A FAMILY-BASED AND CASE-CONTROL ASSOCIATION STUDY OF THE DOPAMINE BETA HYDROXYLASE GENE POLYMORPHISM IN ATTENTION DEFICIT HYPERACTIVITY DISORDER (한국인 주의력결핍 과잉행동장애와 Dopamine Beta Hydroxylase 유전자의 관련성 : 가족기반 연구 및 환자-대조군 연구)

  • Objective : Attention deficit hyperactivity disorder (ADHD) is the most common childhood psychiatric disorder, affecting $3-5\%$ of school age children. Although the biological basis of ADHD is unknown, family studies provide strong evidence that ADHD has a genetic basis. Recent genetic studies have suggested associations between ADHD and Taq I polymorphism of dopamine beta hydroxylase gene(DBH) . The aim of this study is to test the association between ADHD and Taq I polymorphism of DBH in Korean population. Method : We processed DNA extraction and genotyping for 106 korean children with ADHD and their parents. Genotyping was additionally performed for 212 age and gender matched normal controls. Case-control association study was applied. And we tested the association using the transmission disequilibrium test (TDT) and haplotype-based haplotype relative risk test (HHRR). Results : There were no statistical differences of genotype distributions between cases and controls. However, we did observe preferential transmission of allele Al of DBH Taq T polymorphism in ADHD. Conclusion : On the whole, our results lend credence to the notion that the relationship between ADHD and DBH is complex. The number of cases and informative transmissions were small, therefore it would be premature to make any conclusions from our study concerning the role of DBH in ADHD. Further work is needed to support these findings.

Fish Fauna and Community Analysis in Heuck Stream Watershed (흑천수계의 어류상 및 군집분석)

  • This study was conducted at 25 sites of 6 mainstreams and 19 tributaries sites within Heuck Stream watershed during May~October 2003 for the survey of fish distributions, compositions, and community characteristics. The survey showed that total fish was identified as 9 family and 26 species in all sites, and Cyprinidae dominated the community as 15 species. The dominant species of >20% of the total were Zacco temminckii (29%), Zacco platypus (22%), and Rhynchocypris oxycephalus (21%) in the watershed. Community analysis, based on the stream spatial gradients, indicated composition differences along the main axis of the stream from the headwater to the downstream; R. oxycephalus predominated in the most headwater zone and Z. temminckii dominated in the lower headwater zone, whereas Z. temminckii -Z. platypus dominated in the mid-to-downstream, and Z. platypus dominated the community in the most downstream zone. Total endemic species was 7 family and 15 species, which is made of 50% in the fish community, so that the high proportion of endemic species indicated a healthy region in terms of fish community, compared to average 23% in Korean peninsula in general. The total number and species of fish increased as the stream order (stream size) increased, indicating that impacts on chemical pollution or habitat disturbance were not so large to the fish community in this watershed. Trophic and tolerance guilds analysis showed that relative proportions of sensitive and insectivore species were >50% in the watershed and decreased as the stream order increased, whereas relative proportions of tolerant and omnivore species showed an opposite results. These outcomes suggest that the natural condition of watershed is preserved relatively and the region should be protected from the chemical and habitat disturbace by agricultural activity and urban developments.

Relation of Various Parameters Used to Estimate Cardiac Vagal Activity and Validity of pNN50 in Anesthetized Humans

  • Objectives: Analysis of heart rate variability (HRV) has been used as a measure of cardiac autonomic function. According to the pNN50 statistic, the percentage of differences between successive normal RR intervals (RRI) that exceed 50 ms, has been known to reflect cardiac vagal modulation. Relatively little is known about the validity of pNN50 during general anesthesia (GA). Therefore, we evaluated the correlation of pNN50 with other variables such as HF, RMSSD, SD1 of HRV reflecting the vagal tone, and examined the validity of pNN50 in anesthetized patients. Methods: We assessed changes in RRI, pNN50, root mean square of successive differences of RRI (RMSSD), high frequency (HF) and standard deviation 1 (SD1) of $Poincar{\acute{e}}$ plots after GA using sevoflurane anesthesia. We also calculated the probability distributions for the family of pNNx statistics (x: 2-50 ms). Results: All HRV variables were significantly decreased during GA. HF power was not correlated with pNN50 during GA (r = 0.096, P = 0.392). Less than pNN47 was shown to have a correlation with other variables. Conclusions: These data suggest that pNN50 can not reflect the level of vagal tone during GA.

