Previously, we reported induced expression of developmentally regulated CTP-binding protein 2 (DRG2) in fish cells at the late stage of rhabdovirus infection. To investigate the biological role of fish DRG2 (fDRG2), we transfected CHSE-214 cells with an expression vector containing complete fDRG2 fused to the N-terminal end of an enhanced green fluorescent protein (EGFP). Low level expression of fDRG2-EGFP did not induce morphological change or cell death. However, a high level expression of fDRG2-EGFP induced cell rounding and caused depletion of the cell population in FACS analysis. Several truncated fragments were fused to EGFP. FACS analysis was conducted to determine the presence of cells expressing high levels of the resulting chimera. While cells expressing a high level of N-terminus were detected, those expressing high levels of the C-terminal fragment 243-290 containing the G4 motif were absent in FACS analysis. Based on these observations, we propose that overexpression of fDRG2 may induce cell rounding, a representative cytopathic effect of virus-infected cells in the late stage of infection and the C-terminus of the fDRG2 is essential for this function.

Also we calculate the probability generating function for the offspring number of genotype produced by a mating of the ith male and jth female under a diploid model of N population with N1 males and N2 females. Finally, we have conditional joint probability generating function of genotype frequencies.

A numerical simulation is presented for investigating the effects of pressure ratio of $D_2$ injector to supersonic nozzle on the population inversion in the DF chemical laser cavity, while a lasing concurrently takes place. The laser beam is generated between the mirrors in the cavity and it is important to obtain stronger population inversion and more uniform distribution of the excited molecules in the laser cavity in order to produce high power laser beam with good quality. In this study, these phenomena are investigated by means of analyzing the distributions of the DF excited molecules and the F atom used as an oxidant, while simultaneously estimating the maximum small signal and saturated gains and power in the DF chemical laser cavity. For the numerical solution, an 11-species (including DF molecules in various excited states of energies), 32-step chemistry model is adopted for the chemical reaction of the DF chemical laser system. The results are discussed by comparison with two $D_2$injector pressure cases; 192 torr and 388.64 torr. Major results reveal that in the resonator, stronger population inversions occur in the all transitions except DF(1)-DF(0), when the $D_2$injection pressure is lower. But, the higher $D_2$injection pressure provides a favorable condition for DF(1)-DF(0) transition to generate the higher power laser beam. In other words, as the pressure of $D_2$injector increases, the maximum small signal gain in the $V_{1-0}$ transition, which is in charge of generating most of laser power, becomes higher. Therefore, the total laser beam power becomes higher.r.

Schisandra nigra Max. has been cultivated far a medical use as well as food. It is an endemic species which has a unique habitat at the altitude of 600-1,400 m in Cheju island. In this study, three natural populations of S. nigra were investigated by using of starch-gel electrophoresis to determine the extent and distribution of genetic diversity. Except 2 monomorphic locus (Mdh-2 and Pgi-1), 4 of the 6 isozyme locus (Idh, Mdh-2, Mnr, and Pgi-2), verified from 4 isozymes, revealed polymorphism in the three populations of S. nigra. The mean number of allele per locus was 1.7 and the percentages of polymorphism loci were 38.9% at 95% level and 50% at 99% level respectively. The observed and estimated heterozygosities were 0.141 and 0.147 respectively. Although plants which were in the face of crisis and distributed in the restricted area, have been known to the very low degree of genetic variation, S. nigra showed higher genetic variation than others. Genetic variation was mostly allocated within population and individuals than that among populations. The result of Wright's F analysis estimates of $F_{IT}$ and $F_{IT}$ showed that S. nigra population revealed Hardy-Weinberg steady state.
