evaluate organic pollution from anthropogenic activities. Sediment toxicity was also examined by bacterial bioluminescence toxicity test (Vibrio fischeri). The concentrations of 16 PAHs in sediments ranged from 67.9 to 425 ng/g dry wt; BTs ranged from 2.79 to 14.1 ng Sn/g dry wt; NP ranged from 20.7 to 2171 ng/g dry wt; and coprostanol, a fecal sterol, ranged from 7.60 to 245 ng/g dry wt. Effective concentration 50% ($EC_{50}$) of sediments ranged from 0.38 to 23.8 mg/mL. Most of the chemicals were present at levels lower than or comparable to the previously reported values from Korea. However, NP levels in the present study were in the high range of levels reported from the Korean coast, and 40% of the measured samples exceeded screening and ecotoxicological values of NP suggested by the Netherlands and Canada. This suggests that an ongoing source of NP is a serious concern in the Gunsan coast. High levels of contaminants were found in the proximity of potential sources, such as the outfall of a wastewater treatment plant for NP, an anthracite-fired power plant for PAHs, and ports for BTs, fecal sterols, and sediment toxicity. This indicates that Gunsan coast has various potential sources of marine sediment contaminants.
The effect of milk in low and high cholesterol diet was invesigated on serum cholesterol metabolism and lipid contents of serum, aorta, liver of rats. Weanling male Sprague-Dawley rats were divided into low(0.01% w/w) and high(1.01% w/w) cholesterol-diets groups. Low cholesterol groups subdivided into four groups ; control group was given water and three milk groups were given low heat milk(LM), ultra-high heat milk(HM), and powder milk(PM), respectivily, instead of water. High cholesterol groups were consisted of three groups ; control, LM, and HM groups. After feeding these experimental diets for six weeks, lipid levels were measured in serum and tissure and dried feces were analyzed for neutral and acidic sterols. Results obtained from this study are as follows : 1) Nutrient intakes, body weight gains and aorta weights did not differ among groups, but liver weights were higher in high cholesterol fed rats than low cholesterol fed rats. 2) Serum protein contents were increased independently by intakes of high cholesterol and milk. 3) Serum total cholesterol and triglyceride levels were increased but phospholipid levels and HDL-cholesterol/total-cholesterol ratios were decreased by high cholesterol in diet. And milk supplementation decreased serum total cholesterol and triglyceride levels but increased phospholipid levels and HDL-cholesterol/total-cholesterol ratios. 4) Contents of cholesterol and triglyceride in aorta and liver were elevated by dietary high cholesterol and lowered by consumption of all three types of milk. 5) Levels of cholesterol and triglyceride among serum, aorta and liver were highly correlated (r=0.7-0.9, p<0.001). 6) Fecal excretion of total sterols was three times high in high cholesterol group, compared with low cholesterol groups and were increased about 20% by milk consumption. 7) The effects of milk were more pronouncely shown in low cholesterol groups and mostly confined to LM and HM groups, rarely shown in PM group. It is concluded from the present study that milk had the hypolipidemic as well as hypocholes terolemic effect, which appears to be mediated through increased fecal bile acid excretion. But the effect is likely to be shaded by excess consumption of dietary cholesterol and was almost absent in powder milk.
Glycyrrhizin, an active constituent of Glycyrrhiza, was chemically extracted and its hypocholesterolemic activities were studied in rats. Following were the results: 1. The Sprague Dowley rats fed with the standard diet were divided into 4 groups, $50{\sim}60$ in each, and glycyrrhizin was given in doses of 0 (control), 0.5, 1.0 and 2.0mg per 100gm body weight daily for 10, 20, 30, and 60 days. Glycyrrhizin showed more or less the hypocholesterolemic activities in all animals. All of the animal groups to which 1. 0mg and 2.0mg of glycyrrhizin were given died before 60 days, while no mortality cases was noted in the group to which 0.5mg of glycyrrhizin was given. 2. Cholesterol administration induced hypercholesterolemia in all experimental animals, however, simultaneous administration of cholesterol with 0.5mg of glycyrrhizin suppressed the rise of the total serum cholesterol levels. The former group did not survive until 56 days, while the latter did. 3. Glycyrrhizin also Suppressed the cholesterol biosynthesis and incorporation of cholesterol in the liver tissues. 4. Glycyrrhizin induced the increase of the fecal excretion of the sterol compounds.
