Bacillus cereus Clinical Isolates : Characteristics, Enterotoxin Production and Antimicrobial Susceptibility (임상 검체에서 분리된 Bacillus cereus의 성상, 장독소 생성 및 항균제 감수성)

  • Biochemical characteristics, enterotoxin production and antimicrobial susceptibility were determined for 30 strains of Bacillus cereus isolated from stool specimens of diarrhea patients at an university hospital in Chulabuk-do province. Positive rate for VP reaction and citrate utilization were lower, (33 % and 40 % respectively) while the rates of acid production from mannitol, arabinose, and xylose were higher (17 %, 13 % and 3 % respectively) than those obtained by other investigators. The enterotoxin gene was detected in 18 of 30 isolates (60 %) by PCR, and the toxin was detected from all of the toxin gene-positive isolates by RPLA test. The agar dilution test showed that all isolates were resistant to penicillin G and 73 % were to cephalothin, but all were susceptible to ciprofloxacin, clindamycin, erythromycin, fusidic acid, gentamicin, rifampin, teracycline and vancomycin. We conclude that B. cereus isolates producing acid from mannitol, arabinose and xylose exist, that PCR can be used to detect enterotoxin genes rapidly and accurately, and that this organism is susceptible to various antimicrobial agents though not penicillin G and cephalothin.

Bacteriological Study of Erysipelothrix rhusiopathiae Isolated from the Pigs in Youngnam Area (영남지방 돼지에서 분리한 Erysipelothrix rhusiopathiae의 세균학)

  • The present study was conducted to investigate biochemical, serologic, and pathogenic characteristic of E. rhusiopathiae isolated from the cases of acute septicemic swine erysipelas in Youngnam provinces during the period from June 1988 to September 1990. The majority of biochemical and cultural properties of E. rhusiopathiae isolated from pigs affected with acute erysipelas were identical to those of the standard strain employed. All of the 45 isolates were serotype la. All isolates were highly susceptible to penicillin G, lincomycin, cephalothin, ampicillin, erythromycin (MIC : 0.025-0.78IU or ${\mu}g$ / ml ), and moderately susceptible to oleandomycin, oxytetracycline, chloramphenicol (MIC : 0.78-25${\mu}g$ / ml ). Kanamycin and sulfadimethoxine showed no activity against the isolates(MIC : >400${\mu}g$ / ml ). The MICs of dihydrostreptomycin presented two distribution peaks ; of 45 strains, 5(11.1%) were resistant to dihydrostreptomycin(MIC : 400${\mu}g$ / ml ). All of 5 selected isolates were pathogenic for mite and $LD_{50}$ was $3.7{\times}10^3$viable cells. Mice immunized subcutaneously with live vaccine did not die after challenge to virulent isolates of E. rhusiopathiae.

  • Mastitis is a common and serious infection of the mammary gland in dairy cattle and has a major economic impact on the production of milk and dairy products. Bacterial mastitis is caused by several pathogens and is most frequently associated with coagulase-negative staphylococci (CNS). Although CNS are typically associated with subclinical or mild mastitis, the importance of CNS has increased as these pathogens have emerged as predominant mastitis-related pathogens in many countries. CNS can cause persistent infections, resulting in increased milk somatic cell counts and thereby affecting milk quality and decreasing milk production. Globally, Staphylococcus chromogenes, S. epidermidis, and S. simulans are the predominant CNS species in dairy cattle mastitis. Antibacterial resistance of CNS varies with species, and most CNS are susceptible to vancomycin and resistant to penicillin and tetracycline. As the most frequently isolated CNS species, some strains of S. chromogenes exhibit phenotypic resistance to ampicillin, erythromycin, oxacillin, penicillin, and tetracycline. Some strains of S. epidermidis and S. haemolyticus are only susceptible to vancomycin and rifampicin. Therefore, more studies are needed to achieve the control and prevention of CNS as environmental pathogens.

