고지방식이 섭취 마우스의 간 조직의 지질 축적에 대한 밀순 추출물의 억제 효과 (Inhibitory Effects of Triticum aestivum L. Extracts on Liver Lipid Accumulation in High Fat-Fed Mice)

  • Fatty liver disease refers to a range of disorders associated with fatty liver, which occur in excessive eating, evident infection or significant consumption of alcohol. This study was to investigate the effects of water and ethanol extracts of Triticum aestivum young leaf on lipid metabolism and accumulation in liver of mice fed with high-fat diet. Male C57BL/6 mice were divided into normal diet group, high fat diet (HFD) group, high fat diet group administrated with 200 mg/kg/day of T. aestivum water extract (HFD-TAWE) and high-fat group administrated with 200 mg/kg/day of T. aestivum ethanol extract (HFD-TAEE). TAWE and TAEE were administrated orally for 5 weeks once at the same time point. Both TAWE and TAEE significantly reduced body weight, food intake and liver tissue weight, which were augmented in high fat-fed mice. The serum levels of triglyceride, total and LDL-cholesterol also were significantly attenuated in both HFD-TAWE and HFD-TAEE groups compared to the HFD group. Moreover, administration of HFD-TAWE or HFD-TAEE reduced the lipid accumulation in liver tissue of mice fed with high fat diet. Levels of total lipids and triglyceride in liver tissues also was significantly reduced in HFDTAWE and HFD-TAEE groups compared to HFD group. The activities of serum ALT and AST revealed in HFD group were remarkedly decreased in HFD-TAEE groups. These results indicate that both water and ethanol extract of T. aestivum may improve the lipid accumulation in liver as well as lipid metabolism in serum, and that in particular, the ethanol extract of T. aestivum may has the potent anti-hyperlipidemic effect, suggesting that it may be a useful candidate for the therapy preventing fatty liver diseases.

  • A chronical intake of Substantial amount of alcohol would disrupt anormal function of liver if not develop liver diseases in relatively short period. In order to out whether ginseng or ginseng plus high protein diet have any protective effects on the liver of chronical alcoholist from developing malfunction enzymatic activities of both glutamic-pyruvic and glutamic-oxaloacetic transaminases were measured on serum of rats maintained with basal low protein diet, basal diet plus 1 percent ginseng and high protein (40%) plus 1 percent ginseng and administered intraperitoneally with a Constant amount of ethanol either periodically or chronically. It was found that, unlike human subject GOT content was exceedingly high and significant difference was found either among treatment or among sexes thus indicating that either ginseng intake or high protein diet plus ginseng has a protective effect on the liver function of ethanol treated rats. From these results, it was suggested that the dietary ginseng might, have a protective effect on the alcohol hepatic disturbance. As one of probable mechanisms for the characteristic pharmacological activity, it was considered that a secondary action of the saponin of the dietary ginseng would result in the anti-inflammatory through the stimulation of de nove synthesis of certain functional proteins in hepatic organs.

Wild Ginseng Prevents the Onset of High-Fat Diet Induced Hyperglycemia and Obesity in ICR Mice

  • Ginseng is a shade-loving perennial herb that is cultivated mainly in Korea, Japan, and China. The ginseng root has been used as a tonic remedy, and its antidiabetic activity has been demonstrated as early as 1920s. Although wild ginseng was anecdotally thought to be superior to cultivated ginseng as far as pharmacological properties were concerned, there have been no prior reports on the antidiabetic effect of wild ginseng. In this study, we investigated the preventative anti-diabetic and anti-obese effects of wild ginseng ethanol extract (WGEE). In the preventive experiment, WGEE co-administered with a high fat diet significantly inhibited body weight gain, fasting blood glucose, triglyceride, and free fatty acid levels in a dose dependent manner. WGEE-treated mice at doses of 250 and 500 mg/kg improved the insulin resistance index by 55% and 61% compared to the high fat diet (HFD) control, respectively. Diameters of white and brown adipocytes were also decreased by 62% and 46% in the WG500-treated group compared to those in HFD fed control mice. Taken together, WGEE has potential as a preventive agent for type 2 diabetes mellitus (and possibly obesity) and deserves clinical trial in the near future.

