A Biochemical Study on the Ethanol Metabolism in the Animal Body (동물 체내에서의 에탄올 대사에 관한 생화학적 연구)

  • Ethanol은 섭취량에 따라 간 대사에 여러가지 영향을 미치는 것으로 알려져 있다. 과량의 ethanol 섭취가 유해한 것은 ethanol 그 자체보다는 산화과정에서 생성된 acetaldehyde와 과량의 수소(NADH)에 기인한다. 과량의 NADH는 간 세포의 화학적 평형을 저해하고 대사이상을 초래한다. 본 연구에서는 in vitro 뿐만 아니라 in vivo에서 alcohol dehydrogenase(ADH), aldehyde dehydroge-nase(ALDH), microsomal ethanol oxidizing system(MEOS)에 미치는 인삼 사포닌의 영향을 조사하고, 간에서의 수소 평형, 간에서의 $[1^(-14)C]$-ethanol의 분포, ethanol의 acetaldehyde와 lipid로의 전환 등을 관찰하였다. 인삼 사포닌은 상기 효소외에도 ethanol 대상에 관련된 다른 효소들의 활성을 증가시키는 것으로 관찰되었으며 이는 동물 체내로부터 acetaldehyde와 과량의 수소를 신속히 제거하는 것으로 사료된다.

  • Ethanol can affect a wide range of organ and organelle systems. Some of its effects are directly due to the action of either ethanol or its metabolites, whereas others are related to nutritional deficencies associated with ethanol intake. Some of the liver damages occurring in alcohol abusers are mainly due to generation of free radical during the metabolism of ethanol and subsequent lipid peroxidation. Acetthrough the formation of free radicals, or depletion of levels of antioxidant substances. When prevent generation of the activity. Several authors have reproted the ethanol-related variation of antioxidants such as $\alpha$-tocopherol, ascorbic acid, selenium and glutathionem which are important thereby promoting deficiencies as well as toxic interactions with vitamin A and $\beta$-carotene. It has been appeared that ethanol-induced hepatotoxicity may be partially prevented by the administration of antioxidants.

  • Allyl alcohol is metabolized in the liver through two steps, first to reactive acrolein by alcohol dehydrogenase (ADH), subsequently to acrylic acid by aldehyde dehydrogenase (ALDH). Since ethanol could compete the same enzymes to be metabolized in the liver, we have determined the plasma concentrations of allyl alcohol and ethanol followed by combined treatment. Pretreatment of rats with 2g/kg ethanol followed by ip administration of 40mg/kg allyl alcohol increased the lethality significantly. Determination of in vivo blood concentrations revealed that ethanol pretreatment caused the apparent decrease in allyl alcohol clearance, whereas acetaldehyde level in blood increased significantly by allyl alcohol treatment, as determined by head space GC analysis. Treatment of 4-methylpyrazole, an inhibitor of ADH, delayed allyl alcohol elimination significantly and reduced its lethality. Collectively, these findings suggested that reduction of allyl alcohol clearance in the presence oj ethanol was mediated through ADH competitive inhibition.

  • The optimum conditions for ethanol fermentation and ethanol productivity of the fusant ESC-14-15 were examined in a mini-jar formentor scale (working volume : 2.5 liters) to assess the possibility of practical application. Addition of yeast extract to fermentation broth greatly enhanced the ethanol productivity and shortened the period of fermentation. The pH 4.2 was more favorable than pH 5.5 with respect to ethanol productivity and fermentation speed. The optimum concentration of liquefied potato starch for ethanol fermentation of FSC-14-15 was 15%(w/v) and the corresponding productivity was 8.7%(v/v) of ethanol with an efficiency of 80.6% to the theoretical maximum. When the fresh fermentation broth containing 20% of liquefied potato starch was inoculated with love(v/v) of inoculum, the fusant FSC-14-75 produced 11.0%(v/v) of ethanol in 4 days, which is considered comparable to that from an industrial process. From the liquefied cassava starch or the equal mixture of liquefied barley and sweet potato starch prepared according to the same method as in the industrial process except saccharification step, the fusnnt FSC-14-75 produced 8.5%(v/v) or 7.6%(v/v) of ethanol in 4 days, respectively.

