Iron Containing Superoxide Dismutase of Streptomyces subrutilus P5 Increases Bacterial Heavy Metal Resistance by Sequestration (Streptomyces subrutilus P5의 철 함유 Superoxide Dismutase의 중금속 격리에 의한 세균의 중금속 저항성 증가)

  • Mitigation of heavy metal toxicity by iron containing superoxide dismutase (FeSOD) of Streptomyces subrutilus P5 was investigated. For E. coli $DH5{\alpha}$, the survival rate in the presence of 0.1 mM lead ions was only 7% after 120 min; however, with the addition of $0.1{\mu}M$ of purified native FeSOD the survival rate increased to 39%. This detoxification effect was also shown with 0.01 mM copper ions (survival increased from 6% to 50%), and the effect was stronger than with the use of EDTA. E. coli M15[pREP4] producing 6xHis-tagged FeSOD was constructed, and this showed an increase in survival rates throughout the incubation time; in the presence of 0.1 mM lead ions,the final increase at 60 min was from 3% to 19%. The FeSOD absorbed about 123 g-atom lead per subunit; therefore, we suggest that FeSOD could sequestrate toxic heavy metals to enhance bacterial survival against heavy metal contamination.

Development of the Purification Method of Ovotransferrin in Egg White (난백 내 Ovotransferrin의 분리방법에 관한 연구)

  • This study was carried out to separate ovotransferrin in chicken egg white by gel chromatography and heparin affinity chromatography. In gel filtration which was performed with 50mM Phosphate buffer (pH 7.2, 0.15M salt) at a flow rate of 2.0 ml/min, ovotransferrin and ovalbumin were eluted together in fraction number 11-16. In order to separate pure ovotransferrin, fraction No. 12-14 of them which have high concentration of ovotransferrin were concentrated and rechromatographed. However, the ovotransferrin did not separated clearly. In heparin affinity chromatography, the separation was performed with 50mM ethylaminetetraacetic acid (EDTA, pH7.2) and 50mM Phosphate buffer (pH 7.2, 0.15M salt contained) on ferrous and ferric ion saturated column at as same flow rate as gel filtration system's. Ovotransferrin and albumin were eluted together at 10-15min (fraction No.3) and 15-20min (fraction No.4), respectively. However, purified ovotransferrin was eluted at 156-165min and 165-175min (tube No.32-33) with 50 mM phosphate buffer (pH 7.2, 0.15M salt free), respectively. Heparin affinity chromatography with ferric ion saturated column was resulted in the best separation of ovotransferrin rather than separation by gel chromatography and ferrous ion saturated heparin affinity chromatography.

Effect of Iron Activators on the Persulfate Oxidation of Polycyclic Aromatic Hydrocarbons (PAHs) in Contaminated Soils (다환방향족 탄화수소(PAHs) 오염토양의 과황산 산화 시 철 활성화제의 영향)

  • PAHs commonly found in industrial sites such as manufactured gas plants (MGP) are potentially toxic, mutagenic and carcinogenic, and thus require immediate remediation. In-situ chemical oxidation (ISCO) is known as a highly efficient technology for soil and groundwater remediation. Among the several types of oxidants utilized in ISCO, persulfate has gained significant attention in recent years. Peroxydisulfate ion (S2O82-) is a strong oxidant with very high redox potential (E0 = 2.01 V). When mixed with Fe2+, it is capable of forming the sulfate radical (SO4) that has an even higher redox potential (E0 = 2.6 V). In this study, the influence of various iron activators on the persulfate oxidation of PAHs in contaminated soils was investigated. Several iron sources such as ferrous sulfate (FeSO4), ferrous sulfide (FeS) and zero-valent iron (Fe(0)) were tested as a persulfate activator. Acenaphthene (ANE), dibenzofuran (DBF) and fluorene (FLE) were selected as model compounds because they were the dominant PAHs found in the field-contaminated soil collected from a MGP site. Oxidation kinetics of these PAHs in an artificially contaminated soil and the PAH-contaminated field soil were investigated. For all soils, Fe(0) was the most effective iron activator. The maximum PAHs removal rate in Fe(0)-mediated reactions was 92.7% for ANE, 83.0% for FLE, and 59.3% for DBF in the artificially contaminated soil, while the removal rate of total PAHs was 72.7% in the field-contaminated soil. To promote the iron activator effect, the effects of hydroxylamine as a reducing agent on reduction of Fe3+ to Fe2+, and EDTA and pyrophosphate as chelating agents on iron stabilization in persulfate oxidation were also investigated. As hydroxylamine and chelating agents (EDTA, pyrophosphate) dosage increased, the individual PAH removal rate in the artificially contaminated soil and the total PAHs removal rate in the field-contaminated soil increased.

