Structure of Epithelium and Gland cells in a Korean leech, Whitemenia edentula (한국산 작은 말거머리 (Whitemenia edentula)의 상피조직과 선세포의 구조적 특성)

  • Dorsal and ventral epithelium of leech, Whitemenia edentula, were observed, using both light and electron-microscope. Results are as follows. Epithelia are composed of simple columnar or irregularly-shaped epithelium, the transveral folds were seen in the cross-sectioned dorsal and ventral epithelia, but only longitudinal folds(depth $500{\mu}m$, width $350{\mu}m$) were discovered in the ventral epithelium. As to muscular tissue under the dorsal epithelium of Whitemenia edentula, that of longitudinal muscle layer is thicker and more developed than that of circular muscle layer. Seven gland cells (type-A, B, C, D, E, F and H cells) and one pigment cell (type-G cell) were discovered in the whitemenia edentula. Those were identified as three kinds of the neutral mucopolysaccharide granoles (type-A, B and F cells), two kinds of acidic mucopolysaccharide granules (type-C and D cells), and two kinds of non-reactive granules respectively (type-E and F cells). The glanules of the type-A and type-B gland cells ae electronly high-dense, and surrounded by the muscular tissue. As to the distribution of gland cells, the type-A, type-B, type-C, type-D, type-I and type-H gland cells were discovered only between the connective tissue and the circular muscle layer, while type-F gland cells were discovered only between tile circular muscle layer and the longitudinal muscle layer.

  • The gingiva consists of an epithelial layer and an underlying connective tissue layer. The oral epithelium is a keratinized, stratified, squamous epithelium. The epithelium can be divided into the following cell layer: basal layer, prickle cell layer, granular cell layer and keratinized cell layer. The desquamative disease of gingiva means exfoliative diseases of epithelial layer on the gingiva. The chronic desqumative gingivitis is usually related to the dematologic disorders that produce cutaneous and mucous membrane blisters. The cicatricial pemphigoid and lichen planus are representative diseases of the dermatologic cases. Patients may be asymptomatic or symptomatic. When symptomatic, their complaints range from a mild burning sentation to an severe pain. The clinical examination must be considered with a thorough history, and routine histologic and immunofluorescence studies. A systemic approach needs to achieve accurate diagnosis and treatment of the gingival desquamative diseases.

Expression of Epidermal Growth Factor Receptor in the Inflamed Gingival Epithelium and the Dental Follicle (염증성 치은 상피와 치낭의 표피성장인자 수용체의 발현 및 실험적 치아이동에 미치는 영향에 관한 연구)

  • Epidermal growth factor(EGF), a single chain polypeptide of 53 amino acids with a molecular weight of 6,045 Da, was first isolated from the male mouse submandibular glands. EGF stimulates cellular proliferation and differentiation in several tissues and accelerates the rate of wound healing. EGF is bound to the specific receptor(EGFR) on the cell membrane of its target cell. EGFR is a transmembrane glycoprotein with a molecular weight of 170,000 Da and is detectable on a large variety of cell types and tissues. The authors investigated the expression of EGFR in the normal and inflamed human gingival epithelium to study the role of EGFR in the inflammation of the gingival epithelium, and the expression of EGFR in the dental follicle by using in situ mRNA hybridization and immunohistochenistry. The results weree as follows : 1. The expression of EGFR mRNA in the normal gingival epithelium on in situ mRNA hybridization was mainly localized on the basal cell layer, and the spinous layer was weakly positive The granular and cornified layers were negative 2. The expression of EGFR protein in the normal gingival epithelium on inmunohistochemistry was localized on the cornified and granular layers, and the spinous layer was weakly positive. The basal cell layer was completely negative 3. The expression of EGFR mRNA in the inflamed gingival epithelium on in situ mRNA hybridization was evenly and homogeneously distributed in the whole layers of the gingival epithelium except the cornified layer. The staining intensity appeared to increase progressively from the basal cell layer to the cornified layer. 4. The expression of EGFR protein in the inflamed gingival epithelium on immunohistochemistry was evenly and homogeneously distributed in the whole layers of the gingival epithelium. The staining intensity appeared to increase progressively from the cornified layer to the basal cell layer. 5. Strong positive reaction was seen in the epithelial cell rests of Malassez, whereas only background staining was seen in other cells of the dental follicle. In conclusion, the up-regulation of EGFR in the inflamed gingival epithelium and the high amounts of EGFR in the epthelial cell rests of Malassez in the dental follicle can be regarded as responses to the possible damages to the oral environment to maintain the homeostatic conditions.

