Effect of Extraction Condition on the Content of EGCG and Caffeine of Green Tea: Comparison with the Inhibitory Activity on Pancreatic Lipase

  • Caffeine and epigallocatechin gallate (EGCG) are major constituents of green tea, the leaves of Camellia sinensis (Theaceae). Although EGCG is well known for diverse beneficial effect, caffeine is sometimes harmful with adverse effects. Therefore, the extraction efficiency was investigated using different extraction method such as extraction solvent, extraction time, extraction method, and repeated extraction. The content of EGCG and caffeine in green tea extract was quantitated by HPLC analysis. The extraction condition exerted difference on the extraction yield. The content of EGCG was also affected by different extraction condition. Especially, the extraction solvent greatly affected the content of EGCG in the extract. However, the content of caffeine was less affected compared to that of EGCG. The inhibitory effect of green tea extract on pancreatic lipase was almost similar regardless of extraction condition. Taken together, optimization of extraction condition will provide best efficacy for further development of green tea as anti-obesity therapeutics.

Optimization of Extraction Condition of Hesperidin in Citrus unshiu Peels using Response Surface Methodology

  • Hesperidin, which is the most abundant flavonoid of Citrus unshiu (Rutaceae), has been reported to possess diverse activities and widely used as functional foods and cosmetics. For the development of functional products, extraction procedure is indispensable. Extraction conditions affect the composition of extract as well as its biological activity. Therefore, we tried to optimize extraction conditions such as extraction solvent, extraction time and extraction temperature for maximum yield of hesperidin using response surface methodology with threelevel-three-factor Box-Behnken design (BBD). Regression analysis showed a good fit of the experimental data and the optimal condition was obtained as ethanol concentration, 59.0%; temperature $71.5^{\circ}C$ and extraction time, 12.4 h. The hesperidin yield under the optimal condition was found to be $287.8{\mu}g$ per 5 mg extract, which was well matched with the predicted value of 290.5 μg. These results provides optimized extraction condition for hesperidin and might be useful for the development of hesperidin as functional products like health supplements, cosmetics and medicinal products.

Effect of extraction conditions on the stability and safety of sericin

  • To assess the feasibility of silk sericin for non-textile application, the storage stability and biological safety of sericin were examined. It was extracted at 37℃, 70℃, 100℃, and 121℃ for 1, 3, and 5 h to elucidate the effect of extraction condition on the stability and safety of silk sericin. The solubility was increased till approximately 26% with extraction temperature of 121℃ for 1 h. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) showed that the molecular weight distribution depended on the extraction conditions. Extracted sericin displayed typical UV absorption bands upon spectrometric analysis. To examine the reproducibility of its obtained conformation, sericin was extracted thrice and its circular dichroism (CD) spectra was measured each time. Most CD spectra showed reproducibility regardless of temperature and time except under 100℃ extraction condition. The diversity of CD spectrum showed gradual reduction and was finally coincident with extraction time from 1 to 5 h. Notably, sericin has a negative peak of approximately 200 nm attributed to random coil conformation, regardless of extraction condition. However, at the 100℃ extraction condition, sericin showed both bands to be negative bands of approximately 200 and 220 nm, respectively. Sericin was centrifuged to determine the stability of storage conditions. The sericin extracted at 100℃ and 121℃ for 1 h was found to form gel rapidly within 1 h, but at 121℃ condition, the gel fraction was approximately 20% within 1 h which retained its phase regardless of storage time. The gel fraction of sericin extracted at 100℃ for 5 h increased with time, however at the 121℃ for 5 h condition, the gel fraction was measured to be less than 10% regardless of increase in storage time. PetriflimTM AC plates test showed that sericin was safe from aerobic bacteria activity by extraction under high temperature.

