Development of Simple Benthic Macroinvertebrates Index (SBMI) for Biological Assessment on Stream Environment (하천환경의 생물학적 평가를 위한 간이저서동물지수(SBMI)의 개발)

  • GPI (Group Pollution Index) using 29 indicator groups of Korean benthic macroinvertebrates was proposed in 1992, a higher category taxa-level index developed for rapid field assessment of organic water pollution. This study was performed to revise the assessment scheme of GPI based on taxonomic performance and ecological information accumulated since 1992. The original GPI was renamed SBMI (Simple Benthic Macroinvertebrates Index), and SBMI was based on saprobic valency of 26 indicator groups composed of higher category taxa (mainly family ~ phylum) excluding some genus or species-level taxa. SBMI revealed highly significant correlation with concentration of 5-day biochemical oxygen demand ($BOD_5$) (correlation coefficient r = 0.78, n = 569 sites), total suspended solids (r = 0.69), and total phosphorus (r = 0.77). Also, SBMI revealed strong correlation with Shannon-Weaver's species diversity (r = -0.85), Margalef's species richness (r = -0.85), and McNaughton's dominance (r = 0.83). Determination coefficient of SBMI to concentration of water quality items and values of community indices such as species diversity was 3 ~ 8 % and approximately 11 ~ 14 % higher than that of GPI, respectively. Correlation between SBMI and water quality factors or community indices such as species diversity did not reveal much difference compared to that of species-level indices, such as BMI (Benthic Macroinvertebrates Index) and ESB (Ecological Score of Benthic Macroinvertebrates). SMBI is a simple-qualitative index with higher category taxa easily identified, and is applicable for rapid field assessment of water environment impairment.

  • This study analyzes the attitude toward new products through case studies. The contents of the case study applied to the study were analyzed about the attitude of the new product to the recently screened movie "Sanguiwon(the royal tailor)". The court was an institution that oversees the clothes of the royal family in the Chosun Dynasty. In a conservative and closed market environment, attitudes toward acceptance of new products are strongly influenced by internal and external factors. Authoritative and conservative attitudes to secure competitive advantage in existing markets ultimately hamper the acceptance of new products or cause externalities. Eventually, innovative resistance to new products emerges. On the other hand, the acceptance of new products in an open market will form an attitude toward new products, once they form a respectful attitude towards their own benefit or attitude. These market leaders, depending on the way they pursue their values, form an attitude to adhere to their products or accept innovation. The validity of this study was verified through other cases. Ultimately, success in the market for new products suggests that success can only be achieved by understanding the delivery of value to consumers.

Heterologous Expression and Characterization of a Thermostable Exo-β-D-Glucosaminidase from Aspergillus oryzae

  • An exo-β-D-glucosaminidase (AorCsxA) from Aspergillus oryzae FL402 was heterologously expressed and purified. The deduced amino acid sequence indicated that AorCsxA belonged to glycoside hydrolase family 2. AorCsxA digested colloid chitosan into glucosamine but not into chitosan oligosaccharides, demonstrating exo-β-D-glucosaminidase (CsxA) activity. AorCsxA exhibited optimal activity at pH 5.5 and 50℃; however, the enzyme expressed in Pichia pastoris (PpAorCsxA) showed much stronger thermostability at 50℃ than that expressed in Escherichia coli (EcAorCsxA), which may be related to glycosylation. AorCsxA activity was inhibited by EDTA and most of the tested metal ions. A single amino acid mutation (F769W) in AorCsxA significantly enhanced the specific activity and hydrolysis velocity as revealed by comparison of Vmax and kcat values with those of the wild-type enzyme. The three-dimensional structure suggested the tightened pocket at the active site of F769W enabled efficient substrate binding. The AorCsxA gene was heterologously expressed in P. pastoris, and one transformant was found to produce 222 U/ml activity during the high-cell-density fermentation. This AorCsxA-overexpressing P. pastoris strain is feasible for large-scale production of AorCsxA.

A Study on Historical Characteristics and Modern Trend of Torajan Traditional Housing in Indonesia (인도네시아 또라자 전통주거의 역사적 특성과 현대적 양상에 관한 연구)

  • The purpose of this study is to investigate the basic historical characteristics and its modern trend of Torajan traditional housing architecture in Sulawesi Province, Indonesia. The Toraja culture belongs to the cosmological culture with Cosmos centric characteristics. A traditional house, being called 'tongkonan' in Toraja region, is more than just a structure, representing the symbol of family identity and tradition. The Torajan architecture is a combination of the myth and cosmos, also regional conditions. With a short description of the general conditions and spiritual values of the Toraja, this paper explains the space-composition, the stylistic characteristics, the ornamental elements, construction, and its modern trend, etc. In general, it is raised on stilts several metres high, with a dramatically boat-shaped roof. Village layout varies according to size. The houses are arranged in a row, side by side, with their front gables facing north. Each house stands opposite its own rice-barn. The houses with their oblong ground-plans, built on piles set on stones. The interior is divided into three or four rooms, having few window. The houses are embellished with carving and paintings, and the facades display engraved and painted geometric and figural designs. The most frequent motif is the buffalo head, ranging from the realistic to the highly stylized. The Torajan traditional housing have experienced radical changes during the Modern period. In spite of the popularity of new modern house-styles, the traditional architectural style is often now constructed as an icon of Toraja identity. This paper will be helpful for understanding regional diversity of the traditional housing in Southeast Asia.

