Phellinus igniarius로부터 분리한 단백다당류의 분리 및 특성 (Characteristics and purification of proteoglycan from Phellinus igniarius)

  • Phellinus igniarius의 배양방법별 균사체 및 단백다당류 생산수율비교 실험에서는 균사체내 다당류의 경우 모두 진탕배양이 효과적이었으나 균사체외 다당류의 경우 다당류 생산에서는 정치배양이 효과적이었다. 다당류의 정제는 조단백다당류를 이용하여 DEAE-cellulose column에 의한 1차 정제를 행하였고, 최종으로 Sepharose 2B를 이용한 2차 정제를 실시하여 최종적으로 균사체내 단백다당류의 탈이온수 분획물(PIIPDG)과 알칼리분획물(PIIPAG), 균사체외 단백다당류의 탈이온수 분획물(PIEPDG) 알칼리 분획물(PIEPAG)을 얻었다. 이때의 정제 수율은 1차 정제에서 40%의 회수율을 나타내었으며 2차 정제에서는 $38{\sim}61%$의 높은 회수율을 보였다. PIEPDG는 총당 79.0%, 총단백질 7.2%, PIEPAG는 총당 56.7%, 총단백질 40.8%, PIIPDG는 총당 64.8%, 총단백질 17.4%, PIIPAG는 총당 56.9%, 총단백질 41.5%으로 측정되었다. 각 단백다당류의 분자량은 PIEPDG 166KDa에서 PIEPAG 565KDa까지 모두 10만이 넘는 거대분자로 나타났다. 각 분획물의 단당류 조성을 볼때, PIEPDG는 glucose, PIIPDG와 PIIPAG는 glucose, inositol, PIEPAG는 glucose, rfructose, inositol이 검출되었다.

Biased G Protein-Coupled Receptor Signaling: New Player in Modulating Physiology and Pathology

  • G protein-coupled receptors (GPCRs) are a family of cell-surface proteins that play critical roles in regulating a variety of pathophysiological processes and thus are targeted by almost a third of currently available therapeutics. It was originally thought that GPCRs convert extracellular stimuli into intracellular signals through activating G proteins, whereas ${\beta}$-arrestins have important roles in internalization and desensitization of the receptor. Over the past decade, several novel functional aspects of ${\beta}$-arrestins in regulating GPCR signaling have been discovered. These previously unanticipated roles of ${\beta}$-arrestins to act as signal transducers and mediators of G protein-independent signaling have led to the concept of biased agonism. Biased GPCR ligands are able to engage with their target receptors in a manner that preferentially activates only G protein- or ${\beta}$-arrestin-mediated downstream signaling. This offers the potential for next generation drugs with high selectivity to therapeutically relevant GPCR signaling pathways. In this review, we provide a summary of the recent studies highlighting G protein- or ${\beta}$-arrestin-biased GPCR signaling and the effects of biased ligands on disease pathogenesis and regulation.

Sequence Analysis and Potential Action of Eukaryotic Type Protein Kinase from Streptomyces coelicolor A3(2)

  • Protein kinase C (PKC) is a family of kinases involved in the transduction of cellular signals that promote lipid hydrolysis. PKC plays a pivotal role in mediating cellular responses to extracellular stimuli involved in proliferation, differentiation and apoptosis. Comparative analysis of the PKC-${\alpha},{\beta},{\varepsilon}$ isozymes of 200 recently sequenced microbial genomes was carried out using variety of bioinformatics tools. Diversity and evolution of PKC was determined by sequence alignment. The ser/thr protein kinases of Streptomyces coelicolor A3 (2), is the only bacteria to show sequence alignment score greater than 30% with all the three PKC isotypes in the sequence alignment. S.coelicolor is the subject of our interest because it is notable for the production of pharmaceutically useful compounds including anti-tumor agents, immunosupressants and over two-thirds of all natural antibiotics currently available. The comparative analysis of three human isotypes of PKC and Serine/threonine protein kinase of S.coelicolor was carried out and possible mechanism of action of PKC was derived. Our analysis indicates that Serine/ threonine protein kinase from S. coelicolor can be a good candidate for potent anti-tumor agent. The presence of three representative isotypes of the PKC super family in this organism helps us to understand the mechanism of PKC from evolutionary perspective.

오디씨 에탄올 추출물의 멜라닌 합성 억제효과 (Melanogenesis Inhibitory Activities of Mulberry Seed Ethanol Extracts)

  • 본 연구는 오디씨 에탄올 추출물(MSE)의 멜라닌 합성 저해 효과를 밝히는 것이다. 먼저 MSE의 melan-a 세포를 이용한 멜라닌 합성 저해 실험결과, 독성을 보이지 않는 $10{\mu}g/mL$의 농도까지 멜라닌 합성 저해 효과를 나타내었다. 또한 tyrosinase, tyrosinase-related protein-1 (TRP-1) 단백질의 발현이 저해되었으며, extracelluar signal-regulated protein kinase (ERK)의 발현을 농도 의존적으로 증가시키는 MSE의 기전을 확인하였다. 뿐만 아니라, 제브라피쉬를 이용한 in vivo 모델의 실험에서도 색소 발생이 저해됨을 관찰하였다. 따라서 오디씨로 부터 획득한 에탄올 추출물이 ERK 단백질의 발현으로 인해 멜라닌 생합성을 억제할 수 있음을 밝혔다.

