Bacterial Brwon Rot of Ivy-aureus (Scindapsus aureus) Caused by Erwinia chrysanthemi (Erwinia chrysanthemi에 의한 Ivy-aureus (Scindapsus aureus)의 세균성 갈색부패병)

  • In 1991, the leaf brown spot of ivy-aureus (Scindapsus aureus) was found in Taejon and Seoul, Korea. The symptoms were appeared as dark-brown spots. The lesions were often surrounded by yellowish halos. These spots were enlarged to circular or elliptical in shape and dark-brown to black in color with slightly elevated in margin and sunken in center. The pathogenic bacteria were isolated from the diseased leaf of ivy-aureus were identified as Erwinia chrysanthemi on the basis of bacterial characteristics therefore, we would like to propose to the name of ivy-aureus disease caused by E. chrysanthemi as“bacterial brown rot of ivy-aureus”hereafter.

  • Based on equal number of cells, supplementation of 10$^{-6}$ M cadimium highly simulated the intracellular amylase. GCT, LDH as well as the glucose and erea content of E carotovora var, carotovaro cells. This was coupled with initiation of highly active GOT, CPK as well as accumulation of cholesterol in the cells. Lanthanum was less active and unable to initiate COT or CPK. Nickel was almost without effect though reduced LDH activity without initiating either enzyme or cholesterol production. Similar stiulations and/or initiations were observed, though to variable extents, when the same concentration of the three elements were supplied to E. carotovora var, citullis or E. toxica. The highest yield of amylase, GPT, GGT or glucose was obtained when E. carotovora var. carotovora was supplemented with Cd + Ni. The highest urea level was recorded in Erwinia carotovara var, cirullis, amended with Cd + La.

Effect of Carbon Sources and Culture Temperature on Pectate Lyase Production in Phytopathogenic Bacteria (탄소원과 배양온도가 식물 병원세균의 Pectate lyase 생산에 미치는 영향)

  • Phytopathogenic bacteria causing soft-rot many vegetables; extracellular enzymes produced by them, pectate lyase(Pel) is important pathogenicity facotrs which cause tissue maceration and cell death. Ten of seventeen plant pathogenic bacteria showed weak Pel activity, four of them showed low Pel activity and Erwinia acrotovora subsp. carotovora, E. chrysanthemi, Pseudomonas marginalis and Xanthomonas campestris pv. campestris showed high Pel activity in the polygalacturonate yeast extract agar (PAY) plate. High Pel activity of the four bacteria species produced the highest Pel activity when pectin or polygalacturonic acid (PGA) was added to minimal salts (MS) medium. Pel activity of the four bacterial species was the highest at 2$0^{\circ}C$ among different temperature conditions. The rate and amount of maceration of potato tuber tissue were highest at 2$0^{\circ}C$ in E. carotovora subsp. carotovora, E. chrysanthemi and P. marginalis, while those were the highest at $25^{\circ}C$ in X. campestris pv. campetris.

  • Fire blight disease caused by the bacterium, Erwinia amylovora, was observed in apple and pear orchards in Korea in 2015. Since then, it has spread, sometimes over long distances to other orchards. Therefore, we examined the ability of E. amylovora to survive in soils and on the surfaces of common materials such as T-shirts, wrist bands, pruning shears, and rubber boots by both conventional PCR (cPCR) and quantitative PCR (qPCR) methods. E. amylovora was detected in all materials tested in this study and survived for sufficiently long periods to cause fire blight disease in new sites. Thus, based on the results of this study, sanitation protocols must be applied to equipment during orchard work.

  • Black shoot blight disease caused by Erwinia pyrifoliae has serious impacts on quality and yield in pear production in Korea; therefore, rapid and accurate methods for its detection are needed. However, traditional detection methods require a great deal of time and fail to achieve absolute quantification. In the present study, we developed a droplet digital polymerase chain reaction (ddPCR) method for the detection and absolute quantification of E. pyrifoliae using a pair of species-specific primers. The detection range was 103-107 copies/ml (DNA templates) and cfu/ml (cell culture templates). This new method exhibited good linearity and repeatability and was validated by absolute quantification of E. pyrifoliae DNA copies from samples of artificially inoculated immature pear fruits. Here, we present the first study of ddPCR assay for the detection and quantification of E. pyrifoliae. This method has potential applications in epidemiology and for the early prediction of black shoot blight outbreaks.

