The study on the anti-allergic effect of Fagopyrum esculentum Moench(Me-mil) Extract. (메밀(蕎麥) 抽出物의 抗알레르기 反應에 對한 實驗的 硏究)

  • This Experimental study was done to research effects of Fagopyrum esculentum Moench(Me-mil) Extract on the anti-allergic effects. The results were obtained as follows : 1. All concentrations of Fagopyrum esculentum Moench Extract(Flours and seeds) inhibit histamine release under the vascular permeability response to intracutanenous injection. The result is proportion to concentration. But, rutin can't get considerable result. 2. All concentrations of Fagopyrum esculentum Moench Extract inhibit histamine release under the vascular permeability response to intraperitoneal injection of SD Rat in comparison with Diphenhydramine which is typical anti-histamine drug. 3. All concentrations of Fagopyrum esculentum Moench Extract inhibit histamine release under the vascular permeability response to per-oral during the three or five days. 4. In the result of quantification of histamine induced Compound $\frac{48}{80}$, Flours and seeds of Fagopyrum esculentum Moench Extract inhibit histamine release. 5. Among the fracination of Fagopyrum esculentum Moench Extract, $CHCl_3$ fraction inhibit histamine release effectively. 6. In the result of genetic manifestative inhibition about the Human mast cell treated PMA and A23187, Fagopyrum esculentum Moench Extract effect in the IL-4 and TNF-${\alpha}$ except IL-5. According to the above results, it is suggested that Fagopyrum esculentum Moench(Me-mil) Extract has anti-allergic effect.

Study of Skin Depigmenting Mechanism of the Ethanol Extract of Fagopyrum esculentum (교맥 에탄올 추출물의 피부 미백기전 연구)

  • The aim of this study was to investigate the effect of ethanol extract of Fagopyrum esculentum on the melanogenesis. To determine whether ethanol extract of Fagopyrum esculentum suppress melanin synthesis in cellular level, B16F10 melanoma cells were cultured in the presence of different concentrations of Fagopyrum esculentum ethanol extract. In the present study, we examined the effects of Fagopyrum esculentum ethanol extract on cell proliferation, melanin contents, tyrosinase activity, expression of melanogenic enzyme proteins including tyrosinase, tyrosinase-related protein 1 (TRP-1) and tyrosinase-related protein 2 (TRP-2). Cell proliferation was slightly increased by treatment with ethanol extract of Fagopyrum esculentum $(25-200 {\mu}g/m{\ell}).$ The ethanol extract of Fagopyrum esculentum effectively suppressed melanin contents at a dose of $100 {\mu}g/m{\ell}).$ It was observed that the color of cell pellets was totally whitened compared with the control. The ethanol extract of Fagopyrum esculentum inhibited tyrosinase activity, regulate melanin biosynthesis as the key enzyme in melanogenesis. Using western blot analysis, the ethanol extract of Fagopyrum esculentum dose-dependently decreased tyrosinase and TRP-1 protein levels, and tyrosinase and TRP-1 were detected in similar manner. ${\alpha}-MSH$ leads to a stimulation of melanin synthesis through increase of tyrosinase activity, melanin contents and cytoplasmic dendricity. In this study, ethanol extract of Fagopyrum esculentum down-regulated the ${\alpha}-MSH$-induced tyrosinase activity, melanin contents and cytoplasmic dendricity. Regarding protein levels of the melanogenic enzymes, the amounts of tyrosinase and TRP-1 was increased after incubation with a-MSH. The treatment of ethanol extract of Fagopyrum esculentum decreased the ${\alpha}-MSH$-induced expression levels of tyrosinase and TRP-1. These results suggest that the ethanol extract of Fagopyrum esculentum exerts its depigmenting effects through the suppression of tyrosinase, TRP-1 and cytoplasmic dendricity. And it may be a potent depigmetation agent in hyperpigmentation condition.

