The poultry red mite (PRM), Dermanyssus gallinae, causes great economic losses to poultry industries in Korea. The molecular epidemiological characterization of PRM has been investigated in some countries, but those analysis has been not conducted yet in Korea. The aim of this study is to determine the genetic diversity of PRMs in Korea compared with those from other countries. Here, 13 PRM samples collected from Korea were analyzed with a part of the mitochondrial cytochrome oxidase subunit I (COI) gene and nuclear internal transcribed spacers (ITS) region. All the samples showed an identical COI sequence, which has also been reported in European countries and Japan. Phylogenetic diversity analysis showed that the mites from Korea were genetically related to those in other countries. The nuclear ITS region sequences were classified into three sequence types. Additionally, one of the ITS sequences was an intermediate type, implying that a hybridization event occurred among the mite populations in Korea. These findings suggested PRMs from Korea showed low genetic diversity with respect to mitochondrial COI gene, but three different populations inhabited in Korea with respect to nuclear ITS region sequences.

Living modified crops are genetically modified living organisms and are widely used in biotechnical research and desired goods. As the reliance on LM products, concerns about safety of LMOs have been continuously increased in South Korea. We established the detection methods for unintentional released LMOs in environmental conditions. To detect six LM event genes of 1 canola, 1 maize and 4 soybeans, PCR conditions were based upon consideration of the Joint Research Centre information. Genomic DNAs were isolated from LM samples and PCR analysis were performed using each event-specific primer pair. Event-specific genes of all events were efficiently recognized by our methods. To investigate the insertion site of LM genes in each genome, we verified PCR product sequence by DNA sequencing. These results suggest that the LM event-specific gene amplification can be efficiently developed. In addition, our detection method is fit for monitoring and post-management of LM crops in the environment.

South Korea opened Incheon international airport(IIA) in march 29, 2001, with high expectations of becoming the distribution hub of Northeast Asia and aiming at a world best air hub. IIA compares quite well with any other leading airports in the world in it's facilities for the movement of people and vehicles. However, with the sequence of events following the September, 2001. terrorist attack and the war in Iraq, South Korea, an ally of the US, cannot be considered a safe haven from terrorism. At a point in time when national security is given utmost importance, it is necessary to reevaluate the security of airports, because international terrorism can only occur via air and seaports. Nowadays all the countries of the world have entered into competition for their national interests and innovation of their images. with the increasing role of international airports also comes an increased likelihood as a terrorist target, because it can affect so many people and countries. From the condition of current international terrorism, we can realize that our IIA is not completely safe from a terrorist attack. The major part of counterterrorism is event control process. It's very important for quickly saves an accident and rescues a life of person, In addition for the normalization which the airport operation is prompt. In conclusion, we should secure the legal responsibility and establish and establish a system under which we can work actively in order to implement counter terror activities from being taken an airport.
Recently personal media were produced in a variety of ways as a lot of smart devices have been spread and services using these data have been desired. Therefore, research has been actively conducted for the media analysis and recognition technology and we can recognize the meaningful object from the media. The system using the media ontology has the disadvantage that can't classify the media appearing in the video because of the use of a video title, tags, and script information. In this paper, we propose a system to automatically classify video using the objects shown in the media data. To do this, we use a description logic-based reasoning and a rule-based inference for event processing which may vary in order. Description logic-based reasoning system proposed in this paper represents the relation of the objects in the media as activity ontology. We describe how to another rule-based reasoning system defines an event according to the order of the inference activity and order based reasoning system automatically classify the appropriate event to the category. To evaluate the efficiency of the proposed approach, we conducted an experiment using the media data classified as a valid category by the analysis of the Youtube video.
Objective: Mitochondrial DNA (mtDNA) is considered a hotspot of mutations in various tumors. However, the relationship between microsatellite instability (MSI) and mtDNA copy number alterations in lung cancer has yet to be fully clarifieds. In the current study, we investigated the copy number and MSI of mitochondrial genome in lung carcinomas, as well as their significance for cancer development. Methods: The copy number and MSI of mtDNA in 37 matched lung carcinoma/adjacent histological normal lung tissue samples were examined by polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) assays for sequence variation, followed by sequence analysis and fluorogenic 5'-nuclease real-time PCR. Student's t test and linear regression analyses were employed to analyze the association between mtDNA copy number alterations and mitochondrial MSI (mtMSI). Results: The mean copy number of mtDNA in lung carcinoma tissue samples was significantly lower than that of the adjacent histologically normal lung tissue samples (p<0.001). mtMSI was detected in 32.4% (12/37) of lung carcinoma samples. The average copy number of mtDNA in lung carcinoma samples containing mtMSI was significantly lower than that in the other lung carcinoma samples (P<0.05). Conclusions: Results suggest that mtMSI may be an early and important event in the progression of lung carcinogenesis, particularly in association with variation in mtDNA copy number.
The DNA sequence immediately following the xynA gene of Bacillus stearothermophilus 236 [about l-kb region downstream from the translational termination codon (TAA) of the xynA gene]was found to have an ability to enhance the xylanase activity of the upstream xynA gene. An 849-bp ORF was identified in the downstream region, and the ORF was confirmed to encode a novel protein of 283 amino acids designated as XaiF (xylanase-activity-increasing factor). From the nucleotide sequence of the xaiF gene, the molecular mass and pI of XaiF were deduced to be 32,006 Da and 4.46, respectively. XaiF was overproduced in the E. coli cells from the cloned xaiF gene by using the T7 expression system. The transcriptional initiation site was determined by primer extension analysis and the putative promoter and ribosome binding regions were also identified. Blast search showed that the xaiF and its protein product had no homology with any gene nor any protein reported so far. Also, in B. subtilis, the xaiF trans-activated the xylanase activity at the same rate as in E. coli. In contrast, xaiF had no activating effect on the co-expressed ${\beta}-xylosidase$ of the xylA gene derived from the same strain of B. stearothermophilus. In addition, the intracellular and extracellular fractions from the E. coli cells carrying the plasmid-borne xaiF gene did not increase the isolated xylanase activity, indicating that the protein-protein interaction between XynA and XaiF was not a causative event for the xylanase activating effect of the xaiF gene.
Sequences of the trnL/F intergenic spacer of chloroplast DNA were used to investigate the intraspecific evoution and phylogeography of Sedum takesimense (Crassulaceae). The trnL/F intergeneric spacer sequences from 32 individuals of S. takesimense were either 291 bp (17 samples "without indel" in the following) or 297 bp (15samples "with indel 1") in length due to an indel of 6 bp. Two main cpDNA haplotypes were detected within S. takesimense. The haplotype with indel was found on Ulleung Island and without indel on Ulleung Island and Dok Island. This confirmed the existence of two cpDNA lineages with different geographical distributions. The cpDNA sequence analysis also suggested a putative long-distance dispersal event between Ulleung Island and Dok Island.

