  • Oxidative stress has been implicated in a range of disease states, including end-stage renal failure treated with hemodialysis [Westhuyzen J. et al, 2003]. Free radicals react with biological molecules and destroy the structure of cells, which eventually causes free-radical induced disease such as cancer, renal failure, aging, etc. Exogenous or endogenously produced nitric oxide (NO) inhibits superoxide-stimulated urea permeability. In the inner medulla, superoxide generation by local oxidases may stimulate urea transport, and the role of endogenous No may be to dampen this effect by decreasing superoxide levels [Zimpelmann J. et al, 2003 (Epub ahead of print)].   • through a literal study upon Arrhythmia between east and west medicine, the results were as follows 1. Arrhythmia is not only irregular cardiac beat but disorder of cardiac impulse making and conduction disturbance. it means almost irregularity of interval between two beat 2. In the east medicine, arrhythmia is represented various category as palpitation, continuous palpitation, dizziness, consumption which give the first consideration by the accompanied symptoms. A separate way in a diagnostics it become a base of diagnosis of diseases and decision of prognosis by the examination of pulse 3. In the west medicine, Arrhythmia is classified as disorder of heart rate, rhythm, conduction disturbance or tachycardia, bradycardia and it is concomitant with fatigue, palpitation, dyspnoea, syncope, chest discomfort 4. The diagnostic study of pulse condition which represent arrhythmia was started from $\mathbb{<}$Nae-Kyung$\mathbb{>}$ and it was revealed as pulse condition of rapid pulse, slow pulse, swift pulse, running pulse, knotted pulse, intermittent pulse etc. Out of them running pulse, knotted pulse, intermittent pulse which obviously are concomitant with irregularity of interval are clinically meaningful in a conditions of disease and decision of prognosis. and the significance of these pulse condition are transformed through the changes of the times 5. According to cause of disease it is classified by Arrhythmia(running pulse, knotted pulse, intermittent pulse) due to heat, cold, phlegm, deficiency(or insufficiency) and There are three categories of etiological factor that is, endogenous, exogenous and non-exo-endogenous factor. the endogenous factor is insufficiency of the heart Ki, deficiency of both Ki and blood, intemal stagnation of phlegm and fluid, stagnation of seven emotions. the exogenous factor is caused by stagnation of Ki and blood by six exogenous pathogenic factor and the non-exo-endgenous factors are improper diet, overstrain, traumatic injury. A cause of arrhythmia in western medicine are a organic and pathological change of the heart itself and malfunction of the autonomic nervous system.

  • Cleft lip and/or palate is the congenital orofacial malformation most commonly occurred in humans, The disease is multifactorial and is probably caused by genetic and/or environmental factors, So, there are many problems in research concerning etiology and in treatment of the disease, Even the most practiced and sophisticated methods of surgical repair are necessarily followed by scar contraction and fibrosis, which result in skeletal defects, dental abnormalities, cosmetic disfigurement, and speech impairment, As a result, Fetal surgery can be considered but practiced rarely when the deformity is not fatal to life, And treatment of cleft palate is performed in the form of medicine projection into uterus in animal experiments, Many studies show that growth factor and its receptor emerge from the developing palate; and the epidermal growth factor receptors have a important role in craniofacial development and in palatal fusion, The palatal morphogenesis of the avine is different from the mammal's; it takes the form of physiologic cleft palate, Recently, cleft palate fusion experiment was performed when the avine were in the period of palate formation through the exogenous TGF-β3 addition, and it showed that the exogenous TGF-β3 makes fusion of divided palate through certain process when cleft palate is occurred in palatal formation, In this study, I had the conformation of the fusion of cleft palate through the addition of TGF-β in case of chicken embryo, and observed the effect of TGF-β in EGF receptor distribution, And the following is the results of this study, 1. In case of the TGF-βl and β3 addition group, there was the decrease of EGFR(Epidermal Growth Factor Receptor) immunoreactivity in mesenchymal cells beneath the medial edge epithelium and also in epithelial mesenchymal interface which is between medial edge epithelium and nasal septum in 72 hours, 2, The immunoreactivity of the control group resembles that of normal chicken embryo palate in development, 3. In the view through fluorescence confocal microscopy, there was confluence in TGF-β3 addition group, This shows that the confluence induced by exogenous TGF-β3 is related to EGFR expression in palate of chicken embryo, which is a physiologic cleft palate model.

