Production of Rapamycin in Streptomyces hygroscopicus from Glycerol-Based Media Optimized by Systemic Methodology

  • Rapamycin, produced by the soil bacterium Streptomyces hygroscopicus, has the ability to suppress the immune system and is used as an antifungal, anti-inflammatory, antitumor, and immunosuppressive agent. In an attempt to increase the productivity of rapamycin, mutagenesis of wild-type Streptomyces hygroscopicus was performed using ultraviolet radiation, and the medium composition was optimized using glycerol (which is one of the cheapest starting substrates) by applying Plackett-Burman design and response surface methodology. Plackett-Burman design was used to analyze 14 medium constituents: M100 (maltodextrin), glycerol, soybean meal, soytone, yeast extract, $(NH_4)_2SO_4$, $\small{L}$-lysine, $KH_2PO_4$, $K_2HPO_4$, NaCl, $FeSO_4{cdot}7H_2O$, $CaCO_3$, 2-(N-morpholino) ethanesulfonic acid, and the initial pH level. Glycerol, soytone, yeast extract, and $CaCO_3$ were analyzed to evaluate their effect on rapamycin production. The individual and interaction effects of the four selected variables were determined by Box-Behnken design, suggesting $CaCO_3$, soytone, and yeast extract have negative effects, but glycerol was a positive factor to determine rapamycin productivity. Medium optimization using statistical design resulted in a 45% ($220.7{\pm}5.7mg/l$) increase in rapamycin production for the Streptomyces hygroscopicus mutant, compared with the unoptimized production medium ($151.9{\pm}22.6mg/l$), and nearly 588% compared with wild-type Streptomyces hygroscopicus ($37.5{\pm}2.8mg/l$). The change in pH showed that $CaCO_3$ is a critical and negative factor for rapamycin production.

Bronchoplastic Procedures for Bronchogenic Carcinoma (폐암 환자에서 기관지성형술)

  • Bronchoplastlc procedures including sleeve lobectomy were initially introduced for patients whose pulmonary function was insufficient to tolerate pneumonectomy In more recent years, sleeve lobectomy has evolved as an alternative to pneumonectomy in carefully selected cases of bronchogenic carcinoma, especially for centrally located lesions. Between 1992 and 1995, bronchoplastic procedures for bronchogenic carcinoma were performed in 15 patients and the majority of operative procedures were sleeve lobectomy (W: 12). All procedures were considered as complete and potentially curative. Mean age was 62.3 years (range 46 to 70 years) and there were 12 males and 3 females. Of 15 patiients, 7 underwent right upper sleeve, 2 underwent right lower sleeve, 5 underwent left upper sleeve, and 1 underwent right sleeve pneumonectomy. Postoperative staging was , stage I in 3, stage ll in 8, stage llla in 3 and stage lII in 1. The postoperative complications included anastomosis site obstruction due to granulation tissue in 1, local recurrence in 3, and wound infection in 1 There were 1 operative death due to sepsis and 2 late deaths. The three-year survival rate was 80%. The significant correlation was observed (r=0.11) between the predicted FEVI (1.851 L) and measured FEVI (1 762L). In conclusion, bronchoplastic procedure will have better prognosis than pneumonectomy in selected lung cancer patients because of preserving good function in remnant lung.

Response of Soil Microbial Communities to Different Cultivation Systems in Controlled Horticultural Land

  • Ester-linked fatty acid methyl ester (EL-FAME) profiles were used to describe differences in soil microbial communities influenced by conventional farming system (CFS), and organic farming system (OFS) in controlled horticultural land. Soil physicochemical properties and soil microbial communities were determined in the experimental fields. Higher organic matter content in OFS reduced soil bulk density which in turn increased the soil porosity. Generally, soil chemical properties in OFS were higher than those of CFS, but EC value in OFS was significantly lower than that of CFS. With the exception of Fe content, other macronutrient contents and pH in both farming system decreased with the soil depth. Soil microbial biomass of OFS was approximately 1.3 times in topsoil and 1.8 times in subsoil higher than those of CFS. Lower ratios of cy17:0 to $16:1{\omega}7c$ and cy19:0 to $18:1{\omega}7c$ were found in the CFS soils than the OFS soils, indicating that microbial stress decreased. The ratio of MUFA to SFA was higher in OFS due to organic input to the soil. In principal components analysis (PCA), the first variable accounted for 54.3%, while the second for 27.3%, respectively. The PC1 of the PCA separated the samples from CFS and OFS, while the PC2 of the PCA separated the samples from topsoil and subsoil. EL-FAMEs with the positive eigenvector coefficients for PC1 were cy17: 0 to $16:1{\omega}7c$ ratio, cy19:0 to $18:1{\omega}7c$ ratio, soil pH, soil organic matter, and soil $NO_3$-N content. Our findings suggest that the shifting cy19:0 to $18:1{\omega}7c$ ratio should be considered as potential factors responsible for the clear microbial community differentiation observed between different cultivation systems and soil depth in controlled horticultural land.

