Development of an Open Sandwich Fluoroimmunoassay Based on FRET (FRET에 기반한 Open Sandwich Fluoroimmunoassay)

  • We have developed a sensitive, one-step, homogeneous open sandwich fluoroimmunoassay (OsFIA) based on fluorescence resonance energy transfer (FRET) and luminescent semiconductor quantum dots (QDs). In this FRET assay, estrogen receptor-$\beta$ (ER-$\beta$) antigen was incubated with QD-labeled anti-ER-$\beta$ monoclonal antibody and AF (Alexa Fluoro)-labeled anti-ER polyclonal antibody for 30 minutes, followed by FRET measurement. The dye separation distance was estimated to be between $80\sim90\;{\AA}$. The present method is rapid, simple and highly sensitive, and did not require the bound/free reagent separation steps and solid-phase carriers. A concentration as low as 0.05 nM (2.65 ng/ml) receptor was detected with linearity ($R^2$ > 0.990). In addition, the assay was performed with commercial antibodies. This assay provides a convenient alternative to conventional, laborious sandwich immunoassays.

Diagnostic Accuracy of the Quidel Sofia Rapid Influenza Fluorescent Immunoassay in Patients with Influenza-like Illness: A Systematic Review and Meta-analysis

  • Background: Although the Quidel Sofia rapid influenza fluorescent immunoassay (FIA) is widely used to identify influenza A and B, the diagnostic accuracy of this test remains unclear. Thus, the objective of this study was to determine the diagnostic performance of this test compared to reverse transcriptase-polymerase chain reaction. Methods: A systematic literature search was performed using MEDLINE, EMBASE, and the Cochrane Central Register. Pooled sensitivity, specificity, diagnostic odds ratio (DOR), and a hierarchical summary receiver-operating characteristic curve (HSROC) of this test for identifying influenza A and B were determined using meta-analysis. A sensitivity subgroup analysis was performed to identify potential sources of heterogeneity within selected studies. Results: We identified 17 studies involving 8,334 patients. Pooled sensitivity, specificity, and DOR of the Quidel Sofia rapid influenza FIA for identifying influenza A were 0.78 (95% confidence interval [CI], 0.71-0.83), 0.99 (95% CI, 0.98-0.99), and 251.26 (95% CI, 139.39-452.89), respectively. Pooled sensitivity, specificity, and DOR of this test for identifying influenza B were 0.72 (95% CI, 0.60-0.82), 0.98 (95% CI, 0.96-0.99), and 140.20 (95% CI, 55.92-351.54), respectively. The area under the HSROC for this test for identifying influenza A was similar to that for identifying influenza B. Age was considered a probable source of heterogeneity. Conclusion: Pooled sensitivities of the Quidel Sofia rapid influenza FIA for identifying influenza A and B did not quite meet the target level (≥80%). Thus, caution is needed when interpreting data of this study due to substantial betweenstudy heterogeneity.

Influence of the Food Intake Ability and the Number of Remaining Teeth on Oral Health Related Quality of Life in Some Elderly People (일부 노인의 저작강도별 음식섭취능력과 잔존치아수가 구강건강관련 삶의 질에 미치는 영향)

  • Eating supports to gain energy and nutrition and improves quality of life. Ageing affects the food intake ability due to loss of natural teeth and the progressive muscle degeneration. Our objective was to investigate how the food intake ability (FIA) and the remaining teeth could influence on oral health related quality of life in the elderly. This study was performed with 503 elderly subjects living in Daejeon, Korea. The questionnaire with the FIA with 30 Korean food and Oral Health Impact Profile 14 (OHIP-14) and oral examination were surveyed. The five groups according to cluster analysis of FIA had the different numbers of remaining teeth and functional posterior teeth with opposing teeth or prosthesis significantly: group 1, $21.78{\pm}8.27$ and $2.80{\pm}2.63$; group 2, $16.75{\pm}7.87$ and $2.16{\pm}2.44$; group 3, $14.68{\pm}9.77$ and $1.73{\pm}2.30$; group 4, $9.93{\pm}8.13$ and $0.78{\pm}1.68$; group 5, $10.18{\pm}8.37$ and $0.51{\pm}1.22$. The more foods the subjects could masticate, the better oral health related quality of life they had. The medium FIA, soft FIA and the number of remaining teeth could explain 46% of OHIP-14, but hard FIA could not in the multiple regression model. We suggested to develop the oral health program for the elderly to be able to eat the food with medium physical property at least be helpful to improve oral health related quality of life.