This study was conducted to provide basic data to develop a smoking prevention and non-smoking education program. Data were collected, using a questionnaire, from 438 male middle school students living in Chunchon, Kwangwon- do. The data collection period was from September 2 to 30, 2000. The questionnaire used to measure the subjects' knowledge and attitude toward smoking was based on the guideline for the contents of tobacco smoking surveys for the general population designed by WHO. The collected data were analyzed using the SPSS-Win program. The results were as follows. 1. The subjects' average score for smoking knowledge was 58.52 out of 100 points. 2. The subjects' average score for attitude toward smoking was 63.54 out of 100 points. 3. Smoking knowledge of the subjects was significantly different by their grade (F=9.187, p=.000), father's smoking behavior (t=2.261, p= .024), aware of harmfulness of smoking (F=8.911, p=.000). The subjects' attitude toward smoking was significantly different by mother's smoking behavior (t=-2.557, p=.011), brother's smoking behavior (t=-3.959, p=.000), having a smoking friend or not (t=-4.422, p=.000), subjects' smoking experience (t=-8.304, p=.000), present smoking behavior (t=7.001, p=.000), quantity of smoking (F=3.297, p= .041), abstinence period of smoking (F=3.858, p=.013), thinking about smoking behavior (F=20.999, p=.000), aware of harmfulness of smoking (F=20.964, p=.000), and amount of drinking (F=8.316, p=.000). 4. There was a significant correlation between subjects' knowledge and attitude toward smoking (r=.514, p=.000).
This study was conducted to examine effects of mating type on the growth traits in an $F_2$ population produced by reciprocal intercrosses between Landrace and the Jeju Native Black pig (JBP). The $F_2$ progeny were produced by two different mating types based on the grand dams of founder breeds JBP (Cross_1) and Landrace (Cross_2). The body weights at 21 days after birth (BW21D) was significantly different between Cross_1 and Cross_2 (P<0.05), showing that the BW21D of Cross_1 has about 0.25 kg heavier than Cross_2. The significant differences were found between males and females for the growth traits including the body weights (BWB, BW21D, BW70D and BW140D) and average daily gains (ADG, eADG and lADG) (P<0.05). Males were heavier BWB, BW21D and BW140D levels, and higher ADG and lADG levels than females. On the other hand, females had heavier BW70D and higher eADG levels than those of males. When considering the mating types and sex simultaneously the Cross_2 males had the heaviest BW140D among the combinations of cross and sex. In conclusion, it is desirable to choose Landrace as grand dams in the reciprocal intercrosses between Landrace and JBP for producing their progeny construction and to plan the production of $F_2$ males for industrial purposes. These results suggested that it may be one of useful strategies to improve the productivity through out selection of the mating type of founder breeds and the progeny sex, especially in Landrace, JBP and their related populations.
Objective: Previously, we reported quantitative trait loci (QTLs) affecting backfat thickness (BFT) traits on pig chromosome 5 (SW1482-SW963) in an F2 intercross population between Landrace and Korean native pigs. The aim of this study was to evaluate glutamate receptor-interacting protein 1 (GRIP1) as a positional candidate gene underlying the QTL affecting BFT traits. Methods: Genotype and phenotype analyses were performed using the 1,105 $F_2$ progeny. A mixed-effect linear model was used to access association between these single nucleotide polymorphism (SNP) markers and the BFT traits in the $F_2$ intercross population. Results: Highly significant associations of two informative SNPs (c.2442 T>C, c.3316 C>G [R1106G]) in GRIP1 with BFT traits were detected. In addition, the two SNPs were used to construct haplotypes that were also highly associated with the BFT traits. Conclusion: The SNPs and haplotypes of the GRIP1 gene determined in this study can contribute to understand the genetic structure of BFT traits in pigs.
