Dietary Aloe QDM Complex Reduces Obesity-Induced Insulin Resistance and Adipogenesis in Obese Mice Fed a High-Fat Diet

  • Obesity-induced disorders contribute to the development of metabolic diseases such as insulin resistance, fatty liver diseases, and type 2 diabetes (T2D). In this study, we evaluated whether the Aloe QDM complex could improve metabolic disorders related to blood glucose levels and insulin resistance. Male C57BL/6 obese mice fed a high-fat diet for 54 days received a supplement of Aloe QDM complex or pioglitazone (PGZ) or metformin (Met) and were compared with unsupplemented controls (high-fat diet; HFD) or mice fed a regular diet (RD). RT-PCR and western blot analysis were used to quantify the expression of obesity-induced inflammation. Dietary Aloe QDM complex lowered body weight, fasting blood glucose, plasma insulin, and leptin levels, and markedly reduced the impairment of glucose tolerance in obese mice. Also, Aloe QDM complex significantly enhanced plasma adiponectin levels and insulin sensitivity via AMPK activity in muscles. At the same time, Aloe QDM decreased the mRNA and protein of $PPAR{\gamma}/LXR{\alpha}$ and scavenger receptors in white adipose tissue (WAT). Dietary Aloe QDM complex reduces obesity-induced glucose tolerance not only by suppressing $PPAR{\gamma}/LXR{\alpha}$ but also by enhancing AMPK activity in the WAT and muscles, both of which are important peripheral tissues affecting insulin resistance. The Aloe QDM complex could be used as a nutritional intervention against T2D.

  • 1. 본 연구 조사대상자인 22명의 Type IIahypercholesterolemia 환자군에 있어서 혈중지질 농도에 영향을 미치는 여러가지 식이성분 섭취량 측정법을 비교했을 때, 24시간회상법은 식이섭취 기록법보다 대체로 높은 수치를 나타냈다. 2. 식품섭취기록기간에 따른 섭취량 측정을 비교할때에 7일간의 기록은 9일간의 기록에 근사한 수치(5% 이내)를 나타냈다. 3. 본 연구 조사대상자에 있어서 주말과 주중의 섭취량에는 큰 차이가 없었다. 단 P/S ratio 만은 주중과 주발을 비교할 때 유의적인 차이가 있었다. 4. 식품섭취기록기간에 따른 섭취량 측정에서는 남녀의 차이가 현저하였다. 여자의 경우, 4일의 기록표는 9일과 5%이내의 차이를 보였으나, 남자의 경우, 4일의 기록표는 9일과 12%의 차이가 있었다. 주중과 주말을 비교할 때에도 남녀의 차이가 있었다. 5. 본 연구에서 분석한 여러 식이성분 중 그 섭취량에 가장 변동이 심한성분은 P/S ratio와 알콜이었다.

Formula Optimization of a Perilla-canola Oil (O/W) Emulsion and Its Potential Application as an Animal Fat Replacer in Meat Emulsion

  • The formulation of an oil/water (o/w) emulsion made up of a mixture of perilla oil and canola oil (30/70 w/w) was optimized using a response surface methodology to find a replacement for animal fat in an emulsion-type meat product. A 12 run Plackett-Burman design (PBD) was applied to screen the effect of potential ingredients in the (o/w) emulsion, including polyglycerol polyricinoleate (PGPR), fish gelatin, soy protein isolate (SPI), sodium caseinate, carrageenan (CR), inulin (IN) and sodium tripolyphosphate. The PBD showed that SPI, CR and IN showed promise but required further optimization, and other ingredients did not affect the technological properties of the (o/w) emulsion. The PBD also showed that PGPR played a critical role in inhibiting an emulsion break. The level of PGPR was then fixed at 3.2% (w/w total emulsion) for an optimization study. A central composite design (CCD) was applied to optimize the addition levels of SPI, CR or IN in an (o/w) emulsion and to observe their effects on emulsion stability, cooking loss and the textural properties of a cooked meat emulsion. Significant interactions between SPI and CR increased the cooking loss in the meat emulsion. In contrast, IN showed interactions with SPI leading to a reduction in cooking loss. Thus, CR was also removed from the formulation. After optimization, the level of SPI (4.48% w/w) and IN (14% w/w) was validated, leading to a perilla-canola oil (o/w) emulsion with the ability to replace animal fat in an emulsion-type meat products.

