Interplay between the Gut Microbiome and Metabolism in Ulcerative Colitis Mice Treated with the Dietary Ingredient Phloretin

  • A growing number of healthy dietary ingredients in fruits and vegetables have been shown to exhibit diverse biological activities. Phloretin, a dihydrochalcone flavonoid that is abundant in apples and pears, has anti-inflammatory effects on ulcerative colitis (UC) mice. The gut microbiota and metabolism are closely related to each other due to the existence of the food-gut axis in the human colon. To investigate the interplay of faecal metabolites and the microbiota in UC mice after phloretin treatment, phloretin (60 mg/kg) was administered by gavage to ameliorate dextran sulfate sodium (DSS)-induced UC in mice. Gut microbes and faecal metabolite profiles were detected by high-throughput sequencing and liquid chromatography mass spectrometry (LC-MS) analysis, respectively. The correlations between gut microbes and their metabolites were evaluated by Spearman correlation coefficients. The results indicated that phloretin reshaped the disturbed faecal metabolite profile in UC mice and improved the metabolic pathways by balancing the composition of faecal metabolites such as norepinephrine, mesalazine, tyrosine, 5-acetyl-2,4-dimethyloxazole, and 6-acetyl-2,3-dihydro-2-(hydroxymethyl)-4(1H)-pyridinone. Correlation analysis identified the relations between the gut microbes and their metabolites. Proteus was negatively related to many faecal metabolites, such as norepinephrine, L-tyrosine, laccarin, dopamine glucuronide, and 5-acetyl-2,4-dimethyloxazole. The abundance of unidentified Bacteriodales_S24-7_group was positively related to ecgonine, 15-KETE and 6-acetyl-2,3-dihydro-2-(hydroxymethyl)-4(1H)-pyridinone. The abundance of Christensenellaceae_R-7_group was negatively related to the levels of 15-KETE and netilmicin. Stenotrophomonas and 15-KETE were negatively related, while Intestinimonas and alanyl-serine were positively related. In conclusion, phloretin treatment had positive impacts on faecal metabolites in UC mice, and the changes in faecal metabolites were closely related to the gut microbiota.

Comparison of Faecal Microbial Community of Lantang, Bama, Erhualian, Meishan, Xiaomeishan, Duroc, Landrace, and Yorkshire Sows

  • The objective of this study was to investigate differences in the faecal microbial composition among Lantang, Bama, Erhualian, Meishan, Xiaomeishan, Duroc, Landrace, and Yorkshire sows and to explore the possible link of the pig breed with the gut microbial community. Among the sows, the Meishan, Landrace, Duroc, and Yorkshire sows were from the same breeding farm with the same feed. Fresh faeces were collected from three sows of each purebred breed for microbiota analysis and volatile fatty acid (VFA) determination. Denaturing gradient gel electrophoresis (DGGE) analysis revealed that samples from Bama, Erhualian, and Xiaomeishan sows, which from different farms, were generally grouped in one cluster, with similarity higher than 67.2%, and those from Duroc, Landrace, and Yorkshire sows were grouped in another cluster. Principal component analysis of the DGGE profile showed that samples from the foreign breeds and the samples from the Chinese indigenous breeds were scattered in two different groups, irrespective of the farm origin. Faecal VFA concentrations were significantly affected by the pig breed. The proportion of acetate was higher in the Bama sows than in the other breeds. The real-time PCR analysis showed that 16S rRNA gene copies of total bacteria, Firmicutes and Bacteroidetes were significantly higher in the Bama sows compared to Xiaomeishan and Duroc sows. Both Meishan and Erhualian sows had higher numbers of total bacteria, Firmicutes, Bacteroidetes and sulphate-reducing bacteria as compared to Duroc sows. The results suggest that the pig breed affects the composition of gut microbiota. The microbial composition is different with different breeds, especially between overseas breeds (lean type) and Chinese breeds (relatively obese type).

