FMN-Based Fluorescent Proteins as Heavy Metal Sensors Against Mercury Ions

  • Bacterial light-oxygen-voltage-sensing photoreceptor-derived flavin mononucleotide (FMN)-based fluorescent proteins act as a promising distinct class of fluorescent proteins utilized for various biomedical and biotechnological applications. The key property of its independency towards oxygen for its chromophore maturation has greatly helped this protein to outperform the other fluorescent proteins such as GFP and DsRed for anaerobic applications. Here, we describe the feasibility of FMN-containing fluorescent protein FbFP as a metal-sensing probe by measuring the fluorescence emission changes of a protein with respect to the concentration of metal ions. In the present study, we demonstrated the mercury-sensing ability of FbFP protein and the possible amino acids responsible for metal binding. A ratiometric approach was employed here in order to exploit the fluorescence changes observed at two different emission maxima with respect to Hg2+ at micromolar concentration. The engineered variant FbFPC56I showed high sensitivity towards Hg2+ and followed a good linear relationship from 0.1 to 3 μM of Hg2+. Thus, further engineering with a rational approach would enable the FbFP to be developed as a novel and highly selective and sensitive biosensor for other toxic heavy metal ions as well.

  • This research was performed to find out the adaptation patterns of maxillary and mandibular posterior teeth to the changes in relationships of vertical skeletal components, which constitute the skeletofacial complex. For this research, 61 adult malocclusion patients were chosen as subjects according to the Hellman's dental age with normally ranged FMN-A-B angle. These subjects were divided into 4 groups in maxilla and 3 groups in mandible according to mesiodistal inclinations of teeth. Following results were obtained after studying the relationships of the vertical skeletal components between each group. 1. Inspire of the fact that the FMN-A-B angle was within a normal range, the degree of mesiodistal inclinations of maxillary and mandibular posterior teeth showed differences in relation to the anteroposterior relationships of maxilla and mandible. In case where the FMN-A-B angle was large, the mesial inclinations of maxillary posterior teeth showed more increase from the posterior to the anterior, whereas in mandible it showed overall decrease. 2. The degrees of mesial inclinations of mandibular posterior teeth were increased when the angulations of lower facial height, occlusal plane angle and mandibular plane angle were greater. 3. The patterns of mesial inclinations of maxillary posterior teeth were varied according to the angulation of lower facial height. If relatively large, it showed more increase from the posterior to the anterior and it was decreased nearly consistent when the angulation was small. 4. The degrees of mesial inclinations of maxillary posterior teeth were decreased as the lower facial height, palatal plane angle, occlusal plane angle and the mandibular plane angle became greater.

  • This investigation was designed to categorize Angle's class III malocclusion groups through analyzing horizontal and vertical components of craniofacial skeleton in Angle's class III malocclusion. The material selected for this study consisted in standard lateral cephalogram of one hundred and fifty five children, seventy four boys and eighty one girls, aged 6 through 15 years, having Angle's class III malocclusion. On the basis of findings of this study, the following results were obtained. 1. In horizontal skeletal classifications,16 groups were classified FMN-A-B, SE-FMN-A according to the Ba-SE-Me, Ba-SE/R. 2. The sequences that have relatively high frequency are as follow; a) Horizontal Group l b) Horizontal Group 3 c) Horizontal Group 5 d) Horizontal Group 9 e) Horizontal Group 4 3. In vertical skeletal classification, 8 groups were classified according to the PMV/PP, PMV/OP, PMV/MP. 4. The sequences that relatively high are as follows; a) Vertical Group 1 b) Vertical Group 3 c) Vertical Group 4 d) Vertical Group 8.

Chemical Modification of Glycolate Oxidase from Spinach by Diethyl Pyrocarbonate. Evidence of Essential Histidine for Enzyme Activity$^\dag$

  • FMN-dependent glycolate oxidase from spinach is inactivated by diethyl pyrocarbonate at pH 7.0. Inactivation of both apo- and holoenzyme by diethyl pyrocarbonate follows pseudo-first-order kinetics and first order with respect to the reagent. A series of difference spectra of inactivated and native enzymes show a single peak at 240 nm, indicating the modification of histidyl residues. No decrease in absorbance at around 280 nm due to formation of O-carbethoxytyrosine is observed. The rate of inactivation is dependent on pH, and the data for pH dependent rates implicate the involvement of a group with a pKa of 6.9. The activity lost by treatment with diethyl pyrocarbonate could be almost fully restored by incubation with 0.75M hydroxylamine. The reactivation by hydroxylamine and the pH dependence of inactivation are also consistent with that the inactivation is due to modification of histidyl residues. Although coenzyme FMN is without protective effect, the substrate glycolate, the product glyoxylate, and two competitive inhibitors, oxalate and oxalacetate, provide marked protection against the inactivation of the holoenzyme. These results suggest that the inactivation of the oxidase by diethyl pyrocarbonate occurs by modification of essential histidyl residue(s) at the active site.

The Characteristics on Water Surface Effects of Air Supported TP 620 Hydrofoil over Water (박용 TP 620익형의 공기 부양 특성)

  • The steady potential flow about a TP 620 hydrofoil, flying in air above a dynamic water surface. is calculated by the one dimensional theory as the clearance-to-length ratio tends to very small. The characteristics on water surfare effects of air supported TP 620 hydrofoil over water are compared NACA 4412, NACA 66 and plate. respectively. It is found that using the TP 620 hydrofoil is superior lift coefficients. Moreover for trailing edge shape of the hydrofoil, high lift coefficients at 1 Fmn can be obtained.

  • Ascorbyl free radical reductase was purified from the white rot fungus, Pleurotus ostreatus. The enzyme contained FMN as a prosthetic group, which was reduced by NADH and reoxidized by ascorbyl free radical. Reduction of ascorbyl free radical by the enzyme was observed by EPR spectroscopy.(omitted)
