신선초 추출물의 아토피 억제활성 (Inhibitory Effect of Angelica keiskei Extract in an Atopic Dermatitis Animal Model)

  • 신선초 추출물의 접촉성 아토피 피부염 완화효과를 확인하고 이의 소재를 개발하기 위한 목적으로 신선초 DW 추출물, 50% 에탄올 추출물, 100% 에탄올 추출물을 DNFB로 아토피를 유도한 마우스 동물모델에 각각 처리하여 in vivo 실험과 in vitro 실험을 실시하였다. 그 결과, 신선초 추출물을 처리한 군의 귀 손상정도가 대조군에 비해 감소되었으며, 귀 조직을 HE 염색으로 전체 귀 두께와 상피 두께를 측정하였을 때에도 대조군에 비해 상당한 감소를 나타냈다. 또한 RT-PCR로 염증 분자마커인 MMP series의 mRNA 발현수준을 검토한 결과, 신선초 추출물은 MMPs에 비의존적으로 마우스 아토피 피부염의 완화작용에 작용하는 것으로 나타났다. 마지막으로 염증반응을 유도한 마우스 비장조직세포에서 염증성 cytokine인 IL-4는 신선초 녹즙에서 농도 의존적으로 감소되었으며, IL-13 또한 신선초 녹즙과 에탄올 추출물에서 상당한 억제효과가 있음을 확인하였다. 이상의 결과로 신선초 추출물의 항아토피 활성을 확인 하였으며, 관련 제품 개발에 유용한 소재가 될 가능성을 확인하였다.

포공영의 항위염 작용 (The Antigastritic Effect of Taraxaci Herba)

  • The methanol extract of Taraxaci Herba was found to have inhibitory effect on the gastric lesion induced by HCl-ethanol. The systematic fractionation of the methanol extract resulted in positive action with water fraction in the gastric lesion. It also showed significant inhibition of gastric lesion induced by HCl-aspirin and absolute ethanol, but did not prevent indomethacin induced gastric lesion. This fraction did not affect basal gastric acid secretion but showed a decrease of pepsin activity in pylorus-ligated rats.

조릿대 잎 분획 추출물의 식중독균에 대한 항균활성 (Antimicrobial Activity of Bamboo (Sasa borealis) Leaves Fraction Extracts against Food Poisoning Bacteria)

  • 본 연구에서는 조릿대 잎의 70% ethanol 조추출물과 이의 분획추출물 5종이 식품부패균과 식중독균 7종에 대해 나타내는 항균활성과 최소저해농도 및 생육억제효과를 수행하였다. 70% ethanol 조추출물과 분획추출물 모두 1% 농도에서는 항균활성을 보이지 않았으나 5% 농도에서는 aqueous분획을 제외하고 대부분의 균주에 대해 항균활성이 나타났다. 즉 70% ethanol 추출물과 chloroform과 ethyl acetate 및 n-butanol 분획은 본 연구에서 실험한 7종의 모든 균에 대해 항균활성을 보였으며, 특히 chloroform 분획은 5종의 균에 대해, ethyl acetate 분획은 3종의 균에 대해 그리고 70% ethanol 추출물은 1종에 대해서 높은 항균활성을 보였다. 반면에 n-hexane 분획은 M. luteus와 B. subtilis 2종의 균에 대해서만 강한 항균활성을 나타냈다. 이들 조릿대 잎 추출물이 항균작용을 나타낸 최소저해농도는 70% ethanol 추출물은 S. aureus를 제외한 6종의 균에 대해 $50{\mu}L$/disc이었고, n-hexane 분획은 항균활성을 나타낸 M. luteus와 B. subtilis에 대해서만 $100{\mu}L$/disc의 최소저해농도를 보였으며, chloroform 분획은 3종의 균에 대해서는 $35{\mu}L$/disc의 낮은 최소저해농도를 보였고 나머지 4종에 대해서는 $50{\mu}L$/disc이었으며, ethyl acetate 분획은 7종 모든 균에 대해 $50{\mu}L$/disc가 최소저해농도이었다. 한편 n-butanol 분획은 S. aureus에 대해서는 $35{\mu}L$/disc가 그리고 나머지 6종에서는 $50{\mu}L$/disc가 최소저해농도이었다. 항균활성이 보이지 않았던 aqueous 분획은 S. aureus와 L. monocytogens에 대해 $200{\mu}L$/disc의 최소저해농도를 나타내었다. Ethyl acetate 분획의 생육저해효과는 0.1% 처리 시에는 2종의 균만 생육을 8시간 이상 저해하였고, 0.25% 처리는 2종에 대해서는 8시간까지 그리고 5종의 균에 대해서는 12시간까지 생육을 저해하였으며, 0.5% 처리는 7종 모든 균의 생육을 24시간까지 저해하였다. 이러한 생육저해효과는 용매로 사용된 70% ethanol의 기여도 있었지만 ethyl acetate 분획추출물에 함유된 항균물질의 작용임이 확인되었다. 결론적으로 본 연구 결과는 조릿대 잎이 천연 항균 소재로서의 활용 가능성이 있음을 확인해주었다.