  • Changes of zinc speciation and dehydrogenase activity in soil were studied before and after a field scale phytoextraction by Fagopyrum esculentum at a paddy soil near a closed zinc mine. The concentrations of zinc in paddy soil, in which Fagopyrum esculentum was planted, ranged from approximately 600 mg/Kg(high Zn soil) to 300 mg/Kg(moderate Zn soil). Despite of severe growth inhibition by Zn at the high Zn region, Fagopyrum esculentum accumulated phytoavailable fraction of Zn absorbed from the soil, and enhanced soil dehydrogenase activity (DHA) that had been inhibited by Zn toxicity. After the plant cultivation of 2 months, the concentrations of phytoavailable Zn in the rhizosphere soil at high and moderate Zn region has decreased 25% and 75%, respectively. The amount of Zn reduction in the rhizosphere soil corresponds to that accumulated in plants (recovery 92$\sim$107%), which implies Fagopyrum esculentum removed Zn from the soil. DHA was inversely correlated to the total Zn concentrations in soil. Before plant cultivation, the DHA in the high Zn soil was twice lower than that in the moderate Zn soil. More than 35% of DHA increase was observed in both soils after the application of phytoextraction with Fagopyrum esculentum.

DPPH Radical Scavenging Effect of the Aerial Parts of Fagopyrum esculentum and Isolation of Bioactive Flavonoids (메밀 지상부의 DPPH 라디칼 소거작용과 활성 플라보노이드의 분리)

  • The inhibitory effect of the aerial parts of Fagopyrum esculentum on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical was examined. The n-butanol fraction from the methanol extract of title plant showed stronger inhibitory effect than other fractions on DPPH radical. Two flavonoids were isolated from n-butanol fraction having the potent activity and elucidated as quercetin-3-O-${\alpha}$-L-rhamnoside and quercetin-3-O-rutinoside on the basis of spectral evidence. The $IC_{50}$ values of these compounds on DPPH radical were 6.56 ${\mu}M$ and 8.37 ${\mu}M$, respectively.

Effect of Microwave Assisted Water Extraction on Insoluble Phenolic Compounds through Bioactivity of Fagopyrum esculentum

  • Fagopyrum esculentum (Buckwheat) is a globally used alternative crop that contains several useful substances with various effects; however, many of these substances (rutin, quercetin, etc.) are water insoluble. To extract these substances, alcohols is required, which is inconvenient because these solvents cause diverse problems. Many studies are underway to achieve effective extraction of these substances with water. Among of these studies, microwave assisted water extraction (MAE) has been performed extensively. In this study, we performed the extraction in various solvents and/or microwave from Fagopyrum esculentum. The analysis of the content of useful substances and the bioactivity were performed and shown to increase in MAE. Liquid chromatography-mass was performed in order to identify of the useful water-insoluble substances. Catechin, quercetin, and rutin, which are all insoluble in water, were hardly extracted with water even on heating (4.4 ppb, 3.9 ppb and 60.3 ppb, respectively). However, MAE was found to extract much more of these substances than water (1204 ppb, 110.8 ppb and 2946 ppb, respectively). Although less efficient than alcohols, MAE showed much higher efficiency than simple water extraction. These results indicate that water extraction using microwave technology is effective in cases where it is difficult to extract useful substances using water.

Prevention of Photoaging and Wrinkle Formation in Hairless Mice by Korean Red Gnseng and Fagopyrum Esculentum Extract Mixture (Hairless mouse에서 홍삼 . 교맥 복합물의 피부주름 생성 예방 효과)

  • UV irradiation causes skin-aging involving coarse wrinkles, thickening, dyspigmentation, and rough skin surface. This study was carried out to develop health & functional food by using Korean red ginsneg and Fagopyrum esculentum extract mixture (RGFE) for prevention of skin wrinkles. The RGFE-treated group showed the more effective collagenase inhibition rate than the red ginseng (RG)-treated group. To investigate photo protective effects of RGFE on UV-induced damaged skin, SKH hairless male mice were orally administerd RGFE and regional treatment and irradiated with UV for up to 8 weeks. In RGFE-treated group, better skin, and less wrinkle formation were observed compared with UV group. Epidermal thickness of hairless mouse was significantly decreased in RGFE, RG, and Fagopyrum esculentum (FE) groups compared with UV group. These results demonstrate RGFE have photo-protective effects on UV-damaged hairless mouse skin.