A Study on the Spray and Combustion Characteristics of Diesel-ethanol-biodiesel Blended Fuels in a Diesel Engine (디젤엔진에서 디젤-에탄올-바이오디젤 혼합연료의 분무 및 연소 특성에 관한 연구)

  • The aim of this study is to analyze the effect of the ethanol blending in diesel-ethanol blended fuels on the spray and combustion characteristics in a common-rail four-cylinder diesel engine. For the analysis of the spray characteristics, the spray images were obtained using a high speed camera with metal-halide lamps. From these spray images, the macroscopic spray characteristics such as the spray tip penetration and spray cone angle were investigated. Also, the combustion characteristics including the combustion pressure and the rate of heat release were studied with the analysis of the exhaust emissions in diesel-ethanol blended fuel driven diesel engine. It can be confirmed from the experiment on spray characteristics of diesel-ethanol blended fuels that the increased ethanol blending ratio induced the decrease of the spray tip penetration after the end of the injection. The spray cone angle slightly increased by the blending of ethanol fuel. In the experiment on atomization characteristics, the ethanol blending caused the improvement of the diesel atomization performance. On the other hand, at the same engine load condition, the increase of the ethanol blending ratio lead to lengthen the ignition delays, and to decrease the peak combustion pressure and the rate of heat release. Totally, the combustion and emission characteristics of ULSD and DE10 showed similar characteristics. However, in the case of DE20, CO and HC rapidly increased, and $NO_x$ decreased. It can be believed that 20% ethanol disturbed the combustion of diesel-ethanol blended fuel due to the low cetane number and evaporation.

Effect of Ethanol Intake on the Concentration of Minerals in Broiler Chicks (알콜섭취가 성장기닭의 장기중 무기질농도에 미치는 영향)

  • The effect of chronic ethanol consumption on the concentration of minerals in tissues and serum was studied in growing broiler chicks. Four different groups of the chicks were fed mixtures of 0(control), 1,2 and 3% ethanol and water respectively for 7 weeks. Body weight gain in 1% ethanol group and liver weight in 3% ethanol group were significantly higher than those of control. Mg, K, Mn, and Zn concentrations in liver were higher in ethanol groups than those in control. In ethanol groups, femoral muscle Mg level was increased while its Na concentration was decreased. The concentrations of Ca, Mg and Na in serum were higher in 3% ethanol group than those in control, 1 or 2% ethanol groups. In femur, Zn and Fe levels in 1% ethanol group and Mn concentration in 2 or 3% ethanol groups were increased. But its weight, length, and ash content were not affected by ethanol intake.

Effects of Alcohol in Intoxicated Patients (알코올이 약물 중독 환자에게 미치는 영향)

  • Purpose: This study was to investigate the effects of ethanol in ingested patients by analyzing data from a single institution's registry, Methods: We conducted a prospective study of 50 patients who has ingested drugs with/without ethanol came to emergency department from January 2004 to May 2004. Only patients over 18 years of age were included. Clinical characteristics, general and specific treatment, laboratory finding, complication, and clinical outcomes were obtained from protocol. Patients were divided into two groups: drug ingested with alcohol (ethanol group, n=18), and ingested without alcohol (non-ethanol group, n=32). Results: The age, the amout of ingestion, the time to treatment, the systolic blood pressure, the diastolic blood pressure and the shock duration were not different between two groups. The AST level with the ethanol group was higher than with the non-ethanol group ($230.94\pm518.88$ U/L vs $43.22\pm63.39$ U/L, p=0.002). The ALT level with the ethanol group was higher than with the non-ethanol group ($97.06\pm152.98$ U/L vs $32.75\pm43.10$ U/L, p=0.001). The lactic acid level with the ethanol group was higher than with the non-ethanol group ($7.40\pm6.33$ mmol/L vs $3.77\pm3.10$ mmol/L, p=0.001). The hospital stay duration and the admission rate were not different between two groups. Conlusions: The ethanol increased the levels of serum AST, ALT and lactic acid in intoxicated patients. But the ethanol dose not increase admission rate and duration of admission stay in intoxicated patients.

A Study on the Spray-atomization Characteristics of Diesel-ethanol Blended Fuels in a High Pressure Diesel Injection System (디젤 고압 분사 시스템에서 디젤-에탄올 혼합연료의 분무 및 미립화 특성에 관한 연구)

  • The purpose of this paper is to analyze the effects of ethanol blending ratio and fuel temperature in diesel-ethanol blended fuel on the spray-atomization characteristics in a high pressure common-rail injection system. In this work, a diesel fuel and three blended fuels were used as test fuels. Blended fuels were made by blending ethanol with a purity 99.9% to diesel fuel, from 0% to 30%. In order to keep diesel-ethanol blending stability, 5% of biodiesel fuel as volumetric ratio was added into test fuels. The fuel temperature was controled in steps with 40K, from 290K to 370K. Macroscopic spray characteristics were investigated by analyzing the spray tip penetration and spray cone angle through spray images obtained from visualization system. In addition, in order to study microscopic spray characteristics of ethanol blended fuels, the droplet diameter, was analyzed using the droplet measuring system. It is revealed that the spray tip penetration is similar regardless of ethanol blending ratio. As ethanol blending ratio is increased, the spray cone angle becomes wider. It is shown that the spray cone angle is affected by low viscosity and density of ethanol. As the fuel temperature increases, the spray tip penetration and spray cone angle become shorter and narrower respectively. The SMD of ethanol blending fuels is smaller than that of diesel fuel because of low viscosity and surface tension of ethanol.