Study on Clothing Life of Korea-Vietnam Multicultural Families - Focus on clothing behaviors and attitudes toward traditional dresses - (베트남 다문화가정 의생활 실태조사 - 의복행동과 전통복식 태도를 중심으로 -)

  • This research is designed to provide basic data to study the life and culture of multicultural families in Korea by taking a look at the clothing behaviors and attitudes toward traditional dresses. To this end, quantitative research was conducted on Korea-Vietnam multicultural families and Korean families. The surveys of the Korean families were completed by 250 married women in Seoul, and those of Korea-Vietnam multicultural families by 104 married Vietnamese women living in Seoul, Gyeonggi-do and Incheon. The data were analyzed using frequency analysis, factor analysis, t-test, crosstabs and ${\chi}^2$-test. The findings are as follows: First, the comparison of clothing behaviors of the Korean families and the Korea-Vietnam multicultural families found that they had statistically significant differences in values towards clothes, clothes shopping orientation and clothes purchasing behaviors. The Korean women were more involved in clothes and fashion-oriented than their Vietnamese counterparts. However, the Vietnamese women in their 20s were likely to rely more on social trends than their own needs when purchasing clothes compared to their Korean counterparts. Korean families preferred to shop in department stores, while the multicultural families relied more on discount stores and outlets. Second, the comparison of the Korean families and the Korea-Vietnam families in attitudes toward their own traditional dresses and how often to wear them showed statistically meaningful differences. The Korean group had more negative attitudes toward Hanbok, the Korean traditional dresses than the Vietnamese group. The Vietnamese women showed more interest in information on Hanbok than their Korean counterparts. They also were wearing the Korean traditional dresses more often than the Korean families. In addition, the Vietnamese women showed a stronger tendency than the Korean women that they took pride in their country's traditional dresses and believed that they were beautiful.

  • Dolbok, the first birthday suit, is a formal suit a person puts on for the first time after birth. It not only a symbolizes of Korean customs or traditions, but also represents transmitted ideas, values and lifestyles of Koreans. In this nation where strong preferences for son prevailed, dolbok for boy was specially important. 1.Styles of weaing Dolbok are classfied into trousers and jeogori, baeja, magoja, durumagi, jeonbok, sagyusam and yongpo styles in accordance with what kind of clothes was finally worn. 2. Accessories of dolbok for boys included dolddi, dol jumeoni, gulre, bokgeon, hogeon and tarae baeseon. 3. Dolbok of trousers and jeogori, baeja or magoja style simply consisted of 2 to 4 units of clothes. It was usually worn by ordinary people, and strongly imaged as common. Dolbok of durumagi or jeonbok style was a combination of 5 to 6 units of clothes and mainly worn by noble and royal families. It was remarkably formal, but quite brilliant. The suit for first birth anniversary which was sagyusam or yongpo was made in accordance with court rules and used by the royal family, It consisted of 7 to 8 units of clothes. Dolbok of sagyusam or yongpo style used invariable, simple patterns and colors, but had strong images of courtesy and dignity. 4. Regarding accessories of dolbok, the long-hanged goreum was a symbol of parental wishes for the $baby^{\circ}{\emptyset}s$ health and longevity and dolddi was joined by dol jumeonisymbolizing the wealth and prosperity of the baby. In addition, hogeon was a hat whose shape was the head of tiger symbolizing wishes for bravery and health and tarae beoseon was childlen's socks that had seams, symbolizing prosperity, on the fore edge. Seon for sagyusam was used as a symbol of preventing diseases or evil spirits to protect the health of the baby.

Docking Studies of Camptothecin Analogues into Human Topoisomerase I-DNA Complex (Camptothecin 유도체의 Human Topoisomerase I-DNA 복합체에 대한 Docking 연구)

  • Human topoisomerase I (Topo I) plays a pivotal role in cell replication, transcription and repair and, therefore, is an important anti-cancer target. 20S-camptothecin (CPT) is a representative Topo I inhibitor. Compounds belonging to CPT family inhibit the religation step of Topo I-DNA by binding to the DNA cleavage site. Computational docking studies with Surflex-Dock were carried out to investigate the binding modes between Topo I-DNA binary complex structure and the ligand such as 20S-CPT and 9,10-substituted 20S-CPT analogues. The docking results demonstrated that most of the compounds with $IC_{50}$ value under $0.5{\mu}M$ intercalated exactly between the -1 and +1 DNA bases, deeply toward the cleavage site. The complex was stabilized by hydrogen-bonding and hydrophobic interactions with both the enzyme and the DNA. The compounds with $IC_{50}$ value above $0.5{\mu}M$ were poorly docked and did not intercalate. In addition, the docking results confirmed the overall correlation between the $IC_{50}$ values and docking scores, indicating the possible use of the modeling for the prediction of biological activity and design of potential inhibitors.