본 연구는 계란 난황을 경구투여 한 랫드에서 혈액 지질감소에 관한 생화학적 대사기전을 규명하기 위하여 수행하였다. 36마리의 Sprague Dawley 숫컷 랫드를 4처리구 3반복으로 완전임의 배치하였다. 6주 동안 동일한 정제고형식이를 급여하면서 매일 한번 씩 난황을 경구투여 하였다. 대조군(C; 생리식염수 1.0 g), T1(삼겹살 기름 1.0 g), T2(난황 1.0 g), T3(삼겹살 기름 1.0g 과 난황 1.0 g을 각각 1주일씩 교체 투여군)으로 구분하였다. 일일평균 증체량은 T1이 가장 높았고 T3, T2, C 순서로 낮게 나타났다. 혈액 중성지방과 총콜레스테롤은 T1이 가장 높았고 C, T3, T2 순서로 낮게 나타났다. HDL-C는 T2가 가장 높았고 C, T3, T1 순서로 낮게 나타났으나 LDL-C는 T1이 가장 높았고 T3, C, T2 순서로 높게 나타났다. 간 기능을 나타내는 지표 AST, ALT는 T1에서 가장 높았고 T2가 가장 낮게 나타났다. 복강지방을 제외한 간, 비장, 신장의 무게는 각 처리구 사이에 차이는 없었다. 복강지방은 T1이 가장 높았으나 C, T2, T3사이의 차이는 없었다. HMG-CoA reductase activity는 T1이 가장 높았고 T3, C 순서로 낮게 나타났으며 T2는 가장 낮았다. 분을 통하여 배설되는 일일 총스테롤, 중성스테롤, 산성스테롤의 배설량은 T2가 가장 높았으나 T1은 가장 낮았다. 본 연구결과는 계란의 섭취가 동물의 간에서 콜레스테롤 합성효소의 활성을 억제함과 동시에 분을 통한 스테롤의 배설을 촉진시켜줌으로써 혈액 지질을 낮춘다는 사실을 확인하였다.
This study was conducted to examine the effect of dietary iron levels on lipid metabolism, antioxidative and antithrombogenic capacities in 16-month-old rats. Thirty-two Sprague-Dawley male 16-month-old rats weighing 618 $\pm$ 6 g were raised for 10 days with medium-iron diet (35 ppm in diet) and blocked into 4 groups according to their body weights. One of groups was sacrificed to obtain initial data and the rest 3 groups were raised for 3 months with experimental diets containing different levels of iron (5 ppm, 35 ppm, and 350 ppm). Total lipid, triglyceride and total chole-sterol concentrations in plasma and liver, HDL-cholesterol concentration in plasma, fecal total lipid triglyceride and total cholesterol excretions, thiobarbituric acid reactive substances (TBARS) level in plasma LDL + VLDL (low density lipoprotein + very low density lipoprotein) fractions, blood-clotting time and eicosanoids levels in plasma were measured. The results are as follows: Plasma total lipid, triglyceride and total cholesterol concentrations, TBARS level in plasma LDL + VLDL fractions were increased and blood-clotting time tended to be shortened during 3 months of experimental period. Low (5 ppm) iron diet improved lipid metabolism via increasing HDL-cholesterol and fecal choles-terol excretion. High (350 ppm) iron diet decreased plasma total lipid, triglyceride and total cholesterol concentrations as compared to medium (35 ppm) iron diet and lowered body weight and epididymal fat pad weight. On the other hand, TBARS level in plasma LDL + VLDL fractions and blood-clotting time were increased with high iron diet. It is plausible that low iron diet improves lipid metabolism, antioxidative and antithrombogenic capacities in 16-month-old rats.
The study was designed to observe the effect of dietary calcium and fats on plasma cholesterol level, hepatic microsomal fluidity and HMG-CoA reductase activity as well as the excretion of fecal bile acids and neutral sterols in 1, 2-dimethylhydrazine(DMH)-treated rats. Male Sprague Dawley rats, at 7 weeks of age, were divided into 2 groups, 0.3% and 1.0% Ca levels and each group again subdivided into 2 groups of corn oil and perilla oil. Each rat was intramuscularly infused with DMH for 6 weeks to give total dose of 180mg/kg body weight and also fed experimental diet containing 15%(w/w) different fit and Ca(0.3% or 1.0%) for 20 weeks. High dietary calcium(1.0%) did not significantly influence on plasma cholesterol as well as hepatic microsomal fluidity and HMG CoA reductase activity, but significantly reduced the excretion of total bile acid per gram of faces and increased the excretion of total neutral sterol. However, high dietary Ca reduced the excretion of secondary bile acid(deoxycholic and lithocholic acids) which was known as promoter for colon cancer. Perilla oil rich in n-3 ${\alpha}$-linolenic acid significantly decreased plasma cholesterol by increasing hepatic microsomal fluidity compared with corn oil, but did not influence on HMG CoA reductase activity. Perilla oil did not influence on fecal excretion of total and primary bile acids, but reduced the excretion of secondary bile acids. Therefore, it could be recommended to consume more fish product and food rich in calcium and use more perilla oil in meal preparation to prevent from coronary hear disease and colon cancer especially when high fit diet has been practiced.