Effects of Dietary Methionine and Folate Supplementation in Ethanol-Fed Rats

  • Chronic alcohol consumption is associated with perturbation of hepatic metabolism of sulphur-containing amino acid. The goal of present study was to evaluate the influence of dietary supplementation of methionine or folate to chronically ethanol-fed mts on the metabolism of sulfur-containing amino acids and one-carbon metabolism. Sprague-Dawley male mts were fed Lieber-Decarli liquid diet with 0% ethanol (control), 36% ethanol (E), 36% ethanol combined with methionine supplement (EM) or folate supplement (EF) for 8 weeks. Hepatic S-adenosylmethionine (SAM) and S-adenosylhomocysteine (SAH), plasma folate and homocysteine (Hcy), urinary excretion of folate and formiminoglutamate were investigated after feeding experimental diets. Growth was retarded by 36% ethanol consupmtion (E, EM and EF) (p<0.01). Liver total fat (p<0.05) and plasma ALT (P<0.01) were increased by methionine supplementation (EM), implicating fatty liver and liver injury. Liver folate was increased slightly by folate supplementation (EF) (p=0.077). Urinary folate loss was increased 2.3 fold by ethanol consumption (E) and 17.2 fold by folate supplementation (EF), while decreased by methionine supplementation (EM) (p<0.000l). Plasma Hcy was increased 1.9 fold by methionine supplementation (EM) in ethanol-fed mts (p<0.05), which was related with decreased methionine synthase activity (p<0.05). Hepatic SAM/SAH ratio was depressed by methionine supplementation in ethanol-fed mts (EM) (p<0.05). Urinary formininoglutamate (Figlu) excretion after histidine loading was increased by ethanol ingestion and reduced by methionine supplementation (p<0.00l). Based on these data, methionine supplementation appears to accelerate histidine oxidation. In conclusion, dietary supplementation of methionine to ethanol-fed mts exacerbates alcoholic liver injury possibly by complicating sulphur-containing amino acid metabolism, as while it may have beneficial effects on folate and histidine metabolism.

Effects of Scolopendra on Ethanol-Induced Erectile Dysfunction in Rats (Ethanol로 발기부전을 유도한 흰쥐의 성기능 개선에 오공(蜈蚣)추출물이 미치는 영향)

  • Scolopendra was known to cure erectile dysfunction. This study was aimed to investigate the effects of Scolopendra on the nitric oxide synthase activity, nitrite level, cyclic-GMP and erectile responses in rat's corpus cavernosum penis. The crushed Scolopendra was extracted 3 times, each time with 3 volumes of methyl alcohol at $60^{\circ}C$ for 24 h. The extract was filtered and evaporated under a reduced pressure using a rotary evaporator to yield 14.2 g. Scolopendra extract was oral-administered 50 mg per 1 kg of body weight for 30 days. First, samples were treated with Scolopendra, and then ethanol-treated rats and L-N-Nitroarginine methyl ester (L-NAME) treated rats were fed with the samples. The level of urethral nitrite and nitric oxide synthase activity in the ethanol-Scolopendra double administered rats were as high as in the normal group, while the one in the ethanol-treated group was decreased. The level of urethral cyclic-GMP and guanylate cyclase actiyity in the ethanol-Scolopendra double administered rats were as high as in the normal group, while the one in the ethanol-treated group was decreased. The level of urethral phosphodiesterase activity in the ethanol-Scolopendra double administered rats was as low as in the normal group, while the one in the ethanol-treated group was increased. The erectile response to a cavernous nerve stimulation in L-NAME $(10^{-4})-treated$ rats was restored by the Scolopendra to the similar level seen in the normal group. The electile response to cavernous nerve stimulation in the ethanol-Scolopendra double administered rats were increased as high as in the normal group while the one in the ethanol-treated group was decreased. Scolopendra was effective in restoring the ethanol-induced or L-NAME-induced erectile dysfunction in rats.