A Study on the Detection Characteristics in Glucose and Fabrication of Bi-Enzyme Electrode using Electrochemical Method (전기화학적 방법을 이용한 다중 효소 전극 제작 및 글루코스 검출 특성에 관한 연구)

  • In this study, the development of biosensors capable of bi-enzyme reactions by including Horseradish peroxidase and glucose oxidase was carried out for detection of glucose. The sensors were manufactured using electro deposition method to reduce production time, and screen printed electrodes (SPE) were used to produce economical sensors. To check the bienzyme effect, the sensor was compared and analyzed with single enzyme biosensor. The characteristics of the sensor were evaluated using scanning electron microscopy(SEM), cyclic voltammetry(CV), electrochemical impedance spectroscopy(EIS), chronoamperometry(CA), and flow injection analysis(FIA). Analysis results from SEM, CV and EIS confirmed that the enzymes are well fixed to the electrode surface. In addition, it was confirmed that bi-enzyme biosensors manufactured from the CA method improved signal performance by 200% compared to single enzyme biosensors. From this results, we were able to explain that HRP and GOD react catalyzed to each other. And the results of FIA showed that the intensity of each current signal was constant when the same concentration of glucose was injected four times. In addition, by analyzing the intensity of current signals for glucose concentrations, the biosensors manufactured in this study showed excellent trends in signal sensitivity, reproducibility and stability.

Analysis Results in Technical Trends of 2018 Farnborough International Airshow via Centrality Analysis (중심성 분석을 이용한 2018년 판보로 국제 에어쇼 참가업체 기술동향 분석)

  • The purpose of this research was to introduce a network analysis method for analyzing technology trends in the aerospace industry at the Farnborough International Airshow (FIA), one of the world's three major airshows. Civil and defense companies and government and military officials from 112 countries and 1,500 agencies convened at FIA 2018 to share and explore recent trends in the aerospace industry. We studied aerospace technologies from 45 countries, 1,108 exhibiters, and 223 technology categories via centrality analysis. The results from the network analysis showed that machining is the center of aerospace technology. However, there were quite different tendencies, depending on the region and country. The centers of aerospace technology are machining in Europe and the United Kingdom, aircraft components in Asia, and engine components/controls in the United States. In Korea, no one key technology was recognized, due to the country's small attendance. We hope this research will be conducive to aerospace technology-and-research planning, and that it will be an appropriate tool to help domestic manufacturers boost their exports.

  • Recently, FET type dissolved oxygen sensor was proposed to overcome the disadvantages of the amperometric Clark-type sensor. The inherent problem of the proposed sensor, however, is the flow dependence of the sensor performances since the proposed sensor detects the pH change in close proximity to the working electrode. In this study, we decided the direction which minimize the flow effect in FIA(flow injection analysis) system. And a hydrodynamic buffer layer which can reduce the flow dependence were proposed. The suggested buffer-layers were formed onto sensing area and working electrode with mixed polymer matrix of TEOS(tetraethylorthosilicate) and DEDMS(diethoxydimethylsilane).

Spectrophotometric Determination of Ultra trace Tri & Hexavalent Chromium by Using on-line Flow Injection Analysis with Dual Pre-concentration Column

  • An on-line flow injection analysis with dual pre-concentration method was developed to determine the ultra trace tri and hexavalent chromium in water. In this system, the cation and anion pre-concentration columns were combined with a 10-port injection valve and then used to separate and concentrate Cr (III) and Cr (VI) selectively. The two species of concentrated chromium were sequentially eluted and determined by using HCl-KCl buffer of pH 1.8 as an eluent. Cr (III) was oxidized by hydrogen peroxide to Cr (VI). It was detected spectrophotometrically at 548 nm by complexation with DPC (diphenylcarbazide). Several factors such as concentration of $H_2O_2$, DPC and coil length in reaction condition were optimized. The linear range for Cr (III) and Cr (VI) was 0.1-50 ${\mu}g$/L. The limit of detections ($3{\sigma}$) of Cr (III) and Cr (VI) were 52 ng/L and 44 ng/L under the optimized FIA system, and their recoveries 98% and 103%, respectively. This method was applied to analyze contamination level of chromium species in tap water, groundwater and bottled water.

