Purification and Characterization of Mouse Liver Rhodanese

  • Rhodanese from mouse liver was purified to near homogeneity by ammonium sulfate precipitation, CM-Sephadex ion exchange, hydroxyapatite and Sephacryl S-200-HR gel filtration chromatographies with a purification of 776 folds. The molecular weight was determined by Sephadex G-150 gel filtration and found to be 34.8 KDa. SOS-PAGE showed molecular weight 34 KDa and two identical subunits splitting by aging for 3 weeks at $-70^{\circ}C$ the molecular weight of which was 17 KDa. The optimal pH of enzyme activity was 9.4 and the pI value of the enzyme was 6.6. Rhodanese showed the optimal reaction temperature of $25^{\circ}C$ and near linear increasing pattern until 10 min. incubation. $K_m$ values of rhodanese for KCN and $Na_{2}S_{2}O_{3}$ as substrates were 12.5 mM and 8.3 mM, respectively. Rhodanese activity was inhibited by more than 70% at a concentration of 100 ${\mu}M$ of $Ni^{2+}$, $Zn^{2+}$, $Cd^{2+}$, $Hg^{2+}$ and $Cu^{2+}$. Other metal ions, such as $Mn^{2+}$, $Mg^{2-}$, $Ca^{2+}$, and $Fe^{2+}$ showed no effect on rhodanese activity.

Purification and Characterization of an Extracellular Protease from Bacillus pumilus CN8

  • The protease produced by a Bacillus pumilus CN8 strain was purified by DEAE-Cellulose-52 ion exchange. It has a molecular weight of approximately 96,920 Dalton. In the present study, this protease showed strong activity over a broad range of pH (6.5-9.5) and temperature from $40^{\circ}C$ to $60^{\circ}C$, and the protease performed the maximal activity at pH 7.3 at $42^{\circ}C$. The effect of metal ions on protease activity showed that $K^+$ could slightly increase the protease activity, and other ions such as $Zn^{2+}$, $Fe^{2+}$, $Na^+$, $Ca^{2+}$, $Mg^{2+}$ had no significant activation or inhibition to the protease (P> 0.05), and the more important is that $Cu^{2+}$, $Mn^{2+}$, $Sn^{2+}$, $Cd^{2+}$ had a strong inhibitory effect on the protease activity.

Chemical Modification of Extracellular Cytosine Deaminase from Chromobacterium violaceum YK 391

  • Essential amino acids involved in the catalytic role of the extracellular cytosine deaminase from Chromobacterium violaceum YK 391 were determined by chemical modification studies. The enzyme activity required the reduced form of Fe (II) ion, since the enzyme was inhibited by ο-phenanthroline. The enzyme activity was completely inhibited by the chemical modifiers, such as p-chloromercuribenzoate (p-CMB), p-hydroxymercuribenzoate, and chloramine-T at 1 mM each. The enzyme activity was also markedly inhibited by pyridoxal-5'-phosphate, diethyl pyrocarbonate, and phenylmethylsulfonyl fluroride at 1 mM each. The inactivation of the enzyme activity with p-CMB was reversed by a high concentration of cytosine. Furthermore, the inactivation of the enzyme activity with p-CMB was also reactivated by 1 mM dithiothreitol, 1 mM 2-mercaptoethanol, 1 mM cysteine-HCI, 10% ethyl alcohol, and 10% methyl alcohol. These results suggested that cysteine and methionine residues might be located in or near the active site of the enzyme, while lysine, histidine, and serine residues might be indirectly involved in the enzyme activity.

Production of 1-Deoxynojirimycin by Streptomyces sp. SID9135

  • To increase the high production of 1-deoxynojirimycin (DNJ) from Streptomyces sp. SID9135, the effect of various carbon sources, nitrogen sources, cationic metal ions, the initial pH of the medium, and agitation speed were investigated. The most effective carbon and nitrogen sources were found to be lactose 2.5% (w/v) and soybean meal 2.0% (w/v), respectively. None of the cationic metal ions examined had any detectable stimulating effect on DNJ production except $Fe^{+2}$ ion. The initial optimum pH for DNJ production ranged from 6-8 and agitation speed was most effective at 400 rpm. In the jar fermentor experiments under optimal culture conditions, the accumulation of DNJ reached about $640{\mu}g$/ml after 5 days of cultivation and the level remained the same for a further two days.

  • Recently, $TiO_2$ nanotubes have considerably researched because of their novel application about photocatalysis, dye-sensitized solar cells (DSSCs), lithium ion battery, etc. In this work, self-standing $TiO_2$ nanotube arrays were fabricated by anodic oxidation method using pure Ti foil as a working electrode in ethylene glycole with 0.3M $NH_4F$ + $2%H_2O$. Growth behavior of $TiO_2$ nanotube arrays was compared according to temperature, voltage and time. The morphology, structure and crystalline of anodized $TiO_2$ nanotube arrays were observed by FE-SEM (field emission scanning electron microscope) and XRD (X-ray diffraction).

