Ginsenosides: the need to move forward from bench to clinical trials

  • Panax ginseng, known as Koran ginseng, one of the most commonly used traditional plants, has been demonstrated to show a wide range of pharmacological applications. Ginsenosides are the major active ingredients found in ginseng and are responsible for the biological and pharmacological activities, such as antioxidation, antiinflammation, vasorelaxation, and anticancer actions. Existing studies have mostly focused on identifying and purifying single ginsenosides and investigating pharmacological activities and molecular mechanisms in cells and animal models. However, ginsenoside studies based on clinical trials have been very limited. Therefore, this review aimed to discuss the currently available clinical trials on ginsenosides and provide insights and future directions for developing ginsenosides as efficacious and safe drugs for human disease.

  • The therapeutic goal in liver fibrosis is to reverse fibrosis and selective clearance of activated hepatic stellate cells (HSCs), which playa central role in liver fibrogenesis, by apoptosis might be essential during resolution of fibrosis. Past several years we screened for natural products which mediate apoptosis in activated HSCs, and among the candidates honokiol, isolated from Magnoliae Cortex, was found to induce apoptotic death in activated rat HSCs in a dose- and time-dependent manner at the concentration between 12.5 microM and 50 microM.   • A liquid chromatography-tandem mass spectrometric (LC/MS/MS) method was developed for the determination of a new anti-angiogenic drug KR-31831 in rat plasma. KR-31831 and internal standard, KR-31543 were extracted from rat plasma with dichloromethane at basic pH. A reverse-phase LC separation was performed on Luna C8 column with the mixture of acetonitrile-ammonium formate (10 mM, pH 4.5) (67:33, v/v) as mobile phase. The detection of analytes was performed using an electrospray ionization tandem mass spectrometry in the multiple-reaction-monitoring mode. (omitted)

Expectation and Satisfaction of Parents with Inpatient Hospital Service (입원 아동 부모의 병원서비스 기대수준과 만족도)

  • Purpose: The purpose of this study was to investigate parent expectation and satisfaction with respect to pediatric inpatient care and to identify the variables related to parent satisfaction. Methods: The study was conducted in pediatric wards of a tertiary children's hospital in Korea. The participants were 361 parents of children who were inpatients. Data were collected using a structured questionnaire (The Pediatric Family Satisfaction Questionnaire) at the time of discharge. Results: The highest parent expectation domain was medical service. The parents were most satisfied with nursing service and least satisfied with general hospital service and accommodation. The parents expressed lower satisfaction with hospital facilities, equipment, noise, cleanliness, and communication by health care professionals. Parents with younger children reported higher expectation from the complete hospital service and those who had a longer length of stay reported higher expectation from the nursing service. Conclusion: To improve the quality of hospital services, we need to understand parent expectation and improve and provide clear communication. In addition, the general hospital service and accommodation should not be overlooked for improvement.

  • This study investigated the effects of three factors-mothers' parenting beliefs; child care-home involvement; and the home learning environment - on the school readiness of 3- to 5-year-olds. The subjects were 366 children who were enrolled in child care centers located in Seoul and the Kyoungki area, and their mothers. The Structural Equation Modeling (SEM) technique was employed to test the pathways to children's school readiness as indicated by the child's abilities in vocabulary, math and reading. The results showed that mothers' stronger beliefs in their responsibilities in their children's academic and behavioral development predicted greater involvement in child care and better quality in the home learning environment. Likewise, the quality of the learning environment predicted the extent of the child's readiness for school. No direct relation was found between child care involvement and the child's school readiness. The results imply that multiple factors - parental, child-care-related, and home environmental- explain the extent to which the child is prepared to adjust to scholastic life.

Optimization of in vitro seed germination of Taraxacum platycarpum

  • Dandelion (Taraxacum platycarpum) has been widely utilized for medicinal purposes. However, the dandelion seeds are relatively difficult to germinate under cultivation conditions, which hampers seedling propagation of dandelion plants and reduces the opportunity of usage of such a useful medicinal plant. Thus, in this study, in vitro conditions for the dandelion seed germination were optimized to enhance the germination rate. In seed washing steps, the sequential treatments with 20% of ethanol, 20% of NaOCl, and distilled water avoided microbial contamination with the highest in vitro germination rate (67.5%) from seeds sown in germination media. The media supplemented with 1.4 g/L of MS salts and 1% of sucrose significantly enhanced the germination rate compared to the media with 4.4 g/L of MS and 3% of sucrose. Sowing the seeds vertically in the optimized media supplement conditions, 1.4 g/L of MS salts and 1% of sucrose, gave the maximum in vitro germination rate (61%), which was almost three times higher than sowing seeds on a soil pot (23%). Our results indicate that the seed washing and sowing methods including germination medium supplements can be optimized to enhance in vitro seed germination of dandelion.