  • Bacterial population densities on soybean pods from Chungnam province ranges 105~106 CFU/$\textrm{cm}^2$, whereas those of bacteria causing sprout rot ranged 0~103 CFU/$\textrm{cm}^2$. Erwinia chrysanthemi, Xanthomonas campestris pv. glycines, Staphylococcus sp., and Micrococcus sp. were identified as pathogenic bacteria causing soybean sprout rot. The population density of X. campestris pv. glycines was higher than those of other bacteria.

  • Bacterial soft rot caused damage to beets massively grown in Pyungchang, Kangwon province, Korea. The affected roots and stems became cream colored and slimy, and turned black, causing the plants to become wilt and die. The casual organism isolated from the infected plants was identified as Erinia carotovora subsp. carotovora based on physiological and biochemical characteristic, and the results of the Biolog program (Biolog Inc., U.S.A.). E. carotovora subsp. carotovora is the first discribed bacterium which causes bacterial soft rot on beet in Korea.

  • The hollow stalk, caused by Erwinia carotovors var. carotovora, is probaly the most devastating disease to burley tobacco in Korea. There are two modes of infection to tobacco plants: the primary infection along the plant stem from soil and the secondary one from the topped end of plants. The secondary infection rate could be reduced to almost zero by applying an antibiotic, streptomycin sulfate(400 ppm solution), right after topping.

  • Biohydrogen production and analysis of microbial community were attempted from the sugar manufacturing wastewater with anaerobic fermentation process. Addtion of nutrients ($N{\cdot}P$) into sugar manufacturing wastewater stimulates hydrogen production from 9.53 to $26.67m{\ell}$ $H_2/g$ COD. Butyric acid, acetic acid, lactic acid, and propionic acid were detected in the sample of the anaerobic fermentation process. Butyric acid/Acetic acid(B/A) ratio was increased 0.50 to 0.92 according to the nutrients addtion into the wastewater. Microbial community was analyzed as Clostridium sp. in the phylum of Firmicutes and Klebsiella sp., Erwinia sp., and enterobacter sp. of the class of $\gamma$-Proteobacteria. As the improvement of hydrogen production, Erwinia sp. was decreased and Klebsiella sp. was increased.

Comparative Genome Analysis Reveals Natural Variations in the Genomes of Erwinia pyrifoliae, a Black Shoot Blight Pathogen in Apple and Pear

  • Erwinia pyrifoliae is a Gram-negative bacterial plant pathogen that causes black shoot blight in apple and pear. Although earlier studies reported the genome comparison of Erwinia species, E. pyrifoliae strains for such analysis were isolated in 1996. In 2014, the strain E. pyrifoliae EpK1/15 was newly isolated in the apple tree showing black shoot blight in South Korea. This study aimed to better understand the similarities and differences caused by natural variations at the genomic level between newly isolated E. pyrifoliae EpK1/15 and the strain Ep1/96, which were isolated almost 20 years apart. Several comparative genomic analyses were conducted, and Clusters of Orthologous Groups of proteins (COG) database was used to classify functional annotation for each strain. E. pyrifoliae EpK1/15 had similarities with the Ep1/96 strain in stress-related genes, Tn3 transposase of insertion sequences, type III secretion systems, and small RNAs. The most remarkable difference to emerge from this comparison was that although the draft genome of E. pyrifoliae EpK1/15 was almost conserved, Epk1/15 strain had at least three sorts of structural variations in functional annotation according to COG database; chromosome inversion, translocation, and duplication. These results indicate that E. pyrifoliae species has gone natural variations within almost 20 years at the genomic level, and we can trace their similarities and differences with comparative genomic analysis.