  • Attempts have been made to define new classes of distributions that provide more flexibility for modelling skewed data in practice. In this work we define a new extension of the generalized gamma distribution (Stacy, The Annals of Mathematical Statistics, 33, 1187-1192, 1962) for Marshall-Olkin generalized gamma (MOGG) distribution, based on the generator pioneered by Marshall and Olkin (Biometrika, 84, 641-652, 1997). This new lifetime model is very flexible including twenty one special models. The main advantage of the new family relies on the fact that practitioners will have a quite flexible distribution to fit real data from several fields, such as engineering, hydrology and survival analysis. Further, we also define a MOGG mixture model, a modification of the MOGG distribution for analyzing lifetime data in presence of cure fraction. This proposed model can be seen as a model of competing causes, where the parameter associated with the Marshall-Olkin distribution controls the activation mechanism of the latent risks (Cooner et al., Statistical Methods in Medical Research, 15, 307-324, 2006). The asymptotic properties of the maximum likelihood estimation approach of the parameters of the model are evaluated by means of simulation studies. The proposed distribution is fitted to two real data sets, one arising from measuring the strength of fibers and the other on melanoma data.

Association Study between Tic Disorder and Dopamine D2 Receptor Gene Polymorphism in Korean Population (틱장애와 도파민 D2 수용체 유전자와의 연합 연구)

  • Objectives : The purpose of the present study was to investigate whether the TaqI A polymorphism of dopamine receptor D2 gene(DRD2) is associated with Tourette syndrome(TS) and chronic motor tic disorder(CMT) in Korean population. Methods : DRD2 TaqI A RFLP genotyping was carried out with DNA extracted from blood samples of 75 patients with tic disorders(47 with TS and 28 with CMT) and 90 healthy subjects. Genotype and allelic frequencies for the DRD2 gene polymorphisms of the tic disorder group as a whole were compared to those of the control group. Separating the TS group, thereafter, the frequency of genotypes and alleles were compared to those of the controls. Results : The results demonstrated that genotype and allele distributions for the DRD2 gene polymorphism in the tic disorder as a whole, TS, and control groups were not significantly different. Conclusion : No association was found for DRD2 gene, TS and CMT. The data suggest that DRD2 gene may not be a useful marker for the prediction of the susceptibility of tic disorder.

Effect of Xeroderma Pigmentosum Complementation Group F Polymorphisms on Gastric Cancer Risk and Associations with H.pylori Infection

  • We conducted a hospital case-control study by genotyping four potential functional single nucleotide polymorphisms (SNPs) to assess the association of Xeroderma pigmentosum complementation group F (XPF) with gastric cancer susceptibility, and role of XPF polymorphisms in combination with H.pylori infection in risk definition. A total of 331 patients with gastric cancer and 355 controls were collected. Four SNPs of XPF, rs180067, rs1799801, rs2276466 and rs744154, were genotyped by Taqman real-time PCR method with a 7900 HT sequence detector system. The gastric cancer patients were more likely to have smoking habit, a family history of cancer and H.pylori infection. We did not find any significant difference in the genotype distributions of XPF rs180067, rs1799801, rs2276466 and rs744154 between cases and controls. However, multivariate logistic analysis showed a non-significant decreased risk in patients carrying rs180067 G allele, rs1799801 T allele or rs2276466 T allele genotypes. A non-significant increased risk of gastric cancer was found in individuals carrying the rs744154 GG genotype. Stratification by H.pylori infection and smoking was not significantly different in polymorphisms of XPF rs180067, rs1799801, rs2276466 and rs744154. The four XPF SNPs did not show significant interaction with H.pylori infection and smoking status (P for interaction was 0.35 and 0.18, respectively). Our study indicated that polymorphisms in rs180067, rs1799801, rs2276466 and rs744154 may affect the risk of gastric cancer but further large sample size studies are needed to validate any association.

A Herbological Study on the Plants of Verbenaceae in Korea (한국산 마편초과 식물에 관한 본초학적 연구)

  • Objective : For the purpose of developing Verbenaceae plants recorded in Korean Herbology in Korea, the literatures of the successive generations have been thoroughly investigated to prepare this article. Methods : Books and research papers about Herbology which published at Korea and other countries were examined. Results : A list was made about Verbenaceae plant which were cultivated or grew naturally in Korea, after investigated the data on domestic and foreign Verbenaceae plants. Out of those lists, serviceable plants were selected and with those available plants, their distributions were analysed and parts which can be used as medicines were divided into 3 categories such as oriental medicine name, scientific name and non-official name. Verbenaceae's properties flavor, channels they use, effects, and toxicity were also noted. Verbenaceae(grew in Korea) were divided into 5 classes with 21 species. Out of those, 5 classes with 8 species were found serviceable which indicates 40% of all. Out of all 20 species of Verbenaceae family, Callicarpa genus were found 10 species, which were shown the most. Among 3 genus of Vitex species, medicinal plants were all one species each. Out of all serviceable parts in Verbenaceae, Folium parts took first place as 5 species. There are no toxic, the survey said. Conclusion : There were totaled to 5 genera and 21 species in Verbenaceae in Korea and among them medicinal plants are 5 genera, 8 species, some 38% in total.

