Antioxidant Activities of Various Extracts from Korean Yam (Dioscorea batatas DECNE.)

  • The objective of this study was to investigate the antioxidant activity of Korean yam (Dioscorea batatas $D_{ECNE.}$) by different extraction solvents including 70% methanol, 70% ethanol and chloroform-methanol mixture (CM, 2:1, v/v). Raw yam was analyzed for its color property, total phenol content and antioxidant activity. Yam possessed high $L^*$ value and $H^{\circ}$ value, which were $81.64{\pm}2.59$ and $83.36{\pm}0.15$, respectively. Raw yam was found to have great antioxidant activity evaluated through ABTS [2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt] radical scavenging activity, DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity, reducing power, and ferric reducing antioxidant power. Total phenol contents of various extracts from raw yam increased in the following order: 70% methanol extract ($37.62{\pm}0.88mg\;CAE/g$), 70% ethanol extract ($43.38{\pm}0.66mg\;CAE/g$) and CM extract ($67.17{\pm}0.12mg\;CAE/g$), respectively. The same trend was also could be found in the ABTS radical scavenging activity, DPPH radical scavenging activity and ferric reducing antioxidant power assays. These results implied that Korean yam might play an important role in antioxidation and serve as the bio-health functional food to take a good part in prevention of human diseases and aging.

북극 지의류 유래 미생물의 항균성 (Antibacterial Properties Associated with Microorganisms Isolated from Arctic Lichens)

  • 북극 지의류로부터 서로 다른 5종의 극지 미생물을 분리하였고, 그들의 생리활성 물질들은 아세톤, 물, 클로로포름, 다이에틸에테르, 에탄올, 에틸아세테이트, 메탄올 및 페트롤륨 에테르 등 다양한 용매를 이용하여 균 배양액으로부터 추출되었다. 이러한 추출물들의 항균성은 Staphylcoccus aureus, Bacillus subtilis, Micrococcus luteus, Enterobacter cloacae, Pseudomonas aeruginosa 및 Escherichia coli 등 6종의 병원체에 대한 디스크 확산법과 최소억제농도 측정법에 의해 조사되었다. 여러 추출 샘플 중, Burkholderia sordidicola S5-$B^T$ (KOPRI 26644) 유사미생물 종의 에틸아세테이트 추출물이 표적박테리아에 대한 높은 항균활성을 나타내었다(억제대, 7-10 mm; 최소억제농도, $57.5{\rightarrow}1000{\mu}g/ml$). 또한, 다양한 추출 용매 중, 클로로포름 추출물에서 약하지만 분명한 활성을 나타내었다.

  • The purpose of this study was to investigate the bioactivity of extracts from the seeds of Gardenia jasminoides Ellis fructus (GJE) found in Namhae, Korea. Extraction was performed using three solvents, 70% methanol, Distilled Water (DW), and Ethyl Acetate (EA). We determined the total phenol and phytic acid contents of the extracts to evaluate their nitrogen oxide scavenging activity, antioxidant activity, reducing power, and lipid peroxidation inhibition ability. The phytic acid content of GJE was found to be 1.157 mg PAE (Phytic Acid Equivalent) /g DW. The yields of the three extraction processes were as follows: DW, 36.61%; 70% methanol, 30.10%; and EA, 20.40%. The physiological activities of the extract solvents increased significantly with increasing concentrations (0.2, 0.4, and 0.6 mg/mL) (p<0.05), but were lower than those of ascorbic acid, BHA, and trolox. Total phenol content was the highest in the 70% methanol extract, followed by DW and EA extracts. Further, nitrogen oxide scavenging activity and antioxidant activity were the highest for the 70% methanol extract followed by DW and EA extracts. Based on these results, the bioactivities of the 70% methanol and DW extracts of GJ seeds were excellent. These extracts can be used as natural antioxidants.

수산화암모늄나이트레이트(HAN)의 습식합성 및 이중 유기용매를 이용한 고체상 추출 (Wet Synthesis of Hydroxylammonium Nitrate (HAN) and Solid Phase Extraction Using Dual Organic Solvents)

  • 수산화암모늄나이트레이트(HAN; NH3OHNO3)는 낮은 용융점과 증기압의 특성을 가지고 있고, 상대적으로 높은 산소균형을 이루고 있는 이온성 화합물이다. 본 연구에서는 높은 함량의 액상추진제를 제조하기 위한 산화제로 활용하기 위하여 이중용매를 적용하여 고체입자상으로 얻었다. 감압 하에서 액상의 HAN으로부터 수분을 증발시킨 후, 반용매로서 아세톤과 에탄올을 적용하여 추출한 입자상의 HAN에는 13.8 wt%의 수분이 포함되어 있었다. 아세톤을 단독으로 적용하였을 때, 합성수율은 질량기준으로 최대 88%이었고, TGA로 측정된 최대 함량은 86.2%, 분해온도는 160~205℃ 범위로 나타났다.