Optimization of Extraction Condition of Methyl Jasmonate-treated Wild Ginseng Adventitious Root Cultures using Response Surface Methodology

  • The usage of wild ginseng (Panax ginseng C.A. Meyer) has been limited due to short supply and high price. Therefore, sufficient production as well as efficient extraction of mountain ginseng are required for the development as products. In this study, wild ginseng adventitious root cultures were prepared for efficient production with advantages of fast growth and stable production. Treatment of methyl jasmonate (MJ) to wild ginseng adventitious root cultures increased the extraction yield and antioxidative activity. Further investigation on effect of extraction conditions suggested the importance of ethanol concentration on antioxidative activity and extraction yield of MJ-treated wild ginseng adventitious root cultures. Optimized extraction condition of MJ-treated wild ginseng adventitious root cultures for maximum extraction yield and antioxidative activity was determined using response surface methodology with three-level-three-factor Box-Behnken design (BBD). Extraction of 1 g MJ-treated wild ginseng adventitious root culture with 30 ml of 9% ethanol at $30^{\circ}C$ produced 310.2 mg extract with 71.0% antioxidative activity at $100{\mu}g/ml$. Taken together, MJ-treated wild ginseng adventitious root culture is valuable source for wild ginseng usage and optimized extraction condition can be used for the development of functional products or folk remedies.

마이크로웨이브 추출방법을 이용한 작약의 유효성분 추출 및 생리활성 측정 (Development of Microwave Extraction Method for the Active Ingredients and Functional Constituents of Paeonia Root)

  • The heat-mediated reflux apparatus extraction of Paeonia lactiflora Pall. has been widely used as a traditional extraction method. In this paper, the microwave apparatus extraction method of Paeonia Radix was performed and the active ingredients and functional constituents were compared with the reference extraction method. The most effective extraction condition of albiflorin using the microwave was 120℃ in 50% methanol, and paeoniflorin was maximally extracted at 60℃. The reduced level of paeoniflorin molecule at high-pressure and high-temperature extraction condition was caused by the molecular instability. Additionally, the microwave extraction of 50% methanol extracts at 150℃ showed the highest functional constituents determined by in vitro DPPH radical scavenging activity, polyphenol concentration, and tyrosinase inhibition assay. The microwave apparatus was adapted as a rapid, low-cost, and environmentally friendly method to extract active ingredients and the practical extraction conditions of Paeonia Radix can be used in industrial applications.

초임계유체 추출에 의한 초피나무 과피 중 신미성분의 추출조건 (The extraction condition of pungent compounds from Zanthoxylum piperitum D.C pericarps by using supercritical fluid extraction)

  • 초임계추출(SFE)를 이용한 초피나무 과피 중 신미성분의 최적추출조건을 조사하였다. 최적 추출 압력은 $300kg/cm^2$ 이었으며 온도는 $60^{\circ}C$, 추출 부피는 $12\;m{\ell}$, flow rate는 3m/min 그리고 modifier로 사용한 methanol의 함량비율은 20%였다. 추출시간은 초반 20분 안에 대부분의 신미성분이 추출되었다. 또한 용매추출법과 비교한 결과 추출효능이 약 40%정도 향상되었고 시간은 20분으로 단축되었으며 유기용매 methanol은 12 ml밖에 소요되지 않을 뿐만 아니라 주요한 추출용매로 무독성 액화$CO_2$를 사용하기에 독성 유기용매의 다량 사용을 피할 수 있는 친환경적이고 경제적인 방법으로 생각되어 진다.

실관막모듈에 의한 중금속염 혼합용액으로부터 Cr(VI) 분리 (Separation of Cr(VI) from Heavy Metal Salts Mixed Solution by using Hollow Fiber Module)

  • This work reports the application of a hollow fiber module(HFM) for Cr(VI) extraction from heavy metal salts mixed solution by using microporous hydrophobic hollow fiber module. In HFM configuration, the organic extraction used for the extraction of Cr(VI) was di-(2-ethyl hexyl) phosphoric acid(D2EHPA) diluted with n-heptane. The study of HFM includes the influence of hydrodynamic and chemical condition, i.e., the flow rate of feed solution, the time of reactive extraction, the concentration of feed solution, and the pH of aqueous phase solutions. Several experiments with synthetic solution of different mixed components system of Cr(VI) solutions established optimum condition to achieve a clean separation of Cr(VI). It was possible to separate Cr(VI) in the presence of metal salts mixed solution, such as Zn(II), Ni(II), Cu(II), and Cd(II) using the HFM technique.