  • This study was carried out a investigate the effect of chemical cleaning of corrosion product in cooling system made of copper and copper alloy as basic material and used cooling water as pure water. We studied chemical cleaning condition that minimizes the influence on basic material by means of EDTA solution so as to eliminate the slurry in cooling system. As a result, we found that the main components of sludge in cooling system produced by corrosion of copper were $Cu_2O$, CuO, Cu, and Fe. The optimum condition of chemical cleaning was 400 ppm EDTA solution at $60^{\circ}C$.

Metallo-collagenase production by Arthrobacter creatinolyticus KP015744

  • Amongst 27 isolates from deteriorated leather samples, Arthrobacter creatinolyticus KP015744 zzx28 was found to be an efficient collagenase producer. Collagenase production of 13.33 µmoles/min was shown at an optimum temperature at 37℃ after 72h and at pH 7.5 by using 2 ml/dL inoculum in 10 mg/ml collagen peptide type I as a substrate. In presence of Hg2+, EDTA and 𝛽-mercaptoethanol the collagenase production by the isolate was strongly inhibited however Fe2+, Ca2+and DMSO enhanced production of the enzyme. Specific activity was found to be 19.46×103 U/mg and molecular weight 66 kD by SDS PAGE. Isolate also has potential to hydrolyze keratin which is another important protein found in leather. Experimental results propose that collagenase can be effectively used as a tool for collagen and keratin rich solid waste treatment.

Ex-situ Remediation of a Contaminated Soil of Fe Abandoned Mine using Organic Acid Extractants (유기산 추출에 의한 철 폐광산 오염토양의 복원)

  • A study on the remediation of heavily for ion contaminated soils from abandoned iron mine was carried out, using ex-situ extraction process. Also, oxalic acid as a complex agent was evaluated as a function of concentration, reaction time and mixing ratio of washing agent in order to evaluate Fe removability of the soil contaminated from the abandoned iron mine. Oxalic acid showed a better extraction performance than 0.1N-HCl, i.e., the concentrations of Fe ion extracted from the abandoned mine for the former at uncontrolled pH and the latter were 1,750 ppm and 1,079 ppm, respectively. The optimum washing condition of oxalic acid was in the ratio of 1:5 and 1:10 between soil and acid solution during l hr reaction. The total concentrations of Fe ion by oxalic acid and EDTA at three repeated extraction, were 4,554 ppm and 864 ppm, respectively. The recovery of Fe ions from washing solution was achieved, forming hydroxide precipitation and metal sulfide under excess of calcium hydroxide and sodium sulfide. In addition, the amounted of sodium sulfide and calcium hydroxide for the optimal revovery of Fe were 15g/$\ell$ and 5g/$\ell$ from the oxalic acid complexes, respectively.

  • 2,3,6-Tribromo-4,5-dihydroxybenzyl methyl ether (TDB) from the methanolic extract of the red alga Symphyocladia latiuscula exhibits major antioxidant activity. In this study, the activity of TDB against oxidative damage in deoxyribose and DNA was investigated in vitro for potential applications in preventing mutagenesis caused by DNA damage. TDB inhibited the oxidation of deoxyribose at concentrations of up to $1{\mu}g$/mL in the presence of $Fe^{+3}$-EDTA/$H_2O_2$. Furthermore, TDB showed no pro oxidant activity as determined by absence of the reduction of bleomycin-$Fe^{+3}$ to bleomycin-$Fe^{+2}$, which leads to DNA damage. Based on these results, TDB demonstrated considerable antioxidant activity without prooxidant properties.

