Cloning of a Gene Specifically Expressed During Early Stage of Fruiting Body Formation in Flammulina velutipes (팽이버섯의 자실체형성 초기과정에서 특이적으로 발현하는 유전자의 클로닝)

  • A cDNA library was constructed using mRNA from the cells of 7-day-old cultures of Flammulina velutipes after induction of fruiting treatment. A cDNA clone, FVFD16 (Flammulina velutipes fruiting body differentiation), was selected by differential screening. The expression property of the FVFD16 gene was examined by Northern blot analysis. FVFD16 represents mRNA that is specifically expressed during differentiation of fruit bodies. The conspicuous accumulation of the FVFD16 mRNA was detected in 4-day-old and 1-day-old cultures. The nucleotide sequence of the FVFD16 gene was determined and the mRNA contained an open reading frame that encoded a putative protein of 128 amino acid residues (13.5 kDa).

Selection of Superior Strains of Cordyceps militaris with Enhanced Fruiting Body Productivity

  • In vitro fruiting bodies were produced from ten different isolates of Condyceps militaris EFCC C-5736, EFCC C-5941, EFCC C-5976, EFCC C-6040, EFCC C-6849, EFCC C-7268, EFCC C-7342, EFCC C-7992, EFCC C-8027 and EFCC C-8549. Single ascospores were isolated from in vitro grown fruiting bodies and used for fruiting body production in brown rice medium by both intra-strain crossing and out-crossing. Length and dry wt. of stromata grown in vitro were measured. Strains producing highest dry wt. of stromata were selected. Both intra-strain crossings and inter-strain crossings of single ascospore strains were found to produce profuse fruiting bodies of C. militaris.

  • We inoculated the spawn of Inonotus obliquus on living trees of Betula platyphylla var. japonica, and could get several fruiting bodies from the inoculations. The fruiting body showed almost the same feature of that collected from natural habitats in Korea, while the size was much smaller than that of natural one. The diameter of the fruiting body ranged from 1.1 cm to 4.8 cm, and the height 0.5 cm to 2.0 cm. We expect that a harvest is possible within 5 years, but need to check the growth rate of the fruiting body to decide suitable time for getting more economic value.

Characteristics of Suillus bovinus fairy rings and genets associated with thinning intensity in Pinus densiflora forests (소나무림에서 간벌강도에 따른 황소비단그물버섯(Suillus bovinus)의 균환과 genet 특성)

  • To study the fairy ring and genet characteristics of Suillus bovinus based on thinning intensity in Pinus densiflora forests, a simple sequence repeat (SSR) analysis was performed on the fruiting bodies of the plant. In pine wood production forests, the thinning strengths applied were 34%, 45%, and 60%. As a result, the number of fruiting bodies in the 34% treatment area was 104, which was higher than that in the other treatment areas. In the 34% treatment area, fruiting bodies occurred in a circular shape, within a diameter of approximately 5 meters (m) of the trees. In the 45% treatment area, the fruiting bodies were randomly distributed between 6 to 7 m from the trees, while in the 60% treatment, fruiting bodies occurred in a narrow oval shape, 6 m from the trees. In the control area, two fruiting bodies were present around the root collar. Hybridity was confirmed in the SSR markers of Sb-CA1 and Sb-CA3. The fruiting bodies in the 34% treatment area had a He / Ho value lower than that in the 60% treatment area. In fruiting bodies of the 34% treatment area, a total of 20 genets were identified, with an average size of 14±11 ㎡; 60% of genets were formed by a single fruiting body. In fruiting bodies of the 45% treatment area, a total of 6 genets were identified and the average size was 11±12 ㎡; 50% of genets were formed by a single fruiting body. In fruiting bodies of the 60% treatment area, a total of 10 genets were identified, with an average size of 1.1±0.8 ㎡; 70% of genets were formed by a single fruiting body. Thus, the formation ratio of a new genet increases when the thinning intensity is increased.

  • Traditionally, Cordyceps species have been used as a part of herbal medicine in Oriental countries, including Korea a for intemal health, vigorosity and to cure different diseases related to heart, lung etc. In the recent years, research on artificial fruiting of different Cordyceps species including C. militaris has been carried out in the world because of their medicinal value. Variationas well as instability occur during artificial fruiting. Instability observed in the in vitro fruiting of C. militaris is reported in the present study.

  • A gene (hpkA), encoding a histidine protein kinase homolog, has been identified in the upstream region of the espAB operon in Myxococcus xanthus. It encodes a 333 amino acid (35,952 Da) protein with a histidine protein kinase domain in the region from amino acid 90 to 317. Null mutations in the hpkA gene caused formation of loose irregular fruiting bodies, while wild-type strains developed tight hemispherical fruiting bodies under developmental conditions. Sporulation of the hpkA mutant was delayed by at least 12 h compared to that of the wild-type. It appeared that the hpkA mutation increased the expression of the espAB operon by more than 2-fold compared with the wild-type under developmental conditions. Expression of the hpkA gene was low under vegetative conditions, but was highly induced under developmental conditions.

