The Behavior of a $CO_2$Fixation Process by Euglena Gracilis Z with a Photobioreactor (광반응기와 Euglena gracilis Z를 이용한 이산화탄소 고정화 공정의 거동 특성)

  • Biological fixation of carbon dioxide using microalgae is known as an effective CO$_2$reduction technology. However, many environmental factors influence microalgal productivity. Optimal cultivation factors were determined for the green alga, Euglena gracilis Z, which offers high protein and vitamin E content for animal fodder. In batch culture in a photovioreactor, it was found that theinitial pH, temperature, CO$_2$concentration in air, and light intensity during the optimal cultivating conditions were 3.5, 27$^{\circ}C$, 5-10% and 520 ${\mu}$mol/㎡/s, respectively. When tap water and freshwater were used as cultivating media unsterilized tap water was found to be effective. A kinetic model was considered to determine the relationship between the specific growth rate and the light intensity. The half-velocity coefficient (K(sub)I) in the Monod model under photoautotrophic conditions was 978.9 ${\mu}$mol/㎡/s.

Effect of Dietary Supplementation of Fresh Water Algae Euglena on the Performance and Fatty Acid Composition of Breast Muscle of Broiler Chickens (육계에서 담수녹조류 Euglena 첨가사료가 생산성 및 흉근의 지방산 조성에 미치는 영향)

  • A feeding trial was conducted with Euglena strains grown under different media. The effect of supplementation of Euglena on the performance, nutrient availability and fatty acid composition of breast muscle was studied. In experiment I, two hundred ten hatched broiler chicks (Ross) were assigned to seven dietary treatments for 5 weeks. Each treatment consisted of 3 replications with 10 birds each. Control diet was formulated to have $22\%$ CP and 3,150 kcal ME/kg for starter diet, $19\%$ CP and 3,200 kcal ME/kg for finisher diet. Euglena gracilis Z. (EG) was added to control diet at the plevel of 0.25, 0.5, $1.0\%$ and Euglena gracilis Z. bleached and DHA enriched (EGBD; a strain mutated by streptomycin and cultivated in DHA enriched medium) at the level of 0.5, 1.0, $2.0\%$ in the diet. In experiment 2, two hundred fifty hatched broiler chicks (Ross) were assigned to five dietary treatments: T1; Control, T2; T1 + Euglena gracilis Z. DHA enriched (EGD; cultivated in DHA enriched medium) $0.5\%$, T3; T1 + EGD $1.0\%$, T4; T1 + EGBD $0.5\%$, T5; T1 + EGBD $1.0\%$. The weight gain and feed consumption were measured weekly. Fatty acid composition of breast muscle was determined. In experiments I and 2, Euglena supplementation had no significant effects on weight gain, feed intake and feed conversion ratio. In experiment 1, EGBD treatments significantly increased DHA concentration but decreased concentration of linoleic acid and arachidonic acid in breast muscle. EGBD 2% treatment showed the highest DHA concentration (14.27%) which is 3.9 times of that of the control ($3.66\%$). In experiment 2, $1.0\%$ EGBD treatment showed highest EPA, lignoceric acid and DHA level in breast muscle (P<0.05). Also, EGD treatments significantly increased DHA and EPA concentration. It was concluded that EGBD and EGD can be supplemented to broiler diet to produce DHA enriched broiler meat.

  • In Euglena gracilis arginine deiminase was located in the mitochondrial matrix. The highly purified enzyme required $Co^{2+}$ for the enzyme reaction with the $K_m$ value of 0.23 nM, and its optimum pH was 9.7 to 10.3. The molecular weight of the native enzyme protein was 87,000 by gel filtration, and SDS-acrylamide gel electrophoresis showed that the enzyme consisted of two identical subunits with a molecular weight of 48,000. Euglena arginine deiminase was inhibited by sulfhydryl inhibitors, indicating that a sulfhydryl group is involved in the active center of the enzyme. It exhibited negative cooperativity in binding with arginine. $L-{\alpha}-amino-{\beta}-guanidino-propionate$, D-arginine, and L-homoarginine strongly inhibited the enzyme while ${\beta}-guanidinopro-pionate$, ${\gamma}-guanidinobutyrate$, and guanidinosuccinate did not. Considerable inhibition was also observed with citrulline and ornithine. We discuss the effects of the unique properties of the Euglena arginine deiminase on the regulation of arginine metabolism in this protozoon.

