Effects of Wax Gourd Extracts on Adipocyte Differentiation and Uncoupling Protein Genes(Ucps) Expression in 3T3-Ll Preadipocytes

  • Although various raw plant materials have been demonstrated to exert anti-obesity effects to a greater or lesser extent in both humans and animals when they are used to supplement the diet, it has not been shown extensively that they influence adipocyte cell differentiation involving lipid metabolic gene expressions. Using a well-established 3T3-L1 preadipocyte differentiation system, we decided to look into molecular and cellular event occurring during adipocyte differentiation when raw plant materials aye included in the process, in an effort to demonstrate the potential use of a screening system to define the functions of traditionally well-known materials. To these ends, the effects of ethanol (EtOH) or EtOH/distilled water (DW) extracts of Wax Gourd were examined using cytochemical and molecular analyses to determine whether components of the extracts modulate adipocyte differentiation of 3T3-Ll preadipocytes in vitro. The cytochemical results demonstrated that EtOH or EtOH/DW extracts did not affect lipid accumulation and cell proliferation, although the degree of lipid accumulation was influenced slightly depending on the extract. EtOH extract was highly effective in apoptotic induction during differentiation of 3T3-Ll preadipocytes (p<0.05). Reverse transcription-polymerase chain reaction (RT-PCR) analysis of lipoprotein lipase (LPL), Uncoupling protein (Ucp) 2, 3 and 4 also showed that while LPL expression was not influenced, Ucp2, 3 and 4 were up regulated in the EtOH extract-treated group and down regulated in the EtOH/DW extract-treated group. These changes in gene expressions suggest that the components in different fractions of Wax Gourd extracts may modulate lipid metabolism by either direct or indirect action. Taking these results together, it was concluded that molecular and cellular analyses of adipocyte differentiation involving lipid metabolic genes should facilitate understanding of cellular events occurring during adipocyte differentiation. Furthermore, the experimental scheme and analytical methods used in this study should provide a screening system for the functional study of raw plant materials in obesity research.

Evaluation of the EtOAc Extract of Lemongrass (Cymbopogon citratus) as a Potential Skincare Cosmetic Material for Acne Vulgaris

  • This study evaluated the biological properties of lemongrass (Cymbopogon citratus) extracts. The EtOAc extract of lemongrass had DPPH, TEAC, and nitric oxide-scavenging activity assay results of 58.06, 44.14, and 41.08% at the concentration of 50, 10, and 50 ㎍/ml, respectively. The EtOAc extract had higher elastase and collagenase inhibitory activities than the 80% MeOH, n-hexane, BuOH, and water extracts and comparable whitening activity toward monophenolase or diphenolase. Also, the EtOAc fraction had higher lipase inhibitory and antimicrobial activities against Cutibacterium acnes among extracts which is known to an important contributor to the progression of inflammatory acne vulgaris, and an opportunistic pathogen present in human skin. Total phenolic and flavonoid concentrations in the EtOAc extract were 132.31 mg CAE/g extract and 104.50 mg NE/g extract, respectively. Biologically active compounds in lemongrass extracts were analyzed by LC-MS. This study confirms that lemongrass extracts have potential use as cosmetic skincare ingredients. Thus, lemongrass can be considered a promising natural source of readily available, low-cost extracts rich in antioxidant, skincare, and antimicrobial compounds that might be suitable for replacing synthetic compounds in the cosmeceutical industry.

오미자(Schisandra chinensis) 착즙박 추출물의 항산화 활성 (Antioxidation activity of residue after omija (Schisandra chinensis) juice extract)

  • 오미자를 착즙하고 남은 착즙박을 이용하여 추출조건에 따른 이화학적 변화를 알아보았다. 즉, 착즙박을 열수, 50% 에탄올, 70% 에탄올 및 95% 에탄올을 추출 용매로 사용하였으며, 추출시간은 1시간에서 10시간까지 1시간 간격으로 $60^{\circ}C$에서 정치 추출하여 여과한 후, 감압 농축하여 제조된 용매별 농축액을 분석시료로 이용하였다. 오미자에 함유된 유효성분 중 하나인 schizandrin 함량은 추출 용매에 따라서 열수: 0.52 g/100 g, 50% 에탄올: 1.79 g/100 g, 70% 에탄올: 2.01 g/100 g 및 95% 에탄올은 2.37 g/100을 나타내었으며, 열수를 이용하여 추출한 경우보다 에탄올 농도가 증가할수록 schizandrin 함량이 증가하는 경향을 나타내었다. 총 폴리페놀 함량은 50% 에탄올 용매 추출물에서 16.70 mg/mL로 가장 높게 나타났으며, 2,2-diphenyl-1-picrylhydrazyl 전자공여소거능에 대한 효과는 분석 시료의 200 mg/mL 농도에서 측정하였을 때, 50% 에탄올 용매 추출물에서 86.16%로 가장 높게 나타내었다. 또한, 안토시아닌 함량은 모든 용매 추출물이 추출 시간이 증가할수록 감소하는 경향을 보였다.