A rare duodichogamous flowering system in monoecious Toona sinensis (Meliaceae)

  • Background: Duodichogamy is the rarest form of dichogamy in angiosperms, which is characterized by flowering in the sequence of male${\rightarrow}$female${\rightarrow}$male. Disentangling factors promoting duodichogamy require the discovery of more duodichogamous species in angiosperms. However, extremely limited information on duodichogamous species makes it difficult to make general conclusions. Given the inflorescence morphology and flowering characteristics, the Meliaceae family is highly likely to contain duodichogamous species. Methods: We selected 48 individuals from 20 populations in Korea and investigated their flower morphology, arrangement of flowers by sexual condition within inflorescences, and flowering phases and duration of male and female flowers of Toona sinensis (Meliaceae) for 5 years (2011-2015) to determine if the species shows duodichogamous flowering. Results: Toona sinensis belonging to Meliaceae possessed functionally unisexual flowers with rudimentary parts of the opposite sex. The floral organs in male were larger than those in female, except for ovary length and width. In dichasium, male flowers were observed on primary or lateral branches, whereas female flowers were borne only on lateral branches. Overall, individuals from six different populations flowered in the male${\rightarrow}$female${\rightarrow}$male sequence, thereby male is blooming far longer than female flowers at the level of individual trees (male vs. female = 17-20 days vs. 2-4 days). Conclusions: This is the first study to report a duodichogamously flowering species, T. sinensis, within Meliaceae. Several flowering characteristics observed from T. sinensis may be important clues used to discover additional duodichogamous Meliaceae species. Short flowering period and relatively small number of female flowers, which is analogous to reduced ovule numbers observed in other duodichogamous species, may intensify male-male competition in T. sinensis. This study contributed to narrowing down potential candidates of duodichogamy based on their geographic distributions and flowering time.

Isolation, Molecular Phylogeny, and Tissue Distribution of Four cDNAs Encoding the Apolipoprotein Multigene Family in Barred Knifejaw, Oplegnathus fasciatus (Teleostei, Perciformes)

  • Lipoproteins are complexes of lipids and specific apolipoproteins that are involved in lipid transport and redistribution among various tissues. In this study, we isolated full-length apolipoprotein cDNA sequences encoding apolipoprotein A-I (apoA-I), apoE, apoC-II, and apo-14 kDa in barred knifejaw, Oplegnathus fasciatus. In addition, we reconstructed phylogenetic trees and investigated mRNA tissue distributions. Alignment analyses of amino acid sequences revealed that secondary structures of the polypeptides apoA-I, apoE, and apoC-II in barred knifejaw are well conserved with their teleostean and mammalian counterparts in terms of characteristic tandem repetitive units forming amphipathic ${\alpha}$-helices. Both the sequence alignment data and cleavage sites of apo-14 kDa indicated a clear differentiation between Percomorpha and Cypriniformes. Meanwhile, the phylogenetic trees of apolipoprotein sub-families suggested that the common ancestor prior to the split of the Actinopterygii (ray-finned fishes) and Sarcopterygii (tetrapods) would have possessed the primordial protein-encoding genes. Tissue distribution of each apolipoprotein transcript determined by semi-quantitative RTPCR showed that barred knifejaw apoA-I transcripts were more or less ubiquitously expressed in the liver, intestines, brain, muscle, spleen, and kidney. The most striking difference from previous observations on barred knifejaw was the ubiquitous expression of apoE across all somatic tissues. Barred knifejaw apoC-II showed tissue-specific expression in the liver and intestines, while the liver and brain were the major sites of apo-14kDa mRNA synthesis.

Body Mass Index Distributions and Sociodemographic Factors Affecting BMI of Children Living in Anyang, Korean

  • Body mass index(BMI) is recognized as one of the most useful indexes for adiposity in children and adults. This study was conducted to provide information on BMI distribution by age and sex in Korean children, and to identify sociodemographic factors that affected BMI among children. The study was conducted on 2376 children(boys 1184, girls 1192) living in a middle-size city in Korea. Subjects were selected from the primary schoolchildren in grades 1-6 using the two-stage sampling method. Their mean age was 9.4 years. The percentile of BMI tended to increase as age increased in both sexes. The 85th percentile for boys, aged 6-12 years, was 19.8kg/㎡, and the 95th percentile was 22.5kg/㎡. The 85th percentile for girls, aged 6-12 years, was 18.9kg/㎡, and the 95th percentile was 21.5kg/㎡. Multiple regression analysis was carried out to predict BMI from percent ideal body weight (PIBW), age and sex. After adjusting PIBW and age, BMI for boys was 0.062kg/㎡ lower than that for girls. One year of age increased BIM by 0.55kg/㎡. The prevalence rate of overweight and obesity based on PIBW was 15.8% and 15.0% for boys, and 14.0% and 11.0% for girls, respectively. Demographic factors such as sex, age, and parents' obesity influenced children's BMI. After adjusting for sex, age and parents' BMI, the presence of another children in family, and mother's employment status showed a strong effect on children's BMI. The results suggest an age-sex specific BMI distribution of Korean children. The present study also provides direct evidence of a correlation between early life environmental factors, such as presence of siblings or mother's employment, and BMI level in Korean children.
