GnRH Agonist Stimulation Test (GAST) for Prediction of Ovarian Response in Controlled Ovarian Stimulation (COH) (난소기능평가를 위한 Gonadotropin Releasing Hormone Agonist Stimulation Test (GAST)의 효용성에 관한 연구)

  • Objectives: The aims of this study are 1) to determine if GAST is a better indicator in predicting ovarian response to COH compared with patient's age or basal FSH level and 2) to evaluate its role in detecting abnormal ovarian response. Design: Prospective study in 118 patients undergoing IVF-ET using GnRH-a short protocol during May-September 1995. Materials and Methods: After blood sampling for basal FSH and estradiol $(E_2)$ on cycle day two, 0.5ml (0.525mg) GnRH agonist ($Suprefact^{(r)}$, Hoechst) was injected subcutaneously. Serum $E_2$ was measured 24 hours later. Initial $E_2$ difference $({\Delta}E_2)$ was defined as the change in $E_2$ on day 3 over the baseline day 2 value. Sixteen patients with ovarian cyst or single ovary or incorrect blood collection time were excluded from the analysis. The patients were divided into three groups by ${\Delta}E_2$; group A (n=30):${\Delta}E_2$<40 pg/ml, group B (n=52): 40 pg/ml${\leq}{\Delta}E_2$<100 pg/ml, group C (n=20): ${\Delta}E_2{\leq}100$ pg/ml. COH was done by GnRH agonist/HMG/hCG and IVF-ET was followed. Ratio of $E_2$ on day of hCG injection over the number of ampules of gonadotropins used ($E_2hCGday$/Amp) was regarded as ovarian responsiveness. Poor ovarian response and overstimulation were defined as $E_2$ hCGday less than 600 pg/ml and greater than 5000 pg/ml, respectively. Results: Mean age $({\pm}SEM)$ in group A, B and C were $33.7{\pm}0.8^*,\;31.5{\pm}0.6\;and\;30.6{\pm}0.5^*$, respectively ($^*$: p<0.05). Mean basal FSH level of group $A(11.1{\pm}1.1mlU/ml)$ was significantly higher than those of $B(7.4{\pm}0.2mIU/ml)$ and C $(6.8{\pm}0.4mIU/ml)$ (p<0.001). Mean $E_2hCGday$ of group A was significantly lower than those of group B or C, i.e., $1402.1{\pm}187.7pg/ml,\;3153.2{\pm}240.0pg/ml,\;4078.8{\pm}306.4pg/ml$ respectively (p<0.0001). The number of ampules of gonadotropins used in group A was significantly greater than those in group B or C: $38.6{\pm}2.3,\;24.2{\pm}1.1\;and\;18.5{\pm}1.0$ (p<0.0001). The number of oocytes retrieved in group A was significantly smaller than those in group B or C: $6.4{\pm}1.1,\;15.5{\pm}1.1\;and\;18.6{\pm}1.6$, respectively (p<0.0001). By stepwise multiple regression, only ${\Delta}E_2$ showed a significant correlation (r=0.68, p<0.0001) with $E_2HCGday$/Amp, while age or basal FSH level were not significant. Likewise, only ${\Delta}E_2$ correlated significantly with the number of oocytes retrieved (r=0.57, p<0.001). All four patients whose COH was canceled due to poor ovarian response belonged to group A only (Fisher's exact test, p<0.01). Whereas none of 30 patients in group A (0%) had overstimulation, 14 patients among 72 patients (19.4%) in group B and C had overstimulation (Fisher's exact test, p<0.01). Conclusions: These data suggest that initial $E_2$ difference after GAST may be a better prognostic indicator of ovarian response to COH than age or basal FSH level. Since initial $E_2$ difference demonstrates significant association with abnormal ovarian response such as poor ovarian response necessitating cycle cancellation or overstimulation, GAST may be helpful in monitoring and consultation of patients during COH in IVF-ET cycle.

Red AGNs becoming normal AGNs

  • Red active galactic nuclei (AGNs) are supposed to be transitional objects becoming normal AGNs in the galaxy evolution scenario. So far, ~200 red AGNs have been found by very red color in optical through NIR wavelength (e.g., r'-K >5 and J-K>1.3; Urrutia et al. 2009). Here, we compare nuclear activities of the red AGNs to those of normal AGNs to verify the evolutionary phase of the red AGNs. In order to study the nuclear activities of the red AGNs, we use broad emission lines of $P{\beta}$ ($1.28{\mu}m$) of which flux is less suppressed by a factor of 100 than the $H{\beta}$ line in the case of the red AGNs with a color excess of E(B-V)=2 mag. We use 16 red AGNs discovered in previous red AGN surveys by using SDSS, 2MASS, and FIRST (Glikman et al. 2007; Urrutia et al. 2009) at z ~0.7 for which $P{\beta}$ lines are redshifted to the sky window at ${\sim}2.2{\mu}m$. The mean Eddington ratio of the 16 red AGNs is 0.562, and that of the normal AGNs is 0.320, which indicates the red AGNs include more active black hole (BH) than the normal AGNs. To test how significantly the nuclear activities of the red AGNs and the normal AGNs are different, we perform a two-dimensional Kolmogorov-Smirnov test (K-S test) on their Eddington ratio distributions. The K-S test shows the maximum deviation between the cumulative distributions, D, is 0.48, and the probability of null hypothesis, p, is even less than 0.001. This result is consistent with a picture of that the red AGNs are in intermediate phase between the stage of merger-driven starburst galaxy and the normal AGN.