Effect of Water Extracts from Root of Taraxacum officinale on Innate and Adaptive Immune Responses in Mice (민들레 뿌리 물 추출물의 마우스 선천 및 획득 면역계에 미치는 효과)

  • Hot-water($100^{\circ}C$) and cold-water($4^{\circ}C$) extracts of Taraxacum officinale root were assessed for the effects of innate and adaptive immune responses in mice. Hot water extracts(TO-100) and cold water extracts(TO-4) did not affect the viability of macrophages at concentrations below to 18 mg/ml and 8 mg/ml, respectively. The thioglycollate-induced macrophages cultured with TO-100 and TO-4 produced a significantly higher quantity of various cytokines, such as IL-6 and IL-12, than those treated with medium. This shows that the extracts potently stimulated the innate immune response. When mice were subcutaneously immunized(sc) with OVA+FIA(Freund's incomplete adjuvant)-emulsified TO-100, TO-100 did not affect the production of IgE, but enhanced the production of IgG1, IgG2a and IgG2b. The culture supernatant obtained from the splenocytes of mice treated with OVA+FIA-emulsified TO-100 also evidenced elevated levels of both OVA-specific Th1-type(IFN-$\gamma$) and Th2-type cytokines(IL-4, IL-6 and IL-10). These results suggested that TO-100 can modulate the immune responses to allergens in mice.

Analysis of $Cr_2O_7^{-2}/MnO_4^{-}$ Mixtures by an Absorption Spectrometry Coupled with Flow Injection Analysis(FIA) (흐름주입분석기법에 접목된 흡수분광분석법에 의한 $Cr_2O_7^{-2}/MnO_4^{-}$혼합물의 분석)

  • An absorption spectrometry coupled with flow injection analysis has been developed and tested for the analysis of the Cr$_2O_7^{2-}$/Nn$O_4^-$ mixtures. Even though one has to inject the sample twice into the FIA system and the process of the sample treatment is required to completely destroy the Mn$O_4^-$ ion for the analysis of the Cr$_2O_7^{2-}$ ion, the new method has definite advantages over the current method. It utilizes only a single analytical wavelength (570 nm) and enables one to construct calibration curves which accurately follow the Beer's law over wide ranges of analytical concentrations of both ions ($2.0{\times}10^{-6}$M∼$8.0{\times}10^{-3}$M for Cr$_2O_7^{2-}$ ion, $2.0{\times}10^{-6}$M∼$4.0{\times}10^{-3}$M for MnO4- ion).

Key Food Selection for Assessement of Oral Health Related Quality of Life among Some Korean Elderly (일부 한국 노인 구강건강 관련 삶의 질 평가를 위한 핵심 음식 선택)

  • Oral health can influence on diverse food intake, and food intake affect oral health related quality of life. The aim of this study was to select key foods to be able to represent oral health related quality of life in Korea. We used the data of 503 Korean older persons to participate in the oral health promotion programme in 2009. The low consumption or low intake foods with criteria in 2012 National Nutrition Statistics were eliminated among 30 foods of food intake ability (FIA) at first. Decision tree model, correlation analysis, factor analysis, and internal reliablity test were used for oral health related quailty of life (OHRQoL) key food selection. We selected 13 foods-hard persimmon, dried peanut, pickled radish, caramel, rib of pork, glutinous rice cake, cabbage kimchi, apple, yellow melon, boiled chicken meat, boiled fish, mandarin, noodles as OHRQoL Key Foods 13. Thirty foods of FIA and OHRQoL Key Foods 13 displayed the same pattern of variation among sociodemographic groups. In a regression model, both of 30 foods of FIA and OHRQoL Key Foods 13 influenced on oral health impact profile-14. The findings suggest that OHRQoL Key Foods 13 have good reliability and validity and be able to use in oral health survey.