Finite element analysis of peri-implant bone stresses induced by root contact of orthodontic microimplant (치근접촉이 마이크로 임플란트 인접골 응력에 미치는 영향에 대한 유한요소해석)

  • Objective: The aim of this study was to evaluate the biomechanical aspects of peri-implant bone upon root contact of orthodontic microimplant. Methods: Axisymmetric finite element modeling scheme was used to analyze the compressive strength of the orthodontic microimplant (Absoanchor SH1312-7, Dentos Inc., Daegu, Korea) placed into inter-radicular bone covered by 1 mm thick cortical bone, with its apical tip contacting adjacent root surface. A stepwise analysis technique was adopted to simulate the response of peri-implant bone. Areas of the bone that were subject to higher stresses than the maximum compressive strength (in case of cancellous bone) or threshold stress of 54.8MPa, which was assumed to impair the physiological remodeling of cortical bone, were removed from the FE mesh in a stepwise manner. For comparison, a control model was analyzed which simulated normal orthodontic force of 5 N at the head of the microimplant. Results: Stresses in cancellous bone were high enough to cause mechanical failure across its entire thickness. Stresses in cortical bone were more likely to cause resorptive bone remodeling than mechanical failure. The overloaded zone, initially located at the lower part of cortical plate, proliferated upward in a positive feedback mode, unaffected by stress redistribution, until the whole thickness was engaged. Conclusions: Stresses induced around a microimplant by root contact may lead to a irreversible loss of microimplant stability.

Cooking Characteristics of Coated Rice with Water Homogenate of Citrus Fruits Peel (감귤과피 물 균질액으로 가공한 유색미의 취반 특성)

  • Cooking characteristics of coated rice with water homogenate of citrus fruit peels (1% for rice) were investigated. The color of the coated rice both before and after cooking was dark yellow. The total content of carotenoids, hesperidin and naringin were 10.74, 2173.12 and 1468.40 mg% for citrus fruit peels, 0.46, 108.65 and 73.35 % for its water homogenate, 0.12, 21.73 and 14.62 mg% for coated rin, and 0.05, 8.67 and 5.87 mg% fur cooked coated rice, respectively. Citrus fruits peel contained 94.22 mg% of asparagine, 24.88 mg% of methionine, 19.64 mg% of alanine, and 15.37 mg% of ${\gamma}$-aminoisobutyric acid as the majority free amino acids, accounting for 70% of the total free amino acids present. Total free amino acid content of the cooked coated rice increased by 15% compared to those of cooked uncoated rice. The majority of minerals in the citrus fruit peels were K and Ca, accounting for 56% of total minerals present. The mineral content of cooked coated rice was generally higher than that of the cooked uncoated rice. The cooked coated rice showed comparable hardness, gumminess and brittleness, but higher cohesiveness and springiness than the cooked uncoated rice. There were no differences in sweet and bitter taste between the cooked uncoated and coated rice. However, the cooked coated rice showed higher sensory scores fur color acceptability, savory taste and overall acceptability than the cooked uncoated rice.

