Effects of the Feral Peach (Prunus persica Batsch var. davidiana Max) Extract on the Lipid Compositions and Blood Pressure Level in Spontaneously Hypertensive Rats (야생 돌복숭아(Prunus persica Batsch var. davidiana Max.) 추출액이 선천성 고혈압 흰쥐의 지질성분 및 혈압 저하에 미치는 영향)

  • The purpose of this study was to observe the effects of the feeding physiological activity substance in feral peach(Prunus Persica Batsch var. davidiana Max.) extract intake on the improvement of the lipid compositions, apolipoprotein and blood pressure level in spontaneously hypertensive rats(SHR, Wistar strain, male) fed the experimental diets for 33 days. Concentrations of total cholesterol, triglyceride(TG), LDL-cholesterol, free cholesterol and atherosclerotic index in serum were significantly lower in the feral peach extract intake groups[groups 5g% Ex.(basal diet+feral peach 5.0g% extract), 10g% Ex.(basal diet+feral peach 10.0g% extract)] than those in the group Control(basal diet+water). In the ratio of HDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration, feral peach 5.0g%, 10.0g% extract intake groups(group 5g% Ex. and 10g% Ex.) were higher percentage than in the group Control. However, concentrations of total cholesterol and TC in liver and brain were significantly lower in the groups 5g% Ex. and 10g% Ex. than those in the group Control. But the concentrations of apolipoprotein (Apo) A-I and Apo A-II in serum were significantly higher in the feral peach 5.0g% and 10.0g% extract intake groups(5g% Ex. and 10g% Ex.) than in the control group. However, concentrations of Apo C-II, Apo C-III, Apo E and ratio of Apo B to Apo A-I in serum were fairly reduced in the groups 5g% Ex. and 10g% Ex. than in the control group. The levels of systolic and diastolic blood pressure were significantly lower in feral peach 5g% Ex. and 10g% Ex. groups than control group. However, no significance was found in the effect of among the groups(groups 5g% Ex. and 10g% Ex.). From these results, physiological activity substance in feral peach(Prunus persica Batsch var. davidiana Max.) extracts were effective on the improvement of the lipid compositions and cardiovascular heart disease, hypertension in spontaneously hypertensive rats. And particularly, feral peach extracts were more effective as a therapeutic regimen for the control of blood pressure in hypertension.

Comparative Analysis of Filtering Techniques for Vegetation Points Removal from Photogrammetric Point Clouds at the Stream Levee (하천 제방의 영상 점군에서 식생 점 제거 필터링 기법 비교 분석)

  • This study investigated the application of terrestrial light detection and ranging (LiDAR) to inspect the defects of the vegetated levee. The accuracy of vegetation filtering techniques was compared by applying filtering techniques on photogrammetric point clouds of a vegetated levee generated by terrestrial LiDAR. Representative 10 vegetation filters such as CIVE, ExG, ExGR, ExR, MExG, NGRDI, VEG, VVI, ATIN, and ISL were applied to point cloud data of the Imjin River levee. The accuracy order of the 10 techniques based on the results was ISL, ATIN, ExR, NGRDI, ExGR, ExG, MExG, VVI, VEG, and CIVE. Color filters show certain limitations in the classification of vegetation and ground and classify grass flower image as ground. Morphological filters show a high accuracy of the classification, but they classify rocks as vegetation. Overall, morphological filters are superior to color filters; however, they take 10 times more computation time. For the improvement of the vegetation removal, combined filters of color and morphology should be studied.

Antioxidant Effect of Garlic Supplement against Exercise-Induced Oxidative Stress in Rats (운동으로 유발된 산화 스트레스와 마늘의 항산화 작용)