The present study was conducted to compare the relative efficiency of four different breeding methods in tobacco varietal development. A single Fl hybrid plant from cross of two flue-cured cultivars of Nicotiana tabacum L Bright Yellow 4(BY4) and NC95, was selfed. F2population above cross was screened for resistance against bacterial wilt caused by Pseudommonas solanacearum E.F.Smith under the naturally infested field conditions, and the 30 lines were developed from F2 individual plant by anther culture (ADH), maternal method utilizing Nicotiana afpicana (MDH), single- seed descent(SSD) and Bulk breeding method, respectively. All characters except content of total alkaloids of ADH and MDH which wore bred by haploid methods reduced more than that of lines bred by conventional methods(SSD & Bulk) : however, the yields were 8% lower than other lines. The total alkaloid content of ADH was higher than that of MDH, and yield was reduced about 4n even though the number of leaf was identical with the MDH. All other characters of ADH were also reduced significantly. In the lines bred by conventional methods, population developed by SSD showed significantly wider leaf width, shorter plant height, later days to flower, and lower in percent reducing sugar than those by Bulk. The populations derived from haploid method showed greater phenotypic variance and wider range of variation than conventionally developed ones. The results obtained indicate that selection will provide a significantly greater genetic gain for leaf number and leaf length in the ADU and MDH populations, and for plant height and days to flower in the SSD and Bulk populations.
The aim of this study was to detect positional candidate genes located within the support interval (SI) regions based on the results of red blood cell, mean corpuscular volume (MCV), and mean corpuscular hemoglobin quantitative trait locus (QTL) in Sus scrofa chromosome 13, and to verify the correlation between specific single-nucleotide polymorphisms (SNPs) located in the exonic region of the positional candidate gene and the three genetic traits. The flanking markers of the three QTL SI regions are SW38 and S0215. Within the QTL SI regions, 44 genes were located, and runt-related transcription factor 1, dual-specificity tyrosine-(Y)-phosphorylation regulated kinase 1A (DYRK1A), and potassium inwardly-rectifying channel, subfamily J, member 15 KCNJ15-which are reported to be related to the hematological traits and clinical features of Down syndrome-were selected as positional candidate genes. The ten SNPs located in the exonic region of the three genes were detected by next generation sequencing. A total of 1,232 pigs of an $F_2$ resource population between Landrace and Korean native pigs were genotyped. To investigate the effects of the three genes on each genotype, a mixed-effect model which is the considering family structure model was used to evaluate the associations between the SNPs and three genetic traits in the $F_2$ intercross population. Among them, the MCV level was highly significant (nominal $p=9.8{\times}10^{-9}$) in association with the DYRK1A-SNP1 (c.2989 G$F_2$ intercross, our approach has limited power to distinguish one particular positional candidate gene from a QTL region.
The difference in the breeding programs and population history may have diversely shaped the genomes of Korean native cattle breeds. In the absence of phenotypic data, comparisons of breeds that have been subjected to different selective pressures can aid to identify genomic regions and genes controlling qualitative and complex traits. In this study to decipher genetic variation and identify evidence of divergent selection, 3 Korean cattle breeds were genotyped using the recently developed high-density GeneSeek Genomic Profiler F250 (GGP-F250) array. The three Korean cattle breeds clustered according to their coat color phenotypes and breeding programs. The Heugu breed reliably showed smaller effective population size at all generations considered. Across the autosomal chromosomes, 113 and 83 annotated genes were identified from Hanwoo-Chikso and Hanwoo-Heugu comparisons, respectively of which 16 genes were shared between the two pairwise comparisons. The most important signals of selection were detected on bovine chromosomes 14 (24.39-25.13 Mb) and 18 (13.34-15.07 Mb), containing genes related to body size, and coat color (XKR4, LYN, PLAG1, SDR16C5, TMEM68, CDH15, MC1R, and GALNS). Some of the candidate genes are also associated with meat quality traits (ACSF3, EIF2B1, BANP, APCDD1, and GALM) and harbor quantitative trait locus (QTL) for beef production traits. Further functional analysis revealed that the candidate genes (DBI, ACSF3, HINT2, GBA2, AGPAT5, SCAP, ELP6, APOB, and RBL1) were involved in gene ontology (GO) terms relevant to meat quality including fatty acid oxidation, biosynthesis, and lipid storage. Candidate genes previously known to affect beef production and quality traits could be used in the beef cattle selection strategies.
