Objectives : Taraxacum coreanum (TC) have been used as a traditional medicine to treat inflammatory diseases and anti-oxidant effect in Korea. However, the anti-inflammatory effect of TC water extract on lipopolysaccharide (LPS)-induced inflammation is not well-known. Therefore, this study was performed to identify the anti-inflammatory effect of TC on LPS induced inflammatory. Methods : RAW 264.7 cells were treated with 500 ng/mL of LPS. Water extracts of TC (0.1, 0.25, 0.5 mg/ml) was treated 1 h prior to LPS. Cell viability was measured by MTT assay. Levels of nitric oxide (NO) were measured with Griess reagent and pro-inflammatory cytokines were measured by enzyme-linked immunosorbent assay (ELISA) and real-time polymerase chain reaction (real-time PCR). We also examined molecular mechanisms such as mitogen-activated protein kinases (MAPKs) and nuclear factor-B ($NF-{\kappa}B$) activation by western blot. Results : Water Extract from TC itself did not have any cytotoxic effect in RAW 264.7 cells. TC treatment inhibited the production of NO production, and pro-inflamamtory cytokines such as interleukin (IL)-6 and $IL-1{\beta}$ on protein and mRNA levels. In addition, TC treatment inhibited the LPS-induced activation of MAPKs such as extracellular signal-regulated kinase1/2 (ERK1/2), p38 kinases (p38), c-Jun $NH_2$-terminal kinase (JNK) and $NF-{\kappa}B$. Conclusions : In summary, our result suggest that treatment of TC could reduce the LPS-induced inflammation. Thereby, TC could be used as a protective agent against inflammation. Also, this study could give a clinical basis that TC could be a drug or agent to prevent inflammation.
We examined that the protective effects of ANX1 on 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced skin inflammation in animal models using a Tat-ANX1 protein. Topical application of the Tat-ANX1 protein markedly inhibited TPA-induced ear edema and expression levels of cyclooxygenase-2 (COX-2) as well as pro-inflammatory cytokines such as interleukin-1 beta (IL-$1{\beta}$), IL-6, and tumor necrosis factor-alpha (TNF-${\alpha}$). Also, application of Tat-ANX1 protein significantly inhibited nuclear translocation of nuclear factor-kappa B (NF-${\kappa}B$) and phosphorylation of p38 and extracellular signal-regulated kinase (ERK) mitogen-activated protein kinase (MAPK) in TPA-treated mice ears. The results indicate that Tat-ANX1 protein inhibits the inflammatory response by blocking NF-${\kappa}B$ and MAPK activation in TPA-induced mice ears. Therefore, the Tat-ANX1 protein may be useful as a therapeutic agent against inflammatory skin diseases.
Objectives : Ecklonia cava is brown alga(Laminariaceae) which grows is sea, it has antioxidant, diarrhea and anticoagulant effect. In this study, the effect of ethanol extract of Ecklonia cava (EC) on peptidoglycan(PGN) -induced NO production in RAW 264.7 cells was investigated. Methods : In the present study, IL-6 production was measured using the enzyme-linked immunosorbent assay(ELISA), prostaglandin $\E_2$($\PGE_2$) production was measured using the EIA kit, and inducible NO synthase(iNOS), cyclooxygenase-2(COX-2), and mitogen-activated protein kinase(MAPK) activation, as determined by western blot analysis and reverse transcription -polymerase chain reaction(RT-PCR). Results : EC inhibited PGN-induced NO and IL-6 production. Consistent with these observations, the protein expression of iNOS and COX-2 were inhibited by EC. Moreover, EC suppressed the phosphorylation of extracellular signal-regulated kinase(ERK) 1/2 in PGN-induced RAW 264.7. Conclusions : These results suggest that EC has inhibitory effects on PGN-induced $\PGE_2$, NO, and IL-6 production, as well as the expressions of iNOS and COX-2 in the murine macrophage. These inhibitory effects occur through blockades on the MAPKs phosphorylation.