흰쥐를 대상으로 한 양파주의 알코올성 지방간 개선 효과 (Protective Effect of Onion Wine on Alcoholic Fatty Liver in Rats)

  • 본 연구에서는 흰쥐를 이용하여 양파주의 섭취가 알코올성 지방간 증상 완화에 미치는 영향을 조사하였다. Lieber-DeCarli 액체 표준식이만 공급받는 동물군을 대조군, 대조군 표준식이의 탄수화물 대신에 에탄올로 열량을 대체한 액체식이를 공급받는 동물군을 에탄올군, 에탄올로 열량을 대체한 식이에 양파주를 추가로 공급받는 동물군을 양파주군으로 설정하여 6주 동안 공급하였다. Lieber-DeCarli 식이공급 전, 3주째 그리고 6주째에 혈액을 채취하였고, 간은 6주째 혈액 채취 직후 적출하였다. 양파주 농축액의 섭취로 인해 혈액의 중성지방과 총콜레스테롤 농도는 에탄올군에 비해 현저히 감소하였다. 혈액의 ALT, AST, ALP 농도 모두 에탄올군에 비해 양파주군에서 감소한 것으로 나타났다. 또한 간 조직의 총지질과 콜레스테롤의 농도 모두 양파주의 공급으로 현저히 감소하였으며, 주요 지방산 비교에서도 대부분의 지방산이 에탄올군에 비해 양파주군에서 농도가 감소한 것으로 나타나 양파주의 공급이 간 조직의 지방산 축적도 억제한 것으로 나타났다. 본 실험을 통해 양파주가 알코올 섭취로 증가한 혈중 지질 수준과 간 기능 지표 수준을 개선하는 효과를 나타냈으며, 지방간 형성이 유의적으로 억제되는 것을 간 조직 지방분석을 통해 확인하였다. 이렇게 개선된 지표들을 바탕으로 양파주의 섭취는 간 조직 보호 효과와 알코올성 지방간의 개선 효과를 유도할 수 있을 것으로 판단된다.

Effects of Carnitine on the Lipid Metabolism in the Ethanol-Fed Rats

  • The effect of dietary carnitine on ethanol-induced fattv liver and hvpertriglyceridemia was examined in an animal model. Consistent with literature, ethanol fed at 5g/Kg of b.w. to rats produced a significant increase in hepatic concentrations of total lipid, kislycerine, phospholi-pid, free cholesterol and esteriaed cholesterol as well as elevated plasma concentrations of triglvceride. It was when the ethanol diet was supplemented with D.L. camitine that there was a singini-cant reduction in the accumulation of lipids in the ethanol-compromised liver. Dietary cacti-tine was also effective in ameliorating ethanol-induced hypeuriglyceridemia. Total protein con-tents in the plasma was not varied among the groups. Ethanol의 대사과정에 관여하는 영향중에 특징적인 것으로 과유지방혈증(hyperlipidemia) 까 지방간(fatty liver)을 거쳐 간경변에 이르는 간에 관계하는 일련의 증상들을 들 수 있다. 본 실험에서는 만성적 ethanol의 지방대사 장해에 대한 D.L.-carnitine의 효과에 대해 고찰하였다. 실험용 횐쥐를 사용하여 실험군(ethanol group)에게 체중 kg당 5g의 ethanol(30% in saline)을 투여하여 알콜유발성 지방간과 과유지방혈증을 일으키고, 그 실험군 흰쥐들에게 carnitine(0.4 mg/g of body weight)을 첨가하여 그 효과를 관찰하였다. 그 결과 carnitine을 첨가하여 투여한 흰쥐들에서 ethanol처리군과 비교하여 볼 때 간과 혈장에서 지방축적이 현저히 감소하는 것을 관찰할 수 있었다.