  • This study was to investigate lipid peroxidation, antioxidant enzyme activity and DNA damage after exercise, and the protective effect of garlic against exercise-induced oxidative stress. Male Sprague-Dawley rats(4 weeks old) were randomly divided into three groups of 6 rats each; control group(Con) without garlic and exercise, Ex group with exercise alone, and Ex-G group with 2% garlic and exercise. For 4 weeks, rats were given diets containing 15% corn oil and 1% cholesterol with or without garlic. The swimming was selected as a model for exercise performance. Rats swam for 40 min a day, for 5 days a week. Group Ex and Ex-G showed significant lowering in body weight gain and fat accumulation compared to control. No significant changes were observed in levels of plasma cholesterol and triglyceride among three groups, demonstrating that exercise and garlic had no effects on changes of blood lipid. This finding of blood lipid seems to be due to higher plant sterol content in corn oil. The DNA tail moment of lymphocytes showed greater tendency in Ex and Ex-G than in control, but garlic supplements failed to suppress DNA damages. Compared to control, Ex had higher plasma TBARS which was lowered to the control's level in Ex-G with 2% garlic supplementation(p<0.05). Ex-G led to a higher hepatic superoxide dismutase(SOD) activity than control and Ex(p<0.05). Activity of hepatic catalase was also increased in Ex-G, while in Ex it was significantly low(p<0.05). It seemed that TBARS levels were related to the activities of SOD and catalase, and that garlic contributed to increasing the enzyme activities and led to decrease of TBARS. These results demonstrate that lipid peroxidation and DNA damage occur as a consequences of oxidative stress after exercise, and that antioxidant defense against oxidative stress could be enhanced by garlic supplementation through the induction of antioxidant enzymes. However, further investigations should be done on the garlic effect on DNA damage.

The effects of genistein and exercise on obesity and adipose tissue inflammation in ovariectomized mice (난소절제 쥐에서 비만과 지방조직 염증에 대한 제니스테인과 운동의 영향)

  • This study investigated the effects of exercise on genistein-induced adipose tissue reduction and anti-inflammation of adipose tissue in ovariectomized mice fed a high-fat diet. Compared to mice fed a high-fat diet (C), both genistein (G) or exercise only (Ex) not only decreased adipose tissue weight and adipocyte size, but also glucose and free fatty acid levels in serum, which were more effectively reduced when genistein and exercise were treated simultaneously (G/Ex). The expression of lipogenesis and inflammatory cytokine genes of both G and Ex in adipose tissue was decreased compared to C. In particular, the decrease of these genes expression in G/Ex was more effective than each treatment alone. Thus, this study revealed that simultaneous treatment of genistein and exercise resulted in more effective improvement of obesity and adipose tissue inflammation than genistein single treatment in ovariectomized mice fed a high-fat diet, and it is a result of positive regulation of lipogenesis genes. This study suggests that the exercise has a beneficial effect on anti-obesity and adipose tissue anti-inflammation of genistein.

Anti-hyperglycemic and Antioxidative Activities of Phenolic Acid Concentrates of Rice Bran and Hydroxycinnamic Acids in Cell Assays (미강 페놀산 농축물과 Hydroxycinnamic Acids의 세포내 항당뇨 및 항산화 활성)

  • Phenolic acid concentrates of rice bran(RB-ex) and hydroxycinnamic acids were investigated for their anti-hyperglycemic activities through glucose uptake and glucokinase activity using HepG2 cells and stimulatory effects on insulin secretion using HIT-T15 cells. RB-ex was prepared as an ethylacetate extract after alkaline hydrolysis and hydroxycinnamic acids, found as major compositions of RB-ex, such as ferulic acid(FA), sinapic acid(SA) and p-coumaric acid(p-CA) were investigated to compare with the properties of RB-ex. The properties of glucose uptake in HepG2 cells were examined in the absence of insulin and two different glucose concentrations(5.5 mM and 25 mM). RB-ex and FA showed anti-hyperglycemic activities through the increase of glucose uptake and the stimulation of glucokinase activity in HepG2 cells. RB-ex exhibited higher glucose uptakes with higher glucose concentrations, whereas FA exhibited the same increasing effects on both concentrations of glucose. RB-ex and FA exhibited doubled glucokinase activities relative to control. In the presence of insulin in the 25 mM glucose-containing medium, the levels of glucose uptake were increased in all treatments compared with control. As stimulatory effects of samples on insulin secretion were estimated, RB-ex and FA stimulated insulin secretion at a concentration of 25 ${\mu}g/m{\ell}$ and in particular, FA showed the highest amount of insulin-release in HIT-T15 cells. Antioxidative effects on HIT-T15 cells, RB-ex and hydroxycinnamic acids, excluding p-CA, showed inhibitory activities of 78% to 80% at a concentration of 100 ${\mu}g/m{\ell}$. On the basis of these results, we conclude that RB-ex and FA could help decrease blood glucose levels and prevent the cell damages via antioxidant activity.