Clinical Study of Empyema Thoracis: a review of 64 cases (농흉의 임상적 고찰64례 보고)

  • A Clinical analysis of 64 patients of thoracic empyema was done who received surgical intervention at Dept. of Thoracic Surgery of the Chosun University Hospital in the period of 3 years from September 1976 to October 1979. Following was the results: 1. Seven cases [10.9%] were under the age of 15 years, 16 cases [25%] was between 15-30 years and 41 cases [64.1%] was above the age of 30 years. A proportion of children and adult was 1:8. 2. Male and female ratio was 3:1. Right and left side pleural cavity ratio was 2.4:1. 3. Predisposing factors were pneumonia [35.9%] and pulmonary tuberculosis [28.1%]. 4. Most frequently encountered symptoms were dyspnea, cough, chest pain and fever in order. 5. Etiologic organisms were confirmed in 39 cases [86.7%] which requested in 45 cases. Staphylococcal infections were 11 cases and streptococcal, pneumococcal pseudomonas infection was infected in order. 6. Pneumothorax was associated with empyema on 21 cases [32.8%]; among those 13 cases [61.9%] were tuberculous in nature. 7. Sensitivity test was revealed that Minocin was most very sensitive drug, and next Erythromycin, Gentamycin and Penbrex in order. But most resistant drugs were Penicillin, Kanamycin, Streptomycin and Tetracycline in order. 8. Treatments were combined with antibiotics therapy and several surgical procedures for empyema. 26 cases [40.6%] were treated with closed thoracotomy drainage, 17 cases [26.6%] with open thoracotomy tube drainage and 9 cases decortication and 9 cases thoracoplasty. 9. 2 death cases occurred in 64 cases of thoracic empyema, and 79.7% cases were discharged with recovery and improvement.

Purification and Characterization of Antistaphylococcal Substance from Pseudomonas sp. KUH-001

  • A bacterium producing unique antistaphylococcal substance (ASS) was isolated from soil samples. The isolated strain KUH-001 was identified to belong to Pseudomonas species from the characteristic properties of its fluorescence and cellular 3-hydroxy fatty acid composition, etc. The ASS component was purified by procedures employing activated carbon adsorption, column chromatography with silica gel, preparative TLC and HPLC. This compound could also be purified mainly by repeating of trituration and precipitation with chilled ether. Purified ASS with a m.p. value of $140~142^{\circ}C$ showed marked stability at high temperature (at $121^{\circ}C$ for 10 min) and extreme pHs (in 1N HC1 and 1N NaOH for 1 day) without significant loss of antibiotic activity. From spectral data of UV, IR, NMR, and FAB-MS, the compound was elucidated as 2-heptyl-4-hydroxyquinoline N-oxide (HHQO). Under the conditions employed, HHQO exhibited a narrow antimicrobial spectrum. active particularly against Staphylococcus aureus including the methicillin resistant strain. Moreover, it did not induce resistance, and besides, interacted synergistically with certain antibiotics such as vancomycin or erythromycin.

Antibiotic resistance and plasmid profile of salmonella spp isolated from swine in Kyoungbuk province (경북지방 돼지유래 salmonella 속균의 약제내성과 plasmid profile)

  • This study was conducted to investigate the antibiotic resistance and plasmid profiles of 58 salmonella spp isolated from mesenteric lymphnodes of slaughter pigs in Kyoungbuk province during the period from September 1997 to June 1998. The results obtained are as follow that all isolates were susceptible to amikacin, gentamicin, ciprofloxacin and enrofloxacin, and the majority of isolates were highly susceptible to norfloxacin, colistin, nalidixic acid and apramycin while they were moderately susceptible to kanamycin, cephalothin, chloramphenicol, ampicillin, trimethoprim and penicillin. The majority of isolates were over 90% resistant rates to lincomycin, sulfadimethoxine, vancomycin, methicillin and erythromycin. The plasmid profiles of 58 salmonella spp are developed 1 to 4 fractions, 0.9 to 29.5 Kb molecular range sizes and U strains (45.5%) were showed plasmid profiles by agarose gel electrophoresis. 5 derby harbored 29.5 Kb and 7 Kb, and S schwarzengrund had 14 Kb and 0.9 Kb harboring sizes. Four of 10 S agona and 2 of 4 S typhimurium were harbored 3.1 Kb and n.5 Kb, respectively. Thirty-five untypable strains are developed variable size fractions its showed small size plasmid profile less than 6 Kb and 22 (62.8%) of them had no detectable plasmids.

