Anticancer Effect of Houttuynia cordata Extract on Cancered ICR Mouse and L1210 Cells With Changes of SOD and GPx Activities (어성초 추출물의 ICR생쥐와 L1210 세포에 대한 항암작용 및 SOD, GPx 효소활성변화)

  • The present investigation was undertaken to examine the anticancer activity of the methanol extract from Houttuynia cordata on ICR mouse with induced abdominal cancer and L1210 cancer cells. When the methanol extract of Houttuynia cordata (10∼200 $\mu\textrm{g}$/$m\ell$) was administered orally to ICR mouse with abdominal cancer, 47.8% of the best life prolonging effect was obtained. In case of cytotoxicity study (inhibition of cell proliferation) of Houttuynia cordata extract against L1210 cells, $IC_{50}$/ was found to be 62.8 $\mu\textrm{g}$/$m\ell$. In contrast to such considerable toxicity against cancer cell line, the toxicity demonstrated by the identical extract against normal lymphocytes was very meagre as shown to be < 5% compared with 86.5% in case of L1210 cells at the same condition. To get an insight into the reaction mechanism undelying the anticancer activity, $O_2$ion quantity and antioxidant enzyme activities such as superoxide dismiutase (SOD) and glutathione peroxidase (GPx) of L1210 cells in the presence of Houttuynia cordata extract were measured. The increased values of SOD and GPx enzyme activities in addition to the augmented generation of $O_2$ ion in L1210 cells implied that the reactive oxygen species induding $O_2$ion which were presumably induced by Houttuynia cordata extract might have participated in the process of L1210 cells cytotoxicity.

  • The production media and enzymatic characteristics of laccase from Ganoderma lucidum was investigated. Potato dextrose yeast extract media was proved to be the best for laccase production. The enzyme has optimum pH of 6.45km value of 6.71 mM and appeared to be stable at wide pH range. The enzyme was inactivated partially by methanol and ethanol and totally by sodium azide but not at all by acetone. Also the enzyme purification was performed and the data is given.

Effect of Extract or Crude Enzyme Extracted from the Fruit of Paper Mulberry (Broussonetia kazinoki Siebold) on Tenderness and Palatability of Jangchorim (닥나무 열매(緖寶子)의 농축액과 조효소가 장조림의 맛과 연화에 미치는 영향)

  • The purpose of this study was to investigate the effect of the addition of the extract (0, 2, 4, 6, 8%) and crude enzyme (0, 0.05, 0.1, 0.5, 1%) extracted from the fruits of paper mulberry (Broussonetia kazinoki Siebold) on the tenderness and palatability of Jangchorim. Addition of up to 8% of the extract diminished the shear force by 4.8∼27%, while the addition of up to 1% of the crude enzyme begot 7.8∼34.2% decrease. Also, as the addition of the fruits of paper mulberry increased, the cooking loss was somewhat decreased, an the redness of cooked beef surface and free amino acid content in the liquid part of Jangchorim were generally increased. The contents of the mineral were in the order of Na, K, P, Ca, and Mg when crude enzyme was added. In sensory evaluation, the 4% crude extract from the fruit of paper mulberry and 0.1% addition in its crude enzyme showed the most favorable response.

Changes in 2,6-dimethoxy-1,4-benzoquinone and Water Extractable Arabinoxylan Content of Wheat Germ Extract by Enzyme Treatment (효소처리에 따른 밀 배아 추출물의 2,6-dimethoxy-1,4-benzoquinone과 수용성 아라비노자일란 함량 변화)

  • This study was carried out using Celluclast 1.5L to increase the content of 2,6-DMBQ and water extractable arabinoxylan in wheat germ extract. Extraction temperatures were 30℃, 45℃ and 60℃. The extraction times were 0, 6, 12, 18, 24 and 30 h. The pH of the extract decreased rapidly from 18 h at 30℃ in both water- and enzyme-treated extracts. 2,6-DMBQ of water- and enzyme-treated extracts increased with the extraction time. At 30-hour extraction time, enzyme-treated extract increased 27.60% at 30℃ extraction temperature than water extraction. Extraction temperatures of 45℃ and 60℃ were increased by 65.03% and 151.05%, respectively. The highest content of water-extractable arabinoxylan was 15.23±0.08 mg/g when the enzyme was treated at an extraction temperature of 60℃ for 30 h. At 30=hour extraction time, enzyme-treated extract increased 7.92% at 30℃ extraction temperature compared to water extraction. Extraction temperatures of 45℃ and 60℃ were increased by 31.20% and 54.38%, respectively.

Isolation of Fibrinolytic Enzyme and β-Glucosidase Producing Strains from Doenjang and Optimum Conditions of Enzyme Production (된장으로부터 혈전용해능 및 β-Glucosidase 활성을 가진 균주 분리 및 효소생산 배지의 최적화)

  • Bacterial strains showing the firinolytic and $\beta$-glucosidase activity were screened from Doeniang. The strain of KH-15 revealed a high level of fibrinolytic and $\beta$-gluocosidase activity. The isolated bacterium was identified and desingnated as Bacillus sp. KH-15. The carbon, nitrogen and salts sgnificantly influenced te fibrinolytic enzyme and $\beta$-glucosidase production. The optimized composition of medium appeared to be 2% glucose, 0.5% yeast extract and 0.1% calcium chloride. The optimum pH and temperature for fibrinolytic enzyme and $\beta$-glucosidase activities were pH 7∼8, 4$0^{\circ}C$ and pH 6∼8, 30∼4$0^{\circ}C$, respectively.