SMW treatment inhibited the production of NO, and interleukin $(IL)-1{\beta}$ which is pro-inflammatory cytokine. And SMW treatment inhibited the LPS-induced activation of MAPKs such as extracellular signal-regulated kinase1/2 (ERK1/2), p38 kinases (p38), c-Jun NH2-terminal kinase (JNK) and $NF-{\kappa}B$. In addition, it also showed reducing the level of $IL-1{\beta}$ on the serum and liver tissue of mice. Also, death of LPS-induced mice was inhibited by SMW. Conclusions: The result suggests that treatment of SMW could reduce the LPS-induced inflammation. Thereby, SMW could be used as a protective agent against inflammation. Also, this study could give a clinical basis that SMW could be a drug or agent to prevent inflammatory diseases.
Oxidative stress have known to be a risk factor for the degenerative processes and closely related to a lot of diseases. It is well established that antioxidants are good in protection and therapeutic means against oxidative damage. There is increasing interest in natural antioxidants and many natural antioxidants have been found and utilized as the possible protection for various diseases and skin aging. We have screened natural antioxidant agents for cosmeceuticals, nutraceuticals, and drugs as therapeutic and preventive means against oxidative stress, and have developed a number of novel antioxidants from various natural sources. A novel melanin synthesis inhibitor, Melanocin A, isolated from the metabolite of a fungal strain Eupenicillium shearii F80695 inhibited mushroom tyrosinase and melanin biosynthesis of B16 melanoma cells with $IC_{50}$ value of 9.0 nM and MIC value of $0.9\;{\mu}M$, respectively. Melanocin A also exhibited potent antioxidant activity by scavenging of DPPH and superoxide anion radicals. UV was found to increase the level of hydrogen peroxides and other reactive oxygen species (ROS) in skin tissues. This increase in ROS may not only alter the structure and function of many genes and proteins directly but may also modulate their expressions through signal transduction pathways and, ultimately, lead to skin damage. We investigated the effect of Melanocin A on UV-induced premature skin aging. Firstly, the effect of Melanocin A on UV-induced matrix metalloproteinase (MMP)-9 expression in an immortalized human keratinocyte cell line, HaCaT in vitro was investigated. Acute UV irradiation induced MMP-9 expression at both the mRNA and protein levels and Melanocin A suppressed this expression in a dose-dependent manner. We then investigated UV-induced skin changes in hairless mice in vivo by Melanocin A. Chronic exposure of hairless mouse dorsal skin to UV increased skin thickness and induced wrinkle formation and the gelatinase activities of MMP-2 and MMP-9. Moreover, Melanocin A significantly suppressed UV-induced morphologic skin changes and MMP-2 and MMP-9 expression. These results show that Melanocin A can prevent the harmful effects of UV that lead to skin aging. Therefore, we suggest that Melanocin A should be viewed as a potential therapeutic agent for preventing and/or treating premature skin aging. Terrein is a bioactive fungal metabolite isolated from Penicillium species. Terrein has a relatively simple structure and can be easily synthesized. However, the biologic effects of terrein are comparatively unknown. We found for the first time that terrein potently inhibit melanin production in melanocytes and has a strong hypopigmentary effect in a spontaneously immortalized mouse melanocyte cell line, Mel-Ab. Treatment of Mel-Ab cells with terrein (10-100 mM) for 4 days significantly reduced melanin levels in a dose-dependent manner. In addition, terrein at the same concentration also reduced tyrosinase activity. We then investigated whether terrein influences the extracellular signal-regulated protein kinase (ERK) pathway and the expression of microphthalmia-associated transcription factor (MITF), which is required for tyrosinase expression. Terrein was found to induce sustained ERK activation and MITF down-regulation, and luciferase assays showed that terrein inhibits MITF promoter activity in a dose-dependent manner. To elucidate the correlation between ERK pathway activation and a decreased MITF transcriptional level, PD98059, a specific inhibitor of the ERK pathway, was applied before terrain treatment and found to abrogate the terrein-induced MITF attenuation. Terrein also reduced the tyrosinase protein level for at least 72 h. These results suggest that terrain reduces melanin synthesis by reducing tyrosinase production via ERK activation, and that this is followed by MITF down-regulation.