Monoglyceride의 지방산(脂肪酸) 조성(組成)이 Ice Cream 물성(物性)에 미치는 영향 (Effect of Fatty Acid Compositions by Monoglyceride on Rheological Properties of Ice Cream)

  • Ice cream용 유화제로 사용되는 monoglyceride의 지방산 조성에 따른 ice cream의 물성에 미치는 영향에 대해 검토하였다. 일반적으로 monolaurin부터 monostearin의 포화지방산 monoglyceride에는 각기 성질에 큰 차이가 없었으나 monocaprin및 불포화지방산인 monoolein에서는 ice cream의 점도가 현저히 상승되어 적합한 overrun과 보형성이 양호한 ice cream이 만들어지며 이의 원인은 지방구의 응집 때문으로 확인되었다. 한편 포화지방산 monoglyceride는 overrun이 freezing time의 초기에 발생되기 쉽지만 시간이 경과함에 따라 차츰 저하되는 반면 불포화지방산 monoglyceride는 액상 또는 저융점으로 인해 기름에 대한 융해성이 커짐으로써 overrun이 초기에는 어려우나 freezing time이 경과함에 따라 지방구가 입체적인 망상구조를 이룰뿐 아니라 지방구의 응집을 나타냄으로써 overrun이 개선되며 ice cream에 보형성을 증진시키는 결과를 얻었다. 또한 monostearin과 monoolein의 비율에 있어서는 30:70의 부근에서 지방구의 응집이 극대점을 나타내고 있음을 확인하였다.

Estimating excess post-exercise oxygen consumption using multiple linear regression in healthy Korean adults: a pilot study

  • [Purpose] This pilot study aimed to develop a regression model to estimate the excess post-exercise oxygen consumption (EPOC) of Korean adults using various easy-to-measure dependent variables. [Methods] The EPOC and dependent variables for its estimation (e.g., sex, age, height, weight, body mass index, fat-free mass [FFM], fat mass, % body fat, and heart rate_sum [HR_sum]) were measured in 75 healthy adults (31 males, 44 females). Statistical analysis was performed to develop an EPOC estimation regression model using the stepwise regression method. [Results] We confirmed that FFM and HR_sum were important variables in the EPOC regression models of various exercise types. The explanatory power and standard errors of estimates (SEE) for EPOC of each exercise type were as follows: the continuous exercise (CEx) regression model was 86.3% (R2) and 85.9% (adjusted R2), and the mean SEE was 11.73 kcal, interval exercise (IEx) regression model was 83.1% (R2) and 82.6% (adjusted R2), while the mean SEE was 13.68 kcal, and the accumulation of short-duration exercise (AEx) regression models was 91.3% (R2) and 91.0% (adjusted R2), while the mean SEE was 27.71 kcal. There was no significant difference between the measured EPOC using a metabolic gas analyzer and the predicted EPOC for each exercise type. [Conclusion] This pilot study developed a regression model to estimate EPOC in healthy Korean adults. The regression model was as follows: CEx = -37.128 + 1.003 × (FFM) + 0.016 × (HR_sum), IEx = -49.265 + 1.442 × (FFM) + 0.013 × (HR_sum), and AEx = -100.942 + 2.209 × (FFM) + 0.020 × (HR_sum).

New Approaches to Production of Turkish-type Dry-cured Meat Product "Pastirma": Salt Reduction and Different Drying Techniques

  • In this study, the possible changes in the quality characteristics of pastirma, Turkish-type dry-cured meat product, produced by using two different salts (NaCl-KCl) in a curing mixture and two different production techniques (natural and controlled condition) were examined. Moisture, pH, salt, sodium, potassium, TBA, fat, water activity, instrumental colour, texture, and sensory analyses were implemented in order to determine the possible effects of these applications. Fat, aw, pH, colour, tiobarbituric acid (TBA), texture, salt, Na and K values may allow these desired modifications in pastirma production to be limited. The substitution of 15% KCl instead of NaCl was acceptable in terms of the sensorial properties of the pastirma. However, the sensory analyses did not allow for using a higher KCl instead of NaCl because both the hardness and chewiness in the texture of the pastirma samples salted with 30% of KCl were not scored positively. Besides this, negative effects, which may occur during the pastirma production under natural conditions, can be eliminated by the production being under controlled conditions.