Morphological Studies on the Epithelium of the Foregut of the Nymph and the Adult in the Grasshopper, Oxya sinuosa (우리벼메뚜기 유충과 성충의 전장 점막상피에 관한 형태학적 연구)

  • The morphological characteristics and changes of the foregut in the grasshopper (Oxya sinuosa Mistshenko) have been studied with the special reference to the developmental stage. In light microscope level, the epithelia are seen the cuboidal shape in the 5th instar nymph and the adult. A number of pigment granules are appeared in the cuboidal epithelium of the 5th instar nymph, however the pigment granules were a few in the adult. Indistinct or undifferentiated folds of the epithelial layer were appeared in the 5th instar nymph, whereas the well-developed folds were in the adult. The well-developed muscular layers are seen in the 5th instar nymph, however in the adult the muscular layers are appeared thin or a few layer except the crop. In electron microscope level, in the foregut epithelium, a few round or oval shaped mitochondria, well-developed rER, Golgi complex, lysosome and a number of vacuoles were appeared in the cytoplasm. Two types of pigment granules, electron dense or homogenous and low electron dense or lamellar profiles, are seen in the esophageal epithelium. The crystalline profiles and plasma membrane infoldings were also appeared. In the foregut epithelium of the 5th instar nymph, however the cell organelles were generally undifferentiated and in the crop two types of pigment granules as the adult esophageal epithelium were also found.

  • The Langerhans cells are dendritic nonkeratinocytes found suprabasally in most stratified squamous epithelia, such as human epidermis and the epithelium of the oral mucosa including that of gingiva. After Paul Langerhans found it in the skin in 1968, there have been sturdies of it's function and distribution . Stingle et al. reported that the Langerhans cells seem able to present antigens and to stimulate T-lymphocytes. Shelley et al. discovered that they can take up contact allergens. Accordingly it has been suggested that Langerhans cells are important elements of p Peripheral cell mediated immune system. In this study, the gingival tissue of a adult periodontitis patient was taken and freeze dried. In one specimen, we used the CD1 monoclonal antbody to staining the Langerhans cell. The other specimen, we embedded in paraffin and staining it with S-100 monoclonal antibody. The purpose of this study was to use these specimens to find out the distribution, orientation, morphology of the Langerhans cell and to discover the increase or decrease of Langerhans cell in an increased inflammatory state. The results were obtained as follows : 1. Langerhans cells were distributed between the basal cell layer and spinous cell layer against the CD1 & S-100 monoclonal antibody. 2. Langerhans cessl were plentiful in the oral eptihelium, and there was very little in the sulcular epithelium. 3. There were no Langerhans cell in the junction epithelium and pocket lining epithelium. 4. The number of Langerhans cells that responsed to the CD1 & S-100 monoclonal antibody had a statistically difference. 5. As the infiltration of the lymphocyte into the connective tissue were increased, the number of Langerhans cells in the epithelium were increased. 6. As the inflammation was increased, Langerhans cells in the spinous cell layer were more increased than those of the basal layer.

Histological Observation on Development of the Small Intestine of the Korean Native Goat (재래산양(在來山羊)의 소장형성(小腸形成)에 관한 조직학적관찰(組織學的觀察))

  • The morphological development of the small intestinal tissues of the Korean native goat were observed by light microscopy. Samples were taken from a 60-, 90-, 120-day-old fetus, a newborn goat and a 30-day-old goat. The results were summarized as follows; 1. In the small intestine of 60-day-old fetus, the apexes and sides of villi were covered with a simple columnar epithelium, and intervilous areas and mucosal ridges were still covered with stratified epithelium of two to six cell layers. Mesenchymal tissues formed lamina propria, circular muscle layer and serosa. The numbers of villi per cross section of the small intestine (NVPCS) were 10 to 18. 2. In 90-day-old fetus, intervillous areas and mucosal ridges of the organ were covered with simple columnar epithelium. Goblet cells in epithelium and outer longitudinal muscle layer often appeared. NVPCS were 35 to 60 and Brunner's glands were appeared. 3. In 120-day-old fetus, Brunner's glands of the duodenum and circular connection of outer longitudinal muscle layer were formed, NVPCS were 50 to 87. 4. In newborn goat, Peyer's patches were fully formed and NVPCS were 50 to 87. 5. In 30-day goat, the small intestine was fully matured and NVPCS were 81 to 102.

Differential expression of glycoconjugates, estrogen receptor-α, c-fos and c-jun in the vagina of normal and ovariectomized rat (흰쥐 발정주기와 난소절제에 따른 질상피의 glycoconjugates, estrogen receptor-α, c-fos 및 c-jun 분포변화)

