전기전도성 고분자 위에 고정된 압타머에 흡착된 테트라브롬페놀프탈레인 에틸 에스테르를 이용한 트롬빈 검출 (Thrombin Detection with Tetrabromophenolphthalein Ethyl Ester Adsorbed on Aptamer-attached Conductive Polymer)

  • 새로운 산화환원 표지자를 이용한 압타머 기반의 전기화학적 트롬빈 검출 바이오 센서를 개발하였다. 1차 지방족 아민(primary aliphatic amine) 으로 개질한 압타머를 전기 전도성 고분자 poly-(5,2':5',2"-terthiophene-3'-carboxylic acid) (polyTTCA) 층 위에 공유결합을 통해 고정하여 센서 표면을 개질하였다. Tetrabromophenolphthalein ethyl ester (KTBPE)를 압타머와 상호 작용시켜 전기화학적인 산화환원 표지자로 사용하였다. 압타머로 개질한 층 위에 KTBPE의 산화반응을 differential pulse voltammetry (DPV)를 사용하여 조사하였으며, 최종 센서의 특성은 voltammetry, QCM, and ESCA 를 사용하여 조사하였다. KTBEF와 압타머 센서와 반응 후, KTBPE의 산화 피크는 감소하였다. 센서의 선형 동적 범위는 10.0 ~ 100.0 nM 이었으며, 이 때 검출 한계는 $1.0{\pm}0.2nM$이었다.

한국산 및 Canada산 High Bush Blueberry 열매의 항산화 활성 비교 (Comparative Anti-oxidant Activity of Korean and Canadian High Bush Blueberry Fructus)

  • Blueberries (Ericaceae) are cultivated worldwide, and are used not only as foodstuff but also for relievement of eyestrain. Bluberry species representatively includes highbush blueberry (V. corymbosum L.), lowbush blueberry (V. angustifolium $A_{IT}$.), rabbiteye blueberry (V. ashei $R_{EADE}$), and bilberry blueberry (V. myrtillus L.). All of these species contain large amounts of phenolics and anthocyanins. In this regard, we isolated six compounds from Korea cultivated blueberry and identified as 3-O-caffeoylquinic acid (1), 5-O-caffeoylquinic acid (2), myricetin-3-O-${\beta}$-D-galactoside (3), quercetin-3-O-rutinoside (4), ethyl-3-O-caffeoylquinic acid ester (5), ethyl-5-O-caffeoyl quinic acid ester (6) by $^1H$-NMR, $^{13}C$-NMR and MS. Anti-oxidative activities of six compounds were verified by anti-oxidant assay such as DPPH, ABTS and Hypoxanthine/xanthine oxidase system. And then, anti-oxidant activities of Korea blueberry and Canadian were compared with each other. These results support that Korean blueberry has also the possibility to be potential supplementary material as healthy food like Canadian blueberry. Therefore, Korean blueberry can be used as a substitute of Canadian blueberry.

  • 토양으로부터 특이적인 광학 이성질체에 활성을 갖는 두 효소원을 선별한 후, 동정하여 Pseudomonas sp. S34와 B. stearothermophilus JY144로 명명하였고, genecloning과 sequencing을 통해 유전자의 특성 및 관련효소들과의 유연관계를 규명하였다. 규명된 정보의 분석과 비교를 통해 ketoprofen ethyl ester에 활성을 갖는 효소들의 구조적 특성을 추론할 수 있었고, 이를 바탕으로 발현시스템을 구축하였다. 대량생산된 효소를 활용한 반응의 결과 높은 수율과 순도를 갖는 각각의 광학이성질체를 경제적으로 생산할 수 있었다.