Paratope Mapping of Anti-Ex-A IgG as Studied by NMR (NMR에 의한 anti-Ex-A IgG의 항원결합부위 해석)

  • The anti-Ex-A IgG was specifically labeled with stable isotopes, DL-His-2,4-$d_2$, L-Phe-$d_5$, L-Trp-$d_5$, L-Tyr-2,6-$d_2$ and L-[1-$^{13}C$]Trp, by growing hybridoma cell in serum-free medium. By use of NMR spectroscopy with selectively labeled Fab fragment, we applied a paratope mapping on antigen-antibody complex. Assignments of the observed carbonyl carbon resonances have been determined by using $^{13}C$-$^{15}N$ double labeling method in order to assign the Trp resonances. Photo CIDNP was also applied to investigate the antigen-binding site(s) on the surface residues of antibody. We found that Trp 36, which is located at the $V_H$ domain, is an important residue to bind to Ex-A, however, two Tyr on the surface of anti-Ex-A IgG plays no crucial role to bind to antigen. On the basis of these results, we demonstrate that stable isotope-aided NMR strategy can be extended to molecular structural analyses of the complex of an Fab fragment and a protein antigen.

Comparison of the effectiveness of extensor muscle strengthening exercise by itself, exercise with polydeoxyribonucleotide injection, and exercise with extracorporeal shockwave therapy in lateral epicondylitis: a randomized controlled trial

  • Background: Extensor muscle strengthening exercises with counterforce braces (EX) is a conventional conservative treatment for lateral epicondylitis (LE) of the elbow. In addition, polydeoxyribonucleotide (PDRN) or extracorporeal shockwave therapy (ESWT) has been recently used for LE. Methods: Sixty-three patients with chronic LE participated in this study and randomly allocated in three groups (G1: EX, G2: EX+PDRN injection, and G3: EX+ESWT). All of the three groups were taught to perform EX at the first out-patient department (OPD) visit. Group 2 was injected with 3 mL PDRN (5.625 mg/3 mL), while group 3 received ESWT at the first OPD visit. Visual analog scale pain score, Mayo elbow performance score (MEPS), and ultrasonographic examination were checked before, 6 weeks, and 12 weeks after the treatments. Results: Overall functional scores and ultrasonographic findings in all three groups improved after treatment. The mean MEPS in group 2 improved more than groups 1 and 3 at 6 weeks (G1, 56.9>62.4; G2, 54.3>65.0; G3, 55.7>62.6), and more than group 1 at 12 weeks (G1, 56.9>67.9; G2, 54.3>73.6). The mean common extensor tendon depth (CETD) on ultrasonography in group 2 increased more than groups 1 and 3 at 6 and 12 weeks (6 weeks: G1, 0.385>0.386; G2, 0.332>0.392; G3, 0.334>0.357; 12 weeks: G1, 0.385>0.409; G2, 0.332>0.438; G3, 0.334>0.405 [cm]). Conclusions: PDRN injections combined with EX exhibited a greater improvement in mean MEPS and mean CETD compared to EX only or EX combined with ESWT for LE within the 12 weeks follow-up.

Effect of Garlic Supplement and Exercise on Plasma Lipid and Antioxidant Enzyme System in Rats (마늘의 섭취와 운동이 혈장지질과 항산화효소계에 미치는 영향)