THE VIRIAL RELATION AND INTRINSIC SHAPE OF EARLY-TYPE GALAXIES

  • Early-type galaxies (ETGs) are supposed to follow the virial relation $M=k_e{\sigma}^2R_e/G$, with M being the mass, σ* being the stellar velocity dispersion, Re being the effective radius, G being Newton's constant, and ke being the virial factor, a geometry factor of order unity. Applying this relation to (a) the ATLAS3D sample of Cappellari et al. (2013) and (b) the sample of Saglia et al. (2016) gives ensemble-averaged factors 〈ke〉 = 5.15 ± 0.09 and 〈ke〉 = 4.01 ± 0.18, respectively, with the difference arising from different definitions of effective velocity dispersions. The two datasets reveal a statistically significant tilt of the empirical relation relative to the theoretical virial relation such that $M{\propto}({\sigma}^2_*R_e)^{0.92}$. This tilt disappears when replacing Re with the semi-major axis of the projected half-light ellipse, a. All best-fit scaling relations show zero intrinsic scatter, implying that the mass plane of ETGs is fully determined by the virial relation. Whenever a comparison is possible, my results are consistent with, and confirm, the results by Cappellari et al. (2013). The difference between the relations using either a or Re arises from a known lack of highly elliptical high-mass galaxies; this leads to a scaling (1 - ϵ ) ∝ M0.12, with ϵ being the ellipticity and $R_e=a\sqrt[]{1-{\epsilon}}$. Accordingly, a, not Re, is the correct proxy for the scale radius of ETGs. By geometry, this implies that early-type galaxies are axisymmetric and oblate in general, in agreement with published results from modeling based on kinematics and light distributions.

Effects of Several Herbal Medicines on the Replication of Hepatitis B Virus (수종(數種)의 한약재(韓藥材)가 B형 간염(肝炎)바이러스 증식억제(增殖抑制)에 미치는 효과(效果))

  • This study was performed to investigate an anti-HBV activities of the aqueous extracts from 10 Korean herbal medicines in the HepG2 2.2.15 cell culture system and the results were as follows: 1. The extracts of 6 plants (Herba Artemisiae Capillaris, Radix et Rhizoma Rhei, Cortex Cinnamomi, Fructus Chebulae, Fructus Rubi and Radix Rubi) decreased, significantly and dose-dependently, the levels of extracellular HBV virion in the concentrations (10, 100, 500 and $1,000\;{\mu}g/m{\ell}$) tested. 2. However, others (Radix lsatidis, Lignum Sappan, Herba Lysimachiae and Fructus Lycii) did not show any effect either on the replication of HBV or on the levels of virion DNA in the culture media of HepG2 2.2.15 cell. 3. Among the 6 plants which showed the inhibitory potency on the production of extracellular HBV virion, Radix et Rhizoma Rhei, Cortex Cinnamomi, Fructus Chebulae, Fructus Rubi and Radix Rubi except Herba Artemisiae Capillaris also showed the inhibition of the replication of intracellular HEV DNA in the range of $100{\sim}500\;{\mu}g/m{\ell}$. Considering the above results, it is thought that 6 plants(Herba Artemisiae Capillaris, Radix et Rhizoma Rhei, Cortex Cinnamomi, Fructus Chebulae, Fructus Rubi and Radix Rubi) possess the anti-HBV activities in the HepG2 2.2.15 cell culture system. We thus suggest that these plants possess a potential as a therapeutic agent for the chronic viral hepatitis. These results might be useful as a basic data for the development of the new preventive drugs for HBV diseases.

Corydalis cornupetala Y.H. Kim et J.H. Jeong: A New Species of Corydalis sect. Corydalis (Fumariaceae) (쇠뿔현호색: 현호색속 현호색절의 1신종)

  • A new species of Corydalis sect. Corydalis (Fumariaceae), Corydalis cornupetala Y.H. Kim et J.H. Jeong is described from Gyeongsan-si, Gyeongsangbuk-do in Korea. This new species is distinguished from the other species of the sect. Corydalis by some flower characters such as, purplish white petal with two dark stripes and horn-shaped apex of outer petal. The linear leaflets of C. cornupetala are similar to the lineariloba type of C. remota Fisch. ex Max. or C. namdoensis B.U. Oh et J.G. Kim, but much longer and narrower. The linear or lanceolate bract of C. cornupetala is also distinctive from the elliptical shape of C. remota and C. namdoensis.

