Inhibition of Melanin Production and Tyrosinase Expression of Ergosterol Derivatives from Phellinus pini

  • Three ergosterol derivatives, ergosta-4,6,8(14),22-tetraen-3-one (1), ergosta-7,24(28)-dien-3-ol (2), and 5,8-epidioxyergosta-6,22-dien-3-ol(3) were isolated from the fruit body of Phellinus pini. Their structures were based on spectroscopic methods including IR, MS, and NMR (1D and 2D). These compounds were evaluated for their activity to decrease melanin production in ${\alpha}$-MSH (melanocyte stimulating hormone) activated B16F10 cells. Compound 1, 2, and 3 reduced melanin content in a dose-dependent manner at concentrations of 5~15 uM. They also suppressed the tyrosinase expression of protein and m-RNA level dose dependently by western blot analysis and RT-PCR experiment in B16F10 murine melanoma cells.

CONTINUOUS HAMILTONIAN DYNAMICS AND AREA-PRESERVING HOMEOMORPHISM GROUP OF D2

  • The main purpose of this paper is to propose a scheme of a proof of the nonsimpleness of the group $Homeo^{\Omega}$ ($D^2$, ${\partial}D^2$) of area preserving homeomorphisms of the 2-disc $D^2$. We first establish the existence of Alexander isotopy in the category of Hamiltonian homeomorphisms. This reduces the question of extendability of the well-known Calabi homomorphism Cal : $Diff^{\Omega}$ ($D^1$, ${\partial}D^2$)${\rightarrow}{\mathbb{R}}$ to a homomorphism ${\bar{Cal}}$ : Hameo($D^2$, ${\partial}D^2$)${\rightarrow}{\mathbb{R}}$ to that of the vanishing of the basic phase function $f_{\underline{F}}$, a Floer theoretic graph selector constructed in [9], that is associated to the graph of the topological Hamiltonian loop and its normalized Hamiltonian ${\underline{F}}$ on $S^2$ that is obtained via the natural embedding $D^2{\hookrightarrow}S^2$. Here Hameo($D^2$, ${\partial}D^2$) is the group of Hamiltonian homeomorphisms introduced by $M{\ddot{u}}ller$ and the author [18]. We then provide an evidence of this vanishing conjecture by proving the conjecture for the special class of weakly graphical topological Hamiltonian loops on $D^2$ via a study of the associated Hamiton-Jacobi equation.

  • 레티노이드 효과가 나타나는 것을 목적으로 하여 다양한 히드록삼산 유도체를 합성하였고, 레티노익산 수용체에 대한 전사 활성을 측정하여 스크리닝하였다. 합성된 화합물 중에서 N-(4-N-hydroxycarbamoyl)phenyl) [4-(tort-butyl)phenyl] carboxamide (2f)가 수용체에 대한 가장 강력한 적합성을 나타내었다. 히드록삼산 구조에서 산도를 나타내는 히드록시기는 쉽게 이온화하여 음이온을 형성한다. 형성된 히드록삼산의 음이온은 레티노익산의 음이온과 유사한 역할을 한다. 이와 같이 히드록삼산 화합물이 레티노이드 효과를 나타내는 예는 이전에 알려진 바가 없다. 화합물 2f의 레티노이드 효과는 레티노이드와 연관된 유전자의 발현 증가 효과로 한번 더 검증하였다. 노화 원료로서의 가능성을 확인하기 위해서 MMP-1의 발현 억제능을 레티노익산, 레티놀과 비교하여 살펴보았다. 10 uM 농도에서 화합물 2f는 MMP-1의 발현을 억제하였다. 이와 같은 결과로 화합물 2f는 항노화 원료로서의 가능성을 가지고 있다.

Dental Age측정에 관한 연구 (A STUDY ON THE MEASUREMENT OF DENIAL AGE)

  • Dental maturity is one of the index of physiological maturity indicators. To investigate the relationship between dental maturity and chronological age, the author took the orthopantomograms of 984 students, aged 7 through 17 years, having normal occlusion. The orthopantomograms were examined and calcification degree of each tooth on the left side was rated according to the method described by Demirjian. On the basis of findings of this study, the following results were obtained. 1. The root completion periods of mandibular permanent teeth were as follows; Central Incisor M $8.32{\pm}1.03\;years$ F $7.96{\pm}1.04\;years$ Lateral Incisor M $9.40{\pm}1.30\;years$ F $9.01{\pm}0.90\;years$ Canine M $12.81{\pm}1.24\;years$ F $11.42{\pm}0.94\;years$ 1st Premolar M $12.76{\pm}1.74\;years$ F $12.19{\pm}1.33\;years$ 2nd Premolar M $13.31{\pm}1.88\;years$ F $12.88{\pm}1.49\;years$ 1st Molar M $9.60{\pm}1.69\;years$ F $9.30{\pm}1.16\;years$ 2nd Molar M $14.38{\pm}1.73\;years$ F $13.96{\pm}1.63\;years$ 2. Sexual differences in same age group at given calcification stage were not significant statistically. 3. The developmental order in mandibular permanent teeth was as follows; a) central incisor, b) lateral incisor, c) 1st molar, d) canine and 1st premolar, e) 2nd premolar, f) 2nd molar.