Over the last five years, we have conducted research on risk assessment of domestically developed genetically modified (GM) crops and found a number of factors which could delay risk assessment process. In this review, we described such cases and discussed the problem of transgene cassette integration, the lack of information on vectors, the poor quality control in seed production and absence of bioinformatic analysis on amino acid sequence homology before GM crop development. To solve these problems, we have suggested the introduction of the screening system of elite event before risk assessment process and quality control strategies for GM seed production. In addition, we suggested that the developers of GM crops should understand the importance of risk assessment and management for the commercialization of those crops and consider the biological and ecological characteristics of host plants. Consistent communications may need to be established between GM crop developers, risk assessors and risk managers at the initial stages of GM crop development to reduce trial-and-errors.

Through molecular phylogenetic analysis using the nef gene sequences of HIV-l isolated from Korean registered in the NCBI GenBank together with 41 reference strains and 94 foreign isolates, we verified that most (${\sim}80%$) of Korean isolates belonged to subtype B and 78% of subtype B were clustered together exclusively of foreign isolates, and this cluster was named Korean clade subtype B ($K_cB$). Similarity study suggested that the $K_cB$ cluster was more homogeneous than and clearly distinctive from the non-Korean subtype B ($NK_cB$). Comparison of the consensus amino acid sequences of the $K_cB\;or\;NK_cB$ revealed characteristic $K_cB$ signature amino acid pattern comprised of 13 amino acid residues. The $K_cB$ signature amino acid residues were critical in separating the $K_cB$ ftom the $NK_cB$, since substitution of the $NK_cB$ sequences with $K_cB$ signature amino acids relocated them to the Koran clade, and vice versa. Synonymous and nonsynonymous substitution rate study suggested positive selection event for the $K_cB$.
For the development of qualitative and quantitative PCR methods of genetically modified (GM) pepper developed in Korea, a capsanthin-capsorubin synthase (CCS) gene was used as the endogenous reference gene. The primer pair ccs-F/R amplifying the pepper endogenous gene gave rise to an amplicon of 102 bp. No amplified product was observed when DNA samples from 16 different plants were used as templates. The construct-specific primer pairs amplifying the junction region of the bar gene and Ti7 introduced in GM pepper gave rise to an amplicon of 182 bp. Quantitative PCR assay was performed using a TaqMan probe and a standard plasmid as a reference molecule, which contained both an endogenous and event-specific sequence. For the validation of this method, the test samples containing 0.1, 1, 3, 5, and 10% GM pepper were quantified.
