Specific and Sensitive Detection of Venturia nashicola, the Scab Fungus of Asian Pears, by Nested PCR

  • The fungus Venturia nashicola is the causal agent of scab on Asian pears. For the rapid and reliable identification as well as sensitive detection of V. nashicola, a PCR-based technique was developed. DNA fingerprints of three closely related species, V. nashicola, V. pirina, and V. inaequalis, were obtained by random amplified polymorphic DNA (RAPD) analysis. Two RAPD markers specific to V. nashicola were identified by PCR, after which two pairs of sequence characterized amplified region (SCAR) primers were designed from the nucleotide sequences of the markers. The SCAR primer pairs, designated as D12F/D12R and E11F/E11R, amplified 535-bp and 525-bp DNA fragments, respectively, only from genomic DNA of V. nashicola. The specificity of the primer sets was tested on strains representing three species of Venturia and 20 fungal plant pathogens. The nested PCR primer pair specific to V. nashicola was developed based on the sequence of the species-specific 525-bp DNA fragment amplified by primer set E11F/E11R. The internal primer pair Na11F/Na11R amplified a 235-bp fragment from V. nashicola, but not from any other fungal species tested. The nested PCR assay was sensitive enough to detect the specific fragment in 50 fg of V. nashicola DNA.

Characteristics Analysis of Class E Frequency Multiplier using FET Switch Model (FET 스위치 모델을 이용한 E급 주파수 체배기 특성 해석)

  • This paper has presented research results for the switching mode class E frequency multiplier that has simple circuit structure and high efficiency. Frequency multiplication is coming from the nonlinearity of the active component, and this paper models the FET active component as a simple switch and some parasitics to analyze the characteristics. The matching component parameters for the class E frequency doubler have been derived with modeling the FET as a input controlled switch and some parasitics. A circuit simulator, ADS, is used to simulate the output voltage and current waveform and efficiency with the variation of the parasitic values. With 2.9GHz input and 2V bias, the drain efficiency has been decreased from 98% to 28% with changing the parasitic capacitance from 0pF to 1pF at 5.8GHz output, which shows that the parasitic capacitance CP has the most significant effect on the efficiency among the parasitics of FET.

  • Bis(phosphine)ruthenium derivatives $({\eta}^5-C_5Me_5)RuCl(PR_3)_2(PR_3=PMe_3,\; PMe_2Ph,\;PEt_3,\;PMePh_2$, 1/2DPPE, 1/2DPPB) (2a${\sim}$2f) have been synthesized by the reaction of $[({\eta}^5-C_5Me_5)RuCl_2]_2$ (1) with excessive phosphine in ethanol. The reaction of complexes $({\eta}^5-C_5Me_5)Ru(PR_3)_2Cl\;with\;NaBH_4$ in ethanol gave the corresponding hydride complexes $({\eta}^5-C_5Me_5)Ru(PR_3)_2H (PR_3=PMe_3, PEt_3, PMePh_2$, 1/2 DPPE, 1/2DPPB) (3a${\sim}$3e). Chloride complexes (2a${\sim}$2f) and hydride complexes (3a${\sim}$3e) were isolated as crystals, which were characterized by IR, $^1H-NMR$ , and elemental analysis.

Serological Studies on the Specific Antibodies Against P-pili of Uropathogenic Escherichia coli (요로 감염환자에서 혈청학적 방법을 이용한 P-pili특이혈중 항체의 조사)

  • Escherichia coli is one of the most common etiological agents in urinary tract infection. An important virulence factor is the adhesive capacity of E. coli to uroepithelial cell, mediated by bacterial fimbriae. The Adhesion property has been regarded as an important virulence determinant in urinary tract infections. A total of 60 patients, who were diagnosed microbiologically as urinary tract infections, were examined by immunoblotting and enzyme-linked immunosorbent assay(ELISA). Uropathogenic E. coli with recombinant plasmid were positive for mannose resistant hemagglutination (MRHA). For identification of p-fimbriae subtype in uropathogenic E. coli, In the immunoblot analysis, specific bands in the range of p-fimbriae molecular weight of 17KD-22KD were identified. For the distribution of p-fimbriae subtype in the patient sera, 34/60(56.7%) were positive for $F7_1$, 28/60(46.7%) were positive for $F7_2$, and 30/60(50%) were positive for F13 with immunoblotting method. similar trends were observed in the enzyme-linked immunosorbent assay. Relatively good specificity(92.6%) and sensitivity(90%) were found in the ELISA test system using mixed antigens of purified $F7_1$, $F7_2$, and F13 p-fimbriae, and 60 sera from patients with urinary tract infections. In conclusion The serological tests were convenient method in diagnosis of urinary tract infections. among those ELISA could be recommended in diagnosis of urinary tract infections.

  • Linear free energy relation ship(LFER) on the insecticidal activity of O,O-diethylphenylphosphate (A) and 3,5-dimethylphenyl-O,O-diethylphosphate (B) derivatives were studied by EHT MO calculation method and regression analysis method. LFER between varying substituent constants and $pI_{50}$ constants of phosphates, (A) & (B) were calculated with applying Hammett, Okamoto-Brown, Taft and Swain-Lupton's DSP equations;percent resonance effect(R) and field effect(F) of (A) were %R=33.5 & %F=66.5 and also that of (B) were %R=2 & %F=98, respectively. On the basis of above findings, the insecticidal activities were similar for both (A) and (B), but (B) have larger field and inductive contribution than (A), due to the 3,5-dimethyl group of (B).