Effect of the Ethanol Extract from the Aleurone Layer of Anthocyanin-Pigmented Rice on Blood Glucose and Lipid Metabolism in Streptozotocin Induced Diabetic Rats

  • The present study was designed to investigate the hypoglycemic activity and lipid metabolism of ethanol (EtOH) extract from the aleurone layer of anthocyanin-pigmented (AP) rice in streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley male rats weighing 210~240 g were divided into 4 groups, normal, diabetic control, and two experimental groups, and diabetes in rats was induced by injection of STZ (45 mg/kg, body weight) into tail vein. The EtOH extract of the powdered aleurone layer of AP rice was administered orally in diabetic rats for 14 days. In order to find the hypoglycemic effects in the animal model, the body weight, plasma glucose levels, cholesterol, HDL-cholesterol, triglyceride (TG), free fatty acid (FFA), aspartate aminotransferase (AST) and alanine amino- transferase activities (ALT) were determined. Oral administration of 1.0 81kg on the EtOH extract for 14 days resulted in a significant reduction in blood glucose, ALT, TG and FFA. However, in the case of 2.0 g/kg, the hypo-glycemic effects were not considerable. This results suggest that the EtOH extract might induce hypoglycemic effects in STZ-induced diabetic rats due to some photochemical components in the aleurone layer of AP rice.

  • The purpose of this study is to investigate the effect of water extract from Artemisiae Argi Folium (WAAF) on hydrogen peroxide production within mouse macrophage Raw 264.7 Cells treated with gallic acid, EtOH, LPS, and acetaminophen. WAAF (0${\sim}$400 ug/mL) was treated with gallic acid, EtOH, LPS, acetaminophen. And the intracellular productions of hydrogen peroxide were measured by dihydrorhodamine 123 (DHR) assay. WAAF showed the restoration of the intracellular productions of hydrogen peroxide which were reduced by gallic acid, EtOH, LPS, and acetaminophen in Raw 264.7 Cells. WAAF could be supposed to have the immunological activity related with macrophage's oxidative burst.

Characterization of Mulberry Root Bark Extracts (Morus alba L.) Based on the Extraction Temperature and Solvent

  • Mulberry root bark is one of potential plant sources for antioxidant materials which can be used for the relief of oxidative stress. To explore the effects of solvent type and temperature on the structural characteristics and antioxidant activity of the root bark extracts, we prepared various extracts of mulberry root bark (Morus alba L.) using 0 - 100 % ethanol (EtOH) at RT - 100℃. EtOH concentration and temperature critically affected the extraction yields, the content of bioactive components, and antioxidant activity of the extracts. Use of high content of EtOH solvent and low temperature resulted in the low extraction yield. Meanwhile, it was revealed that the extract prepared using absolute EtOH at room temperature contained polyphenols and flavonoids with the highest contents among other extracts. Interestingly, the temperature differently affected the polyphenol and flavonoid contents according to the solvent types. In the case of 30% EtOH solvent, polyphenol and flavonoid contents increased with an increase in temperature, whereas in the case of 70 and 100 % EtOH, these contents decreased. Using the radical scavenging assay, it was confirmed that the 100% EtOH extracts had higher antioxidant activity compared to distilled water (DW) extracts regardless of temperature. Also, heating might extract more antioxidant components from the root bark. Especially, the extract prepared using 30% EtOH solvent at 100℃ showed the highest antioxidant activity. Taken together, these experimental results imply that the extraction parameters should be designed carefully considering the productivity, the extracted bioactive components, and antioxidant activity.

체지방 감소 고기능성 발현 떫은 감 소재 개발 연구 (Research for Reduction of Body Fat and Development of High Functionality in Young Persimmon Fruit)

  • Objectives : This study was conducted to evaluate the antioxidant activity and inhibitory activity of pancreatic lipase in vitro using natural products sangju dungsi, chungdo bansi, kabju baekmok three kind of young persimmon fruit extracts and to develop a high - functional anti - obesity substance with reduced body fat. Methods : The antioxidative effect of the three kind of young persimmon fruit extracts was determined by hot water extraction, 30% EtOH extraction and 70% EtOH extraction. The antioxidant activities were evaluated through radical scavenging assays using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radicals. Furthermore, we examined total phenol and flavonoids contents. And the pancreatic lipase inhibition was measured by selecting the sangju dungsi extract with antioxidant effect. Results : The yield of water, 30% and 70% EtOH extract from young persimmon fruit was sangju dungsi (water 8.3%, 30% EtOH 18.1%, 70% EtOH 16.9%), chungdo bansi (water 16.2%, 30% EtOH 16.1%, 70% EtOH 11.6%), kabju baekmok (water 16%, 30% EtOH 16.8%, 70% EtOH 11.5%). The sangju dungsi 30% EtOH extract eminent DPPH ($IC_{50}$, $19.25{\pm}2.94{\mu}g/m{\ell}$) and ABTS ($IC_{50}$, $19.94{\pm}4.07{\mu}g/m{\ell}$) radical scavenging activity, and total phenol $52.37{\pm}0.78mg/g$ and flavonoid $7.97{\pm}0.13mg/g$ contents were also excellent. It also showed excellent pancreatic lipase inhibition efficacy. Conclusion : The extracts from the three kind of young persimmon fruit extracts showed excellent antioxidant activity. sangju dungsi 30% EtOH extract may be a new anti-obesity material by reducing fat absorption through the inhibition of pancreatic lipase.