  • Amphoteric surfactants were synthesized by the cyclization of 1-(2-hydroxyethyl)-2-undecyl-2-imidazoline [I] with acrylic acid ethyl ester. Compound [I] was easily hydrolyzed with water, especially in the presence of a alkali, to afford amidoamines. After [I] was hydrolyzed, the reaction mixture was allowed to react with acrylic acid ethyl ester and then soapoinfied. Only sodium salts of N- -(2-carboxyethyl)-N'-(2-hydroxyethyl)aminoethyl]dodecanoyl amide[III] was obtained. However, when the reacton of [I] with acrylic acid ethyl ester was carried out in the presence of water, followed by soapnification, ring cleavage of [I] occurred at 2, 3 position, different from hydrolysis of [I] where the cleavage occurred at 1, 2 position, to give sodium salts of N-[N'-(2-carboxyethyl)aminoethyl]-N-(2-hydroxyethyl)dodecanoyl amide [IV] and N-[N', N'-bis(2-carboxyethyl)aminoethyl]-N(2-hydroxyethyl)dodecanoyl amide [V] as main products.

Simultaneous Quantitative Determination of Monosaccharides Including Fructose in Hydrolysates of Yogurt and Orange Juice Products by Derivatization of Monosaccharides with p-Aminobenzoic Acid Ethyl Ester Followed by HPLC

  • We have determined the contents of monosaccharides in the hydrolysates of some yogurt and orange juice products by derivatizing monosaccharides with p-aminobenzoic acid ethyl ester (ABEE). The separation of the ABEE-derivatized monosaccharides was efficiently carried out by HPLC using a microcolumn packed with the Alltima $C_{18}$ stationary phase. The concentrations of monosaccharides were determined based on the measured peak area/height counts. ABEE derivatization of fructose and its detection have never been successfully carried out before this work. In this study, two peaks were observed in a fixed ratio for ABEE-fructose, and the ratio was maintained over a wide range of fructose concentration. In order to prove the validity of the above method, we compared the concentrations of glucose, galactose and fructose determined by ABEE derivatization and UVD (ultraviolet detector) chromatography with those determined by RID (refractive index detector) chromatography without derivatization. The determined concentrations of monosaccharides obtained from the two chromatographic methods were found close to each other within acceptable error ranges.

  • The synthesis is described of new pepsin substrates of benzyloxycarbonyl-glycyl-L-tyrosyl-L-phenylalanyl-glycine ethyl ester and benzyloxycarbonyl-glycyl-L-tyrosyl-L-phenylalanyl-glycine for studies on the specificity of pepsin, and thin layer chromatographic examination of the peptides prepared showed the new substrates are homogeneous and also, same examination of the incubation mixtures showed that two synthetic substrates are cleaved by pepsin at the L-tyrosyl-L-phenylalanyl bond and hydrolysis of these substrates by pepsin is achieved without transpeptidation. It is found that synthetic peptides are moderately soluble with the amount of the substrate up to a concentration of 0.7 mM in aqueous sodium citrate buffers (0.04 M) in the pH range 1.8-4.0, thus obviating the necessity for the adding of an organic solvent in the assay mixture. The kinetic parameters for synthetic substrates are tabulated in the following table. The data in the table indicate that the susceptibility of synthetic peptides to peptic hydrolysis are relatively large and the change of the carboxyl-terminal group of synthetic substrate from glycine ethyl ester to glycine causes a small decrease in the susceptibility of the L-tyrosyl-L-phenylalanyl bond.

  • Recently pathotoxicological study of lymphoid organs by administration of some phthalate ester in rats, indicated marked effect of architechure of thymus, spleen, and lymphonodes. Dioctyl phthalate (DOP), one of the phthalate ester, caused statistically significant reduction in the weights of various lymphoid organs. This senstivity of the lymphoid organ to phthalate toxicity which could lead to adverse effects on the immune response and also suppression of immune system. Therfore it is possible the presense of di-(2-ethylhexyl) phthalate (DEHP), one of the phthalate ester as well as DOP, in spleen and other organs might have some moderately effect on the function of the immune system, So our present study was proceeded to assess the effect of DEHP on the immunotoxicity in mouse. In the immune response of DEHP administered mice, HA, HY, Arthus reaction and Rosette forming cell were decreased but DTH was increased. Furthermore, in the DEHP plus ethanol group, HA, HY, Arthus reaction and Rosette forming cell were remarkably decreased and elevation of DTH was inhibited.

  • Methylene protons of benzyl and ethyl ester in ethyl 1-benzyl-2, 3-dioxo-cyclopenta[b]-pyrrole-3a-carboxylate (A) exhibited opposite$^{1}$H-NMR spectral patterns mutually between magnetic equivalence and nonequivalence depending on concentration and temperature. The diastereotopic spectral data of compound A were reported with brief interpretation.