Dependence of Gas Pressure on Cr Oxide Thin Film Growth Using a Plasma Focus Device (플라즈마 포커스를 이용한 크롬 산화물 박막 성장의 분위기 기체 압력 의존성 연구)

  • Chromium oxide thin films have been deposited on silicon substrates using a tabletop 9kJ mathertyped plasma focus (PF) device. Before deposition, pinch behavior with gas pressure was observed. Strength of pinches was increased with increasing working pressure. Deposition was performed at room temperature as a function of working pressure between 50 and 1000 mTorr. Composition and surface morphology of the films were analyzed by Auger Electron Spectroscopy and Scanning Electron Microscope, respectively. Growth rates of the films were decreased with pressure. The oxide films were polycrystalline containing some impurities, Cu, Fe, C and revealed finer grain structure at lower pressure.

Hot Petroleum Drying Method to the Preparation of Multicomponent Oxide Ceramic Material (다성분계 산화물의 요업재료 제조를 위한 석유 증발 건조 방법)

  • As a wet chemical drying process "hot petroleum drying method" was applied and developed for preparing uniformly fine oxide powder with high purity and sinterreactivity. Using this method solution of sulfates was dried in hot petroleum bath (~17$0^{\circ}C$) to sulfate powder from which corresponding mullite doped by Fe3+ ion was formed. Particle size, shape, decomposition by heat, and phase identification of sulfate andoxide powders determined by DTA, TGA, X-ray diffraction, analysis and electron microscopy: sulfate powder prepared by this drying method is an intimate mixture of the amorphous form of uniformly and finely distributed spherical particles (0.05-0.1$\mu$). Mullitization with the sulfate powder occurs at 110$0^{\circ}C$ in air. The morphology of mullite particle made by firing the sulfate powder at 135$0^{\circ}C$ in oxygen atmosphere is granular of 0.1-0.3$\mu$ in size. This drying process proved to be a very effective method for preparing fine, homogeneous, and highly sinterreactive multicomponent oxide powder without conventional ceramic process of mixing, milling, and granulating. Catalytic oxidation kinetics of iron-containing carbon particles generated from diesel-sprayed hydrogen-air diffusion flame (디젤-분무 수소-공기 확산화염에서 생성된 철-함유 탄소입자의 촉매 산화반응 특성)

  • In this study, we devoted to kinetic measurement of the catalytic oxidation of iron-containing flame soot particles and better understanding the role of catalytic particles on carbon oxidation in particular at low temperature, targeting on autothermal regeneration of diesel particulate filter by diesel exhaust gas. Carbon-based Fe-containing particles generated by spraying ferrocene-doped diesel fuel in an oxy-hydrogen flame are tested and compared with a commercial carbon black powder for thermogravimetric analysis (TGA), secondary ion mass spectrometry (SIMS), Fourier-transform infrared spectroscopy (FTIR), Induced coupled plasma-Atomic emission spectroscopy (ICP-AES), and High-resolution transmission electron microscopy (HR-TEM). As a result, we found that a small amount of the ferrocene addition led to significant reductions in a on-set temperature and an activation energy of the carbon oxidation as well. An oxygenated surface complex forming at the particle surface could be thought as active species that would be readily consumed in particular at low temperature.

Isolation of Iron-Binding Peptides from Sunflower (Helianthus annuus L.) Seed Protein Hydrolysates (해바라기씨박 단백질 가수분해물로부터 철분 결합 펩타이드의 분리)

  • Proteins from sunflower seeds were hydrolyzed with Alcalase and Flavourzyme to isolate iron-binding peptides. The optimal hydrolysis conditions were determined. Hydrolysates were filtered under a 3 kDa membrane and iron-binding peptides separated from the hydrolysates using ion exchange and gel permeation chromatographic methods. A fraction with the highest iron-binding activity (Fe/peptide, 0.69), F22, was obtained. These results suggest that fractions isolated from sunflower seed protein hydrolysates can be applied toward the production of iron supplements.

Hydrochemical Characteristics and Changes by Rainfall in the Jungrang River (강우에 의한 중랑천의 수질 특성 변화 연구)

  • Effects of a rainfall event (July 28, 2005) on the hydrochemical characteristics of the Jungrang river, the biggest tributary of the Han river, was investigated. Significant spatial variations in the hydrochemical characteristics were observed. At JR2 location, concentrations of T-N and T-P were relatively low indicating occurrence of active oxidation in the stepped drop structure. At JR3 location, concentrations of Na, K, Cl, $NH_4-N$ and EC were elevated suggesting increased discharge from the nearby waste-water treatment plant and tributaries. The rain event diluted major dissolved ion concentrations in the river by 12~52%. The $NO_3-N$ levels were preserved during the rain then increased about twofold after rainfall, suggesting increased discharge of nitrate-contaminated groundwater. Heavy metals including Cd, Co, Cr, Cu and Pb were not detected in all water samples and the leachates from surface sediment samples. Concentrations of Fe, Mn, Al and Zn were below the Korean Drinking Water Guideline. Results of this study suggested that establishment of water-quality monitoring protocols describing temporal and spatial variations in parameters sensitive to rainfall events, relatively steady factors, and contaminant sources is required.