Ethyl Acetate와 Methanol을 이용한 블루베리 추출물 대사체 분석 (Metabolomic Analysis of Ethyl Acetate and Methanol Extracts of Blueberry)

  • 본 연구에서는 LC-MS/MS를 이용한 블루베리의 methanol과 ethyl acetate 추출 분획에 존재하는 대사체의 분석을 통해 효율적인 대사체 profiling의 가능성을 탐색하였다. LC-MS/MS에서 검출되는 대사체를 통계 처리한 결과, methanol 추출 분획에서는 5-O-feruloylquinic acid, malvidin hexoside, malvidin-3-arabinoside, petunidin-3-arabinoside, delphinidin hexoside, delphinidin, petunidin hexoside와 같은 안토시아닌 계열의 화합물들이 존재하였고, ethyl acetate 분획에서는 chlorogenic acid, chlorogenic acid dimer, 6,8-di-C-arabinopyranosylluteolin, luteolin과 같은 플라보노이드 계열의 화합물이 검출되었다. 본 연구는 기존 연구와 달리 대사체학 기법을 이용한 블루베리 추출물 전체 대사물질의 profiling을 시도한 최초의 연구로서 블루베리에 함유된 유용 성분의 스크리닝 등 향후 응용 연구에 유용한 기반으로 이용될 수 있을 것으로 기대된다.

추출용매와 부위에 따른 고추 추출물의 세포독성 (Cytotoxicity of Extracts from Korean Pepper (Capsicum annuum L.) by Extraction Solvents and Plant Parts)

  • In order to find out anticancer activity of Korean pepper (Capsicum annuum L.), the cytotoxicity against 8 cell lines including 293 (normal kidney cells) and A-431 (epidermoid carcinoma cells) of extracts by extraction solvents and plant parts were investigated using MTT assay. Also the correlation between content of capsaicin known as anticancer ingredient and cytotoxicity of extracts from pepper were analyzed. The distilled water extracts from seed and germinated seed showed very high cytotoxicity against 6 cancer cell lines including A549 (lung carcinoma cells), AGS (stomach adenocarcinoma cells), HeLa (cervix adenocarcinoma cells), HepG2 (hepatoblastoma cells), HT-29 (colon adenocarcinoma cells), and MCF-7 (breast adenocarcinoma cells). But 80% ethanol and methanol extracts showed cytotoxicity against 293 and AGS. The $RC_{50}$, that was, the concentration of sample required for 50% reduction of cell viability, of seed and germinated seed extracts against AGS were $33.4{\sim}389.1{\mu}g/m{\ell}$ and $63.9{\sim}1,316.7{\mu}g/m{\ell}$, respectively, so anticancer activity was higher in seed than in germinated seed. In capsaicin contents, seed with high cytotoxicity and pericarp with a little cytotoxicity contained $47.4{\sim}1,260.0{\mu}g/g$ and $58.3{\sim}1,498.0{\mu}g/g$, respectively. As these results, the correlation was not between cytotoxicity and capsaicin content.

머위(Petasites japonicus) 추출물의 항산화와 항암활성 효과 (The Antioxidant and Anticancer Effects of Butterbur (Petasites japonicus) Extracts)

  • 머위의 뿌리, 줄기, 잎을 사용하여 추출물에 따른 항산화 활성을 분석하였고, 추출물을 분획 정제하여 암세포 생장 억제성을 실험하였다. SOD 유사활성은 머위 잎의 에탄올 추출물이 96.7%의 가장 높은 활성을 보였으며, 추출 용매에 따른 머위의 SOD 유사활성은 물 추출보다는 에탄올 추출물이 더욱 높은 항산화 활성을 나타냈다. 총폴리페놀 함량은 물 추출물에서 전체적으로 높은 함량을 나타내었으며, 머위 잎이 223mg/g dry weight으로 가장 높았다. 시료별로 전자공여능 효과를 살펴보면 머위 뿌리의 물 추출물에서 61.5%, 머위 잎의 에탄올 추출물에서 34.9%로 가장 높은 전자공여능을 나타냈다. 암세포 생존 억제성의 실험에서 머위 추출물의 분획물들은 정상세포 DC2.4의 생육에는 크게 영향을 미치지 않으면서도 위암세포주 SNU-719의 생육은 41.9%로 크게 억제시켰고, 간암세포주 Hep3B의 생육은 72.7% 억제시켰다. 본 연구 결과들을 종합해 볼 때 머위를 이용한 새로운 기능성 식품 소재 및 약용자원식물로 개발이 가능하다.