알칼리에 의한 천마 단백질 추출의 최적 pH 조건 (Optimized pH condition of protein extraction of Gastrodia elata Blume by alkaline method)

  • 본 연구에서는 알칼리 사용에 따른 갈변을 최소화하고 영양적으로 우수한 천마 단백질을 보다 효율적으로 추출하기 위하여 용출 및 침전 pH에 따른 단백질의 갈변도와 함량을 측정함으로써 탈지 천마 단백질 알칼리 추출의 최적 pH 조건을 설정하고자 하였다. 천마의 수분은 약 15%로 측정되었으며, 대부분 탄수화물로 구성되어 있었고, 단백질의 함량이 높은 것으로 확인되었다. 이러한 단백질을 다양한 pH에서 용출시킨 결과 pH가 증가함에 따라 용출된 단백질의 양이 증가하였으며, 갈변도 또한 증가함을 보였다. 침전 pH에 따른 단백질 함량은 pH 4에서 침전된 pellet이 가장 많은 함량을 나타내었으며, 상등액의 단백질 함량 또한 대부분 pH 4로 침전시킨 경우 가장 적은 것으로 확인되었다. 뿐만 아니라 단백질의 회수율도 침전 pH가 4일 때 가장 높은 값을 나타내었다. 따라서 갈변도와 단백질 함량을 고려한 탈지 천마 단백질 추출을 위한 최적조건은 용출 pH 9와 침전 pH 4로 결정되었다.

상압조건의 마이크로웨이브 추출공정에서 가용성 인삼성분의 추출조건 최적화 (Optimization of Microwave-Assisted Extraction under Atmospheric Pressure Condition for Soluble Ginseng Components)

  • 인삼 성분의 열불안정성과 실용성을 고려하여 상압 조건에서 마이크로웨이브 추출실험을 실시하면서 반응 표면분석에 의해 추출조건의 최적화 및 추출특성을 모니터링하였다. 가용성 성분의 수율은 에탄올 농도가 낮고 microwave power가 높을수록, 조사포닌 함량은 에탄올 농도가 높을수록 증가하였다. 그리고 추출물의 가용성성분 함량, 조사포닌 함량, 페놀성화합물 함량, 전자공여작용 등의 추출물 특성을 모두 만족시키는 최적 추출조건을 예측하여 보았을 때, 에탄올 농도 $54{\sim}60%$ 범위, microwave power $41{\sim}90$ W 범위, 추출 시간 4분 이내로 나타났다. 예측회귀식의 신뢰성을 확인하기 위하여 예측된 추출조건의 임의의 최적점에서 실제 추출실험을 실시해 본 결과, 각 반응변수들의 예측값은 실제값과 유사하였다.

인삼 다당체 추출 공정 개발을 위한 인삼의 추출 조건 및 원료에 따른 총당 변화 (Analysis of Total Sugar by Extraction Condition and Material to Develope the Extraction Process of Ginseng Polysaccharide)

  • 인삼 속에 다량으로 함유되어 있는 다당체 성분인 진산에 대한 효능이 연구되고 있는 시점에서 산업적으로 이용할 수 있는 대량 추출공정을 위한 추출조건을 분석하였다. 인삼의 종류는 홍삼(5년근)이 $32\%$로 총당의 함량이 가장 높았으며 금산 수삼(5년근)은 $31\%$$1\%$ 정도 조금 적게 나타났다. 그러나 홍삼 제조 공정에 따른 효율을 비교해 본 결과 금산 수삼(5년근)과 별 차이가 없어 금산 수삼을 사용하였다. 수삼부위 중 주근, 최적온도 $80^{\circ}C$, 최적 추출시간 6시간 추출 시 총당의 추출 효율이 가장 높았다. 또한 총당의 추출효율을 높이기 위해 인삼 추출물에 protease, ${\alpha}-amylase$, cellulase 효소 처리했을 때 추출 효율이 $20\%$까지 높은 것으로 나타났으며, 인삼 추출액에 주정 알콜농도 $70\%$, 동결건조 방법이 총당 회수율이 높았다.