당귀 잎의 항산화 활성 (In vitro Antioxidant Activity of the Aqueous of Angelicae gigas Nakai Leaves)

  • Angelicae gigas Nakai has been used as a traditional medicine as well as an edible vegetable in South Korea. In this study, the total phenolic and flavonoid content and antioxidants of A. gigas Nakai leaves were examined in vitro via hydroxyl-radical-scavenging activity, reducing power activity, metal chelating assay, and DPPH-free -radical-scavenging assay. Among all the extracts from A. gigas Nakai leaves, the ethanol extract showed the strongest effects in all of the assays. The $EC_{50}$ values for the DPPH-radical-scavenging activities of ethanol, methanol, and water extracts were 31.47, 42.14, and $58.47{\mu}g/mL$, respectively. Among the extracts from A. gigas Kakai leaves, the ethanol extract had the highest levels of total phenolics ($7.84{\pm}1.46$ mg TAN/g) and total flavonoids ($4.23{\pm}0.03$ mg QE/g), which correlated strongly with the individual phenolic-compound (p-hydroxybenzoic acid, vanillin, and trans-ferulic acid) contents. The ethanol extract also showed stronger antioxidant activity than tocopherol in hydroxyl- radical-scavenging activity assay. These results indicate that the ethanol extract of A. gigas Kakai leaves possesses significant antioxidant properties, which suggests its great potential as a functional-food ingredient in the food industry.

내소승마탕(內消升麻湯)의 항암효과(抗癌效果) 연구(硏究) (Anti-tumor activities of Naesosungma-tang)

  • This study was carried out to evaluate antioxidative, cytotoxic and chemosensitizing effects for anti-tumor activities of Naesosungma-tang (NST). The results were as follows ; 1. The oxidative effects were measured by DPPH radical scavenging activity. NST water extract was showed more effective than ethanol extract and also various solvent fractions from NST showed effective in the following order : butanol fraction > ethyl acetate fraction > hexane fraction > aqueous fraction. 2. NST water extract was showed cytotoxic effects on the human cancer cells Calu-6 cell and SNU-601 cell, but not on MCF-7 cell. 3. The viability of cells was measured in 4 kinds of solvent fractions from water extract of NST by MTT assay. The results were as followings : 1) On the NIH3T3 cell, $IC_{50}$ value was $200{\sim}300\;{\mu}g/m{\ell}$ in three fractions except aqueous fraction. 2) On the Caco-2 cell, effective only in the butanol fraction. 3) On the Calu-6 cell, most effective in hexane fraction($IC_{50}:80\;{\mu}g/m{\ell}$) and effective in ethyl acetate and butanol fraction($IC_{50}:100{\sim}120\;{\mu}g/m{\ell}$). 4) On the MCF-7 cell, $IC_{50}$ value was similar in three fractions except aqueous fraction. 5) On the SNU-601 cell, $IC_{50}$ value was within $100\;{\mu}g/m{\ell}$ in three fractions except aqueous fraction. 6) On the HCT-1l6 cell, $IC_{50}$ value was $150{\sim}200\;{\mu}g/m{\ell}$ in three fractions except aqueous fraction. 7) On the AML-2/WT cell, not effective in four kinds of solvent fractions. 4. In combined effects of NST and vincristine on AML-2/D100 cell, water and ethanol extracts of NST and hexane fraction of water extracts of NST more effectively inhibited proliferation of AML-2/D100 cell. These results Suggest that NST has antioxidative and cytotoxic effects against Caco-2, Calu-6, MCF-7, SNU-601, HCT116 and NIH3T3 cell especially Calu-6 and SNU-601 cell, and also chemosensitizing effects against AML-2/D100 cell in combined vincristine.

Anti-oxidative, Acetylcholinesterase Inhibitory Activities and Acute Toxicity Study of Nepeta sibirica L.