The Effects of Lead Exposure on Glutamatergic Nervous System in Rat Cerebellar Cells

  • Changes in glutamate release and uptake on cerebellar cells after the chronic exposure to lead were investigated. Rats were received 0.25% lead acetate in drinking water from the beginning of the pregnancy. The control group was given 0.125% sodium acetate in drinking water. The cerebellar cells from 7 or 8 day-old pups were cultured. Amino acid release from cerebellar granule cells and the glutamate uptake into cerebellar glial cells were measured using HPLC-ECD. Basal glutamate release and NMDA-induced glutamate release didn't show significant difference. However, the other amino acids in the granule cells obtained from lead exposed pups were less released than the control after the stimulation by NMDA (50$\mu$M). SNAP-induced (50$\mu$M) glutamate release was significantly reduced in granule cells prepared from lead exposed pups. The basal glutamate uptake in glial cells didn't show any difference. However, the uptake in glial cells prepared from lead exposed pups was significantly less blocked by PDC (24$\mu$M) compared to the control group. These results indicate that lead exposure to the mother might affect the Excitatory amino acid system during the development of the offspring.

Prediction of rice growth parameters by X-band automatic scatterometer system (X-band 자동관측시스템을 이용한 벼 생육인자 추정)

  • 본 연구에서는 기후 등의 영향을 받지 않고 레이더 산란 측정을 할 수 있는 X-band antenna 기반의 자동관측 시스템을 이용하여 벼 생육시기에 따른 후방산란계수와 벼 생육인자와의 관계를 분석하여 후방산란계수를 이용한 벼 생육인자를 추정한 것을 목적으로 하였다. 2008년도 국립농업과학원 시험포장 ($37^{\circ}$15'28.0"N, $126^{\circ}$59'21.5"E)에서 추청벼를 대상으로 생육시기별 후방산란계수를 관측하였는데 모든 편파별 후방산란계수가 벼 유수형성기 (7월 말경)까지 증가하다가 그 후 감소하다가 수확기가 가까워지는 9월 중순이후 다시 증가하는 dual-peak 현상을 보였고 특히 W-편파의 경우 9월 초순부터 후방산란계수 증가가 다른 polarization에 비해 크게 나타났다. 후방산란계수와 작물생육인자와의 관계를 분석한 결과 고주파수인 X-band는 상대적으로 바이오메스, 엽 면적지수와의 상관이 낮게 나타났지만 이삭 건물중은 VV-편파 후방산란계수와 상관관계를 보였다. 이삭 건물중과 상관관계가 높게 나타난 X-band의 W-편파 후방산란계수를 이용하여 수확기 이삭 건물중을 추정하였는데 VV-편파 후방산란계수와 이삭 건물중과는 결정계수 $(R^2)$가 0.85이었고, 이삭 건물중 실측값과 추정값을 비교해 본 결과 1:1 line에 근접하게 분포하였다 ($R^2$=0.85).

A Study on the Engineering Design for 20kW-Grade Waste Gas Heat Recovery (20kW급 폐열회수 시스템 공정 설계에 관한 연구)

  • This study is collects design data through the process design of the organic Rankine cycle, which can produce 20kW of electric power through the recovery of waste heat. In this study, the simulation was conducted by using APSEN HYSYS in order to make the model for the process design of the 20kW class waste heat recovery system. For the thermodynamic model, the test was conducted with hot water as the heat source, with the water steam used as the cooling water for the cooler and the refrigerant R245fa in the cycle. In Case 1 and Case 2, it was expected and found that the cycle efficiency was 10.6% and that 36.86kw was produced, considering the margin of 84% of 20kW. In Case 3 and Case 4, it was expected and found from the simulation that the cycle efficiency was 12% and that 30.0kw was produced, considering the margin of 84% of 20kW.