Entomopathogenicity of Simplicillium lanosoniveum Isolated in Korea

  • Fruiting bodies similar to those of the ascomycete fungi Podostroma cornu-damae and Cordyceps militaris were collected from Mt. Seunghak in Busan, Korea on August 21, 2012. The fruiting bodies were cylindrical, with tapered ends and golden red in color. The fruiting bodies contained abundant conidiophores bearing single-celled conidia, but no perithecia or asci. Pure culture of the fungal isolates was obtained through single-spore isolation. Analyses of morphological characteristics, including conidia shape, and phylogenetic traits, using internal transcribed spacer sequences, showed that these isolates belonged to the species Simplicillium lanosoniveum. Although this fungal species is known to be mycoparasitic, the isolates obtained in this study were unable to infect fungi. However, silkworms (Bombyx mori) inoculated with the fungal isolates died during the larval or pupal stages, as has been shown for the strongly entomopathogenic fungus Beauveria bassiana. This study is the first report of the entomopathogenicity of S. lanosoniveum and indicates its potential for use in biological control of insects.

Comparison of Productivity and Temperature type of fruiting body of Lentinula edodes strains on the Sawdust Cultivation (표고 톱밥재배용 교배균주의 자실체 온도형 및 생산성 비교)

  • We cultivated 117 hybrid strains of Lentinula edodes in greenhouse. 5 hybrid strains(07-55, 07-66, 07-84, 07-93 and 07-117) were selected from comparison of their characteristics of fruiting body and productivity. Biological efficiency of 5 hybrid strains was investigated 36.1(07-66), 35.6(07-117), 27.1(07-93), 25.7(07-84) and 6.5%(07-55), respectively. Selected strains 07-66, 07-117 and 07-93 showed high biological efficiency but observed commercially low quality fruiting body. Temperature type of fruiting body of 5 hybrid strains observed as follows high temperature type(07-117), mid temperature type(07-66 and 07-84) and low temperature type(07-55 and 07-93). Temperature type of fruiting body of 07-93 strain has wide range relatively. In the results, 07-84 strain was selected finally with consideration for quality of fruiting body, productivity and temperature type($8-21^{\circ}C$) in farm test.

Relationship between the sexual and the vegetative organs in a Polygonatum humile (Liliaceae) population in a temperate forest gap

  • Background: The aim of this study was to clarify the relationship between the sexual reproduction and the resource allocation in a natural Polygonatum humile population grown in a temperate mixed forest gap. For this aim, the plant size, the node which flower was formed, the fruiting rate, and the dry weight of each organ were monitored from June 2014 to August 2015. Results: Firstly, in 3-13-leaf plants, plants with leaves ${\leq}8$ did not have flowers and in plants with over 9 leaves the flowering rate increased with the number of leaves. Among plants with the same number of leaves, the total leaf area and dry weight of flowering plants were larger than those of non-flowering plants. The minimum leaf area and dry weight of flowering plants were $100cm^2$ and 200 mg, respectively. Secondary, the flowers were formed at the 3rd~8th nodes, and the flowering rate was highest at the 5th node. Thirdly, cumulative values of leaf properties from the last leaf (the top leaf on a stem) to the same leaf rank were greater in a plant with a reproductive organ than in a plant without a reproductive organ. Fourthly, fruit set was 6.1% and faithful fruit was 2.6% of total flowers. Biomasses of new rhizomes produced per milligram dry weight of leaf were $0.397{\pm}190mg$ in plants that set fruit and $0.520{\pm}0.263mg$ in plants that did not, and the difference between the 2 plant groups was significant at the 0.1% level. Conclusions: P. humile showed that the 1st flower formed on the 3rd node from the shoot's base. And P. humile showed the minimum plant size needed in fruiting, and fruiting restricted the growth of new rhizomes. However, the fruiting rate was very low. Thus, it was thought that the low fruiting rate caused more energy to invest in the rhizomes, leading to a longer rhizome. A longer rhizome was thought to be more advantageous than a short one to avoid the shading.

Breeding and Cultural Characteristics of Newly Bred Lentinula edodes Strain 'Sanjanghyang' (신품종 표고버섯 '산장향'의 교배 육성 및 재배 특성)

  • A new cultivar 'Sanjanghyang' was bred from monokaryotic strains of 'Sanbaekhyang' and 'Jangan 1ho'. Pileus was flat, round, and reddish brown. The diameters of the pilei and stipe length of the fruiting bodies were 67.1 mm and 16.9 mm, respectively. The scales were white or slightly brown and distributed evenly. The gill density was sparse and showed a rippled texture. The stipe was cream in color and the fluff was medium. 'Sanjanghyang' had a short cultivation period and fruiting bodies occurred sporadically. Temperature for fruiting body formation was a medium, between 15 to $19^{\circ}C$. 'Sanjanghyang' was different from 'Sanbaekhyang' with regard to its pileus diameter (67.1 mm) and autumn and spring fruiting body production period. 'Sanbaekhyang' had pileus diameter of 74.7 mm, and fruiting body formation occurred in spring and autumn. The rate of fruiting body formation was 89% (first flush), 4% (second flush), and 7% (third flush).
