Studies on the Chelating Agent-Impregnated Resins for the Adsorption and Separation of Metal Ions (Ⅰ). 8-Hydroxyquinoline-Impregnated Resins (금속이온 흡착 및 분리를 위한 킬레이트 시약-침윤수지에 관한 연구 (제1보). 8-Hydroxyquinoline-침윤수지)

  • The adsorption behavior of 8-hydroxyquinoline (8HQ) on Amberlite XAD-4 and-7 resins was investigated by measuring its distribution coefficients under various experimental conditions, such as shaking time, pH and concentration of methanol in the medium. The application of 8HQ-impregnated-XAD resins for the absorption and separation of metal ions was studied. The maximum adsorption of 8HQ on XAD resins was observed in the 30% methanol solution having pH range from 6.0 to 9.0. The impregnation capacities of XAD resins for 8HQ were 3.81${\times}$10-2mmol, 8HQ/g, XAD-4 resins and 2.60${\times}$10-2mmol, 8HQ/g, XAD-7 resin, respectively. The 8HQ-impregnated-XAD resins were stable in pH range from 6.0 to 10.0 and the amount of 8HQ leached from XAD-4 resin by eluting with hydrochloric acid(above 5M) was negligible. The optimum pH range for the adsorption of metal ions on 8HQ-impregnated XAD resin was also 6.0 to 10.0, and the adsorption mole ratio of metal ion to 8HQ were 1 : 2 for Cu(II), Cd(II) and Ni(II), and 1 : 3 for Fe(III) at the above pH range. It was found that the absorbed metal ions on 8HQ-impregnated-XAD resins were recovered quantitatively with 5M HCl and 8HQ-impregnated-XAD-4 resin could be reusable over 5 times without decrease in its impregnation capacity.

  • Carboxypeptidase B from Salmo Salar eggs was purified by CM-cellulose, 0.5 ammonium sulfate saturation, DEAE-cellulose, and Sephadex G-75 gel filtration and its enzymatic properties were investigated. Optimum temperature was 55$^{\circ}C$, pH optima were 4.0 and 7.0 at 37$^{\circ}C$, and the enzyme was stable at pH 2.0∼3.0 and 5.5∼7.0 for 1.5h. This enzyme showed substrate specificity hydrolyzing the peptide bond of glycyl-L-arginine. Its K$_m$ values was 0.21mM, and the enzyme activity was stimulated by Cu$^{2+}$ and Fe$^{3+}$, while inhibited by Zn$^{2+}$. The lysine was found to be competitive inhibitor and its K$_i$ value was determined to be 4.3mM. Molecular weight of this enzyme was determined to be 36,400 daltons by SDS-PAGE and the enzyme was monomeric protein composed of 19 kinds of amino acid residues.

Purification and Characterization of a Maltopentaose-producing Amylase from Bacillus megaterium KSM B-404. (Bacillus megaterium KSM B-404으로부터 생산되는 Maltopentaose생성 Amylase의 정제 및 특성)

  • An amylase that hydrolyzes starch into maltopentaose as a main product was found in the culture supernatant of a strain of Bacillus megaterium KSM B-404 isolated from local soil. The enzyme was purified 129-fold by ammonium sulfate precipitation, DEAE-Toyopearl and Superdex 75 HR 10/30 column using a FPLC system. The molecular weight of the amylase was determined as about 68 kDa by using SDS-PAGE. Optimum pH and temperature of amylase were found to be $50^{\circ}C$ and pH 6.0~7.0, respectively. The enzyme was stable up to $60^{\circ}C$ by addition of $Ca^{2+}$ and its pH stability was in the range of 6.0~10.0. The activity of enzyme was inhibited by $Cu^{2+}$ $Hg^{2+}$ , and $Fe^{3+}$ and maintained by $Ca^{2+}$ and $Mg^{2+}$ . EDTA and pCMB also showed inhibitory effect to the enzyme. TLC and HPLC analysis of the products of the enzyme reaction showed the presence of maltopentaose(52%), maltotriose (25%), maltose (11%), glucose, and maltotetraose in the starch hydrolysates.

Biochemical Properties of Eggplant Fruit Lectin. (가지 열매 lectin의 생화학적 성질)

  • Biochemical characterization including hemagglutination of erythrocytes, molecular weight, optimum temperature, thermal stability, optimum pH, carbohydrate specificity, and inhibitory effect of metal ion were studied in lectin of eggplant (Solanum melongena L.) fruit prepared by ammonium sulfate fractionation and affinity chromatography. This lectin was agglutinated by trypsin-treated rat blood erythrocyte. The molecular weight of this lectin by SDS-PAGE was estimated to be approximately 19.3 kDa of a single band. This lectin has no activity by 7 carbohydrates containing D-glucose. The optimum range of temperature and pH were $10-20^{\circ}C$ and pH 6.2-7.2, respectively. This lectin was relatively stable at $20-70^{\circ}C$. And the activity of this lectin was not inhibited by $Ca^{2+},\;Co^{2+},\;Cu^{2+},\;Fe^{2+},\;Mg^{2+}$, and $Mn^{2+}$.

  • Mitochondria in Pleurotus ostreatus was purified by stepped sucrose density gradient centrifugation. The activity of mitochondrial ATPase has been investigated during various times of illumination at each wavelength in the range of 400 nm to 700 nm. The mitochondrial ATPase activity was simulated 1,7 fold by 580 nm illumination compared with the broad wavelength group. The mitochondrial ATPase activity according to various times of illumination was stimulated 2.2 fold for 10 seconds at 580 nm compared with the broad wavelength group. The optimum pH and temperature of the mitochondrial ATPase were 7.4 and $60^{\circ}C$, respectively. The activity of this enzyme was stimulated by 5 mmol $Fe^{3+}$, 5 mmol $Mg^{2+}$, 0.1 mmol $Ca^{2+}$ and 5 mmol $Fe^{2+}$ ion, but inhibited by 5 mmol $Na^{+}$ ion.