문둥이박쥐(Eptesicus serotinus)의 배설물을 이용한 먹이원 분석 (The Diet Composition of the Serotine Bat, Eptesicus serotinus Revealed by Faecal Analysis)

  • 문둥이박쥐(Eptesicus serotinus)의 먹이원 구성을 파악하기 위하여 총 25개체의 암컷 성체로부터 배설물을 수집하여 분석하였다. 박쥐의 포획은 경북 안동시의 문둥이박쥐 집단 서식지가 확인된 교량을 대상으로 하였으며, 2014년 5월부터 9월까지 매월 5개체를 대상으로 하였다. 총 100개의 배설물이 수집되었으며, 고배율 현미경(50 ${\times}$ binocular microscope)을 이용하여 곤충의 목(Order)별로 구기(mouthpart), 다리(leg), 날개(wing), 더듬이(antenna), 딱지날개(elytra) 등 구분할 수 있는 특징을 이용하여 동정하였다. 배설물 분석결과 문둥이박쥐의 먹이원으로 이용된 분류군은 딱정벌레목(Coleoptera), 벌목(Hymenoptera), 파리목(Diptera), 노린재목(Hemiptera) 등 4개였다. 먹이원 가운데 딱정벌레목은 6월을 제외하고 5, 7, 8, 9월 모두에서 확인되었으며, 특히 5월과 7월에는 오직 딱정벌레목만 확인되었다. 그 외 노린재목, 파리목, 벌목은 각각 9월, 6월, 8월에 먹이원으로 이용되는 것으로 나타났다.

Predicting N-alkane Concentration in Pastures and Deer Faeces for Dietary Composition and Digestibility Measurement Using Near Infrared Spectroscopy

  • The alkanes in plant materials can potentially be used as markers to estimate composition and digestibility of diet of deer for the development of feeding strategies, but the analysis of alkanes in plant materials and deer faeces is time-consuming and expensive. In this study, 242 faecal samples and 119 pasture samples were scanned using near infrared spectroscopy (NIR) and the concentrations of alkanes in these samples were analysed to develop calibrations for predicting alkane concentrations in pastures and deer faeces. The $R^2$ values for NIR calibrations were <0.6 for $C_{24}$, $C_{26}$, $C_{32}$ and $C_{36}$, but were >0.8 for other alkanes for faecal samples. The $R^2$ values were >0.87 for alkanes with chains from $C_{27}$ to C35 for pasture samples. However, NIR was unable to predict concentrations of alkanes with chains of $C_{24}$, $C_{26}$, $C_{32}$ and $C_{36}$ in faecal samples and $C_{24}$, C25, $C_{26}$ and $C_{36}$ in pasture samples. While the use of these NIR calibrations will accelerate the estimation of diet digestibility, dietary components and botanical composition of pastures, the influence of the type of pasture on NIR calibration will require further examination.

MEASUREMENTS OF METABOLIC PARAMETERS IN LACTATING DAIRY COWS WITH NEAR INFRARED SPECTROSCOPY ANALYSIS USING CATTLE FAECAL SAMPLES

  • The knowledge of the nutrition in animal production is difficult to assess due of difficulty to determine the amount and quality of feeds intake, mainly if forages are the most important compound in the diet. It must be able to know responses to the metabolic process in lactating dairy cows earring out feeds evaluation trials. These metabolism studies with cows, requires measurements of: I) The amount of all feeds consumed. II) Excretion of faeces. III) Excretion of urine. Taking as a basis these trials, forage and total intake, dietary digestibility and balances of nitrogen and energy can be calculated. However, these feeds evaluation experiments with animals are very time consuming and expensives. The faeces excreted by animals containt undigested residues of the diet consumed. For this reason, their analysis can be an successful tool to determine the amount and quality of feed intake and other important biological parameters. The aim of this work was to know if faeces analysis by NIR could be used to determine with enough accuracy some attributes of different lactating dairy cows diets, using a global equation, developed on an heterogeneus population. For this purpose a total of 79 faecal samples from eight grass, three grass silages and two maize silages metabolic trials, on six cows each one, were used to constitute the initial population. The calibration equations were developed to predict forage and total intake, organic matter digestibility, digestibility coefficient of crude energy and digestibility energy. The combination of several trials with different diets and animal conditions gave promising results.