구조방정식 모델을 이용한 사회경제적 수준과 치주질환과의 연관성 (Relationship between socioeconomic status and periodontal disease using Structural Equation Modeling)

  • Objectives: The purpose of this study was to investigate the relationship between socioeconomic status and periodontal disease. Methods: The data used for the analysis were obtained from the sixth Korean National Health and Nutrition Examination Survey (2015). A total of 5,632 adults, aged over 19 years, were included in the study. This study analyzed data relating to socioeconomic status and health behaviors. The socioeconomic status was defined as an exogenous factor. Periodontal disease was the endogenous variable. Health behavior was a mediating variable. The study hypotheses were tested using Structural Equation Modeling analysis with Mplus 7.0 software. Results: According to the model fit test, the associated root-mean-square error was 0.05 (90% CI: 0.046-0.059); comparative fit index was 0.93; Tucker-Lewis index was 0.88; and standardized root-mean-square residual was 0.03. We found that socioeconomic status had significant effects on health behaviors (${\beta}=0.304$, p<0.001) and periodontal disease (${\beta}=-0.289$, p=0.001). Moreover, health behaviors had significant effects on periodontal diseases (${\beta}=0.143$, p<0.001). The analysis of a possible relationship between socioeconomic status and periodontal disease using path analysis demonstrated health behaviors difficulties, along with direct and indirect effects, further increased the risk of periodontal disease. Conclusions: The findings suggest that future interventions for the prevention of periodontal disease should consider health behavior.

Effects of Tissue Factor, PAR-2 and MMP-9 Expression on Human Breast Cancer Cell Line MCF-7 Invasion

  • Objective: This study aimed to explore the expression of tissue factor (TF), protease activated receptor-2 (PAR-2), and matrix metalloproteinase-9 (MMP-9) in the MCF-7 breast cancer cell line and influence on invasiveness. Methods: Stable MCF-7 cells transfected with TF cDNA and with TF ShRNA were established. TF, PAR-2, and MMP-9 protein expression was analyzed using indirect immunofluorescence and invasiveness was evaluated using a cell invasion test. Effects of an exogenous PAR-2 agonist were also examined. Results: TF protein expression significantly differed between the TF cDNA and TF ShRNA groups. MMP-9 protein expression was significantly correlated with TF protein expression, but PAR-2 protein expression was unaffected. The PAR-2 agonist significantly enhanced MMP-9 expression and slightly increased TF and PAR-2 expression in the TF ShRNA group, but did not significantly affect protein expression in MCF-7 cells transfected with TF cDNA. TF and MMP-9 expression was positively correlated with the invasiveness of tumor cells. Conclusion: TF, PAR-2, and MMP-9 affect invasiveness of MCF-7 cells. TF may increase MMP-9 expression by activating PAR-2.

크론씨병(극한성(局限性) 장염(腸炎))에 관(關)한 한의학적(韓醫學的) 고찰(考察) (The oriental-western literal study of Crohn's disease)

  • We arrived at the following conclusion after we have studied crohn's disease through the literatures of western and oriental medicine. 1. Crohn's disease has a characteristic of granulomatous necrosis and cicatricial inflammation which is called by segmental enteritis, granulomatous enteritis. This falls under the category of "Diarrhea", "Dysentery" and is similar to "small intestinal diarrhea", "acute temesmus", "splenic diarrhea" in oriental medicine. 2. In western medicine, the cause of crohn's disease indefinites, but it is presumed immunological unbalance of alimentary canal. In oriental medicine, it is summarized as the abnormal ascending and descending circulation of stomach and splenic energies, the hepatic stagnation, being the lower part of cleaning qi by exogenous disease, dyspeptic convulsion. 3. The presenting symptoms of crohn's disease are intermittent chronic diarrhea, fever, weight loss, abdominal spastic pain or abdominal discomfort. When anyone has a abdominal mass, a rectal abcess, and a rectal constriction by physical examination, we can doubt crohn's disease. 4. The methods of western medical treatment are a suppression of intestinal toxic contents and inflammatory mediator, a supply of nutritive substanceus to intestinal epithelial cell. Oriental medical treatments of these are "inducing diuresis", "warming kidney to reinforce yang", "nourishing qi to invigorate spleen", "eliminatin dampness by cooling" according to syndrome differentiations. As mentioned above, we can confirm possibility of oriental medical treatment that induces recovery of immunologic control function and we need advanced experiment, study, and clinical approach.