The effect of supplementation of Euglena on the laying performance, egg quality and fatty acid composition of egg yolk was studied. In experiment I, two hundred eighty 32-wk-old ISA Brown layers were randomly assigned to seven dietary treatments for 4 wks. Each treatment consisted of 4 replications with 10 birds each housed in two birds cages. Control diet was formulated to have $17\%$ CP and 2,750 kcal ME/kg. Euglena gracilis Z. (EG) was added to control diet at the level of 0.25, 0.5, $1.0\%$ and Euglena gracilis Z. bleached and DHA enriched (EGBD; a strain mutated by streptomycin and cultivated in DHA enriched medium) at the level of 0.5, 1.0, $2.0\%$ in the diet. In experiment 2, three hundred 84-wk-old ISA brown layers were randomly assigned to five dietary treatments: T1; Control, T2; T1 + EGBD $0.5\%$, T3; T1 + Euglena gracilis Z. DHA enriched (EGD; cultivated in DHA enriched medium) $0.5\%$, T4; T1 + EGD $1.0\%$, T5; T1 + EGD $2.0\%$. Each treatment had 5 replication of 12 birds each housed in two birds cages. In experiments 1 and 2, Euglena suppplementation did not significantly affect egg production but increased egg weight and feed intake. In experiment 1, EG was more effective in increasing egg yolk color score than EGBD. Egg yolk color of EG $1\%$ treatment showed the highest score. EGBD supplementation increased DHA concentration of egg yolk. EGBD $2\%$ treatment showed the highest DHA and the lowest palmitic and stearic acids concentration in the egg yolk. In experiment 2, EGBD $0.5\%$ treatment showed highest DHA level in egg yolk (P<0.05). It was conducted that EGBD is a single cell protein source rich in DHA, that can be used to produce DHA enriched eggs.

  • The biological carbon dioxide fixation using microalgae and photo-bioreactor has been known as an effective carbon dioxide reduction technology. As algae has many other environmental factor for its growth, the desirable cultivation factors were investigated using a green alga, Euglena gracilis Z. In this study, Euglena gracilis Z showed good $CO_2$ fixation ability in high $CO_2$ concentration of 10-20% and it contained the high protein and vitamin E enough to be used as fodder. For the mass cultivation, the continuous and semi-continuous cultivation methods were employed. The optimum hydraulic retention time (HRT) for the continuous cultivation was 4 days at carbon dioxide concentration of 10%. In this condition, the final cell number was $3.57{\times}10^6/m{\ell}$. The growth of Euglena gracilis Z increased according to the light intensity.

  • The concentrations of dissolved vitamin $B_{12}$, thiamine and biotin in the water of Gyokpo coast, were determined by microbiological assay methods. Also the relations between the distribution of B group vitamin and other environmental factors were studied. Vitamin $B_{12}$ was assayed with Euglena gracilis strain Z, thiamine with Cryptococous albidus and biotin with Achromo bacter sp. yH-51. It was found that the concentration of B group vitamin in the water of Gyokpo coast were normal level : vitamin $B_{12}$; 1.36-3.95 ng/l, thiamine ; u-0.4 ng/l and biotin; 1.40-14.60 ng/l. The concentration of B group vitamin was high in summer than in winter. In the water of Gyokpo coast during summer, B group vitamin occurred slightly lower level than normal, the concentration suficiently neccessary for phytoplankton development. The concentration of biotin was positively correlated with abundance of phytoplankton, but not aerobic heterotrophic bacteria. It was suggested that the concentration of biotin in water might be much more influenced with the growth of phytoplankton and any environmental factors than bacteria and the other vitamin, especially.

  • Concentration of dissolved $vitamin\;B_{12}$ in the water of Okjeong lake have been determined by microbiological assay methods on the 20th of August, 1983. Also the relations between the $vitamin\;B_{12}$ and other physical, chemical and biological environmental factors were investigated. $Vitamin\;B_{12}$ was assayed with Euglena gracilis Z strain. The results obtained in this investigation were as follows : 1. Generally, the values of COD and $Cl^-$ were higher than values reported in 1981. 2. The concentration of nutrient salts in the lake water was found to be at a higher level: above the general criterion in a eutrophic lake viz., phosphorous 0.175-0.225 mg/l. 3. Chlorophyll a contents in this lake were $1.1-8.7{\mu}g/l$, 4. The concentration of $vitamin\;B_{12}$ in the water fairly high: 1.25-6.0 ng. It is apparent that concentrations of the $vitamin\;B_{12}$ as well as nutrient salts were high in the water of Okjeong lake. It was found that Okjeong lake was in eutrophic.