  • Effects of garlic powder supplementation on blood lipid profile and antioxidant system were investigated in rats with and without swimming exercise. Sprague-Dawley rats of four experimental groups were fed for 4 weeks diets containing $15\%$ beef tallow and $1\%$ cholesterol; control without garlic and exercise, Go with $2\%$ garlic alone, Ex with exercise alone, GoEx with $2\%$ garlic and exercise. Rats were trained 40 min a days a days a week. Group Ex and GoEx showed significant lowering in body weight gain and fat accumulation. In Go, Ex and GoEx, plasm TG and LDL-C were lower and HDL-C was higher, although not significantly, compared to levels in control. Total cholesterol was significantly reduced in group Go, and Ex and GoEx were lower than control. The total/HDL cholesterol ratio was also found to be significantly different, decreasing the ratios in Go, Ex and GoEx. The hepatic TBARS increased significantly in group Ex $(51.7{\pm}3.43nM/g\;liver)$, while TBARS in Go and GoEx were low $(35.68{\pm}3.61,\;39.30{\pm}5.55nM/g\;liver)$ and similar to control's one. The activity of hepatic SOD in Go and GoEx tended higher than control and Ex without garlic. The hepatic catalase showed significantly the highest activity in Go. Activity of GSH-px was significantly low in Ex with $0.14{\pm}0.03$ unit/mg protein, and control, Go and GoEx had higher activities of $0.23{\pm}0.08,\;0.20{\pm}0.07,\;0.22{\pm}0.01\;unit/mg$ protein, respectively. Lower activities of antioxidant enzymes in Ex are likely to associated with the highest level of TBARS. It seems that a decrease in TBARS in GoEx relative to Ex was related to the increase in GSHpx and SOD with garlic supplemented, which led to compensate the oxidative stress from exercise. The results suggests that exercise or garlic supplement exerts blood lipid attenuating effect. In adition, garlic supplementation could strengthen the antioxidant potential against exercise-induced oxidants, partly by modulating oxidant enzyme activity. These effects of garlic may make it a beneficial agent on CVD.

Protective effects of an ethanol extract of Angelica keiskei against acetaminophen-induced hepatotoxicity in HepG2 and HepaRG cells

  • BACKGROUND/OBJECTIVE: Although Angelica keiskei (AK) has widely been utilized for the purpose of general health improvement among Asian, its functionality and mechanism of action. The aim of this study was to determine the protective effect of ethanol extract of AK (AK-Ex) on acute hepatotoxicity induced by acetaminophen (AAP) in HepG2 human hepatocellular liver carcinoma cells and HepaRG human hepatic progenitor cells. MATERIALS/METHODS: AK-Ex was prepared HepG2 and HepaRG cells were cultured with various concentrations and 30 mM AAP. The protective effects of AK-Ex against AAP-induced hepatotoxicity in HepG2 and HepaRG cells were evaluated using 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide, lactate dehydrogenase (LDH) assay, flow cytometry, and Western blotting. RESULTS: AK-Ex, when administered prior to AAP, increased cell growth and decreased leakage of LDH in a dose-dependent manner in HepG2 and HepaRG cells against AAP-induced hepatotoxicity. AK-Ex increased the level of Bcl-2 and decreased the levels of Bax, Bok and Bik decreased the permeability of the mitochondrial membrane in HepG2 cells intoxicated with AAP. AK-Ex decreased the cleavage of poly (ADP-ribose) polymerase (PARP) and the activation of caspase-9, -7, and -3. CONCLUSIONS: These results demonstrate that AK-Ex downregulates apoptosis via intrinsic and extrinsic pathways against AAP-induced hepatotoxicity. We suggest that AK could be a useful preventive agent against AAP-induced apoptosis in hepatocytes.

Making an Index of Mobile Telecommunications Service Quality - An Application to OECD Countries - (이동통신 통화품질의 지표화 - OECD 국가들을 중심으로 -)

  • This paper attempts to develop an index of mobile telecommunications service quality through econometric analysis using data of the leading telco in each OECD country. CapEx of the operator was selected as a proxy variable to represent the quality of the service. To eliminate the part which is dependent on the population and size from CapEx, however, CapEx was regressed on population, area, 3G coverage, Herfindahl-Hirschman Index (HHI) of mobile market, EBITDA margin, and market share of the leading operator. Then, the part of CapEx, which was explained by size and population, was subtracted from CapEx, and the remaining part was regarded as the quality index of mobile telecommunications service. Comparisons among quality indexes of each country revealed that the service quality in Korea and Japan is better than that of France, UK and Germany.