MHEMT를 이용한 광대역 특성의 밀리미터파 Cascode 증폭기 연구 (Research on Broadband Millimeter-wave Cascode Amplifier using MHEMT)

  • 본 논문에서는 밀리미터파 대역에서 광대역 특성을 갖는 MHEMT (Metamorphic High Electron Mobility Transistor) cascode 증폭기를 설계 및 제작하였다. Cascode 증폭기 제작을 위해 먼저 $0.1{\mu}m$ InGaAs/InAlAs/GaAs MHEMT를 설계 및 제작하였다. 제작된 MHEMT는 드레인 전류 밀도가 670 mA/mm이고, 최대 전달컨덕턴스(gm)는 688 mS/mm이며, 주파수 특성으로 전류이득 차단 주파수($f_T$)는 139 GHz, 최대 공진 주파수($f_{max}$)는 266 GHz의 특성을 나타내었다. 설계된 cascode 증폭기는 회로의 발진을 막기 위해서 저항과 캐패시터를 commom gate 소자의 드레인이 병렬로 연결하였다. Cascode 증폭기는 CPW (Coplanar Waveguide) 전송선로를 이용하여 광대역 특성을 얻을 수 있도록 정합회로를 설계하였다. 설계된 증폭기는 본 실험실에서 개발된 MHEMT MMIC 공정을 이용해 제작되었다. 제작된 cascode 증폭기의 측정결과, 3 dB 대역폭이 20.76$\sim$71.13 GHz로 50.37 GHz의 넓은 대역 특성을 얻었으며, 대역내에서 평균 7.07 dB 및 30 GHz에서 최대 10.3 dB의 S21 이득 특성을 나타내었다.

용담댐 운영이 하류 기준유량 설정에 미치는 영향 (Effect of Yongdam Dam Operation to Level of Reference Flows Downstream)

  • The Ministry of Environment is determining reference flows and goal water qualities in many stations over all around riverbasin to control TMDL. Reference flow is now defined to 10 years averaged 275th minimum flow$(Q_{275})$. Dam operation takes direct effect on flows downstream. The Yongdam mutipurposed dam was constructed in 2002 and TMDL managing stations between the Daecheong dam and the Yongdam dam are the Geumbon B, C, D, E, and F in main stream of the Geum river. Geumbon F is the Daecheong dam site. Observed flows are ideal to be used to set reference flows, but simulated flows are more practical to be used to set reference flows from the cause of the Yongdam dam's operation. A system for simulating daily storages of the Yongdam dam was constructed and the DAWAST model was selected to simulate daily streamflows. Analysis period was selected for 10 years from 1996 to 2005. Scenario was set as follows; Firstly, observed outflows from the Yongdam dam are used from 2002 to 2005 and the Yongdam dam does not exist from 1995 to 2001. Secondly, the Yongdam dam existed also from 1995 to 2001 and simulated outflows from the Yongdam dam are used from 1996 to 2005 with provision of constant outflow of $7.0m^3/s$ and water supply to the Jeonju region outsided watershed of $900,000m^3/day$. In case of scenario 1 reference flows at the Geumbon B, C, D, E, F are 4.52, 6.69, 7.96, 11.17, and $13.21m^3/s$, respectively. And in case of scenario 2 reference flows at the Geumbon B, C, D, E, F are 6.27, 8.48, 9.58, 12.73, and $15.12m^3/s$, respectively.

  • 비이온성 계면활성제 수용액 중에서 $Na_2SiO_3$$H_2SiF_6$의 빠른 반응을 통하여 메조포러스 분자체를 제조하였다. 침전은 수초 이내에 형성되었으며, 시료의 XRD분석은 d-spacing이 3.1-5.8 nm인 잘 발달된 피크를 나타내어 메조포러스분자체임을 확인할 수 있었다. 비표면적은 비이온 계면활성제에 따라 $290-1,018m^2/g$의 큰 값을 나타내었으며, 기공분포는 2.5-3.1 nm의 일정한 값을 보여주었다. SEM을 통해 관찰한 입자의 모양은 크기가 ${\sim}0.5{\mu}m$로 균일하고 잘 분리된 구형이었으며, TEM은 기공의 형태가 일정크기의 벌레구멍 모양임을 보여주었다.