  • Nepeta sibirica L. or Siberian catmint is a medicinal plant species used in Mongolian traditional medicine for curing human different disorders and veterinary practices. The previous study of the whole plant concentrated on the determination of its essential oil composition and reported that the major ones are sesquiterpenes, including nepetalactone. The aim of this study was to reveal a new biological activity of the above-ground parts of N. sibirica L. and compare the activity of different extracts correlating with the content of biologically active compounds and evaluate their toxicity. For this purpose, anti-oxidative and acetylcholinesterase inhibitory activities of the above-ground parts of N. sibirica L. aqueous and ethanol (EtOH) (40%, 70%) extracts were assayed spectrophotometrically. The aqueous extract showed positive anti-oxidative activity by both tested DPPH and FRAP assays with IC50 134.24 ± 1.42 mg/mL and FRAP value 1385.15 ± 8.12 µmol/L at 200 ㎍/mL, in contrast to 40% and 70% EtOH extracts. The 70% EtOH extract presented the highest acetylcholinesterase inhibitory activity (IC50 77.29 ± 0.38 mg/mL) followed by 40% EtOH extract (176.72 ± 0.35 mg/mL) and aqueous extract (275.41 ± 0.23 mg/mL). Total phenolics were found to be gallic acid equivalent, % 3.74 ± 0.05 (70% EtOH), 3.94 ± 0.04 (40% EtOH), and 3.79 ± 0.16 (aqueous), whereas the total flavonoids as a rutin equivalent, % as 2.01 ± 0.12, 1.44 ± 0.17 and 1.99 ± 0.02, each. The aqueous extract showed the best anti-oxidative and lowest activity against the acetylcholinesterase; however, the 70% EtOH extract showed the opposite effects than that of the aqueous. No mortality incidence was visible at various doses, indicating that the oral median lethal dose of aqueous and 70% EtOH extracts were considered greater than 5000 mg/kg. N. sibirica L. belongs to the non-toxic category of the OECD 423 classification.

에탄올 농도에 따른 돌외 잎 차 추출물의 생리활성 (Physiological activity of the extract from Dolwoe (Gynostemma pentaphyllum Makino) leaves tea by different ethanol concentrations)

  • 본 연구는 페놀 및 플라보노이드의 총 함량과 생리 활성에 대한 다양한 농도의 에탄올 추출 효과를 비교하기 위해 수행하였다. 추출물의 총 페놀 함량은 35.54 ~ 71.52 mg GAE / g 범위였다. 용매의 에탄올 농도가 증가함에 따라 페놀 함량이 높았고 80%와 99.5%의 에탄올 추출물에서 최고 함량이 확인되었다. 추출물의 총 플라보노이드 함량은 페놀 함량의 경향과 유사하였다. 항산화 실험으로 DPPH, nitric oxide, superoxide, hydroxyl radical 소거능과 TEAC, FRAP, ORAC를 측정하였다. Radical 소거능은 물이나 99.5% 에탄올 추출물 보다 에탄올 수용액 추출물의 활성이 우수하였다. TEAC와 FRAP은 에탄올 농도 의존적인 경향으로 높은 값이 나타났고 ORAC는 40 ~ 80% 에탄올 추출물이 높은 활성을 보였다. 돌외 잎 차의 항산화능을 검토한 결과 측정 방식에 따라 다른 경향이 확인되었다. 그리고 대부분의 실험에서 물 또는 99.5% 에탄올 추출물의 활성이 상대적으로 낮았다. ${\alpha}$-Glucosidase 저해활성은 80%, 99.5% 에탄올 추출물에서 가장 높은 활성이 확인되었고, 미생물 저해활성 또한 동일한 결과를 보였다. 따라서 차의 항산화 활성과 생리활성을 복합적으로 고려할 때 80% 에탄올로 추출하는 것이 적절한 것으로 사료된다. 이러한 결과를 토대로 돌외 차의 식품개발에 있어 기초자료로 활용될 수 있다고 생각된다.