Anti-diabetic Effects of Ethanol Extract from Bitter Melon in Mice Fed a High-fat Diet

  • Present study aimed to determine the effect of 'bitter melon', a popularly used fruit in Bangladesh and several other Asian countries, on high-fat-diet-induced type 2 diabetes. To investigate the effect, ethanol extract from bitter melon (BME) as a dietary supplement with mouse chow was used. BME was found to significantly attenuate the high-fat diet (HFD) -induced body weight and total fat mass. BME also effectively reduced the insulin resistance induced by the HFD. Furthermore, dietary supplementation of BME was highly effective in increasing insulin sensitivity and reducing hepatic fat and obesity. These results indicate that BME could be effective in attenuating type 2 diabetes and could therefore be a preventive measure against type 2 diabetes.

비알콜성 지방간 동물모델에서 갈화추출물의 간보호 효과 (Hepatoprotecive Effects of Puerariae Flos Extract on Experimental Non-alcoholic Fatty Liver Disease Models)

  • Objectives : The aim of this study is to investigate the preventive effect of Puerariae Flos ethanol extract (PE) on methionin and choline deficient (MCD)-diet-induced Nonalcoholic fatty liver disease (NAFLD) in C57BL/6J mice. Methods : In the in vivo experiments, C57BL/6J mice were divided into 4 groups; Normal group, Control group, MCD+PE 100 group, and MCD+PE 300 group. After 4 weeks, body weight, liver weight, biochemical parameters for liver function test, histological changes, reverse transcription polymerase chain reaction (RT-PCR), and Western blot were assessed. Results : Mice lost body weight with the MCD-diet and the MCD+PE 100 group and MCD+PE 300 groups lost less than the control group, though showed no statistical significance. Liver weights were decreased by the MCD diet, but MCD+PE 300 groups were increased significantly. In the liver function test, all the values were decreased with the MCD-diet, MCD+PE 100 group and MCD+PE 300 groups were increased significance. In histological findings of the livers, MCD-diet induced severe fatty accumulation in the livers, but this fatty change was reduced in the MCD+PE 100 group and MCD+PE 300 groups was inhibited respectively. In lipid accumulation factors (such as SREBP-1c, $C/EBP{\alpha}$, PPAR-${\gamma}$), MCD+PE 100 group and MCD+PE 300 groups showed inhibitory effect on liver lipogenesis by reducing associated gene expressions caused by MCD diet. Conclusions : We were able to know that Puerariae Flos ethanol extract (PE) shown hepatoprotective effects via a decrease on the hepatic lipogenesis factors in the experimental NAFLD Models.

추출 조건을 달리한 감잎과 녹차의 물 및 에탄올 추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향 (Effect of Water and Ethanol Extracts of Persimmon Leaf and Green Tea Different Conditions on Lipid Metabolism and Antioxidative Capacity in 12-month-old Rats)

  • This study was performed to investigate effects of dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in 12-month-old rats. Fifty-four male Sprague-Dawley rats weighing 542$\pm$4.5g were blocked into groups according to their body weight and were raised for four weeks with the diets containing 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) and green tea(Camellia Sinensis O. Ktze), water or 75% and 95% ethanol extracts from same amount of each dried tea powder. Food intake was not significantly different among all groups, but weight gain of green tea powder group was significantly lower than that of control group. Plasma and liver lipid levels of all the tea diet groups were lower than those of control group. Especially, 75% ethanol extract of persimmon leaf decreased total lipid and triglyceride concentrations in plasma and 95% ethanol extract of persimmon leaf decreased liver total lipid level. However, there was no difference between 75% ethanol extracts groups and 95% ethanol extracts groups in lipid metabolism. Superoxide dismutase(SOD) and catalase activities in erythrocyte were remarkably increased by all the green tea diets. SOD, catalase and glutathione peroxidase activities in liver were increased by the feeding of ethanol extracts from green tea and persimmon leaf powder. Liver xanthine oxidase activity was not different among all groups. Plasma Thiobarbirutic acid reactive substance(TBARS) concentrations of all the green tea diet groups were significantly low. It was thought that high flavonoids in green tea inhibited plasma lipid peroxidation by promoting SOD, catalase activities in erythrocyte. 95% ethanol extract of persimmon leaf also inhibited plasma lipid peroxidation by high vitamin E and beta-carotene. Persimmon leaf powder decreased liver TBARS concentration by vitamin E, betacarotene and vitamin C and by increasing activities of antioxidative enzymes with flavonoids. In conclusion, dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea were effective in lowering lipid levels and inhibiting lipid peroxidation in 12-month-old rats. Above all, ethanol extracts of persimmon leaf decreased plasma and liver lipid levels and persimmon leaf powder effectively inhibited liver lipid peroxidation. Extracts of green tea leaf inhibited plasma lipid peroxidation. In lowering lipid levels and inhibiting lipid peroxidation, ethanol extracts were more effective than water extracts, but there was no difference between 75% ethanol extracts and 95% ethanol extracts in lipid metabolism. Chronic saponin treatment attenuates damage to the pancreas in chronic alcohol-treated diabetic rats