Effects of pH and slow mixing conditions on heavy metal hydroxide precipitation (pH와 완속교반 조건에 따른 중금속 수산화물 화학침전 특성)

  • Conventional coagulation-gravity settling processes in heavy metal removal have a problem in coagulant cost and instability of the settling efficiency. The authors investigated the effects of pH and slow mixing conditions on heavy metal hydroxide precipitation and the particle size distribution of the precipitate for a precipitation-membrane separation process. The optimum pH values for the hydroxide precipitation ranged from 9 to 10. The addition of $FeCl_3$ did not enhance the heavy metal removal. 20 min of slow mixing at 70 rpm showed the maximum heavy metal removal to meet the water quality criteria for effluent discharge. More than 99.9% of the heavy metal precipitate particles were bigger than $2{\mu}m$.

  • Ninty four strains of lysine producing micro-organisms in culture broth during fermentation have been isolated from soil and other sources. From the comparison of the amounts of lysine produced, 6 strains have been selected as the potentially useful strains, and identified tentatively as Micrococcus sp. (S-16-4), Corynebactcrium sp. (S-27-12, S-281-3, CBY-4) and Brevibacterium sp. (M-6-71, F-629-2), respectively. From the further studies with Corynebacterium sp., S-27-12, its maximum yield was found to be 4mg lysine/ml of synthetic medium, consist of glucose(7.5%), urea(0.6%), $KH_2PO_4(0.2%)$, $Na_2HPO_4(0.05%)$, $MgSO_4{\cdot}7H_2O(0.03%)$, $MnSO_4{\cdot}4H_2O(0.001%)$ and $FeSO_4{\cdot}7H_2O(0.0005%)$ at pH 7.2 and $30^{\circ}C$ after 4 days.

Isolation of Dye-degrading Microbes for the Treatment of Dyeing Wastewater and Dye Decoloring (염색폐수 처리를 위한 미생물 선별 및 염료 탈색)

  • 87 microbes were isolated from dyeing wastewater collected at Dongducheon and Banweol industrial complex. Five microbes showed excellent ability of color removal and were identified as Shewanella putrefaciens, Aeromonas salmonicida(3 different strains), and Pseudomonas vesicularis. Five identified strains had optimal pH and optimal temperature as 7.0 and 30$^{\circ}C$ for cultivation, and showed morphological characteristics of Gram negative, oxidase negative, rod shape, and non-motility, but their biochemical characteristics were distinguishable. Each single strain of five microbes were tested in the 500 mL flask to treat dyeing wastewater, and achieved about 35% color removal efficiency in average. When two strains were selected and applied to the treatment at same time, color removal efficiency was increased up to 65%. While three or more associations of each strain did not show the improvement of color removal. Inhibition effects by $Mn^{2+}\;and\;Fe^{3+}$ on the dye degradation were tested and resulted in no effect under 70 ppm concentration.

Chemical Components and Physiological Activities of Young Mulberry(Morus alba) Stem (뽕나무 어린줄기의 화학성분 및 생리활성)

  • In order to exploit as a new functional decocted beverage, chemical components, antioxidative and antimicrobial activities young stems of mulberry(Morus alba) were surveyed. The proximate composition was composed of crude fiber 51.12%, ash 13.46%, total sugar 10.38%, crude fat 9.10% and crude protein 5.01%. The P(295.9 mg%) was the highest mineral found in young stem of mulberry and Ca 289.6 mg%, K 209.6 mg%, Na 58.3 mg%, Mg 45.0 mg% and Fe 4.6 mg% in that order. Free sugars was composed of glucose 1.08%, galactose 022%, sucrose 0.20% and fructose 0.16%. Eight fatty acids in stem of mulberry were identified and the major fatty acids were linoleic acid(46.10%), palmitic acid(27.84%) and linolenic acid(10.85%). Among the 17 amino acids detected, total amino acid was 2,450.5 mg% and proline(313.7 mg%) was the most predominant. Methanol extract and ethyl acetate fraction showed stronger activity of the hydrogen donating activities, each of 77.24% and 80.08%, respectively. The methanol extract from young stem of mulberry showed the strongest antimicrobial activities to Bacillus subtitis and Bacitus cereus. Chloroform and ethyl acetate fractions from methanol extract of young stem showed a inhibition zone of 9.0∼19.0 ㎜ in diameter against pathogen bacteria.

Magnetite Nanoparticles Containing Nanoporous Carbon for the Adsorption of Ibuprofen (마그네타이트 나노입자를 포함한 탄소나노세공체 합성과 아이부프로펜 흡착거동)

  • Preliminary studies on the synthesis of magnetic nanoparticles including nanoporous carbon materials have been done via a direct carbonization process from resol, ferric nitrate and triblock copolymer F127. The results show that the nanoporous magnetite/carbon ($Fe_3O_4$/carbon) with a low $Fe_3O_4$ content (1 wt%) possesses an ordered 2-D hexagonal (p6mm) structure, uniform nanopores (3.6 nm), high surface areas (up to 635 $m^2/g$) and pore volumes (up to 0.48 $cm^3/g$). Magnetite nanoparticles with a small particle size (10.2 nm) were confined in the matrix of amorphous carbon frameworks with superparamagnetic property (7.7 emu/g). The nanoporous magnetite/carbon showed maximum adsorption amount (995 mg/g) of ibuprofen after 24 h at room temperature. The nanoporous magnetite/carbon was separated from solution easily by using a magnet. The nanoporous magnetite/carbon material is a good adsorbent for hydrophobic organic drug molecules, i.e. ibuprofen.