Anti-oxidative, Acetylcholinesterase Inhibitory Activities and Acute Toxicity Study of Nepeta sibirica L.

  • Nepeta sibirica L. or Siberian catmint is a medicinal plant species used in Mongolian traditional medicine for curing human different disorders and veterinary practices. The previous study of the whole plant concentrated on the determination of its essential oil composition and reported that the major ones are sesquiterpenes, including nepetalactone. The aim of this study was to reveal a new biological activity of the above-ground parts of N. sibirica L. and compare the activity of different extracts correlating with the content of biologically active compounds and evaluate their toxicity. For this purpose, anti-oxidative and acetylcholinesterase inhibitory activities of the above-ground parts of N. sibirica L. aqueous and ethanol (EtOH) (40%, 70%) extracts were assayed spectrophotometrically. The aqueous extract showed positive anti-oxidative activity by both tested DPPH and FRAP assays with IC50 134.24 ± 1.42 mg/mL and FRAP value 1385.15 ± 8.12 µmol/L at 200 ㎍/mL, in contrast to 40% and 70% EtOH extracts. The 70% EtOH extract presented the highest acetylcholinesterase inhibitory activity (IC50 77.29 ± 0.38 mg/mL) followed by 40% EtOH extract (176.72 ± 0.35 mg/mL) and aqueous extract (275.41 ± 0.23 mg/mL). Total phenolics were found to be gallic acid equivalent, % 3.74 ± 0.05 (70% EtOH), 3.94 ± 0.04 (40% EtOH), and 3.79 ± 0.16 (aqueous), whereas the total flavonoids as a rutin equivalent, % as 2.01 ± 0.12, 1.44 ± 0.17 and 1.99 ± 0.02, each. The aqueous extract showed the best anti-oxidative and lowest activity against the acetylcholinesterase; however, the 70% EtOH extract showed the opposite effects than that of the aqueous. No mortality incidence was visible at various doses, indicating that the oral median lethal dose of aqueous and 70% EtOH extracts were considered greater than 5000 mg/kg. N. sibirica L. belongs to the non-toxic category of the OECD 423 classification.

  • Antioxidant and anticoagulant activities of water extract and 70% ethanol extract of Phyllostachys pubescence leaf (PPL) produced in Geoje were investigated. Total polyphenol contents of PPL-water extract and PPL-70%, EtOH extract were measured as $80.6{\pm}3.2$ and $71.3{\pm}2.7$ mg/g, respectively. DPPH radical scavenging activity of PPL-water extract and PPL-70% EtOH extract were 50% and 90%, respectively. The anticoagulant activity, in intrinsic pathway of activated partial thromboplastin time (APTT) showed a concentration-dependency. This results showed that PPL-water extract and PPL-70% EtOH extract have high antioxidant and anticoagulant activities.

토우슬(土牛膝), 회우슬(懷牛膝)의 항신경염증 및 신경세포 보호 효과 비교 (Comparative Study of Achyranthes japonica Nakai and Achyranthes bidentata Blume on Anti-Neuroinflammatory and Neuroprotective Effects)

  • Objectives : Achyranthes japonica Nakai (AJ) and Achyranthes bidentata Blume (AB) have been used without distinguishment. Moreover, comparative studies of AJ and AB on physiological activity in the organism levels remain fully understood. In this study, we aimed to evaluate and compare the effects of AJ and AB on anti-neuroinflammatory and neuroprotective effects. Methods : AJ and AB were extracted with distilled water (DW) and 70% ethanol (EtOH) extract. For the evaluation of anti-neuroinflammatory effects, we measured the production of nitric oxide (NO) in lipopolysaccharide (LPS)-treated BV2 microglial cells. To evaluate the neuroprotective effects, we assessed cell viability against toxicity, including hydrogen peroxide (H2O2), 6-hydroxydopamine (6-OHDA), and amyloid-beta (A𝛽), respectively, in PC12 or HT22 cells. Results : DW and 70% EtOH extracts of AJ and AB inhibited LPS-induced NO production in BV2 cells, with no significant differences between the origins and extraction solvents. Additionally, AJ and AB had no cytotoxicity, and exhibited the similar neuroprotective effects against H2O2 and 6-OHDA toxicities in PC12 cells, showing stronger activity in 70% EtOH extract compared to the DW extract. Furthermore, 70% EtOH extracts of AJ and AB protected neuronal cell against A𝛽 toxicity-induced cytotoxicity in HT22 cells. Conclusions : We demonstrated that AJ and AB have anti-neuroinflammatory and neuroprotective effects in the 70% EtOH extract compared to DW extract, with no significant differences between the species. These results suggested that AJ and AB would be the potential candidates for neurodegenerative diseases.