  • Characteristics of ethanol fermentation were investigated during the stationary culture of a killer yeast, Saccharomyces cerevisiae B15-1. Specific ethanol production rate reached the maximum level, 1.203 g-EtOH/g-cell-hr, at 150 g/l of the initial glucose concentration. No big differences were obtained in ethanol fermentability based on the initial sugar concentration below 150 g/l. When 200 g/l of sugar was used, fermentability dropped significantly. Although the final cell mass and the amount of ethanol produced were increased, their increase rates were declined according to the increase of initial sugar concentration. It was found that most of the sugar used below 150 g/l of concentration could be changed to ethanol. However, when 200 g/l of sugar was used, some of them remained in the media even after increase of cell mass and fermentation stopped. The ethanol yield was decreased when initial sugar concentration was high, and were increased when the amount of ethanol produced was increased and finally reached the plateau over 60 g/l of ethanol concentration.

Functional Analysis of Spectinomycin Biosynthetic Genes from Streptomyces spectabilis ATCC 27741

  • The function of genes related to spectinomycin biosynthesis (spcD, speA, speB, spcS2) from Streptomyces spectabilis ATCC 27741, a spectinomycin producer, was analyzed. Each gene was subcloned from a spectinomycin biosynthetic gene cluster and overexpressed in E. coli BL21 (DE3) using pET vector. After incubating each purified protein with its possible substrates, the final products were analyzed using high-performance liquid chromatography (HPLC). From these results, spcD, speA, and speB have been identified to be dTDP-glucose synthase, myo-inositol monophosphatase, and myo-inositol dehydrogenase, respectively. In addition, the results suggest that the spcS2 gene product functions downstream of the speB gene product in the biosynthetic pathway of spectinomycin. Taken together, the present study elucidates the early steps of the biosynthetic pathway for 6-deoxyhexose (6-DOH) part (actinospectose) and aminocyclitol part (actinamine) of spectinomycin.

Expression of Acetohydroxyacid Synthase from Bacillus anthracis and Its Potent Inhibitors

  • Acetohydroxyacid synthase (AHAS, EC 2. 2. 1. 6) is the enzyme that catalyses the first step in the common pathway of the biosynthesis of the branched chain amino acids, valine, leucine and isoleucine. For the first time, the AHAS gene from Bacillus anthracis was cloned into the expression vector pET28a(+), and was expressed in the E. coli strain BL21(DE3). The purified enzyme was checked on 12% SDS-PAGE to be a single band with molecular weight of 65 kDa. The optimum pH and temperature for B. anthracis AHAS was at pH 7.5 and 37 oC, respectively. Kinetic parameters of B. anthracis were as follows: Km for pyruvate, K0.5 for ThDP and Mg2+ was 4.8, 0.28 and 1.16 mM respectively. AHAS from B. anthracis showed strong resistance to three classes of herbicides, Londax (a sulfonylurea), Cadre (an imidazolinone), and TP (a triazolopyrimidine). These results indicated that these herbicides could be used in the search for new anti-bacterial drugs.

  • This paper deals with the Triozidae of Korea A total of 20 species belonging to 5 genera are recognized, among them 6 species are new to science as: Bactericera (S.) distinctissima n. sp., B. (S.) miyatakei n. sp., B. (S.) yamagishii n. sp., Heterotrioza (D.) noknamui n. sp., Trioza breviata n. sp., and T. mayicola n. sp., 4 species are newly recorded from Korea as: Epitrioza yasumatsui Miyatake, 1978, Bactericesa (S.) calcarata (Schaefer, 1949), B, (S.) striola (Flor, 1861), and Trioza abdominalis Flor, 1861, and a new combination is made to: Heterptrioza (D.) chilgia (Park et Lee, 1980) n. comb. Keys are provided to the genera and species respectively Some illustartions of head, forewing, male and female genitalic characters are given.

A Study on the Initial Hull Form Design by Using Form Parameters (형상계수에 의한 초기선형설계에 관한 연구)

  • This paper describes a method for generating an initial hull form by using form parameters. As a mathematical representation of curves, B-spline curves are used as well as the polynomials used by Durand et al. The five basic control curves and the centerline contour are defined to give the boundary conditions for body plan by using above mentioned mathematical models. From these curves body plan is determined. Two additional curves which are concerned the position of matching point between the cylindrical form and the water line are proposed to get the preliminary faired water lines.