  • A thermophilic soil isolate Bacillus sp. Y-127 was selected for the production of thermostable amylase. The strain was used for the enzyme production and the thermostable amylase was characterized. The optimum cultural conditions for the enzyme production were 6$0^{\circ}C$ at pH 7.0 for 32 hours using a mineral medium containing 2% soluble starch and 0.2% yeast extract. The extra-cellular enzyme was purified about 123-folds with about 6% recovery. The purified enzyme was stable at pH between 4.0 and 7.0, and temperature up to 6$0^{\circ}C$.

Fibrinolytic Enzyme Production by Bacillus subtilis KH-4 Isolated from Deonjang

  • A strong fibrin-specific fibrinolytic enzyme was produced from Bacillus subtilis KH-4 isolated from Deonjang, a Korean fermented soybean paste similar to Japanese miso. The addition of glucose as a carbon source resulted in the highest levels of caseinolytic and fibrinolytic activities. Likewise, the addition of yeast extract as the nitrogen source resulted in the highest caseinolytic and fibrinolytic activities (3473.2 unit and 47.4 munit, respectively), It was observed that out of all metal ion sources only calcium (chloride) enhanced caseinolytic and fibrinolytic activities, with increases of 4949.3 unit and 58.2 unit/mg, respectively. The optimal temperature for the production of the enzyme was found to be 4$0^{\circ}C$ in the optimal medium (glucose 20 g, yeast extract 5 g, CaCl$_2$l g, and NaCl 2 g). The maximum fibrinolytic activity was observed at the late stationary phase. B. subtilis KH-4 produced a fibrinolytic enzyme at 4$0^{\circ}C$, after 30 h growth, which increased up to 54 h and then remained constant. These results suggest that Deonjang has potential as a source of physiologically active anti-thromotic enzymes.

Influence of Gami-oryungsan on bromobenzene-induced liver injury in experimental animal (Bromobenzene독성(毒性)에 의한 간기능손상(肝機能損傷)에 미치는 가미오령산의 영향(影響))

  • Objective : To investigate the hepatoprotective effects of Gami-oryungsan on the liver damage induced by bromobenzene. Method : The development of fibrosis and acute liver injury was examined by the chemical analysis of AST, AL T, ${\gamma}$-GTP . and epoxide hydrolase glutathione S-transferase glutathione peroxidase enzyme activity, lipidoperoxide levels, glutathione levels were measured and oberved. Results : The increasing levels of lipidoperoxide was decreased proportionally according to dose of extract GO. Epoxide hydrolase glutathioneS-transferase glutathione peroxidase enzyme activity highly increased in GO pre-acupunctured group compared with the group treated with only bromobenzene. The increase of serum AST, AL T, ${\gamma}$-GTP enzyme activity of mice by bromobenzene was inhibited by the administration of GO. Lipidoperoxide levels in rat's liver decreased compared to the case of bromobenzene-treated group. The levels of Glutathione decreased by bromo benzene were increased highly in GO pre-acupunctured group. Conclusion : These results suggest that GO extract recovers the damage of liver due to bromobenzene intoxication by decreasing the lipid peroxidation AST AL T ${\gamma}$-GTP enzyme activity and increasing epoxide hydrolase glutathioneS-transferase glutathione peroxidase enzyme activity, glutathione levels.

  • Production and properties of invertase from Aspergillus niger were investigated. Inulin and sucrose were best carbon source and yeast extract was most suitable for the production of the enzyme among tested carbon and nitrogen sources. The enzyme among tested carbon and nitrogen sources. The enzyme was maximally produced by cultivating the organism at medium of pH 4.5 and temperature of 3$0^{\circ}C$ The optimum pH and temperature for the enzyme activity were pH 5.0 and temperature of 5$0^{\circ}C$, respectively. Among tested metal ions. Hg++, Cu++ and Ag+ ions Inhibited the enzyme activity drastically.

  • A strain producing an extracellular cytosine deaminase was isolated from soil samples. The enzyme obtained from the strain possessed the substrate specificity to both cytosine and 5-fluorocytosine. From the results of its morphological, cultural, physiological, and biochemical properties, the strain was thought to be the genus Arthrobacter. Therefore, it was named as Arthrobacter sp. JH-13. The composition of optimum medium for the enzyme formation was 0.5% of peptone, 0.5% of meat extract, 0.5% of soluble starch, and 0.1% of KCl. The optimum pH for the enzyme formation was 8.0. When the microoganism was cultured aerobically in the above medium, enzyme production reached at maximum in 54 hours at $30^{\circ}C$.