Microbiome-Linked Crosstalk in the Gastrointestinal Exposome towards Host Health and Disease

  • The gastrointestinal exposome represents the integration of all xenobiotic components and host-derived endogenous components affecting the host health, disease progression and ultimately clinical outcomes during the lifespan. The human gut microbiome as a dynamic exposome of commensalism continuously interacts with other exogenous exposome as well as host sentineling components including the immune and neuroendocrine circuit. The composition and diversity of the microbiome are established on the basis of the luminal environment (physical, chemical and biological exposome) and host surveillance at each part of the gastrointestinal lining. Whereas the chemical exposome derived from nutrients and other xenobiotics can influence the dynamics of microbiome community (the stability, diversity, or resilience), the microbiomes reciprocally alter the bioavailability and activities of the chemical exposome in the mucosa. In particular, xenobiotic metabolites by the gut microbial enzymes can be either beneficial or detrimental to the host health although xenobiotics can alter the composition and diversity of the gut microbiome. The integration of the mucosal crosstalk in the exposome determines the fate of microbiome community and host response to the etiologic factors of disease. Therefore, the network between microbiome and other mucosal exposome would provide new insights into the clinical intervention against the mucosal or systemic disorders via regulation of the gut-associated immunological, metabolic, or neuroendocrine system.

ABA Increases Susceptibility of Pepper Fruits to Infection of Anthracnose by Collectotrichum acutatum

  • To examine the relationship between plant hormones and the development of pepper anthracnose, we investigated the effects of several plant hormones on the progression of disease symptoms. Of the five plant hormones examined, abscisic acid (ABA) increased the lesion length and disease incidence on detached fruits of Capsicum annuum cv. Nokkwang. The simultaneous application of ABA with inoculation of Colletotrichum acutatum JC24 resulted in increased lesion length, depending the concentration of ABA applied. Additionally, application of ABA caused the development of pepper anthracnose in fruits of Capsicum baccatum cvs. PBC80 and PBC81, which were previously resistant to the disease. Furthermore, ABA administration rendered increased pathogenicity of other isolates of C. acutatum BAC02063, PECH10, and TCBNU3 obtained from the Chinese matrimony vine, peach, and tea tree, respectively. Our data suggest that exogenous ABA may result in the suppression of defense mechanisms of pepper fruits against anthracnose, which leads to a change in the susceptibility of pepper fruits and the development of pepper anthracnose.

Lack of Association between Polymorphisms in Genes MTHFR and MDR1 with Risk of Childhood Acute Lymphoblastic Leukemia

  • Background: Acute lymphoblastic leukemia (ALL) is a complex disease caused by interactions between hazardous exogenous or/and endogenous agents and many mild effect inherited susceptibility mutations. Some of them are known, but their functional roles still requireinvestigation. Age is a recognized risk factor; children with disease onset after the age of ten have worse prognosis, presumably also triggered by inherited factors. Materials and Methods: The MDR1 gene polymorphisms rs1045642, rs2032582 and MTHFR gene polymorphisms rs1801131 and rs1801133 were genotyped in 68 ALL patients in remission and 102 age and gender matched controls; parental DNA samples were also available for 42 probands. Results: No case control association was found between analyzed polymorphisms and a risk of childhood ALL development. Linkage disequilibrium was not observed in a family-based association study either. Only marginal association was observed between genetic marker rs2032582A and later disease onset (p=0.04). Conclusions: Our data suggest that late age of ALL onset could be triggered by mild effect common alleles.

In silico discovery and evaluation of phytochemicals binding mechanism against human catechol-O-methyltransferase as a putative bioenhancer of L-DOPA therapy in Parkinson disease

  • Levodopa (L-DOPA) therapy is normally practised to treat motor pattern associated with Parkinson disease (PD). Additionally, several inhibitory drugs such as Entacapone and Opicapone are also cosupplemented to protect peripheral inactivation of exogenous L-DOPA (~80%) that occurs due to metabolic activity of the enzyme catechol-O-methyltransferase (COMT). Although, both Entacapone and Opicapone have U.S. Food and Drug Administration approval but regular use of these drugs is associated with high risk of side effects. Thus, authors have focused on in silico discovery of phytochemicals and evaluation of their effectiveness against human soluble COMT using virtual screening, molecular docking, drug-like property prediction, generation of pharmacophoric property, and molecular dynamics simulation. Overall, study proposed, nine phytochemicals (withaphysalin D, withaphysalin N, withaferin A, withacnistin, withaphysalin C, withaphysalin O, withanolide B, withasomnine, and withaphysalin F) of plant Withania somnifera have strong binding efficiency against human COMT in comparison to both of the drugs i.e., Opicapone and Entacapone, thus may be used as putative bioenhancer in L-DOPA therapy. The present study needs further experimental validation to be used as an adjuvant in PD treatment.