Effects of Bacillus subtilis KN-42 on Growth Performance, Diarrhea and Faecal Bacterial Flora of Weaned Piglets

  • This research focused on the effects of different doses of Bacillus subtilis KN-42 on the growth performance, diarrhea incidence, faecal bacterial flora, and the relative number of Lactobacillus and Escherichia coli in faeces of weaned piglets to determine whether the strain can serve as a candidate antimicrobial growth promoter. A total of 360 piglets (initial body weight $7.14{\pm}0.63$ kg) weaned at $26{\pm}2$ days of age were randomly allotted to 5 treatment groups (4 pens per treatment with 18 pigs per pen) for a 28-day trial. Dietary treatments were basal diet without any antimicrobial (negative control; NC), basal diet supplemented with 120 mg/kg feed of neomycin sulfate (positive control; PC) and basal diet supplemented with $2{\times}10^9$ (L), $4{\times}10^9$ (M) and $20{\times}10^9$ (H) CFU/kg feed of B. subtilis KN-42. During the overall period, average daily gain and feed efficiency of piglets were higher in groups PC, M, and H than those in group NC (p<0.05), and all probiotics and antibiotics groups had a lower diarrhea index than group NC (p<0.05). The 16S rDNA gene-based methods were used to analyze faecal bacterial flora on day 28 of experiment. The result of denaturing gradient gel electrophoresis analysis showed that supplementation of B. subtilis KN-42 to the diet changed the bacterial communities, with a higher bacterial diversity and band number in group M than in the other four groups. Real-time polymerase chain reaction analysis showed that the relative number of Lactobacillus were higher in groups PC and H than in group NC (p<0.05), and the supplemented B. subtilis KN-42 to the diet also reduced the relative number of E. coli (p<0.05). These results suggest that dietary addition of B. subtilis KN-42 can improve the growth performance and gastrointestinal health of piglets.

  • 돼지의 이유자돈 설사병을 효과적으로 치료, 예방하고자 Enrofloxacin(Baytril premix)을 사료에 혼합투여하여 설사 발생여부, 증체량, 사료요구율, 돈분내 대장균수의 변화, 분리한 대장균의 항균물질에 대한 감수성을 조사하여 다음과 같은 결과를 얻었다. 1. 이유자돈에 Baytril premix를 급여하였던 바 대조구보다 62일령에서 체중은 3.2kg의 통계적 유의차이를 보여(p<0.01) 현저한 증체효과 나타내었으며, 사료요구율도 계선되었고 설사발생도 예방할 수 있었다. 2. Baytril Premix를 급여한 자돈의 분변내 대장균수는 3.14$\times$$10^{8}$에서 5일후 2$\times$$10^{7}$ 개/g 10일후 1$\times$$10^{5}$개/g으로 현저히 감소하였다. 3. 공시농장에서 분리한 E. coli 10중의 항균물질에 대한 감수성은 baytril, gentamicin 및 trimeoprim+sulfamethoxazole에 감수성이 우수하였다.다.

Effect of dietary mannanoligosaccharide supplementation on nutrient digestibility, hindgut fermentation, immune response and antioxidant indices in dogs

  • Background: Use of prebiotics in companion animal nutrition is often considered advantageous over probiotics because of the ease of handling, ability to withstand processing and storage etc. While most of the studies on prebiotic use in dogs have been done with processed food as basal diet, the response in relation to homemade diet feeding is not very well explored. Methods: The study was conducted to evaluate the effects of dietary mannanoligosaccharide (MOS) supplementation on nutrient digestibility, hindgut fermentation, immune response and antioxidant indices in dogs. Ten Spitz pups were divided into two groups: control (CON) with no supplementation, and experimental (MOS) wherein the basal diet was supplemented with MOS at 15 g/kg diet. All dogs were fed on a home-prepared diet for a period of 150 days. The study protocol included a digestion trial, periodic blood collection and analysis for lipid profile and erythrocytic antioxidants. Immune response of the animals was assessed towards the end of the feeding period. Results: Results revealed no significant (P > 0.05) variations in palatability score, intake and apparent digestibility of nutrients between the groups. Faecal score, faeces voided, faecal pH, concentrations of ammonia, lactate and short-chain fatty acids were comparable (P > 0.05) between the two groups. Cell-mediated immune response, assessed as delayed-type of hypersensitivity response, was significantly higher (P < 0.05) in the MOS group. The percent of lymphocyte sub-populations CD4+ and ratio of CD4+:CD8+ were also significantly (P < 0.05) higher in MOS group. The serum IgG levels were similar (P > 0.05) in both the groups. Supplementation of MOS lowered (P < 0.05) serum total- and LDL- cholesterol levels, when compared with the control group. The erythrocytic antioxidant indices were similar (P > 0.05) between the two groups. Conclusions: The results indicated that supplementation of MOS at the rate of 15 g/kg in the diet of dog augmented the cell-mediated immune response and serum lipid profile without any influences on digestibility of nutrients, hindgut fermentation and antioxidants indices.