Prophylactic effect of aqueous extract of Sesamum indicum seeds on ethanol-induced toxicity in male rats

  • The liver is vulnerable to alcohol-related injury because it is the primary site of alcohol metabolism. Additionally, a number of potentially dangerous by-products are generated as alcohol is broken down in the liver. However, dietary supplements may prevent or relieve some of alcohol's deleterious effects. Therefore, this study was conducted to evaluate the prophylactic effect of aqueous extract of Sesamum indicum (SI) on ethanol induced toxicity in rats. Male Wistar albino rats were divided into control, ethanol, pre-treatment, simultaneous and post-treatment groups. In the prophylactic experiment, Sesamum indicum, (200 mg/kg body weight) was administered by oral gavage for 28 days; two hours before, simultaneously with or two hours after ethanol exposure. Toxicity was induced by administering 45% ethanol (4.8 g/kg bw) by oral gavage. Lipid peroxidation (TBARS) and reduced glutathione (GSH) levels and catalase (CAT), glutathione peroxidase (GPx), superoxide dismutase (SOD) and gluthathione-S-transferase (GST) activities were then determined in the liver, serum triglyceride (TG) levels, alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities were monitored and histological examination was carried out. The results revealed that ethanol administration led to significant elevation of TBARS level while depleting in the level of GSH as well as CAT, GPx, SOD and GST activities. Similarly, TG level and ALT and AST activities were elevated. The SI pre-treated group significantly inhibited TBARS, restored GSH level, enhanced CAT, GPx, SOD and GST activities and significantly decreased the elevated level of serum TG, ALT and AST activities. SI treatment (simultaneously with ethanol) exhibited similar effects to those of the SI pre-treated groups, while the SI post-treated group did not show the same protection as the Pre-treated group. S. indicum possesses antioxidant and hepatoprotective properties, that eliminate the deleterious effects of toxic metabolites of ethanol.

아가리쿠스 버섯 균사체 추출물 및 분획물의 항돌연변이원성 및 세포 독성 효과 (The Antimutagenic Effects and Cytotoxic Activities of Agaricus blazei Murill Mycelium Extracts and Fractions)

  • This study was performed to observe the antioxidative effects, antimutagenic capacity, and cytotoxic activity of the 70% ethanol extract, and fractions, of Agaricus blazei Murill mycelium, using DPPH free radical scavenging ability, the Ames test, and SRB assay, respectively. Among the fractions, ethyl acetate showed the most effective antioxidative capacity according to the $RC_{50}$(73.6 $\mu$g/mL) of the scavenging effect on the DPPH radical. The inhibition rate of both the aqueous fraction and 70% ethanol extract(200 $\mu$g/plate) toward the Salmonella typhimurium TA100 strain was 94.6%, and it was 89.4% against the mutagenesis induced by MNNG(0.4 $\mu$g/plate). In addition, an identical concentration of the 70% ethanol extract in the TA98 strain, and the ethyl acetate fraction in the TA100 strain, showed inhibition rates of 80.3% and 76.9%, respectively, the highest activities against the mutagenesis induced by 4NQO(0.15 $\mu$g/plate). The cytotoxic effects of the 70% ethanol extract and its fractions increased with increasing sample concentration against human cervical adenocarcinoma(HeLa), human hepatocellular carcinoma(Hep3B), human breast adenocarcinoma(MCF-7), human stomach adenocarcinoma(AGS), and human lung carcinoma (A549). A 1 mg/mL concentration of the ethyl acetate fraction showed cytotoxicities of 77%, 83.8%, 82.1%, 83.1%, and 92.6% against HeLa, Hep3B, MCF-7, AGS and A549, respectively.

담배연기응축물로 유도된 돌연변이와 구절초 추출물의 억제 효과 (Inhibitory Effect of Chrysanthemum zawadskii var. latilobum Kitamura Extracts against Mutagenicity of Cigarette Smoke Condensates (CSC))

  • This study was carried out to investigate whether Chrysanthermum zawadskii var. latilobum Kitamura (C. zawadskii) extracts has an inhibitory effect against the mutagenicity by cigarette smoke condensates (CSC). C. zawadskii was extracted with 70% ethanol and the yield was 18.5%. We further fractioned 70% ethanol extract sequentially to diethylether, chloroform, dichloromethane, and aqueous water, and gained the yield of 17.5%, 5.6%, 5.8%, 32.8% and 35.5%, respectively. In the Ames test, there was no mutagenic effect of crude extract and its solvent fractions up to 2 mg/plate toward Salmonella typhimurium TA 98 with or without S-9 mix metabolic activations. On the contrary, the crude extract showed an inhibitory activity against the mutagenicity of CSC in the presence of S-9 mix metabolic activation. Diethyl ether layer among five solvent fractions showed the highest inhibitory activity. The inhibitory activity of diethyl ether fraction was also increased in a dose-dependent manner and the inhibitory rate was about 97.7% at the concentration of 1 mg/plate. In this study, we conclude that crude extract of C. zawadskii itself is potentially safe for mutagenicity, and the diethyl ether fraction has an inhibitory effect against the mutagenicity of CSC.
