Flow cytometry of cell-cycle on Flavin mononucleotide (1,4-butanediamine) Pt(II) Complex and Cisplatin and Their Biochemical Analysis of Nephrotoxicity in ICR Mice (Flavin mononucleotide (1,4-butanediamine) Pt(II) Complex와 Cisplatin의 세포주기에 대한 유세포 분석 및 ICR계 생쥐에서의 신장독성에 대한 생화학적 분석)

  • Flavin mononucleotide (1,4-butanediamine) Pt(II) complex (7FMN) was synthesized and screened anticancer activity [J. Pharm. Soc. Korea 43(6),762-770 (1999)]. 7FMN have good water solubility and moderate anticancer activiy In this paper cell-cycle specificity and nephrotoxicity were studied. Interaction of DNA with cisplatin and synthesized 7FMN was analyzed by flow cytometry and showed G2 arrest in L1210 cell line. It means that cell-cycle on L1210 was inhibit in S phase by cisplatin and 7FMN. In order to biochemically analyze nephrotoxicity of cisplatin and 7FMN, after injecting each agent intraperitoneally, blood was exsanguinated after 6 hours, 1 day, 3 days and 7 days, respectively. Then, serum was separated from the blood. The serum level of BUN, creatinine and uric acid in cisplatin and 7FMN administated mice (25~35 g, ICR strain, a dose each 8,12 and 16 times of the $IC_{50}$/ value, cisplatin; 7 times) were determined by autochemistry analyzer. In cisplatinadministered mice group, BUN level was elevated than normal control group at 3rd day and repaired at 7th day. In 7FMN administrated group was not elevated. Creatinine and uric acid level were no difference with the normal control group. Therefore synthesized 7FMN is less toxic than cisplatin in nephrotoxiciaty.

  • Azotobacter vinelandii cell extracts and its variety of purified fractions with regard to their ability to form the redox state of the Shethna Flavoprotein (free radical form FPH.) were studied. A fluorescent flavoprotein (protein I) and a brown protein (protein II) were the most active proteins which were isolated in purified form. The free radical formation activity was substantially decreased during the purification and was completely lost upon storage in a week under nitrogen in a frozenstate. The presence of free flavin (FMN) with NADH enhanced the rate of free radical formation. The reaction of FMN and NADH was found to be catalysed by various cell fractions. A possible role of FMN as a substrate for free radical shethna flavoprotein was investigated. Slower reaction rate of $FMNH_2+Flavoprotein\;(FP){\to}FPH+FMN$ than $FMN+NADH{\to}FMNH_2$, accumulation of $FMNH_2$ ocurred which subsquently caused FPH.

  • A series of vitamin-containing Pt(II) complexes of the type [Pt (FMN) (L)] (FMN=flavin mononucleotide, L=ethylenediamine, 1,3-propanediamine, 1,4-bu-tanediamine) was synthesizd and characterized by IR, electronic absorption, elemental analysis and FAB=Mass. The coordination sites of FMN to Pt(II) ions were determined to be N(5) and O(6) with resultant chelate ring formation. Theses compounds have much better water solubility (30-35 mg/ml) than cisplatin (1 mg/ml). The anticancer activity of this vitamin-containing Pt(II) series was investigated by MTT assay against mouse and human leukemia cell lines in vitro. Among these compounds, FMN (1,4-butanediamine) Pt(II) having seven-membered ring structure as amine ligand showed moderate anticancer activity.

  • NADH-quinone reductase was purified 22-fold from the cytosolic fraction of Streptomyces sp. Imsnu-1 to apparent hemogenity, with an overall yield of 9%, by the purification procedure consisting of ammonium, sulfate precipitation and DEAE Sephacryl S-200 and DEAE 5 PW chromatographies. Thes molecular mass of the enzyme determined by gel filtration chromatography was found to be 110 kDa. SDS-PAGE revealed that the enzyme consists of two sugunits with a molecular mass of 54 kDa. The enzyme contained 1 mol of FMN per subunit as a cofactor. The $A_{272}$ A$_{457}$ ratio was 6.14 and the molar extinction coefficients were calculated to be 20, 800 and 25, 400M$^{-1}$ $cm^{-1}$ / AT 349 AND 457 nm, respectively. The N-terminal sequence of the enzyme contained the highly conserved fingerprint of ADP-binding domain. The enzyme used NADH as an electron donor and various quinones as electron acceptors. Cytochrome c was practically inactive. Air-stable flavin semiquinone was produced by the addition of NADH to the enzyme. Also, naphthosemiquinone was detected in the reaction mixture containing the enzyme.