Hypothetical protein predicted to be tumor suppressor: a protein functional analysis

  • Litorilituus sediminis is a Gram-negative, aerobic, novel bacterium under the family of Colwelliaceae, has a stunning hypothetical protein containing domain called von Hippel-Lindau that has significant tumor suppressor activity. Therefore, this study was designed to elucidate the structure and function of the biologically important hypothetical protein EMK97_00595 (QBG34344.1) using several bioinformatics tools. The functional annotation exposed that the hypothetical protein is an extracellular secretory soluble signal peptide and contains the von Hippel-Lindau (VHL; VHL beta) domain that has a significant role in tumor suppression. This domain is conserved throughout evolution, as its homologs are available in various types of the organism like mammals, insects, and nematode. The gene product of VHL has a critical regulatory activity in the ubiquitous oxygen-sensing pathway. This domain has a significant role in inhibiting cell proliferation, angiogenesis progression, kidney cancer, breast cancer, and colon cancer. At last, the current study depicts that the annotated hypothetical protein is linked with tumor suppressor activity which might be of great interest to future research in the higher organism.

KMU001 조효소에 의한 목질계 바이오매스, 볏짚의 효소당화성에 관한 연구 (Enzymatic saccharification of rice straw, a lignocellulosic biomass by the extracellular enzyme from KMU001)

  • This study shows that lignocellulosic biomass saccharification work has been carried out with rice-straw by the extracellular enzyme from KMU001, and the enzymes produced in 5%(w/v) wood biomass were characterized by protein and various enzyme activity measurements. Several cellulases such as Endoglucanase(EG), $\beta$-D-1,4-Glucosidase(BGL), Cellobiohydrolase(CBH), and $\beta$-D-1,4-Xylanase (BXL) were detected. Saccharification of rice-straw by the enzyme yielded about 233mg/g of glucose after 48hrs.

Shotgun Phage Display of Lactobacillus casei BL23 Against Collagen and Fibronectin

  • Lactobacilli are normal constituents of the intestinal microbiota, and some strains show the capacity to bind to extracellular matrix proteins and components of the mucosal layer, which represents an adaptation to persist in this niche. A shotgun phage-display library of Lactobacillus casei BL23 was constructed and screened for peptides able to bind to fibronectin and collagen. Clones showing binding to these proteins were isolated, which encoded overlapping fragments of a putative transcriptional regulator (LCABL_29260), a hypothetical protein exclusively found in the L. casei/rhamnosus group (LCABL_01820), and a putative phage-related endolysin (LCABL_13470). The construction of different glutathione S-transferase (GST) fusions confirmed the binding activity and demonstrated that the three identified proteins could interact with fibronectin, fibrinogen, and collagen. The results illustrate the utility of phage display for the isolation of putative adhesins in lactobacilli. However, it remains to be determined whether the primary function of these proteins actually is adhesion to mucosal surfaces.

Trichoderma viride 균체외 효소로 부터 Xylanase의 정제 및 Xylan의 분해 (Purification of an Xylanase from the Extracellular Xylanolytic Systems of Trichoderma viride and Hydrolysis of Xylan)

  • The endo-1,4-${\beta}$-xylanase was extracted and purified from the extracellular xylanolytic systems of Trichoderma viride. The crude enzyme was chromatographed with ion-exchange reins of DEAE Sepharose CL-6B, Sepharose, S-Sepharose CL-6B and the resulting xylanase was turned out to be a single protein as 20KD hy SDS-polyacrylamide gel electrophoresis. The xylooligomers were obtained from xylan by incubation with the purified xylanase up to 50%. The ${\beta}$-xylosidase lost its activity completely by incubation of crude enzyme for 24hr with buffer solution of pH 2.8 at $27^{\circ}C$. And also, the xylooligomers were obtained from xylan as a main product by incubation with the crude enzyme treated with acidic buffer.

Amine Oxidase Activity of the Human Lysyl Oxidase-Like 3 (LOXL3) Protein

  • Lysyl oxidase (LOX) catalyzes the lysine-derived cross-links of fibrillar collagens and elastin in the extracellular matrix. Recent molecular cloning has revealed existence of a LOX family consisting of LOX and four lysyl oxidase-like proteins (LOXL, LOXL2, LOXL3 and LOXL4). Pathological conditions associated with impaired LOX activity in several heritable and acquired disorders lead to severe structural and functional abnormalities of cardiovascular tissues, such as occlusion of coronary arteries and aneurysms, suggesting an essential role for the LOX family proteins in the maintenance of the cardiovascular system. However, the specific roles of the lysyl oxidase-like proteins in normal and pathological conditions of the cardiovascular tissues have not been established yet. Here, I report that LOXL3, a novel member of the LOX family, is predominantly expressed in the aorta, with an amine oxidase activity toward collagen and elastin, suggesting an essential role of LOXL3 in the development and maintenance of the aorta.

Preparation of Dopamine Transporter-specific Antibodies Using Molecular Cloned Genes

  • Dopamine transporter (DAT) plays the most important role in terminating the actions of dopamines released into the synaptic cleft. DAT is also the target of various psychotropic drugs such as cocaine and amphetamine. In this study were prepared DAT-specific antibodies using the 2nd extracellular loop of rat DAT as an antigen. The 2nd extracellular loop of the rat DAT was expressed in bacterial as a fusion protein with glutathione-S-transferase, and injected ito rabbits to raise antibodies. Produced antibodies clearly recognized the rat DAT in ELISA, immunoblotting, and immumoprecipitation. As expected from the high sequence homology between the rat and human DAT, the antibodies raised for the rat DAT cross-reacted with the human DAT in the immunoblotting. Considering the specificity for DAT with wide range of applications such as ELISA, immunoblotting, and immunoprecipitation, these antibodies would be valuable tool for understanding the pharmacological actions of dopamine transporter and drug addition.