Effects on Bond Strength between Zirconia and Porcelain according to Etching Treatment and Low Temperature Degradation (지르코니아 표면에칭처리와 저온열화현상이 지르코니아와 전장도재의 결합강도에 미치는 영향)

  • The purpose of this study was to investigate the influence of etching surface treatment and aging treatment of zirconia on the shear bond strength between zirconia core and veneered ceramic. Four groups of zirconia-ceramic specimens were prepared; 1) NEZ group (no etching zirconia), 2) EZ group (etching zirconia), 3) ANEZ group (aging and no etching zirconia), 4) AEZ group (aging and etching zirconia). The shear bond strength between zirconia and porcelain was measured using Instron Universal Testing Machine. Surface texture with crystalline structure of zirconia surface was examined by the field emission scanning electron microscopy (FE-SEM) with ingredient analysis. The fractured surfaces of specimens were examined to determine the failure pattern by a digital microscope. The mean${\pm}$standard deviation of shear bond strengths were $23.47{\pm}3.47$ Mpa in NEZ, $28.30{\pm}4.34$ Mpa in EZ, $21.85{\pm}4.65$ Mpa in ANEZ, $24.65{\pm}3.65$ Mpa in AEZ group, respectively, and were significantly different (p<0.05). The average shear bond strength was largest in EZ group, followed by AEZ, NEZ, and ANEZ groups. Most specimens in NEZ group showed adhesive failure and most specimens in EZ, AEZ, and ANEZ group showed mixed failure. Surface of etching treatment group (EZ and AEZ) showed complex micro-structure and irregular surface texture which may facilitate mechanical interlocking, while untreated zirconia surface presented simpler micro-structure. In conclusion, an etching treatment improved bonding strength between zirconia and porcelain by forming mechanical interlocking.

Distribution of Sulfate-reducing Bacteria in Landfill Leachate and their Role on Insolubilization of Heavy metals (폐기물매립지 침출수에서 황산염환원균의 분포와 중금속 불용화역할)

  • This study, collaborated Gifu University, Japan, was performed to analyze chemical pollutants and microorganism and to clarify the distribution of sulfate-reducing bacteria and their insolubilization of heavy metal ions in leachates sampled seasonally between 1994 and 1996 from Nanjido waste landfill site, sampled 4 times between 1995 and 1996 from Pusan and Daejeon waste landfill site, and sampled 1 time between 1992 and 1994 from Hokkaido, Nagoya, Osaka and Hukuoka waste landfill site in Japan. The results were as follows: 1. The temperatures of internal leachate and leachate effluent were 40$\circ$C and 30$\circ$C, respectively, and the pH values of both leachates were about 8.0 at Nanjido waste landfill site. The concentration of SO$_4^{-2}$ gradually increased with the degree of stabilization and that of NO$_3$-N was detected in a part of sampling sites at one and half years, and in all sampling sites at 3 years after completion of landfill. 2. The organic substances in leachate of Nanjido waste landfill site decreased with the degree of stabilization and they were very fluctuated with measuring point and time. The concentration of organic substance and heavy metals in internal leachate were higher than in leachate effluent and those of Cd, Hg, and Pb were lower than detection limit except a part of samples in 1996. 3. APCs in internal leachate and leachate effluent were not much different and the minimum of APCs in internal leachate and leachate effluent were $1.0\times 10^4$/ml and $4.0\times 10^1$/ml, respectively. 4. The maximums of SRBs in Nanjido, Pusan, and Daejeon waste landfill site were 9180 MPN/ml, 24000 MPN/ml, and 348 MPN/ml, respectively and the maximum of SRBs in Japan waste landfill site was 9300 MPN/ml. 5. During 2-week-SRB culture, the values of MPN were high at 50$\circ$C for initial culture period and at 30$\circ$C for last culture period. MPN started to appear at first day and rapidly increased between 7th day and 9th day. 6. Cadmium and copper were insolubilized by SRB within 6 hr and iron and zinc were done within 48 hr. The rates of insolubilization of Cd, Cu, Fe, Zn, T-Cr were 100%, 99.5%, 95.0%, 99.8%, 16.1% after 48 hr treatment with SRB, respectively.