  • Ca $F_2$ films have superior gate insulator properties than conventional gate insulator such as $SiO_2$, Si $N_{x}$, $SiO_{x}$, and T $a_2$ $O_{5}$ to the side of lattice mismatch between Si substrate and interface trap charge density( $D_{it}$). Therefore, this material is enable to apply Thin Film Transistor(TFT) gate insulator. Most of gate oxide film have exhibited problems on high trap charge density, interface state in corporation with O-H bond created by mobile hydrogen and oxygen atom. This paper performed Ca $F_2$ property evaluation as MIM, MIS device fabrication. Ca $F_2$ films were deposited at the various substrate temperature using a thermal evaporation. Ca $F_2$ films was grown as polycrystalline film and showed grain size variation as a function of substrate temperature and RTA post-annealing treatment. C-V, I-V results exhibit almost low $D_{it}$(1.8$\times$10$^{11}$ $cm^{-1}$ /le $V^{-1}$ ) and higher $E_{br}$ (>0.87MV/cm) than reported that formerly. Structural analysis indicate that low $D_{it}$ and high $E_{br}$ were caused by low lattice mismatch(6%) and crystal growth direction.   • Recently multiple process communicate together. They share resource and information for cooperation in complicated programming environment. Kernel provides IPC (Inter -Process Communication) for communication with each other process. Shared Memory is a technique that many processes can access to identical memory area in the Linux environment. In this paper, we propose a performance improvement method of shared memory in the dual-core embedded linux system which is consist of different core and different operating system. We construct the MPC2530F (ARM926F+ARM946E) linux system and measure the performance therein. We attempt a performance enhancement in each CPU for each process which uses a shared memory.

  • 상사실험법(analogous experimental method)이라 함은 물리적현상을 다른 물리적현상으로 변환 하여, 후자를 실험적으로 측정하여 전자의 제반 물리량을 얻는 과정을 말한다. 이 때 두 물리량 사이에는 수학적 상사관계, 특히 미분방정식 상의 유사관계가 성립함을 전제로 한다. 일반적으로 임의형상의 내부응력을 실험적으로 해석하는 데는 탄소성 변위를 직접 전기적저항으로 바꾸어 측정하는 방법(strain gauge method)이나 광파의 간섭무늬(fringe)로 가시화하는 광탄성 법(photoelastic method), 또는 전자계산기를 이용하여 분할요소해석의 연계집적으로서 얻는 유 한요소법(F.E.M) 등이 널리 사용되고 있으나, 이들은 다같이 그 나름대로의 장단점을 지니고 있다. 전기저항식은 변형을 직접 측정할 수 있어 측정의 오차를 줄일 수 있고, 특히 실물측정과 동하중 해석에는 큰 강점이 있으나, 점해석(point by analysis)이기 때문에 전시야적인 분포를 파악하기 어렵다. 또한 광탄성법은 명료한 전시야적 분포를 얻을 수 있지만 모형해석(model analysis)이기 때문에 정밀한 모형제작의 어려움이 수반되며, F.E.M.(B.E.M.도 포함)은 복잡한 형상에서의 요소분할이 매우 어렵고, 경계조건의 정확한 설정에 문제가 있다. 따라서 여러 실험적 방법은 실측대상에 따라 그 장단점을 감안하여 선택되어야 하며, 이 글에서 논술하고자 하는 상사실험법에 의한 응력해석도 이러한 관점에서 지금까지의 일련의 연구결과를 종합하여 그 효 용적인 용도, 응용 및 그 전망과 더불어 장차 해결하여야 할 2,3의 문제를 제시하고자 한다.

Effects of H-type Sleeper on the Track Behavior (궤도거동에 대한 H형 침목의 영향)

  • The demand for improved rail transportation safety is growing due to the increased speeds of current railways. In addition, freight trains with heavier axle loads are required to run on conventional railways. In order to meet these requirements, increased rail weights and the application of a CWR system have been introduced on conventional railways. The H type sleeper, which has higher lateral resistance than the existing mono sleepers, has been recently developed to increase track stiffness and reduce track irregularities. In this paper, the effects of a H-type sleeper on track behavior were investigated by 3-Dimensional F.E. analysis.

A Prospective Study on an Association between Apolipoprotein E ${\varepsilon}4$ and Cognitive Change in Community-Dwelling Elders with Alzheimer's Disease (일 지역 알츠하이머병 노인에서 Apolipoprotein E ${\varepsilon}4$와 인지변화의 연관에 대한 전향적 연구)

  • Objectives : The aim of this study was to examine the prospective impact of the apolipoprotein E (APOE) ${\varepsilon}4$ on cognitive performance in the community-dwelling elderly individuals with Alzheimer's disease (AD). Methods : The total number of subjects was 30 (12 men and 18 women) who were diagnosed with AD from a Korean project of "Early Detection of Dementia". People aged 65-85 years were included in the analysis. The eight neuropsychological domains from the Korean version of Consortium to Establish a Registry of Alzheimer's Disease (CERAD-K) were conducted to test subjects. They have been followed at 24-month intervals with the same assessments at each interval. Their cognitive performance at 2 year intervals was compared by the occurrence of the APOE ${\varepsilon}4$. Results : The impact of ${\varepsilon}4$ allele was significant in the Word List Memory Test (WLMT, F = 4.345, df = 1, p = 0.021) and Word List Recall Test (WLRT, F = 5.569, df = 1, p = 0.033). Conclusions : The APOE ${\varepsilon}4$ allele was significantly correlated especially with verbal episodic memory domain in community-dwelling elders diagnosed with AD.