Helicobacter pylori Infection and Risk Factors in Relation to Allergy in Children

  • Purpose: To analyze presence of Helicobacter pylori infection and environmental risk factors among children with and without allergy. Methods: Parents of children at primary health care centres/kindergartens and allergologist consultation were asked to answer a questionnaire and to bring a faecal sample. H. pylori infection was detected by monoclonal stool antigen test. Prevalence of H. pylori infection and risk factors were compared between individuals with and without allergy using ${\chi}^2$ test, ANOVA test and logistic regression. Results: Among 220 children (mean age, 4.7 years; ${\pm}standard$ deviation 2.3 years) H. pylori positivity was non-significantly lower among patients with allergy (n=122) compared to individuals without allergy (n=98): 13.9% (17/122) vs. 22.4% (22/98); p=0.106. In logistic regression analysis presence of allergy was significantly associated with family history of allergy (odds ratio [OR], 8.038; 95% confidence interval [CI], 4.067-15.886; p<0.0001), delivery by Caesarean section (OR, 2.980; 95% CI, 1.300-6.831; p=0.009), exclusive breast feeding for five months (OR, 2.601; 95% CI, 1.316-5.142; p=0.006), antibacterial treatment during the previous year (OR, 2.381; 95% CI, 1.186-4.782; p=0.015). Conclusion: Prevalence of H. pylori infection did not differ significantly between children with and without allergy. Significant association of allergy with delivery by Caesarean section and antibacterial therapy possibly suggests the role of gastrointestinal flora in the development of allergy, while association with family history of allergy indicates the importance of genetic factors in the arise of allergy.

Use of Dual-Energy X-ray Absorptiometry in Children with Inflammatory Bowel Disease: A Large Single Centre Study

  • Purpose: Low bone mineral density (BMD) is a complication in children with inflammatory bowel disease (IBD). There are limited data evaluating dual-energy x-ray absorptiometry (DXA) as a screening tool for low BMD in children with IBD. We performed a single site retrospective analysis of DXA use. Methods: Children aged 5-18 years with IBD diagnosed between 2013 to 2017 at the Royal Children's Hospital, Australia, were included. Patient demographics, measures of disease activity, DXA scores, and factors related to BMD were collected. Results: Over a median follow up of 5.1 (4-6.4) years, 72/239 (30.1%) children underwent DXA, and 28/239 (11.7%) children had a second DXA. Our DXA practice differed to consensus guidelines regarding initial screening based on height and/or body mass index (BMI) z-score (8/17 [47.1%]), and repeat surveillance (13/42 [31.0%]). Children had a median lumbar spine (LS) z-score -0.80 (-1.65-0.075). Children with LS z-score≤-2.0 (n=14) had lower weight (6.57 [1.78-23.7] vs. 51.1 [26.5-68.7], p=0.0002) and height centiles (3.62 [1.17-17.1] vs. 42 [16.9-67.1], p=0.0001), and higher faecal calprotectin (FCP) (3041 [1182-4192] vs. 585 [139-2419], p=0.009) compared to children with LS z-score>-2.0. No fractures were reported. Of 28 children who underwent a second DXA 1.6 (1.1-2.2) years following initial DXA, no significant change in z-scores occurred. Conclusion: Children with IBD had low BMD. In addition to height centile and weight centile, FCP was associated with lower BMD, and should be considered in DXA screening guidelines. Greater clinician awareness of DXA consensus guidelines is required. Future prospective studies are required.