Present Status, Difficulties and Support Needs of Multi-cultural Families Raising Children under 3 Years Old Enrolled at Child Care and Education Centers (어린이집 재원 다문화 가정 영아의 양육 현황과 어려움 및 지원요구)

  • Objective: This study intended to analyze parenting characteristics, difficulties and support needs of multi-cultural families that had children under 3 years old, in order to develop their supporting resources. Methods: This study performed semi-structured interviews with 7 mothers from multi-cultural families that had their young children enrolled at child care and education centers in Gyeonggi Do. The interview data transcribed were analyzed using the successive contents analysis, and led to 4 themes and 12 sub-themes. Results: First, the mothers from multi-cultural families were married without understanding the Korean language and culture. Second, the multi-cultural mothers didn't properly respond to their children's needs in interaction and language aspects. As a result, the multi-cultural support centers and child care and education centers gave help to them. Third, in the view of the difficulties facing mothers, parenting problems caused by the lack of information concerning child education, the biased burden of parenting, and the different parenting values of Korean families were all expressed by the mothers. Fourth, regarding support needs, providing individual professional support for children and child-raising support for mothers, customized language and employment support for mothers and parenting education for fathers and family education on multi-culturalism are needed. Conclusion/Implications: Suggestions for decreasing the difficulties facing multi-cultural families that had children under 3 years old were discussed.

진흙버섯의 항인플루엔자 활성 및 활성성분 규명

  • Influenza viruses are RNA viruses that belong to the Orthomyxoviridae family, and those can be divided into three types; A, B, and C, which based on the differences of the inner nucleoproteins and genomic structures. All three genera differ in their genomic structure and nucleoprotein content, they are further classified into various serotypes based on the two surface glycoproteins, hemagglutinin (HA) and neuraminidase (NA). These glycoproteins play crucial roles in viral infection and replication. Hemagglutinin mediates binding of virions to sialic acid receptors on the surfaces of target cells at the initial stage of infection. Neuraminidase cleaves the glycosidic bonds of sialic acids from the viral and cell surfaces to release the mature virions from infected cells, after viral replication. Because NA plays an important role in the viral life cycle, it is considered an attractive therapeutic target for the treatment of influenza. The methanolic extracts of Phellinus baumii and Phellinus igniarius exhibited significant activity in the neuraminidase inhibition assay. Polyphenolic compounds were isolated from the methanolic extracts. The structures of these compounds were determined to be hispidin, hypholomine B, inoscavin A, davallialactone, phelligridin D, phelligridin E, and phelligridin G by spectroscopic methods. Compounds inhibited the H1N1 neuraminidase activity in a dose-dependent manner with $IC_{50}$ values of 50.9, 22.9, 20.0, 14.2, 8.8, 8.1 and $8.0{\mu}M$, respectively. Moreover, these compounds showed anti-influenza activity in the viral cytopathic effect (CPE) reduction assay using MDCK cells. These results suggests that the polyphenols from P. baumii and P. igniarius are promising candidates for prevention and therapeutic strategies against viral infection.

Association of Homocysteine Levels With Blood Lead Levels and Micronutrients in the US General Population

  • Objectives: Even though several epidemiological studies have observed positive associations between blood lead levels and homocysteine, no study has examined whether this association differs by the levels of micronutrients, such as folate, vitamin B6, and vitamin B12, which are involved in the metabolism of homocysteine. In this study, we examined the interactions between micronutrients and blood lead on homocysteine levels. Methods: This study was performed with 4089 adults aged ${\geq}20$ years old in the US general population using the National Health and Nutrition Examination Survey 2003-2004. Results: There were significant or marginally significant interactions between micronutrients and blood lead levels on mean homocysteine levels. Positive associations between blood lead and homocysteine were clearly observed among subjects with low levels of folate or low vitamin B6 (p-trend <0.01, respectively). However, in the case of vitamin B12, there was a stronger positive association between blood lead and homocysteine among subjects with high levels of vitamin B12, compared to those with low levels of vitamin B12. In fact, the levels of homocysteine were already high among subjects low in vitamin B12, irrespective of blood lead levels. When we used hyperhomocysteinemia (homocysteine>15 ${\mu}mol/L$) as the outcome, there were similar patterns of interaction, though p-values for each interaction failed to reach statistical significance. Conclusions: In the current study, the association between blood lead and homocysteine differed based on the levels of folate, vitamin B6, or vitamin B12 present in the blood. It may be important to keep sufficient levels of these micronutrients to prevent the possible harmful effects of lead exposure on homocysteine levels.