재조합 lysozyme-HJP34 단백질의 다양한 병원성 세균에 대한 항균 효능 및 자돈 사료첨가제의 가능성 평가 (Antibacterial evaluation of recombinant lysozyme-HJP24 proteins against various bacterial pathogens, and of its possibility test as a feed additive in piglets)

  • The recombinant lysozyme-HJL34 proteins were expressed and purified using commercial Escherichia (E.) coli expression system. Stx2e+ F18+ E. coli, Actinobacillus pleuropneumoniae (APP), Streptococcus (S.) suis, and Clostridium (C.) perfringens strains were isolated from pigs. The minimum inhibitory concentrations (MICs) of the recombinant lysozyme-HJP34 proteins were examined by means of the microtiter plate method, according to the NCCLS recommendations. The possibility of its as the alternatives to antibiotics was tested in piglets. The MICs were determined as 75 ㎍/mL, 300 ㎍/mL, 75 ㎍/mL, 35.5 ㎍/m against Stx2e+ F18+ E. coli, APP, S. suis, C. perfringens, respectively. A total of 25 piglets were divied 5 groups. The piglets in group A~C were fed with commercial feed and those in groups D, E were fed with commercial feedstuff. All piglets in groups B~E were challenged with virulent Stx2e+ F18+ E. coli, APP, S. suis strains. Groups C and D were treated with antimicrobial from 24 h after challenge. All piglets in group B died within 3 days after challenge. Among 5 piglets in groups C and D piglets, 80% survived after challenge. Among group E piglets, 60% were alive until the end of this study. Therefore, this study indicates that recombinant lysozyme-HJP34 proteins is a suitable possibility as a feed additive for reduction of diseases by bacterial pathogens in piglet feed.

Effects of tktA, $aroF^{FBR}$and aroL Expression in the Tryptophan-Producing Escherichia coli

  • In order to analyze the effects of tktA, $aroF^{FBR}$, and aroL expression in a tryptophan-producing Escherichia coli, a series of plasmids carrying the genes were constructed. Introduction of tktA, $aroF^{FBR}$, and aroL into the E. coli strain resulted in approximately 10-20 fold increase in the activities of transketolase, the feedback inhibition-resistant 3-deoxy-D-arabinoheptulsonate-7-phosphate synthase, and shikimate kinase. Expression of $aroF^{FBR}$ in the aroB mutant strain of E. coli resulted in the accumulation of 10 mM of 3-deoxy-D-arabinoheptulsonate-7-phosphate (DAHP) in the medium. Simultaneous expression of tktA and $aroF^{FBR}$ in the strain further increased the amount of excreted DAHP to 20 mM. In contrast, the mutant strain which has no gene introduced accumulated 0.5 mM of DAHP. However, the expression of tktA and $aroF^{FBR}$ in a tryptophan-producing E. coli strain did not lead to the increased production of tryptophan, but instead, a significant amount of shikimate, which is an intermediate in the tryptophan biosynthetic pathway, was excreted to the growth medium. Despite the fact that additional expression of shikimate kinase in the strain could possibly remove 90% of excreted shikimate to 0.1 mM, the amount of tryptophan produced was still unchanged. Removing shikimate using a cloned aroL gene caused the excretion of glutamate, which suggests disturbed central carbon metabolism. However, when cultivated in a complex medium, the strain expressing tktA, $aroF^{FBR}$, and aroL produced more tryptophan than the parental strain. These data indicate that additional rate-limiting steps are present in the tryptophan biosynthetic pathway, and the carbon flow to the terminal pathway is strictly regulated. Expressing tktA in E. coli cells appeared to impose a great metabolic burden to the cells as evidenced by retarded cell growth in the defined medium. Recombinant E. coli strains harboring plasmids which carry the tktA gene showed a tendency to segregate their plasmids almost completely within 24h.

Galaxy identification with the 6D friends-of-friend algorithm for high resolution simulations of galaxy formation

  • Galaxy/Halo finding based on the friends-of-friend (FoF) algorithm has been widely adopted for its simplicity and expandability to the phase-space. However, cosmological simulations have been progressively bigger in size and more accurate in resolutions, resulting in that galaxy/halo finding gets computationally expensive more and more. In fact, we confirm this issue through our exercise of applying the 6-dimensional (6D) FoF galaxy finder code, VELOCIraptor (Elahi et al.2019) on the NewHorizon simulation (Dubois et al. 2021), in which typical galaxies with about 1e11 Msun (107 particles) are identified with very low speed (longer than a day). We have applied several improvements to the original VELOCIraptor code that solve the low-performance problem of galaxy finding on a simulation with high resolutions. Our modifications find the exact same FoF group and can be readily applied to any tree-based FoF code, achieving a 2700 (12) times speedup in the 3D (6D) FoF search compared to the original execution. We applied the updated version of VELOCIraptor on the entire NewHorizon simulation (834 snapshots) and identified its galaxies and halos. We present several quick comparisons of galaxy properties with those with GALAXYMaker data.