Aspergillus oryzae 누룩으로 담금한 탁주 술덧의 발효 과정 중 휘발성 향기성분 (Volatile Flavor Components in Mash of Takju prepared by using Aspergillus oryzae Nuruks.)

  • Aspergillus oryzae균을 파종하여 만든 누룩으로 담금한 탁주 술덧의 발효과정 중 휘발성 향기성분을 GC와 GC-MS를 사용하여 분석 동정한 결과 ester 24종, alcohol 21종, acid 10종, aldehyde 9종, 기타 4종 등 68종의 향기 성분이 동정되었다. 동정된 향기성분 수는 담금일에 ester 13종, alcohol 12종을 비롯한 총 36종이었으나, 발효 2일에 ester 10종, alcohol 8종을 포함한 29종이 추가 검출되어 65종으로 증가되었다. 발효 12일에는 68종으로 향기 성분수가 최대에 달하였다. ethanol, 3-methyl-1-butanol, 2-methyl-1-propanol, benzeneethanol등 alcohol류 12종, ethyl acetate, ethyl caprylate, ethyl butyrate 등 ester류 13종, acetaldehyde, isobutyl aldehyde등 aldehyde 4종, pentanoic acid , acetic acid등 acid 6종 등 총 35종은 발효 전과정을 통하여 검출되었다. 향기 성분의 면적비율은 ethanol이 $79.86{\sim}89.54%$로 발효 전기간을 통하여 탁주 술덧에서 타 성분 보다 월등히 높았다. ethanol을 제외하고 발효 전과정을 통하여 면적비율이 높은 성분은 ethyl caprylate, 3-methyl-1-butanol, benzeneethanol이었다. 이외 발효기간에 따라서 2-methyl-1-propanol, 1-hexanol, 2,3-butanediol(D.L), 1-dodecanol, 2-phenylethyl acetate, ethyl acetate, monoethyl butanoate, diethyl succinate, ethylbenzoic acid, acetic acid, isobutylaldehyde등의 성분도 동종 계열간의 향기 성분중 면적비율이 다소 높게 나타나기도 하였다.

  • Asp. kawachii균을 파종하여 만든 누룩으로 담금 한 탁주 술덧의 발효과정 중 휘발성 향기성분을 GC와 GC/MS를 사용하여 분석 동정한 결과 ester 22종, alcohol 20종. acid 10종, aldehyde 8종, 기타 3종 등 63종의 향기 성분이 동정되었다. 동정된 향기성분 수는 담금 일에 ester 13종, alcohol 13종을 비롯한 총 32종이었으나, 발효 2일에 ester 6종, alcohol 3종을 포함한 13종이 추가 검출되어 45종으로 증가되었다. 발효 12일에는 63종으로 향기 성분수가 최대에 달하였다. Ethanol, 3-methyl-1-butanol, 2-methyl-1-propanol, benzeneethanol등 alcohol류 12종, ethyl acetate, ethyl caprylate, ethyl butyrate 등 ester류 12종, acetaldehyde, iso-butyl aldehyde등 aldehyde 3종, octanoic acid, 1,2-benzenedicarboxylic acid 등 acid 2종, 총 29종은 발효 전 과정을 통하여 검출되었다. 향기 성분의 면적비율은 ethanol이 72.18-91.32%로 발효 전 기간을 통하여 탁주 술덧에서 타 성분보다 월등히 높았다. Ethanol을 제외하고 발효 전 과정을 통하여 면적비율이 높은 성분은 ethyl caprylate, 3-methyl-1-butanol, benzeneethanol 이었다. 이외 발효기간에 따라서 2-methyl-1-propanol, 1-hexanol, 2, 3-butanediol(D.L), 1-dodecanol, 2-phenylethyl acetate, ethyl acetate, monoethyl butanoate, diethyl succinate, ethylbenzoic acid, acetic acid, iso-butylaldehyde 등의 성분도 동종 계열간의 향기성분 중 면적비율이 다소 높게 나타나기도 하였다.