Objectives : Angelicae Gigantis Radix (AG) is a plant of the Ranunculus family. AG have been reported to have various pharmacological effects on human health which include uterine growth promotion, anti-inflammatory, analgesic, and immune enhancement. However, research on dermatitis disease is insufficient. Therefore, we investigated the effects of AG on tumor necrosis factor-α (TNF-α)/interferon-γ (IFN-γ) stimulated HaCaT cell. Methods : To investigate the effect of AG on HaCaT cell, HaCaT cells were pre-treated with AG for 1 hour and then stimulated with TNF-α/IFN-γ. After 24 hours, media and cells were harvested to analyze the inflammatory mediators. Concentration of human interleukin-1beta (IL-1β), monocyte chemoattractant protein-1 (MCP-1), granulocyte-macrophage colony-stimulating factor (GM-CSF), and TNF-α in the media were assessed by ELISA. mRNA expression of human thymus and activation-regulated chemokine (TARC), IL-6, and IL-8 were analyzed by RT-PCR. Additionally, the mechanisms of mitogen-activated protein kinases (MAPKs) and nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) signaling pathway were investigated by Western blot. Results : The treatment of AG inhibited gene expression levels of IL-6, IL-8, and TARC and protein expression levels of IL-1β, MCP-1, and GM-CSF. Also, AG significantly reduced extracellular signal-regulated kinase (ERK) phosphorylation and NF-κB translocation in TNF-α/IFN-γ stimulated HaCaT cell. Conclusions : Taken together, these results demonstrate that AG can alleviate inflammatory diseases such as atopic dermatitis by regulating the expression of inflammatory cytokines. Also, it suggest that AG may a promising candidate drug for the treatment of inflammatory disease such as atopic dermatitis.
갯질경이(Limonium tetragonum)는 질경이과에 속하는 여러해살이 풀로 습지에 자생하는 염생식물의 일종이며, 항산화, 항종양 및 간보호 효능이 있는 것으로 알려져 있다. 본 연구에서는 갯질경이 추출물로부터 용매 극성에 따라 분획한 분획물($H_2O$, n-BuOH, 85% aq. MeOH 및 n-Hexane)을 이용하여 지방세포내 중성지방 생성 및 지방세포 분화조절 인자 발현에 미치는 영향을 검토하였다. 마우스 유래 지방전구세포3T3-L1을 지방세포로 분화하여 Oil Red O염색법으로 지방 세포 분화정도를 확인한 결과, 갯질경이 분획물에 의해 지방세포의 형성이 농도의존적으로 억제되었다. 또한 지방생성조절에 관여하는 전사인자 $PPAR{\gamma}$, $C/EBP{\alpha}$ 및 SREBP-1c의 발현을 mRNA와 단백질 수준에서 확인한 결과 갯질경이 분획물 처리시 지방세포 분화 인자의 발현이 유의적으로 감소하였다. 지방세포 분화에 관여하는 것으로 알려진 MAPK 신호 전달 경로를 확인한 결과 갯질경이 분획물 처리군에서 p38, ERK 및 JNK의 인산화가 억제되었다. 용매 분획물중에서 $H_2O$ 및 n-Hexane 분획물이 가장 우수한 지방생성 억제활성을 나타내었는데 이는, 분획물 중 페놀 또는 지방 유도체에 의한 것으로 사료된다. 본 연구 결과로부터 갯질경이 분획물의 MAPK 신호전달 경로 억제를 통한 항비만 효과를 확인하였으며, 나아가 건강기능성 식품 소재로서의 개발 가능성이 기대된다.