Effects of Purslane Extract on Obesity and Diabetes in High-Fat Diet-Induced Obese Mice

  • The frequency of obesity has risen dramatically in recent years but only few safe and effective drugs are currently available. In addition, obesity can induce type 2 diabetes (T2DM), hyperlipidemia and fatty liver disease. Recently, protective effect of purslane extract (PE) on obesity has been reported, but little is known about the role and mechanism of PE in obesity. This study aimed to evaluate the effect of PE on obesity and diabetes in obese mice. In addition, the effect of PE was compared with anti-obesity and diabetes drugs. High-fat diet (HFD)-induced obese mice were treated for 8 weeks with drugs as follows: PE, orlistat, metformin, voglibose or pioglitazone. While PE mixed with normal diet did not have any effects on BW in non-obese mice, PE mixed with HFD significantly reduced BW gain, insulin resistance, and glucose intolerance, without affecting food intake and appetite in obese mice. The effect was comparable to the effects of anti-obesity and diabetes drugs. Furthermore, PE significantly increased the activity of hepatocellular anti-oxidant enzymes, leading to protection of liver from oxidative stress in obese mice. These results suggest that PE treatment may be a useful tool for preventing obesity and complication of obesity.

여성 비만의 유발유형별 일반 증상과 검사 특성 연구 (Characterizing of Four Obesity Types in Obese Women Based on the Questionnaire of Diseases and Physical Tests)

  • Objectives : To characterize four types of obesity and to effectively improve the treatment of obesity through Oriental medicine Methods : At 00 Oriental Medical Center, 203 female subjects who intented to lose weight were requested to complete a questionnaire. These participants were also given physical tests. The Questionnaire consisted of questions both about general life style and obesity in oriental medicine framework. The physical tests were blood tests, a body composition via Inbody 2.0, and physical strength tests. One-way analysis of variance was done to compare the means of items and physical tests between four types of obesity. Duncan was used by post hoc test. Results : 1. Significant differences between obesity type III and obesity type IV in ever tried to lose weight, childhood obesity and excercise times were observed in the questionnaire of general life style(p<0.05). 2. Ducan test showed significant differences between four obesity types in diseases (p<0.05). 3. Significant differences between four obesity types in height, % body fat muscular endurance, soft lean mass, fat mass, Trigliceride, Total cholesterol and ${\gamma}-GTP$ were observed (p<0.05). Conclusions : Further clinical research is necessary in the four types of obesity explored. The diagnosis and treatment based on these types should be further studied.

지수치를 이용한 노년 여성의 하반신 체형 유형화에 관한 연구 (Lower Body Somatotype Classification and Discrimination of Elderly Women According to Index)

  • The purpose of this study is to provide the basic data on the development of ready-to-wear clothing for the elderly women as the population of the elderly has been constantly increasing as well as the purchasing power of the aged. The body measurements of 318 elderly women were taken. whose ages were over 60 years and enrolled in colleges for the elderly. sports centers. or business sites in Seoul and the neighboring districts. A total of 39 features in the lower body were used for the anthropometric measurement and analysis. The results of the study are as follows: 1. Indices of height and weight were used for factor analysis. cluster analysis, and discriminant analysis in order to 'classify lower body somatotype according to shape, excluding size factors. From the results of the factor analysis. the 5 factors showed the cumulative sum of square at 75.63%. 2. Somatotype were classified into two types according to a cluster analysis using height and weight dices. Type 1 is the group is relatively tall and has somewhat fat lower limbs. Type 2 is considered fat and has obesity factors around waist and abdomen area. The hit rate for the classified two groups showed the result at 95.9%.

질려자 추출물의 피부 볼륨 증진 및 주름개선 효과 (Skin Volume Augmentation and Anti-wrinkle Effects of Tribulus terrestris Fruit Extract)

  • Aging of the face is mainly related to the features that are sagging or loss of elasticity of the skin by reducing the volume around the eyes or cheek. Intrinsic aging can be seen to cause thinner dermis, reduction of extracellular matrix and subcutaneous fat. This study was carried out to investigate the skin volume augmentation and anti-wrinkle effects of Tribulus terrestris fruit extract. Skin anti-aging effect of Tribulus terrestris fruit extract was evaluated by using lipid accumulation, expressin of type I procollagen and elastin in preadipocytes and human dermal fibroblasts. Tribulus terrestris fruit extract augmented preadipocytes differentiation about 56% at 100 µg/mL. The type I procollagen and elastin were increased about 35% and 25% by treatment 20% Tribulus terrestris fruit extract, respectively. The clinical study also showed that skin sagging, skin elasticity, and dermal density improved without adverse effect following 4 week application of cream containing 2% Tribulus terrestris fruit extract. We suggest that Tribulus terrestris fruit extract can have the good possibility as skin volume augmenting, skin elasticity and wrinkle improving agent.