  • Background: Chronic heavy alcohol consumption may raise the risk of developing type 2 diabetes mellitus. Saponins inhibit apoptosis of pancreatic islet cells and reduce lipid parameters. The present study was designed to investigate the effect of saponin on chronic ethanol-treated diabetic rats. Methods: Long-Evans Tokushima Fatty (LETO) and Otsuka Long-Evans Tokushima Fatty (OLETF) rats were pair-fed a Lieber-DeCarli diet with and without 5% ethanol for 12 wks. Two weeks after starting the pair-feeding with the Lieber-DeCarli diet, intraperitoneal injection of saponin was performed for 10 wks. To perform the experiments, rats were divided as follows: LETO-Control (LC), LETO-Ethanol (LE), LETO-Ethanol-Saponin (LES), OLETF-Control (OC), OLETF-Ethanol (OE), and OLETF-Ethanol-Saponin (OES). Results: The weights of epididymal and mesenteric fat tissue in LES and OES rats were the lightest from among the LETO and OLETF groups, respectively. The secretion of alanine aminotransferase and cholesterol in OES rats decreased significantly compared to their secretion in OC and OE rats, respectively. The islets of the pancreas in LE and OE rats showed clean, unclear, and smaller morphology compared to those of LC, LES, OC, and OES rats. In addition, the expression of insulin in the islets of the pancreas in LC, LES, OC, and OES rats was higher than in LE and OE rats. Conclusion: Saponin may not only be helpful in alleviating the rapid progress of diabetes due to chronic alcohol consumption in diabetic patients, but may also show potential as an antidiabetic drug candidate for diabetic patients who chronically consume alcohol.

Effect of Dietary Vitamin A on Plasma Membrane Property and Ultrastructure in Ethanol -administered Rat Liver

  • This study was conducted to investigate the effect of dietary supplementation of vitamin A on the membrane property and ultrastructure in ethanol-administered rat livers. Male Sprague-Dawley rats weighing of 130 ~150g were fed with experimental diets for 7 weeks. The diets contained different types of vitamin A which were $\beta$-carotene, retinyl acetate and retinoic acid. After feeding theexperimental diets for 7 weeks, a dose of 3.0g ethanol (30%, W/V)/kg B.W was injected to rats intraperitoneally. Control rats received 0.9% saline containing isocaloric sucrose instead of ethanol. Plasma membrane fluidity of liver decreased in rats fed with vitamin a -Deficient diet with ethanol as compared to that of control rats. Fluidity change of liver plasma membrane that ethanol had induced was influenced by dietary supplementation of vitamin A, but not influenced by the type of supplemented vitamin. A . The ultrastructural changed of hepatic mitrochondria were observed in some rats such as vitamin A-deficient rats with ethanol. Inadequate consumptionof vitamin A contributed to ultrastructural changes such as swelled mitochondria occurred by ethanol-induced hepatotoxicity. Although accurate mechanism involved in the plasma membrane-stabilizing effect of vitamin A is still unclear, dietary supplementation of vitamin A such as retinyl acetate is neede to modulate this change. The direct involvement of membrane property on the cell damage caused by ethanol treatment remains to be established.