  • We investigated the compositional changes of glycoconjugates (GCs) and expression of estrogen receptor (ER)-$\alpha$, c-fos and c-jun in the vagina of normal and ovariectomized rats by histochemical and immunohistochemical methods. The mucinous transformation of the superficial layer that occurred from late diestrus to proestrus was accompanied with extensive enrichment of GCs. According to the cyclic changes of the vagina, distinct reactivity patterns such as SBA affinity in the diestrus and Con A affinity in the diestrus and estrus phase was observed. However, weak staining for GCs was detected in the atrophied vaginal epithelium of ovariectomized rats. ER-$\alpha$ immunoreaction was mainly demonstrated in the basal layer of epithelium and estrus cycle-related variation in the number of ER-$\alpha$ immunoreaction were not pronounced. But the stromal cells showing ER-$\alpha$ immunoreaction were abundantly observed from diestrus to estrus phase. The most numerous c-fos immunoreactive cells were observed in the basal and intermediate layer of epithelium and stromal fells from the proestrus to estrus phase and c-jun in the basal layer of epithelium during estrus phase. The c-jun immunoreaction of stromal cells expressed only in the estrus phase. In the ovariectomized rats, a few of ER-$\alpha$, c-fos and c-jun immunoreactive cells were observed in the vaginal epithelium and no immunoreaction were found in the stromal cells. ER-$\alpha$ and c-fos immunoreaction fully expressed in the proestrus coincident with the cell proliferation, mutinous transformation and cornification of vaginal epithelium. These data indicate that vagina epithelium and stromal reals express multiple protein such as ER-$\alpha$, c-fos and c-iun by estrogen that may function in process of cells proliferation and differentiation of vagina epithelium.

The Papillar and Pelvic Epithelia of the Bat: A Scanning and Transmission Electron Microscopic Study (집박쥐 신유두(腎乳頭) 및 신우상피(腎盂上皮)에 미세구조(微細構造))

  • The anatomy of the renal papilla and pelvis of the bat(Pipistrellus abramus abramus Temminck) and their ultrastructure of the epithelium has been studied by means of light, scanning and transmission electron microscopy. The bats were captured at Ondal cave located in Yangchun-Myun, Tanyang-Kun, Chungchongpuk-Do, Korea in the active(July) and hibernating(January) phases. A conical renal papilla with several papillary foramina at the apex was curved toward the ureter. At the margin of the funnel-shaped renal pelvis, the fornix extended deeply into the kidney parenchyme was formed between the outer zone of the medulla and the cortex. The epithelium of the papilla covered the inner zone of the medulla. The epithelium of the fornix medially overlayed the outer zone of the medulla, and laterally the cortex. The surface epithelium of the papilla was simple columnar in shape with the scanty organelles and the short microvilli on their luminal surface. The fornix was Jined with $1{\sim}2$ cell layered squamous epithelium. On the medial surface of the fornix, two types of the superficial epithelial cells were identified; one had numerous short microvilli and the other had the microplicae and plaques on their luminal surface. The lateral surface of the fornix lined with the epithelial cells showed microridges and plaques on the luminal surface. The epithelial cells on the medial and lateral surfaces of the fornix were similar to their internal ultrastructures. The pelvic epithelium was a typical transitional type, which was composed of a layer of basal cells, one or two layers of intermediate cells, and a layer of superficial(facet) cells. The fusiform vesicles were observed in the cytoplasm of the intermediate and facet cells. There were no noticeable morphological changes in the epithelium of the papilla, fornix and pelvis except the medial epithelium of the fornix in the hibernating cycle. The microvillar cells were more numerous in the hibernating phase than in the active phase. The morphology, of the renal papilla and fornix of bats, indicates that a functional relationship may exist between the pelvic urine and the renal parenchyme.

Histological Studies of the Infundibulum of the Oviduct of the Korean Native Pheasants(Phasianus colchicus korpowi) (한국산 꿩 난관깔때기의 조직학적 연구)

  • This study was conducted for the histological observation of the infundibulum of the oviduct of the laying Korean native pheasants. The results are as follows : 1. The infundibular wall is composed of the epithelium, lamina propria, muscle layer(inner circular and outer longitudinal muscle), and serosa. The funnel lip is divided into the inner, and outer lip of the epithelium and muscle layer. 2. The epithelium of the funnel lip and most region of the cranial part of the funnel are composed of ciliated columnar cells. In the surface and lateral part of the folds, ciliated cells and non-ciliated secretory cells tend to alternate in the epithelium of the caudal funnel and the necks, but are also found in groups of the simple cuboidal epithelium at the bases of the grooves between the ridges and tubular glands found in the subepithelium. 3. The secretory material of the non-ciliated secretory cills contains PAS-positive and alcian blue-positive granules, and these materials show purple colour in the basic fuchsin-methylene blue stain. 4. The cells of the glandular groove and tubular gland of the neck portion of the oviduct mostly show weak PAS-positive, and alcian blue stain negative reaction. The tubular gland cells of the infndibulum contain pink of purple colour granules, and without reaction in the anterior neck portion of the infundibulum in basic fuchsin and methylene blue stain.

  • The auther observed the aging of oral epithelium in gingiva of edentulous jaw. The observed staning methods were PAS reaction of the epithelivm of the gingive were increased in aging group, the basal prickle and granulor cell layer presented a strong reaction. The collagenous fiber in subepithelical layer was, quantitalively inrcred accouding to aging.