Potential Role of $Ca^{++}$ on the Differentiation of Erythroid Progenitor Cells

  • In ordedr to gain insight into the mechanisms byl which erythropoietin promotes erythropoiesis, effects of various inhibitors on the erythropoietin-propmoted differentiation of erythroid progenitor cells and on the erythroid progenitor cells and on the erythropoietin-promoted $Ca^{++}$ uptake in the progenitor cells were determined, and the relationship between the inhibitory activity of each inhibitor cells were determined, and he relationship between the inhibitory activity of each inhibitor toward the differentiation and channel blocker (varapamil), a $Ca^{++}$ chelator (EDTA) and a protein kinase C inhibitor (stauroporine). All of these agents inhibited both the erythropoietin-mediated differentiation of the erythroid progenitor cells, as determined by the incroporation of $^{59}Fe$ into heme, and $Ca^{++}$ uptake in a concentrtion dependent manner. In the cases of varapamil and EDTA, the half-miximal inhibitory concentration $(IC_{50})$ values for differentiation of the progenitor cells may be theconsequence of the inhibition of the $Ca^{++}$ uptake in a concentration dependent manner. In the cases of varapamil and EDTA, the half-miximal inhibitory concentration dependent manner. In the cases of verapamil and EDTA, the half-miximal inhibitory concentration $(IC_{50})$ values for differentiation of the progenitor cells may be the consequence of the inhibition of the $Ca^{++}$ uptake by the inhibitor. On the other hand, in the cases of genistein and stauroporine, the $IC_{50}$ values for inhibition of differentitation were significantly different from that for inhibition of $Ca^{++}$uptake. These results suggest that the mechanism of inhibition of differentiation by these two inhibitors in complex. However, taken all together, the above results support the proposition that $Ca^{++}$ uptake may play a role in the erythropoietin-mediated differentiation of erythoid progenitor cells.

  • The potential candidates for IT-SOFCs cathode materials, $Ba_{0.5}Sr_{0.5}Co_{0.8}Fe_{0.2}O_{3-{\delta}}$ (BSCF) and $La_{0.6}Ba_{0.4}Co_{0.2}Fe_{0.8}O_{3-{\delta}}$ (LBCF) powders, were synthesized by a EDTA-citrate combined method from $Sr(NO_3)_2$, $Ba(NO_3)_2$, $La(NO_3)_3{\cdot}6H_2O$, $Co(NO_3)_2{\cdot}6H_2O$, $Fe(NO_3)_3{\cdot}9H_2O$, citric acid and $EDTA-NH_3$. The cathode performance of symmetrical electrochemical cells consisting of BSCF-GDC or LBCF-GDC composite electrodes and a GDC electrolyte was investigated using by AC impedance spectroscopy at the temperature range of 500 to $700^{\circ}C$. It was found that a single phase perovskite could be successfully synthesized when the precursor is heated at $850^{\circ}C$ for 2 h. Due to thermal expansion mismatch between BSCF and GDC, the composite cathodes with lower GDC content than 45 wt% were peeled off from the GDC electrolyte and their electrode polarization resistance was estimated to be high. The thermal expansion coefficient of BSCF-GDC composites was decreased with increasing the GDC content and the electrode peeling off did not occur in BSCF-45 and 55 wt% GDC composites. BSCF-45 wt% GDC composite electrode showed the lowest area specific resistances (ASR) of 0.15 and $0.04{\Omega}{\cdot}cm^2$ at 600 and $700^{\circ}C$, respectively. On the other hand, LBCF-GDC composite cathodes showed higher ASR than the BSCF-45 and 55 wt% GDC and their cathode performance were decreased with the GDC content.

Biochemical studies of the siderophore A3 produced by pseudomonas synxantha A3 (Pseudomonas synxantha A3가 생성하는 siderophore A3에 관한 연구)

  • A yellow-green, fluorescent siderophore A3 was extracellularly produced under iron-limited growth conditions from Pseudomonas synxantha A3. The physicochemical and biological properties of siderophore A3 were examined. The approxiamte molecular weights of the Fe(III)-siderophore A3-1 complex and Fe(III)-siderophore A3-2 complex were estimated to be about 1,300 and 1,100, respectively, by Bio-gel P2 gel exclusion chromatography. The molar ratio between the siderophore and the Fe(III)was 1.08 mole. The molecular weight of the complex could be calculated with this ratio and the new values were 1,150 and 960, respectively. The binding constant(K) between thesiderophore A3 and Fe(III) that determined by displacing the iron from the Fe(III)-siderophore complex with EDTA was 4.12*10$^{18}$ at pH 5.0. Siderophore A3 appeared to have antibacterial activity on several bacterial strains, however, ferric siderophore Ae complex did not show that activity. The cytotoxicity of siderophore A3 was obtained from Human Chronic Myelogenous Leudemia K562 cells. Inhibition concentration (50%)($IC_{50}$ ) was $0.17\mu$\{g/ml}.