  • This study was carried out to investigate the prevalence and antimicrobial resistance patterns of Campylobacter jejuni isolated from duck feces. In total, 112 (32.9%) isolates of C. jejuni were identified from 430 duck feces samples from September to December, 2010. All isolates were susceptible to telithromycin, whereas majority of the C. jejuni isolates were resistant to azithromycin (18.8%), ciprofloxacin (86.6%), erythromycin (0.9%), gentamicin (15.2%), tetracycline (80.4%), florfenicol (3.6%), nalidixic acid (87.5%), clindamycin (7.1%). As a result, appropriate protocols for antimicrobial agents and strategies to reduce antimicrobial resistance will be needed in the future.

  • Objectives : The purpose of our study was to evaluate penicillin as a still drug of choice for severe endodontic infection, by analyzing the antimicrobial susceptibilities from endodontic infections with swelling to figure out appropriate antibiotics as empirical treatment. Materials and methods : This study involved 18 patients who attended for emergency treatment because of facial or periapical swelling associated with root canal infections. Identification and antimicrobial susceptibility test of each pathogen were performed by Vitek2 Systems (bioM$\acute{e}$rieux, Marcy l'Etoile, France). Results : The most frequent bacteria was Streptococcus spp.(77%), and the resistance against penicillin was 35% in overall patients, followed by clindamycin and erythromycin (17%), which was much higher than previous studies. Conclusions : In our study, the higher resistance made penicillin alone not to be chosen as the first antibiotic drug for severe endodontic infections. Combinations with other drug, penicillin with wider spectrum of activity, or changing to other antibiotics was considered while remembering the increased risk of resistant microorganism.

Studies on partial characteyization and pathogenicity of atypical Aeronionas salmonicida EL-I isolated from diseased cultrued Eel (뱀장어에서 분리한 불정형 Aeromonas salmonicida EL-I의 일부 특성과 병원성에 관한 연구)

  • A partial characteristics and pathogenicity of Aeromonas salmonicida strain isolated from diseased cultured eel were studied. On the biochemicial characteristics of the isolate strain was classified into atypical Aeromonas salmonicida : the characteristics of the isolate strain differed from those of the subspecies of Achromogenes, masoucida and salmonicida. It strain was named atypical Aeromonas salmonicida EL-1 and it grew at optimal pH 7.5 and l% Nacl. In the pathogenicity test, at one to two days post infection, the motality of artificially infected eels with $1{\times}10^8$ and $1.0{\times}10^7$ cfu/fish group was 100%, and at three days pos-infection, in the $1.0{\times}10^6$ /cfu /fish infected group, the epizootic haemorrhagic ulcer was developed, and haemorragic red sports were formed around abdomen and fins and its died (40%). Total cell protein peptide bands of the A. salmonicida EL-1 were presented betweeon 11.8~102 Kd in molecular weight by the electrophoretic analysis and appeared 28 bands in all. Drug sensitivity of atypical A. salmonicida EL-1 strain was sensitivity to chlorampenicol, gentamicin, kanamycin, streptomycin and sulfamethoxazole /trimethoprim and intermidiate to ampiclin and neomycin resistant to cephalotin, erythromycin, lincomycin and tetracycline.

Biochemical properties and antimicrobial drug susceptibility of Pasteurella haemolytica isolated from pneumonic calves and goats (호흡기 증상을 나타낸 송아지 및 산양에서 분리한 pasteurella haemolytica의 생화학적 특성 및 약제감수성)

  • The present study was conducted to investigate biochemical properties and antimicrobial drug susceptibilities of 57 strains of Pasteurella haemolytica (P haemolytica) isolated from pneumonic calves and goats in Youngnam province during the period from September 1989 to February 1991. P haemolytica was isolated from 95 of 142(31.7%) pneumonic calves of 1 to 6 months of age and from 9 of 24(37.5%) pneumonic goats of 2 to 10 months of age. Seasonal isolation frequency of P haemolytica in calves and goats varied from 16.6 to 35.7%, and it was higher in winter. The majority of biochemical and cultural properties of P haemolytica isolated from calves and goats were identical to those of the reference strains employed. All isolates were susceptible to ceftiofur, erythromycin, kanamycin, gentamicin, and chloramphenicol (MIC: <$25{\mu}g/ml$), some of them were resistant to amikacin, sulfadimethoxin, and streptomycin (MIC: >200IU or ${\mu}g/ml$).