  • This study was performed to define the adaptation patterns of each skeletal components to the flexures of cranial bases, using 91 males from the ages of 17 to 36 and 64 females from the ages of 16 to 34, without orthodontic or prosthetic treatment experiences and with pleasant profiles as subjects. The conclusions are as follow: 1. When considering the changes of flexure of cranial base (Ba-SE-FMN) in both sexes, changes in the anterior cranial base angle to the PM Vertical line (SE-FMN/PMV) were greater than the changes in the posterior cranial base angle to the PM Vertical line (Ba-SE/PMV). Subsequently the nasomaxillary complex showed antero-superior rotating effect as the cranial base angles were increased and postero-inferior rotating effect as they were decreased. 2. Horizontal mandibular angle (Ba-SE-Me) was increased in both sexes as cranial base angle increases (Ba-SE-FMN) and it decreased as the latter was decreases. There by indicating compensatory effects. 3. Maxillary angle (SE-FMN-A) was decreased in both sexes as cranial base angle (Ba-SE-FMN) increases and it increased as the latter was decreased. There by indicating compensatory effects. 4. Mandibular ramus angle to posterior cranial base was decreased in both sexes as cranial base angle increases. There by indicating compensatory effect to anteriorly displaced maxilla and the mandibular ramus angle was increased as the cranial base angle decreases. There by indicating compensatory effect to posteriorly displace maxilla. 5. The length of posterior upper facial height was decreased in both sexes as the cranial base angle increases and it increased as the latter was decreased.

Determination of vitamin $B_2$ by the lumiflavin fluorometric method and FMN, FAD, FR by the paper chromatography in the feces and urine (Lumiflavin 형광법(螢光法)에 의한 Rat 분뇨중(糞尿中)의 vitamin $B_2$와 FMN, FAD, FR의 정량(定量))

  • After ablactation, wistar strain white male rats, weighing 270g and 340g, were fed with a diet of CLEA for three months. The whole daily excretion of each feces and urine were collected, and extracted with water($80^{\circ}C$ hot water). The combined extraction were filtered and the $B_2$ was determined with the parts of the filterates by the lumiflavin fluorometric method, and the FMN, FAD and FR with the rest of the filterates by paper chromatography. The following results were obtained; 1. $B_2$ contents in the feces were $27.52{\gamma}$ per 100 grams per body weight, and $83.93{\gamma}$ per each rat per day. 2. $B_2$ contents in the urine were $18.47{\gamma}$ per 100 grams per body weight, and $56.33{\gamma}$ per each rat per day. The total daily excretion of $B_2$ contents in the feces were 1. 5 times as much as in the urine. 3. Among the total daily $B_2$ excretion of one white wistar strain rat in the feces were the following ; FAD, 81.0% ; FMN, 14.9% ; FR, 3.3%. Therefore the order of the contents were FAD>FMN>FR.

  • This research was carried out in order to study the effects of Rapid Maxillary Expansion on maxilla and it's surrounding skeletal structures. The sample for this study consists of 14 patients who were in retention period after rapid maxillary expansion. Following results were obtained after performing comparative analysis of cephalograms taken before and after rapid maxillary expansion. 1. In almost every cases, inferior displacement of palatal planes with concurrent changes in their inclination were observed. 2. Changes in the inclination of palatal plane can be categorized into inferiorty inclined group, superiorly inclined group and constant group in relation to the states before rapid masillary expansion. 3. Decrease in < Se-FMN/PMV and increase in < Se-FMN-A were evident in the superiorly inclined group and vice versa for the inferiorly inclined group.

  • The daily variation in the total quantity of flavin compounds (FAD, FMN, FR) in greenbean during its germination were measured by Yagi's methods. The results obtained were as follow. 1. The total quantity of flavin compounds of the greenbean sprouts was increased with growth of sprouts until the 8th days. After this period its contents were decreased slightly. 2. The flavin compounds of greenbean sprouts were comfirmed to be synthesized as free riboflavin during its germination. 3. FAD was the major pat of total flavin compounds in cotyledon but there was significant variation in the contents of FAD, FMN and FR in hypocotyl.

  • 1) The variations of flavin adenine dinucleotide(FAD), flavin monomucleotide(FMN) and free riboflavin in control and 600r X-ray irradiated soybean sprout were estimated by the paper chromatographic method. 2) It was found that the formation of riboflavin in soybean sprout was mainly in FAD. 3) There were no great variation of FMN and free riboflavin in soybean sprout during the germination. 4) The moderate amount (600r) of X-ray irradiation accerelated the formation of FAD in soybean sprout during the germination.

  • The arsH gene is one of the arsenic resistance system in bacteria and eukaryotes. The ArsH protein was annotated as a NADPH-dependent flavin mononucleotide (FMN) reductase with unknown biological function. Here we report for the first time that the ArsH protein showed high ferric reductase activity. Glu104 was an essential residue for maintaining the stability of the FMN cofactor. The ArsH protein may perform an important role for cytosolic ferric iron assimilation in vivo.