본 연구에서는 LPS로 자극을 유도한 RAW 264.7 대식세포에서 지칭개 추출물의 항염증 효능을 알아보기 위해 이와 연관된 다양한 인자(NO, PGE2, IL-6, 및 TNF-α)와 MAPK 신호전달 경로에 대해서 조사하였다. 먼저HL 처리에 따른 세포 생존율에 대해 조사한 결과, HL을 농도별로 처리했을 때 저농도인 25 ㎍/mL에서부터 고농도인 100 ㎍/mL까지 모두 90% 이상의 생존율이 나타났으며, LPS를 처리한 RAW 264.7 대식세포에서도 90% 이상의 생존율을 나타내 실험에 사용된 HL의 농도가 RAW 264.7 대식세포에서 무독성임을 확인할 수 있었다. 이러한 무독성 조건에서 HL의 항염증 활성을 확인하기 위하여 염증 매개체(inflammatory mediators)로 잘 알려진 NO와 PGE2의 생성 변화를 확인한 결과, LPS로 염증반응이 유도된 RAW 264.7 대식세포에서 NO와 PGE2의 생성이 농도 의존적으로 억제됨을 확인하였다. HL이 NO와 PGE2를 억제하는 항염증 효과가 있음을 관찰하였고, 이에 전 염증성 사이토카인 분비에도 유의성 있는 효과를 나타낼 것이라 판단되어 전 염증성 사이토카인(IL-6와 TNF-α)에 어떠한 영향을 미치는지 조사하였다. HL은 LPS로 유도된 RAW 264.7 대식세포의 염증반응에서 IL-6의 생산을 유의적으로 억제함을 관찰할 수 있었다. 한편 다른 전 염증성 사이토카인인 TNF-α생산에는 아무 영향을 주지 않았다. 이러한 결과는 HL이 전 염증성 사이토카인 중 TNF-α조절에 관여하지 않고, IL-6 생성을 억제하여 염증 매개체의 생성을 억제한다고 추측할 수 있었다. HL이 NO, PGE2, 및 IL-6의 조절에 작용하는 메커니즘이 상위 시그널인 MAPK cascade의 억제를 통해 나타나는 효과인지 알아보기 위해 염증과 관련된 MAPK 시그널인 p38, ERK, 및 JNK의 발현 변화를 관찰하였다. HL은 p38과 ERK의 발현 활성화를 상당히 약화시켰지만 JNK는 p38과 ERK 보다 덜 민감하게 조절함을 관찰할 수 있었다. 이상의 결과를 종합해 보면 LPS로 유도된 RAW 264.7 대식세포에서 HL은 MAPK 신호경로인 JNK 발현에 유의적인 영향을 주지 못해서 JNK와 관련된 TNF-α생산에 영향을 주지 못한 것으로 판단된다. 다른 MAPK 신호경로인 p38과 ERK의 발현을 약화시킴으로써 그 다음 기작인 IL-6, NO, 및 PGE2의 생산을 억제시켜 염증반응을 억제한 것으로 추측되어 진다. 이상의 결과를 종합해 보면 HL이 항염 활성을 가지고 있음을 확인할 수 있었으며, 이를 기반으로 HL의 MAPK 신호경로를 통한 염증성 사이토카인과 염증 매개체와의 연관성에 대한 기초자료로 활용할 수 있는 근거 자료가 될 수 있을 것으로 생각된다. 또한, 다양한 경로를 통한 염증 조절 기전 연구는 추가적으로 이루어져야 할 것으로 사료된다.
