Expression of Laminin During the Differentiation of F9 Teratocarcinoma Stem Cell (F9 Teratocarcinoma Stem Cell의 분화에 따른 라미닌의 발현)

  • In order to investigate the retinoic acid indticed-differentiation of F9 teratocarcinoma stem cell, we have analyzed the change of cell morphology and laminin expression after exposure to retinoic add and cyclic AMP. It is shown that undifferentiated F9 stem cells grow as closely packed colonies, and it is difficult to distinguish cell-cell boundaries. After retinoic add and dibutyryl cyclic AMP treatment, F9 cells assume a flat morphology characterized by perinuclear granules and arrest growth. According to Northern blot analysis, laminin expression was increased markedly after retinoic acid treatment. Laminin Bi gene expression was increased at least 30-fold and laminin B2 gene expression was increased approximately 20-fold during differentiation process. Employing immunofluoresence analysis, it was proved that the synthesis of laminin protein was low level in F9 stem cell whereas it became high level in retinoic acid treated F9 cell and the laminin protein was largely accumulated in the cell surface. Our results suggest that induction of laminin Bi and B2 genes m F9 cells is retinoic acid-mediated control, and morphological change and differentiation of F9 cells might be associated with laminin gene expression.

Increased Expression of c-jun in the Bile Acid-Induced Apoptosis in Mouse F9 Teratocarcinoma Stem Cells

  • Ursodeoxycholic acid (UDCA) and lithocholic acid (LCA), secondary bile acids, have been shown to have a cell differentiation activity in mouse F9 teratocarcinoma stem cells. Treatment with bile acids induced morphological changes, including cytoplasmic and nuclear membrane blebbing, aggregation of organelles, and chromatin condensation, corresponding to apoptosis. Moreover, the bile acids induced intemucleosomal DNA fragmentation, a hallmark of apoptosis. In addition, the expression of c-jun was increased, but that of c-myc and laminin was decreased during apoptosis induced by the bile acids in F9 cells. These results suggest that the bile acids can induce apoptosis in F9 cells. Furthermore, the c-jun expression may be related to the apoptosis induced by UDCA or LCA in F9 cells.

Differentiation Effect of Marine Natural Compounds on F9 Teratocarcinoma Stem Cells (F9 기형암종 세포에 대한 해양천연물질의 분화 유도 작용)

  • It has been known that many kinds of cancer are caused by continued proliferation or abnormal differentiation. Thus, recent approaches to anticancer therapy have been focused on developing drugs that induce differentiation of cancer cells to normal cells. A typical differentiation agent, all trans-retinoic acid, is unsuitable for anticancer drug because all trans-retinoic acid produces unfavorable side effects and cytotoxicity in normal cells. Therefore, we have screened some new differentiation-inducing compounds obtained from marine organisms using F9 teratocarcinoma stem cells as a model system. We observed that fatty acid. glycolipid, saponin, sphingosine and sterol compounds of marine organisms had differentiation-inducing activity in F9 cells, were determined by morphological changes and Northern blot analysis. The expression of differentiation marker genes, such as laminin B1, type IV collagen and retinoic acid receptor beta were induced by treatment with those compounds.

Identification of Nuclear Receptors by RT-PCR in F9 Cells Induced by Ginsenosides

  • Ginsenosides $Rh_1$ and $Rh_2$ Induced the differentiation of F9 teratocarcinoma stem cells. These agents are structurally similar to the steroid hormones, therefore, we speculated that the steroid receptor (s) or novel nuclear receptor (s) could be involved in the differentiation process induces by them. Based on this speculation, we tried to alone new nuclear receptors with reverse transcription-polymerase chain reaction (RT-PCR) method by isolating RNA from F9 teratocarcinoma cells induced by ginsenosides. By using RT-PCR with degenerated primers from highly conserved DNA binding domain of nuclear receptors, we identified several nuclear receptors. In northern blot analysis we found that these clones are transcriptionally regulated by ginsenoside Rhl or Rh2 treatment. Further characterizations of these clones are needed to identify the mechanism of gene expression, which has an important role in the differentiation of F9 cells induced by ginsenosides.

F9 기형암종 세포의 분화에 따른 small GTP-binding protein변화

  • 세포분화에 따른 Small GTP-binding protein의 역할을 밝히기 위하여 Retinoic acid(RA)와 dibutyryl cyclic AMP(dbcAMP)로 분화를 유도한 F9 기형암종세포의 형태적인 변화와 함께 Small GTP-binding protein의 분포를 조사하였다. RA와 dbcAMP를 처리한 세포는 분화유도 5일경(초기 분화 단계)에 분명한 세포의 경계를 보이기 시작하여 7일경(분화 후기 단계)에는 거의 모든 세포가 등근 분화된 형태로 전환되었다. 이 분화과정 동안 세포막에는 많은 microvilli와 lamellopodia 같은 구조물이 나타났다. 아울러 초기 분화 단계에 많은 량의 laminin이 발현되었으며 분화 후기에 microtubule의 재분포가 관찰되었다. 세종류의 Small GTP-binding protein(25 23, 21 KD)이 F9 세포의 막성분과 세포질에서 관찰되었으며 분화가 진행됨에 따라서 세단백질 모두 증가되는 양상을 보였다 이러한 결과는 Small GTP-binding protein이 F9 세포의 분화에 특별한 기능을 가지고 있음을 시사해 주고 있다.