Simultaneous Analysis of Cholesterol Oxidation Products (COPs) in Powdered Milk Using HPLC/UV-Vis

  • Cholesterol and cholesterol oxidation products (COPs) may accumulate in foods of animal origin during processing or storage. An effective and sensitive analytical method was developed by increasing the UV absorption of compounds through derivatization by attaching a chromophore to the functional groups of cholesterols (cholesterol, 20-hydroxycholesterol, 7-ketocholesterol, cholestane-$3{\beta}$-$5{\alpha}$-$6{\beta}$-triol, 25-hydroxycholesterol, and $5,6{\alpha}$-epoxycholesterol). The influences of the reaction time, volume of reaction solvent, amounts of derivatizing reagent, and extraction solvents were investigated, as they may influence the reaction and extraction yield. The derivatized COPs were analyzed simultaneously on a C18 column (2.1 mm i.d. ${\times}$ 100 mm length, $3.5{\mu}m$ particle size) using a gradient elution with water and acetonitrile. The derivatized COPs showed increased sensitivity and selectivity in HPLC/UV-Vis. The LOD and LOQ were in the concentration ranges of 0.018-0.55 mg/kg and 0.059-1.84 mg/kg from the powdered milk. And the accuracy and precision were 78.1-116.7% and 1.1-9.9%, respectively.

초임계 이산화탄소를 이용하여 멸치(Engraulis japonicus)로부터 회수된 고농축 단백질의 특성 (Characterization of Concentrated Proteins Recovered from Anchovy Engraulis japonicus using Supercritical Carbon Dioxide)

  • Supercritical carbon dioxide ($SCO_2$) extraction was used to recover concentrated proteins and to remove lipids and odor causing compounds from anchovy. Engraulis japonicus $SCO_2$ was used as the solvent for extraction, which was performed in a semi-batch flow reactor. The experimental conditions used were pressure, 15-25 MPa; temperature, $40-60^{\circ}C$ and sample size, 500 ${\mu}m$. The proteins obtained under these conditions performed well in a sensory evaluation; moreover, effective lipids and odor removal was achieved. The stability and characteristics of the proteins recovered with different solvents were also evaluated. The samples were sterilized by processing with $SCO_2$. Escherichia coli was not detected after storage for several days. The sensory characteristics were found to be superior to those of a sample produced by hexane extraction. Thus, the protein concentrate was obtained at $60^{\circ}C$ and 25 MPa was deemed valuable as a foodstuff.

Extraction of Water-Soluble Porphyrin and Metalloporphyrins into Acetonitrile by Salting-out

  • A cationic water soluble porphyrin (5,10,15,20-tetrakis (l-methyl-pyridinium-4-yl)porphyrin, $H_2tmpyp^{4+}$) and its metalloporphyrins (MP) were easily extracted into acetonitrile separated by addition of sodium chloride ($4mol\;dm^{-3}$) in the presence of sodium perchlorate, where M denotes $Zn^{2+}$, $Cu^{2+}$, $Co^{3+}$, $Fe^{3+}$, and $Mn^{3+}$ and $P^{2-}$ is porphyrinate ion. The extracted ion-pair complexes were completely dissociated to $[MP(ClO_4)_3]^+$, and $[MP(ClO_4)_2]^{2+}$. The extraction and the dissociation constants were determined by taking into account of the partition constant of sodium perchlorate ($K_D=1.82{\pm}0.01$). The chemical properties of the separated acetonitrile phase as $E_{T(30)}$ and $D_{II,I}$ were determined and compared with other water miscible solvents (acetone, actonitrile, 1,4-dioxane, tetrahydrofuran, 1-propanol and 2-propanol). Furthermore, a sensitive and selective method was proposed for the determination of a subnanogram amount of copper(II) in natural water samples by using the present salting-out method and the porphyrins.