생삼을 쪄서 건조시킨 홍삼은 전통적으로 사용되고 있는 약재로서 면역, 내분비 및 중추신경계 작용을 증진시키며 염증을 억제하는 효과가 있는 것으로 알려져 있다. 본 연구에서는 그람 양성균의 세포벽 성분인 lipoteichoic acid (LTA)에 의한 염증반응에 홍삼추출액(RGE)이 항염증 효과를 가지는지 관찰하고 그 작용 기전을 연구하였다. BV-2 소교세포에서 RGE는 세포에 독성을 유도하지 않으면서 LTA로 인한 nitric oxide (NO)의 생성과 inducible nitric oxide synthase (iNOS) 발현을 억제하였으며, NF-kB p65의 핵으로의 이동과 IkB-a의 분해 또한 억제하였다. 한편, RGE는 농도의존적으로 heme oxygenase-1 (HO-1)의 발현을 유도하였으며, HO-1 siRNA를 처리했을 때는 RGE가 iNOS의 발현을 억제하지 못하였다. RGE는 HO-1의 발현에 관여하는 전사인자인 nuclear factor E2-related factor 2 (Nrf2)를 핵으로 이동을 촉진시켰다. 또한 RGE에 의한 HO-1의 발현은 phosphatidylinositol-3-kinase(PI-3K) 및 MAPK 억제제에 의해 감소되었으며, RGE가 Akt와 ERK, p38, JNK의 인산화를 유도하였다. 이상의 결과를 종합해보면, RGE는 PI-3K/Akt 및 ERK, p38, JNK 신호전달과정을 통해 HO-1의 발현을 유도함으로써 NO와 같은 염증매개물질의 생성을 억제한다는 것을 알 수 있다. 그러므로 홍삼추출액은 그람 양성균에 의한 신경염증과 염증관련 신경계 질환의 치료제로서 사용될 수 있을 것이라 사료된다.
황금찰수수 종자의 항염증 활성을 알아보기 위해, 국내에서 재배되고 있는 3가지 수수 품종(황금찰, 현찰, 찰)의 종자로부터 에탄올 추출물을 확보하여, 지질다당류(LPS)에 의해 자극된 마우스 대식세포주 RAW264.7를 이용하여 염증반응 관련 일산화질소(NO) 생성에 대한 억제 활성을 확인하였다. 세가지 품종 중에서 황금찰수수 종자의 억제활성이 가장 강하게 확인되었다. 황금찰수수 종자의 EtOH 추출물은 n-헥산, 메틸렌클로라이드(MC), 에틸아세테이트(EtOAc), 및 n-부탄올로 순차적으로 분획하였다. 이후 활성이 강하게 확인된 MC 분획을 이용하여, LPS-자극 RAW 264.7 세포에서 항염증 효능을 구체적으로 조사한 결과, LPS-자극에 의해 유도되는 염증반응관련 iNOS, COX-2 및 친염증성 사이토카인(IL-1β, IL-6, TNF-α)의 발현을 현저히 감소시켰다. 또한 MC 분획은 LPS-자극에 의해 염증관련 현상으로 유도되는 p38 mitogen-activated protein kinase (MPAK), c-Jun N-말단 키네이스(JNK) 및 세포외 신호조절 키네이스(ERK)의 활성화를 억제하는 것으로 나타났다. HPLC 분석으로 MC분획을 더 조사한 결과, 주요 구성 페놀화합물로서 gentisic acid와 naringenin을 확인하였다. Gentisic acid와 naringenin은 유사한 수준으로 LPS처리애 의해 RAW264.7 세포에서 유도되는 NO 생성에 대한 강력한 억제 활성을 나타내었다. 이상의 연구결과들은 황금찰수수 종자의 항염증 활성과 관련된 gentisic acid 및 naringenin의 작용을 잘 보여주며, 아울러 황금찰수수 종자가 곡물 기반 기능성 식품 개발에 적용될 수 있는 유익한 건강 효과를 가지고 있음을 시사한다.

ID의 비밀번호 관리소홀, 부정사용에 의하여 발생하는 모든 결과에 대한 책임은 회원에게 있습니다.

제 4 장 서비스의 이용
제 12 조 (서비스 이용 시간)
① 서비스 이용은 당 사이트의 업무상 또는 기술상 특별한 지장이 없는 한 연중무휴, 1일 24시간 운영을
원칙으로 합니다. 단, 당 사이트는 시스템 정기점검, 증설 및 교체를 위해 당 사이트가 정한 날이나 시간에
서비스를 일시 중단할 수 있으며, 예정되어 있는 작업으로 인한 서비스 일시중단은 당 사이트 홈페이지를
통해 사전에 공지합니다.
② 당 사이트는 서비스를 특정범위로 분할하여 각 범위별로 이용가능시간을 별도로 지정할 수 있습니다. 다만
이 경우 그 내용을 공지합니다.
제 13 조 (홈페이지 저작권)
① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