Geochemistry of Shallow gases taken from the core sediments in the southeastern Ulleung Basin (울릉분지 남동부 시추 퇴적물 내에 함유되어 있는 천부가스의 특성)

  • Chemical and isotopic compositions of hydrocarbon gases were analyBed to characterize the properties of the shallow gases distributed in the southeastern part of the Ulleung Basin, offshore Korea. Sediments from the core were also analyzed to determine the characteristics and relationship to shallow gases. Hydrocarbon gases in the sediments consisted of methane (697.9-6054.4 ppm), ethane, propane, butane and hexane. The total carbon content of the sediments ranges from 1.84fe to $5.11{\%}$ and the total organic carbon content ranges from $0.29{\%} \;to\; 2.65{\%}$. High C/N ratio (>10) indicates that input of terrestrial organic matter was prevalent at the time of deposition. The methane content and stable isotopic data indicate that hydrocarbon gases from the sediments are identified to be thermogenic gas and mixture of both biogenic and thermal gases. Based on the Rock-Eval and carbon isotopic data, the level of thermal maturity of organic matter in the sediments $(Tmax<425^{\circ}C)$ is lower than that of gas. It suggests that thermal gases in the sediments migrated from the deeper sediments than the penetrated depth.

  • These studies were carried out to investigate the effects of rice straw on the leaching of chemical constituents in paddy soil. Rice plants were cultured in paddy soil to which rice straw was applied and then chemical properties of percolated water were analysed. The results were as follows. The leaching of calcium and magnesium was affected by rice straw application in the early stages of plant growth and by rice root activity in the late stages. The application of the straw promoted the reduction of the soil followed by increasing the leaching of iron and manganese. The leaching of potassium, ammonium-nitrogen and chloride was not due to the application of rice straw and the leaching of carbon dioxide increased with the application of rice straw, at the same time chemical properties seemed to be affected by rice root activity. Generally, cation and anion leached in the percolated water were equivalent. Calcium, magnesium, Fe as cation and $HCO_3$, $SO_4{^{-2}}$ as anion were important constituents in the percolated water from paddy soil.

Effect of Varying Levels of Dietary Minerals on Growth and Nutrient Utilization in Lambs

  • Hisardale male lambs (n=24, 4-5 month of age) were maintained on a conventional ration for a month, to equilibrate the body mineral status. Six lambs were sacrificed after one month of feeding; the samples of organs were analyzed to ascertain mineral status. The remaining 18 lambs were divided into 3 groups of 6 each on body weight basis. Three dietary treatments containing 100 ($T_1$), 110 ($T_2$) and 120% ($T_3$) of minerals (Ca, P, S, Zn and Mn) as specified by NRC (1985) were formulated and each treatment was alloted ad libitum to a group for 120 days. Blood and wool samples were collected at monthly intervals. At the end of the experiment a balance trial of 5 days duration was conducted to study the balance of mineral elements. The effective intake of minerals was Ca, 111 and 120, P, 110 and 122; S, 112 and 129; Zn, 112 and 126 and Mn, 109 and 123 percent in $T_2$ and $T_3$, respectively, in comparison to $T_1$ (100). The additional mineral supplementation had no significant effect on dry matter intake. The average daily weight gain was higher (p<0.01) in $T_2$ and $T_3$ than the control ($T_1$). The lambs under treatments $T_2$ (8.72) and $T_3$ (8.47 kg) consumed apparently lesser amount of dry matter per unit gain as compared to $T_1$ (10.81 kg). Significantly higher (p<0.05) dry matter and crude protein digestibility (%) were observed in $T_2$ and $T_3$ than in $T_1$. The mean balances for different elements were Ca, 1.14, 1.68 and 1.67 g; P, 1.70, 1.95 and 2.18 g; S, 0.54, 0.92 and 1.11 g; Zn, 22.56, 25.30 and 28.71 mg; Cu, 7.94, 5.71 and 5.53 mg; Fe, 33.19, 32.94 and 31.03 mg and Mn, 8.24, 14.40 and 16.07 mg/lamb/day. The retention of supplemental minerals increased (p<0.01) while that of Cu decreased (p<0.01) due to supplementation of minerals (Ca, P, S, Zn and Mn). Retention as per cent of intake increased statistically for S and Mn while that of Cu decreased. It can be concluded that supplementation of minerals (Ca, P, S, Zn and Mn) higher than the recommended level improved body weight gain and feed to gain ratio. The retention of minerals increased due to supplementation. Therefore, an additional supplementation of deficient minerals (Ca, P, S, Zn and Mn) by 10% was beneficial for Hisardale male lambs under tropical condition in India.
